RESUMO

Several studies have evaluated the human exposure of phthalate esters (PAEs) in PM2.5 via inhalation route, however, inhalation bioaccessibility and the lung cell penetration of PAEs were barely considered in risk assessment. In the present study, PM2.5 samples collected from indoor environments were investigated for inhalation bioaccessibility of PAEs using two simulated lung fluids (gamble's solution (GMB) and artificial lysosomal fluid (ALF)). The results showed that the inhalation bioaccessibility of PAEs (except for diethyl phthalate) under healthy state (GMB: 8.9%-62.8%) was lower than that under the inflammatory condition (ALF: 14.5%-67.6%). Lung cell permeation and metabolism of three selected PAEs (diethyl phthalate, di(n-butyl)phthalate and di-2-ethylhexyl phthalate) was tested using equivalent lung cell (A549) model. The inhalation bioavailability obtained by combination of the bioaccessibility of PAEs in indoor PM2.5 and permeability data of A549 cell ranged from 11.7% to 51.1% in health condition, and 13.5%-55.0% in inflammatory state. The calibration parameter (Fc) based on the inhalation bioavailability was established in present study and could provide a reference for a more accurate risk assessment of PM2.5-bound PAEs.

Numerous studies have studied the health risk assessment of human exposure to As or bioaccessible As via rice intake; however, the bioaccessibility of different As species in rice is seldom reported. In the present study, 31 rice samples were collected from markets or individual growers to investigate the speciation and bioaccessibility of As. Five different species (AsIII, AsV, DMA, MMA, and AsB) were detected in rice samples from different regions, among which AsIII accounted for the largest proportion (62.95% in average), followed by DMA and AsV. In addition, the cooking method could facilitate the release of As from rice into gastric and intestinal juice, and subsequently increase the bioaccessibility of As. The bioaccessibility of inorganic As in cooked rice ranged from 71.83 to 100%, and that of organic As ranged from 31.69 to 61.04%. Non-carcinogenic and carcinogenic risk assessment of children and adults exposure to As via rice intake considering the bioaccessibility of cooked rice was carried out. The target hazard quotient (THQ) of iAs and total As for children ranged from 0.21 to 1.61 and 0.48 to 2.26, respectively, while those for adults ranged from 0.12 to 0.88 and 0.26 to 1.23, respectively. Incremental lifetime cancer risk (ILCR) for children and adults ranged from 9.57 [Formula: see text] 10-5 to 7.25 [Formula: see text] 10-4 and 5.21 [Formula: see text] 10-5 to 3.95 [Formula: see text] 10-4, respectively. The results of risk assessment indicated that children would face a higher health risk than adults when they took the same type of rice as their staple food.

Aiming at the ecological footprint model, the traditional trade adjustment method only considered the international trade process at the urban scale, ignoring the trade footprint generated by domestic trade and indirect trade in various products. This paper adopts the urban-scale ecological footprint model based on the macro-trade adjustment method to calculate the trade adjustment coefficient of biological products and the energy trade adjustment coefficient respectively to correct the trade footprint. The results showed that the per capita ecological deficit showed a straight upward trend, from 0.07351 hm2 in 2013 to 0.15472 hm2 in 2018. From 2013 to 2018, the per capita ecological footprint of Guangdong Province was greater than the per capita ecological carrying capacity, and the ecological economic system of Guangdong Province was in an unsustainable state. According to the trade ecological footprint, Guangdong Province was a completely foreign resource and service exporting city, which was consistent with Guangdong Province's own economic development direction; the analysis results of the ecological product trade footprint were more consistent with the current city positioning of biological resource products of each city, and the energy indirect trade footprint. The improved ecological footprint model could more accurately assess the true status of ecological vitality above the urban scale.

Inhalation bioaccessibility of polycyclic aromatic hydrocarbons (PAHs) in PM2.5 was assessed in numerous studies, however, the lung cell uptake and penetration of PAHs was seldom taken into account in risk assessment. In the present study, eighteen indoor PM2.5 samples collected from Guangzhou, China were analyzed for the inhalation bioavailability of PAHs combining the inhalation bioaccessibility and cell absorption of PAHs. Two simulated epithelial lung fluid mimicking the healthy condition (as represented by gamble's solution (GMB), pH = 7.4) and the inflammatory condition (as represented by artificial lysosomal fluid (ALF), pH = 4.5) were employed to evaluate the inhalation bioaccessibility. The results indicated that the bioaccessibility of PAHs under the inflammatory condition (1.28%-87.7%) was higher than that under healthy condition (0.88%-87.6%). Naphthalene, phenanthrene, pyrene and benzo[a]pyrene were selected for absorption assay of lung epithelial cells (A549). The absorption rate of PAHs ranged from 64.7 to 90.7% and it was inversely proportional to the number of aromatic rings. Taken together, the inhalation bioavailability based on the bioaccessibility of PAHs and the lung cell absorption ratio ranged from 9.9 to 56.9% under the healthy state, from 12.7 to 65.6% under inflammatory condition. The correction parameter (Fc) was thus established and can be used to improve the risk assessment of human exposure to PAHs via PM2.5 inhalation in future work.

Numerous studies have focused on assessing the risk of human exposure to polycyclic aromatic hydrocarbons (PAHs) in indoor dust via dermal contact. However, the dermal bioaccessibility and dermal absorption of PAHs in indoor dust have seldom been reported. In the present study, the effects of temperature, sweat ratio, solid-liquid ratio and incubation time on the dermal bioaccessibility of PAHs were examined. Naphthalene, phenanthrene, pyrene and benzo[a]pyrenewere selected for examination in an absorption assay with keratinocyte cells. The results showed the release of PAHs from indoor dust fitted a first-order one-compartment model. Naphthalene had the highest rate of release, which was consistent with the bioaccessibility assay results. In addition, the absorption rate of naphthalene and phenanthrene by keratinocytes was higher than that of pyrene and benzo[a]pyrene, with the latter being of higher molecular weight. These results indicated that low molecular weight PAHs were much more easily absorbed via dermal contact than were high molecular weight PAHs. The dermal bioavailability of PAHs in indoor dust was estimated by multiplying the bioaccessibility of PAHs in indoor dust by the ratio of dermal absorption by skin cells, and ranged from 0.12 to 51.0%. These data will be useful in risk assessments.

OBJECTIVE: To assess the impact of visceral obesity quantified by preoperative computed tomography on short-term postoperative outcomes compared with body mass index (BMI) in stage I-III colon adenocarcinoma patients. METHODS: In this retrospective study, 107 patients treated with radical colectomy for stage I-III colon adenocarcinoma were classified as obese or non-obese by computed tomography-based measures or BMI (obese: BMI ≥28 kg/m2 , visceral fat area (VFA) to subcutaneous fat area ratio (V/S) ≥0.4, and VFA ≥100 cm2 ). Clinical variables, operation time, estimated blood loss, pathologic stage, histologic grade, postoperative complications, postoperative stay and hospitalization expenses were compared. RESULTS: Obese patients by VFA were more likely to have higher postoperative complication rate (32.9 versus 11.8%, P = 0.021), have longer operation time (184.6 ± 49.5 versus 163.1 ± 44.1 min, P = 0.033), postoperative stay (15.21 ± 7.59 versus 12.29 ± 5.40 days, P = 0.047) and cost more ($10 758.7 ± 3271.7 versus $9232.0 ± 2994.6, P = 0.023) than non-obese. CONCLUSION: Visceral obesity graded by VFA is associated with increased postoperative morbidity, operation time, postoperative stay and hospitalization expenses for colon adenocarcinoma patients and may be superior to BMI or V/S for the prediction of colon surgery.

The bioaccessibility of polybrominated diphenyl ethers (PBDEs) in indoor dust was estimated by a series of in vitro digestion methods. However, the absorption of PBDEs by intestinal cells after in vitro digestion was seldom studied. In the present study, the bioaccessibility of BDE-28, 47, 99 and 153 in indoor dust was firstly investigated by using the in vitro digestion method. Bioaccessibility in intestinal phase (BDE-28: 24.5-30.1%; BDE-47: 6.99-13.0; BDE-99: 1.61-14.2%; and BDE-153 5.97-24.4%.) was higher than that in gastric phase (BDE-28: 38.3-58.0; BDE-47: 9.62-30.9%; BDE-99: 9.71-24.3%; and BDE-153: 13.8-57.4%). The organic matter contents in indoor dust showed variable influence on the bioaccessibility of PBDEs. For the Caco-2 uptake assay, the BDE-28 showed greatest transport rate from medium to cell (Kmc: 0.525h-1), followed by -47, -99 and -153. The Kmc of PBDEs was significantly negative correlated with its corresponding KOW value. Similar pattern was found for the maximum uptake flux (Ju, max) and the transport rate from cell to medium (Kcm). The combination of bioacessibility and the absorption factor by Caco-2 cells could be used to estimate human intake of PBDEs via indoor dust would avoid overestimate the health risk.

There is limited study to simultaneously determine the relative bioavailability of heavy metals such as Cd, Pb, Cu, Cr(VI), and Ni in soil samples. In the present study, the bioaccessibility of heavy metals using in vitro assay was compared with the relative bioavailability of heavy metals using in vivo mouse model. The bioaccessibility of heavy metals ranged from 9.05 ± 0.97 % (Cr) to 42.8 ± 3.52 % (Cd). The uptake profile of heavy metals in soil and solution samples in mouse revealed that the uptake kinetics could be fitted to a two-compartment model. The relative bioavailability of heavy meals ranged from 34.8 ± 7.0 % (Ni) to 131 ± 20.3 % (Cu). Poor correlation between bioaccessibility and relative bioavailability of heavy metals was observed (r (2) = 0.11, p > 0.05). The relative bioavailability of heavy metals was significantly higher than the bioaccessibility of heavy metals (p < 0.05). The present study indicated that the in vitro digestion method should be carefully employed in risk assessment.

BACKGROUND: There is limited information on prevalent and incident atrial fibrillation in Chinese. We aimed to investigate the prevalence, incidence, management and risks of atrial fibrillation in an elderly Chinese population. METHODS: In a population--based prospective study in elderly (≥ 60 years) Chinese, we performed cardiovascular health examinations including a 12-lead electrocardiogram at baseline in 3,922 participants and biennially during follow-up in 2,017 participants. We collected information on vital status during the whole follow-up period. RESULTS: The baseline prevalence of atrial fibrillation was 2.0 % (n = 34) in 1718 men and 1.6 % (n = 36) in 2204 women. During a median 3.8 years of follow-up, the incidence rate of atrial fibrillation (n = 34) was 4.9 per 1000 person-years (95 % confidence interval [CI], 3.4-6.9). In univariate analysis, both the prevalence and incidence of atrial fibrillation were higher with age advancing (P < 0.0001) and in the presence of coronary heart disease (P ≤ 0.02). Of the 104 prevalent and incident cases of atrial fibrillation, only 1 (1.0 %) received anticoagulant therapy (warfarin). These patients with atrial fibrillation, compared with those with sinus rhythm, had significantly higher risks of all-cause (n = 261, hazard ratio [HR] 1.87, 95 % CI, 1.09-3.20, P = 0.02), cardiovascular (n = 136, HR 3.78, 95 % CI 2.17-6.58, P < 0.0001) and stroke mortality (n = 44, HR 6.31, 95 % CI 2.81-14.19, P = 0.0003). CONCLUSIONS: Atrial fibrillation was relatively frequent in elderly Chinese, poorly managed and associated with higher risks of mortality.

There is limited study focusing on the bioaccumulation of heavy metals in vegetables and human exposure to bioaccessible heavy metals in soil. In the present study, heavy metal concentrations (Cr, Ni, Cu, Pb and Cd) were measured in five types of vegetables, soil, root, and settled air particle samples from two sites (at a domestic waste incinerator and at 20km away from the incinerator) in Guangzhou, South China. Heavy metal concentrations in soil were greater than those in aerial parts of vegetables and roots, which indicated that vegetables bioaccumulated low amount of heavy metals from soil. The similar pattern of heavy metal (Cr, Cd) was found in the settled air particle samples and aerial parts of vegetables from two sites, which may suggest that foliar uptake may be an important pathway of heavy metal from the environment to vegetables. The highest levels of heavy metals were found in leaf lettuce (125.52µg/g, dry weight) and bitter lettuce (71.2µg/g) for sites A and B, respectively, followed by bitter lettuce and leaf lettuce for sites A and B, respectively. Swamp morning glory accumulated the lowest amount of heavy metals (81.02µg/g for site A and 53.2µg/g for site B) at both sites. The bioaccessibility of heavy metals in soil ranged from Cr (2%) to Cu (71.78%). Risk assessment showed that Cd and Pb in soil samples resulted in the highest non-cancer risk and Cd would result in unacceptable cancer risk for children and risk. The non-dietary intake of soil was the most important exposure pathway, when the bioaccessibility of heavy metals was taken into account.

In the present study, the polycyclic aromatic hydrocarbons (PAHs) in indoor dust and outdoor dust including road and window dust around the traffic road in Hunan Province, China, were sampled and detected. The ∑PAHs in indoor dust ranged from 5007-24,236 ng g(-1), with a median of 14,049 ng g(-1). The ∑PAHs in road dust ranged from 3644-12,875 ng g(-1), with a median of 10,559 ng g(-1). The ∑PAHs in window dust ranged from 803-12,590 ng g(-1), with a median of 5459 ng g(-1). Similar pattern of PAHs was observed in road and window dust except in H3W and H4W samples, which was dominated by naphthalene (Nap), benzo(b+k)fluoranthene (B(b+k)F), phenanthrene (Phe), and fluorine (Fle). Indoor dust showed slightly different PAHs profiles, which was dominated by Nap, fluoranthene (Fla) and Phe. Risk assessment indicated that dermal contact and dust ingestion exposure pathways were more important than the inhalation pathway. Cancer risk of PAHs via dust varied from 2.73 × 10(-8)-8.04 × 10(-6), with a median of 2.06 × 10(-6) for children, and from 2 × 10(-8)-5.89 × 10(-6), with a median of 1.52 × 10(-6) for adult. Probit model showed that 76 and 71 % of samples in the sampling area would result in the risk of children and adult exposure to PAHs via dust higher than the acceptable level (1 × 10(-6)), respectively.

The major objective of this study was to evaluate the human health risks of agricultural land use conversion to other purposes in Hong Kong, based on the levels of polychlorinated dibenzo-p-dioxin and polychlorinated dibenzofuran (PCDD/Fs) and determined dioxin-like activity in soil using ethoxyresorufin-O-deethylase (EROD) bioassay. Hazard quotient showed soils of open burning site (OBS) and electronic waste open burning site (EW (OBS)) exert a relatively higher non-cancer risk on adults (50.9 and 8.00) and children (407 and 64.0) via the pathway of accidental ingestion of soil particles than other types of land use. In addition, the levels of 17 PCDD/Fs congeners in OBS and EW (OBS) soils indicated high and moderate (1654 and 260 in one million people) cancer risks through the above pathway. Furthermore, the biologically derived TCDD concentrations (TEQbio) were also significantly correlated to the chemically derived toxic equivalent concentrations of dioxin-like chemicals (TEQcal (sum of chemically derived 2,3,7,8-TeCDD toxic equivalent concentrations (TEQPCDD/F) and chemically derived dioxin-like PAHs toxic equivalent concentrations (TEQPAH)) (r = 0.770, p <0.05). PCDD/Fs (95.4 to 99.9%) were the major stressor to the TEQcal in the soil samples, indicating higher concentrations of PCDD/Fs derived from chemical analyses may reflect a higher potency of inducing EROD activity.

Numerous studies have reported polychlorinated biphenyl (PCB) concentrations in soil, root, and aerial parts of vegetables. However, few studies have measured the contribution of PCBs bound to particles in air in relation to uptake by vegetables. In the present study, PCB concentrations were measured in five types of vegetables, soil, and settled air particle samples from two sites (at a domestic waste incinerator and at 20 km away from the incinerator) in Guangzhou, South China. ∑PCB concentrations in rhizosphere soil samples from the two sites ranged from 17.2-77.7 to 5.48-25.57 ng/g, respectively. ∑PCB concentrations in aerial parts of vegetables were greater than those in rhizosphere soils and roots with median values of 108 and 47.08 ng/g, respectively. Among the five types of vegetables studied, the highest concentration of PCBs was found in bitter lettuce. No significant correlation between PCBs in soil and roots or aerial parts of vegetables was observed. However, principal component analysis indicated that settled air particles were the dominant source of PCBs in the aerial parts of vegetables. In addition, similar PCB congener profiles were found in the aerial parts of vegetables and settled air particles. This suggests that foliar uptake of PCBs is an important pathway. Risk assessment indicated that human exposure to PCBs by way of dietary intake of vegetables from incinerator sites would result in high risk.

Human exposure to bioaccessible PCBs via indoor dust is limited around the world. In the present study, the workplace dust sample from commercial office, hospital, secondary school, shopping mall, electronic factory and manufacturing plant were collected from Hong Kong for PCBs analyses. Total PCBs concentrations ranged from 46.8 to 249 ng g(-1), with a median of 107 ng g(-1). Manufacturing plant showed the highest concentration among all of sampling sites. PCB 77 was found as the dominant congeners. The bioaccessibility of PCBs in small intestinal juice ranging from 8.3% to 26.0% was significantly higher than that in gastric condition, ranging from 4.8% to 12.4%. In addition, significant negative correlations (p<0.05) were observed between KOW and bioaccessibility for all workplace dust samples. Risk assessment indicated that the averaged daily dose of dioxin-like PCBs via non-dietary intake of workplace dust, considering the bioaccessibility of PCBs, were much lower than the TDI of dioxins (2.3 pg WHO-TEQ kgbw(-1)d(-1)) established by Joint FAO/WHO Expert Committee on Food Additives.

Indoor and outdoor dusts from two urban centers in the Pearl River Delta, China, were analyzed and phthalate esters varied from 4.95 to 2,220 µg g(-1) in indoor dust, significantly higher than outdoor dust (1.70-869 µg g(-1)). Di-2-ethylhexyl phthalate (DEHP) was the dominant phthalate found and the highest distribution factor (DF) (1.56 ± 0.41) was noted in the <63 µm fraction (p<0.05). In vitro cytotoxicity of dust extract on human T cell lymphoblast leukemic cell line (CCRF-CEM) indicated by Lethal Concentration 50 (LC50) decreased with particle size. The power model was found as a better fit for explaining the relationship between LC50 and phthalates (R(2)=0.46, p<0.01). Bioaccessibility of phthalates in dust varied with different particle sizes, with the greatest bioaccessible fraction (2.49-38.6%) obtained in <63 µm. Risk assessment indicated that indoor dust ingestion accounted for the major source for DEHP exposure (81.4-96.4% of non-dietary exposure and 36.5% of total exposure), especially for toddlers. The cancer risks associated with DEHP via home dust were high (10(-6)-10(-4)), with 10% of houses estimated with unacceptable risks (>10(-4)). After corrected with the bioaccessibility of phthalates, the cancer risks of dust exposure were moderate (10(-7)-10(-5)).

There is limited information on the bioaccessible fractions of phthalate esters in indoor dust in order to estimate human exposure. In the present study, workplace dust and settled house dust samples from Hong Kong, Shenzhen, and Guangzhou, the three major cities scattered around the Pearl River Delta (PRD) were collected. Chemical analyses showed that the phthalates in workplace dust ranged from 144 to 1810 µg/g, with dust from shopping malls containing the highest level, and in home dust ranged from 181 to 9240 µg/g. The most abundant phthalate ester found was bis(2-ethylhexyl) phthalate (DEHP) in both workplace dust and home dust, followed by di-n-butyl phthalate (DBP) and di-iso-butyl phthalate (DIBP). Principal Components Analysis (PCA) indicated that indoor dust around PRD showed similar phthalate esters patterns of composition. A significant correlation was observed between total phthalate esters concentrations in home dust and the number of year of house construction (p < 0.05). The oral bioaccessibility of phthalate esters in indoor dust ranged from 10.2% (DEHP) to 32% (DMP). Risk assessment indicated that the dominant exposure routes varied in different phthalate esters exposure profiles and the dermal contact exposure pathway was identified as an important route for indoor DEHP exposure.

Ninety-six urban surface dust samples collected from Guangzhou, a typical urban center in South China, were analyzed for 16 PAHs (2-6 rings). ∑PAHs concentrations in the urban surface dust ranged from 0.84 to 12.3µg/g with a mean of 4.80µg/g. High molecular weight compounds (4-6 rings) contributed to 62 to 94% of ∑PAHs mass in the surface dust samples. Four hotspots with highest ∑PAHs were identified via kriging prediction mapping, representing the highly-urbanized regions: central downtown, highway and industrial area. Two major origins of PAHs inputs to urban surface dust were identified as vehicle emissions (51.9%) and coal combustion (26.8%). The 95% UCL of Incremental Lifetime Cancer Risk (ILCR) due to human exposure to urban surface dust PAHs in central South China was 3.03×10(-6) for children and 2.92×10(-6) for adults.

This study investigated total mercury (THg) and methylmercury (MeHg) concentrations in five species of freshwater fish and their associated fish pond sediments collected from 18 freshwater fish ponds around the Pearl River Delta (PRD). The concentrations of THg and MeHg in fish pond surface sediments were 33.1-386 ng g(-1) dry wt and 0.18-1.25 ng g(-1) dry wt, respectively. The age of ponds affected the surface sediment MeHg concentration. The vertical distribution of MeHg in sediment cores showed that MeHg concentrations decreased with increasing depth in the top 10 cm. In addition, a significant correlation was observed between %MeHg and DNA from Desulfovibrionacaea or Desulfobulbus (p<0.05) in sediment cores. Concentrations of THg and MeHg in fish muscles ranged from 7.43-76.7 to 5.93-76.1 ng g(-1) wet wt, respectively, with significant linear relationships (r=0.97, p<0.01, n=122) observed between THg and MeHg levels in fish. A significant correlation between THg concentrations in fish (herbivorous: r=0.71, p<0.05, n=7; carnivorous: r=0.77, p<0.05, n=11) and corresponding sediments was also obtained. Risk assessment indicated that the consumption of largemouth bass and mandarin fish would result in higher estimated daily intakes (EDIs) of MeHg than reference dose (RfD) for both adults and children.

The mutagenicity and genotoxicity of workplace dust including commercial office, secondary school, shopping mall, hospital, electronics factory and manufacturing plant in Hong Kong and settled house dust from Hong Kong, Shenzhen and Guangzhou were measured. Results indicated that indoor dust contained both frameshift and base pair substitution mutagens. Dust from manufacturing plant showed highest mutagenic potency on TA98±S9 and TA100±S9 activation, whereas, electronics factory showed highest genotoxicity with and without S9 activation. TA100 (-S9) mutagenic potency was significantly correlated with genotoxicity expressed as SOSIP (-S9) of workplace dust (r(2)=0.37, p<0.01). The total PAHs concentration of settled house dust from PRD ranged from 1.63 to 29.2µg/g. Linear regression analyses indicated that the PAHs likely accounted for about 45% of the TA98 with S9 mutagenic activity of workplace dust. TA98 (-S9) mutagenicity (r(2)=0.27, p<0.05) and SOSIP (-S9) of house dust (r(2)=0.41, p<0.01) were both significantly correlated with the number of inhabitants in the house. To achieve a more accurate cancer risk assessment, the oral bioaccessibility of B(a)A, Chry, B(b+k)F, B(a)P, D(ah)A and I(cd)P in different dust ranging from 1.3% to 17% was taken into account. Risk assessments indicated that about 26% of house dust samples resulted in unacceptable cancer risk (>1×10(-6)) for preschool children.
