INTRODUCTION: Meta-analyses of genetic association studies are affected by biases and quality shortcomings of the individual studies. We previously developed and validated a risk of bias tool for use in systematic reviews of genetic association studies. The present study describes a larger empirical evaluation of the Q-Genie tool. METHODS AND ANALYSIS: MEDLINE, Embase, Global Health and the Human Genome Epidemiology Network will be searched for published meta-analyses of genetic association studies. Twelve reviewers in pairs will apply the Q-Genie tool to all studies in included meta-analyses. The Q-Genie will then be evaluated on its ability to (i) increase precision after exclusion of low quality studies, (ii) decrease heterogeneity after exclusion of low quality studies and (iii) good agreement with experts on quality rating by Q-Genie. A qualitative assessment of the tool will also be conducted using structured questionnaires. DISCUSSION: This systematic review will quantitatively and qualitatively assess the Q-Genie's ability to identify poor quality genetic association studies. This information will inform the selection of studies for inclusion in meta-analyses, conduct sensitivity analyses and perform metaregression. Results of this study will strengthen our confidence in estimates of the effect of a gene on an outcome from meta-analyses, ultimately bringing us closer to deliver on the promise of personalised medicine. ETHICS AND DISSEMINATION: An updated Q-Genie tool will be made available from the Population Genomics Program website and the results will be submitted for a peer-reviewed publication.

UNLABELLED: This systematic review was performed to compare the diagnostic accuracy of vertebral fracture assessment (VFA) with that of spinal radiography for identification of vertebral fractures (VFs). VFA appeared to have moderate sensitivity and high specificity for detecting VFs when compared with spinal radiography. INTRODUCTION: VFs are recognized as the hallmark of osteoporosis, and a previous VF increases the risk of a future fracture. Therefore, the timely detection of VFs is important for prevention of further fractures. This systematic review examined the diagnostic accuracy of VFA using dual X-ray absorptiometry (DXA) to identify VFs. METHODS: We searched for potentially relevant studies using electronic databases, including Ovid-Medline, Ovid-EMBASE, Cochrane library, and four Korean databases, from their inception to May 2013. We compared the diagnostic accuracy of VFA with that of spinal radiography for detection of VFs by analyzing the sensitivity and specificity using a 2 × 2 contingency table. Subgroup analyses were also performed on studies with a low risk of bias and applicability. RESULTS: Twelve studies were analyzed for the diagnostic accuracy of VFA. The sensitivity and specificity were 0.70-0.93 and 0.95-1.00, respectively, analyzed on a per-vertebra basis, and 0.65-1.00 and 0.74-1.00 on a per-patient basis. The sensitivity and specificity of five studies in subgroups with a low risk of bias in the intervention test were 0.70-0.84 and 0.96-0.99, respectively. In studies with a low risk of bias in the patient selection, those based on a per-vertebra basis in three studies were 0.70-0.93 and 0.96-1.00, respectively. CONCLUSIONS: VFA had moderate sensitivity and high specificity for detecting VF when compared with spinal radiography. However, the present findings are insufficient to assess whether spinal radiography should be replaced by VFA.

In an intercomparison exercise, the Monte Carlo codes most commonly used in gamma-ray spectrometry today were compared with each other in order to gauge the differences between them in terms of typical applications. No reference was made to experimental data; instead, the aim was to confront the codes with each other, as they were applied to the calculation of full-energy-peak and total efficiencies. Surprising differences between the results of different codes were revealed.

Software technology enables computerized analysis to offer second opinion in various screening and diagnostic tasks to assist the clinicians. Yet, the performance of these computerized methods for medical images is questioned by experts in CAD research, owing to the use of different databases and criteria for evaluating the computer results for comparison. This paper intends to substantiate this statement by illustrating the effects of such issues with the use of 1D physiologic data and multiple databases. For this purpose, the detection of desaturation events in Sp02 and spike events in EEG are used. This is the first time that comparison between different algorithms on a common basis is carried out on an individual effort. The appraisal for all the algorithms is made on the same databases and criteria. It is surprising to find that issues for 2/3D images concur with those found in 1D data here. In evaluating the accuracy of a new algorithm, a single independent database gives results fast. This paper reveals weaknesses of such an approach. It is hoped that the supportive evidence shown here is enough for researchers to innovate a better platform for credibility in reporting performance comparison of computerized analysis algorithms.

TRIPOLI-4.3 Monte Carlo transport code has been used to evaluate the QUADOS (Quality Assurance of Computational Tools for Dosimetry) problem P4, neutron and photon response of an albedo-type thermoluminescence personal dosemeter (TLD) located on an ISO slab phantom. Two enriched 6LiF and two 7LiF TLD chips were used and they were protected, in front or behind, with a boron-loaded dosemeter-holder. Neutron response of the four chips was determined by counting 6Li(n,t)4He events using ENDF/B-VI.4 library and photon response by estimating absorbed dose (MeV g(-1)). Ten neutron energies from thermal to 20 MeV and six photon energies from 33 keV to 1.25 MeV were used to study the energy dependence. The fraction of the neutron and photon response owing to phantom backscatter has also been investigated. Detailed TRIPOLI-4.3 solutions are presented and compared with MCNP-4C calculations.

Monte Carlo code TRIPOLI-4.3 has been used to evaluate the N50 neutron slab monitor detection efficiency benchmark organised by the non-destructive assay (NDA) working group of European Safeguards Research and Development Association (ESARDA). The lattice geometry of TRIPOLI-4.3 was applied to simplify the modelling of the N50 monitor, which consists of four 3He tubes embedded in a high-density polyethylene (HDPE) moderator. Using additional HDPE slabs and/or Cd sheets, 10 geometry configurations with different states of moderation were considered in this study. Two cross section libraries from ENDF/B-VI.4 and JEF2.2 and three representations of the 252Cf fission spectrum were investigated and the calculation experiment (C/E) ratios were discussed. The perturbation option was used in order to study the sensitivity to the HDPE density. The S(alpha,beta) thermal scattering in HDPE was also tested to quantify its impact on the N50 detection efficiency calculations.

Although the epicentres of probiotic research in the past decade have been Japan and Europe, researchers in the Asia-Pacific region have actively contributed to the growing understanding of the intestinal microbial ecosystem, and interactions between gut bacteria, diet and health of the human host. A number of new probiotic strains have been developed in the region that have been demonstrated to have beneficial impacts on health in animal and human trials, including improved protection against intestinal pathogens and modulation of the immune system. Probiotics targeted to animals, including aquaculture, feature heavily in many Asian countries. Developments in probiotic technologies have included microencapsulation techniques, antimicrobial production in fermented meats, and synbiotic combinations. In particular, the impact of resistant starch on the intestinal environment and fermentation by intestinal bacteria has been intensively studied and new probiotic strains selected specifically for synbiotic combinations with resistant starch. This paper provides an overview of probiotic research within Australia, New Zealand and a number of Asian countries, and lists scientists in the Asia-Pacific region involved in various aspects of probiotic research and development.

BACKGROUND: The blood cells of crustaceans are involved in phagocytosis of invading microorganisms, contributing to their defense mechanisms. In this study, phagocytic activity of hemocytes of the prawn, Penaeus merguiensis, was quantitated by means of flow cytometric analysis. METHOD: This study was done in vitro. Hemolymph, which was extracted from prawns, was mixed with an equal volume of anticoagulant. Heat-killed Escherichia coli prestained with propidium iodide (PI) was then added. Hemocytes were fixed at various time intervals for flow cytometric analysis. This study was supplemented with electron micrographs using transmission electron microscopy (TEM), which showed three populations of hemocytes. RESULTS: It was observed that those hemocytes that were more active engulfed and digested bacteria readily, thus having higher red fluorescence intensity. The phagocytic activity was expressed as fluorescence unit or engulfed E. coli number per hemocyte. CONCLUSIONS: With this approach, the phagocytic and cellular activity of individual hemocyte populations could be studied quantitatively.
