RESUMO

Crown removal revitalises sand-fixing shrubs that show declining vigour with age in drought-prone environments; however, the underlying mechanisms are poorly understood. Here, we addressed this knowledge gap by comparing the growth performance, xylem hydraulics and plant carbon economy across different plant ages (10, 21 and 33 years) and treatments (control and crown removal) using a representative sand-fixing shrub (Caragana microphylla Lam.) in northern China. We found that growth decline with plant age was accompanied by simultaneous decreases in soil moisture, plant hydraulic efficiency and photosynthetic capacity, suggesting that these interconnected changes in plant water relations and carbon economy were responsible for this decline. Following crown removal, quick resprouting, involving remobilisation of root nonstructural carbohydrate reserves, contributed to the reconstruction of an efficient hydraulic system and improved plant carbon status, but this became less effective in older shrubs. These age-dependent effects of carbon economy and hydraulics on plant growth vigour provide a mechanistic explanation for the age-related decline and revitalisation of sand-fixing shrubs. This understanding is crucial for the development of suitable management strategies for shrub plantations constructed with species having the resprouting ability and contributes to the sustainability of ecological restoration projects in water-limited sandy lands.

Parathyroid hormone (PTH) type 1 receptor (PTH1R), as a typical class B1 G protein-coupled receptor (GPCR), is responsible for regulating bone turnover and maintaining calcium homeostasis, and its dysregulation has been implicated in the development of several diseases. The extracellular domain (ECD) of PTH1R is crucial for the recognition and binding of ligands, and the receptor may exhibit an autoinhibited state with the closure of the ECD in the absence of ligands. However, the correlation between ECD conformations and PTH1R activation remains unclear. Thus, this study combines enhanced sampling molecular dynamics (MD) simulations and Markov state models (MSMs) to reveal the possible relevance between the ECD conformations and the activation of PTH1R. First, 22 intermediate structures are generated from the autoinhibited state to the active state and conducted for 10 independent 200 ns simulations each. Then, the MSM is constructed based on the cumulative 44 µs simulations with six identified microstates. Finally, the potential interplay between ECD conformational changes and PTH1R activation as well as cryptic allosteric pockets in the intermediate states during receptor activation is revealed. Overall, our findings reveal that the activation of PTH1R has a specific correlation with ECD conformational changes and provide essential insights for GPCR biology and developing novel allosteric modulators targeting cryptic sites.

The microtubule-associated protein tau (MAPT) has a critical role in the development and preservation of the nervous system. However, tau's dysfunction and accumulation in the human brain can lead to several neurodegenerative diseases, such as Alzheimer's disease, Down's syndrome, and frontotemporal dementia. The microtubule binding (MTB) domain plays a significant, important role in determining the tau's pathophysiology, as the core of paired helical filaments PHF6* (275VQIINK280) and PHF6 (306VQIVYK311) of R2 and R3 repeat units, respectively, are formed in this region, which promotes tau aggregation. Post-translational modifications, and in particular lysine acetylation at K280 of PHF6* and K311 of PHF6, have been previously established to promote tau misfolding and aggregation. However, the exact aggregation mechanism is not known. In this study, we established an atomic-level nucleation-extension mechanism of the separated aggregation of acetylated PHF6* and PHF6 hexapeptides, respectively, of tau. We show that the acetylation of the lysine residues promotes the formation of ß-sheet enriched high-ordered oligomers. The Markov state model analysis of ac-PHF6* and ac-PHF6 aggregation revealed the formation of an antiparallel dimer nucleus which could be extended from both sides in a parallel manner to form mixed-oriented and high-ordered oligomers. Our study describes the detailed mechanism for acetylation-driven tau aggregation, which provides valuable insights into the effect of post-translation modification in altering the pathophysiology of tau hexapeptides.

OBJECTIVE: The study explores the application value of three-dimensional arterial spin labeling magnetic resonance imaging (3D pCASL) in early assessment of radiation encephalopathy (REP) in patients with nasopharyngeal carcinoma (NPC). METHODS: A retrospective analysis of 39 cases of NPC was performed. Routine enhanced MRI scan and 3D pCASL imaging were used to examine the apparent diffusion coefficient (ADC) and brain blood flow (CBF) before and after treatment with intensity-modulated radiotherapy (IMRT). Dosimetric analysis of irradiation was performed. Receiver operating characteristic curve (ROC) was used to analyze diagnostic performance of two imaging methods. RESULTS: There was no statistically significant difference between the two methods for the measurement of temporal white matter ADC, but statistically significant difference was found in CBF. 3D pCASL imaging showed more sensitivity, specificity and higher accuracy than conventional MRI enhanced scan in showing REP. The maximum dose of the temporal lobe was at the enhanced area. CONCLUSION: The present study demonstrates that 3D pCASL scan at month 3 can reflect blood flow perfusion differences in NPC patients after IMRT and can accurately assess the possibility of REP at early stage. Enhanced areas have a higher probability of REP than the surrounding areas. ADVANCES IN KNOWLEDGE: There is few magnetic resonance angiography studies used to evaluate arterial circulation on its application on potential REP after radiotherapy for NPC. In our study, we evaluate the application value of 3D pCASL in the early assessment of potential REP in patients with NPC after radiotherapy. The study was to provide an improved understanding of the early specific characteristics on MRI imaging and evolution of potential radiation encephalopathy using 3D pCASL technique, which can quantitatively evaluate the changes of blood flow in tissues at early stage and help to diagnose and treat potential radiation encephalopathy as early as possible.

Language switching is encountered commonly and inevitably in bilingual society and often induces costs for target language's production. However, for auditory words' comprehension at sentence level, the limited research showed divergent findings. Some research observed comprehension costs when the language of target words was switched with the code of sentential contexts kept constant; but a potential switch advantage was also showed in non-dominant targets' processing when sentential context switches occurred. Additionally, it's well documented that the words' emotional connotations play a key role in both L1 and L2 word comprehension. Therefore, we aimed to explore which switch effect would occur when bilinguals comprehended L1/L2 emotional target nouns in auditory modality at behavioral and neural level through a visual object selection task. Behaviorally, switch related costs occurred in L1 targets' comprehension, whereas advantage effects were found in L2. Moreover, greater switch advantage occurred for positive and negative targets than for neutral ones. Consistently, larger LPC (Late Positive Component) defection was elicited for L2-Switch trials relative to L2-Nonswitch trials and the differences of LPC's amplitude could predict the behavioral advantageous effects of switching in nondominant targets' comprehension, which suggest that language switching lead to deeper re-analyses for emotional words. Taken together, it's suggested that bilinguals can adaptively utilize top-down (sentential prediction) and bottom-up (words' emotional information) cues to access weaker L2 representations.

Background: With the popularity of mobile Internet and social networks, an increasing number of social media-based health management systems (SocialHMS) have emerged in recent years. These social media-based systems have been widely used in registration, payment, decision-making, chronic diseases management, health information and medical expenses inquiry, etc., and they greatly facilitate the convenience for people to obtain health services. Objective: This study aimed to investigate the factors influencing sustained health engagement of SocialHMS by combining the theory of planned behavior (TPB) with the big-five theory and the trust theory. Method: We completed an empirical analysis based on the 494 pieces of data collected from Anhui Medical University first affiliated hospital (AMU) in East China through structural equation modeling and SmartPLS (statistical analysis software). Results: Openness to new experience has a significantly positive influence on attitude (path coefficient = 0.671, t = 24.0571, R2 = 0.451), perceived behavioral control (path coefficient = 0.752, t = 32.2893, R2 = 0.565), and perceived risk (path coefficient = 0.651, t = 18.5940, R2 = 0.424), respectively. Attitude, perceived behavioral control, subjective norms, and trust have a significantly positive influence on sustained health engagement (path coefficients = 0.206, 0.305, 0.197, 0.183 respectively, t = 3.6684, 4.9158, 4.3414, and 3.3715, respectively). The explained variance of the above factors to the sustained health engagement of SocialHMS is 60.7% (R2 = 0.607). Perceived risk has a significantly negative influence on trust (path coefficient = 0.825, t = 46.9598, R2 = 0.681). Conclusions: Attitude, perceived behavioral control, subjective norm, and trust are the determinants that affect sustained health engagement. The users' personality trait of openness to new experience and perceived risk were also found to be important factors for sustained health engagement. For hospital managers, there is the possibility to take appropriate measures based on users' personality to further enhance the implementation and utilization of SocialHMS. As for system suppliers, they can provide the optimal design for SocialHMS so as to meet users' needs.
