RESUMO

Perennial monocarpic mass flowering represents as a key developmental innovation in flowering time diversity in several biological and economical essential families, such as the woody bamboos and the shrubby Strobilanthes. However, molecular and genetic mechanisms underlying this important biodiversity remain poorly investigated. Here, we generated a full-length transcriptome resource incorporated into the BlueOmics database (http://blueomics.iflora.cn) for two Strobilanthes species, which feature contrasting flowering time behaviors. Using about 112 and 104 Gb Iso-seq reads together with ~185 and ~75 Gb strand-specific RNA seq data, we annotated 80 971 and 79 985 non-redundant full-length transcripts for the perennial polycarpic Strobilanthes tetrasperma and the perennial monocarpic Strobilanthes biocullata, respectively. In S. tetrasperma, we identified 8794 transcripts showing spatiotemporal expression in nine tissues. In leaves and shoot apical meristems at two developmental stages, 977 and 1121 transcripts were differentially accumulated in S. tetrasperma and S. biocullata, respectively. Interestingly, among the 33 transcription factors showing differential expression in S. tetrasperma but without differential expression in S. biocullata, three were involved potentially in the photoperiod and circadian-clock pathway of flowering time regulation (FAR1 RELATED SEQUENCE 12, FRS12; NUCLEAR FACTOR Y A1, NFYA1; PSEUDO-RESPONSE REGULATOR 5, PRR5), hence provides an important clue in deciphering the flowering diversity mechanisms. Our data serve as a key resource for further dissection of molecular and genetic mechanisms underpinning key biological innovations, here, the perennial monocarpic mass flowering.

BACKGROUND AND AIMS: Flowering plants show a high diversity of pollen morphology, assumed to reflect not only variations in the underlying design, but also stress imposed by ecological conditions related to pollen survival and germination. Both components are expected to constrain the accumulation of pollen disparity. However, this assumption has rarely been tested using empirical data. METHODS: This study is designed to test this hypothesis by inferring the accumulation of pollen disparity in Ericaceae, a large eudicot family with recent, ongoing radiations, with focus on three functionally significant pollen characters using a dated phylogeny. KEY RESULTS: Multiple lines of evidence supported the hypothesis that pollen disparity in Ericaceae did not evolve steadily but rather pulsed over time, clearly decoupling from the relative constant rate pattern of species diversification inferred. In a 3-D pollen morphospace, most major clades appear to occupy distinct neighbouring regions, whereas the subfamily Epacridoideae overlaps extensively with other subfamilies. No evidence for correlations was found between dimension of pollen disparity and species diversity at either the subfamily or generic level. Furthermore, the distribution of species in present pollen morphospace showed a strong central tendency, with the core compartment containing a large number of species from species-rich genera. CONCLUSIONS: The recovered evidence fits well with the expectations of limitations on available pollen morphological disparity, and suggests that innovation of pollen germination traits may have little effect on species diversification.

ETHNOPHARMACOLOGICAL RELEVANCE: Three species of yews Taxus contorta Griff., Taxus mairei (Lemée & Lév.) S.Y. Hu ex T.S. Liu and Taxus wallichiana Zucc. distributed in the Hindu Kush-Himalayan (HKH) region have been commercially exploited in recent decades to extract an anticancer chemotherapeutic drug 'Taxol'. Additionally, indigenous people of this region are using yews for several other purposes including gastro-intestinal disorders, respiratory problems, skeletal system disorders, and as edible fruit, fodder, fish poison, traditional veterinary medicine, among others. AIM OF THE STUDY: The study was designed to document and evaluate knowledge concerning uses of yews among indigenous communities of Mongol and Caucasian origins. MATERIALS AND METHODS: Ethnobotanical knowledge from 10 major ethnic/caste groups of Mongol and Caucasian origins in the Nepal Himalayas was documented in 2010 and 2011 from 27 sites covering the extant distribution range of the three species of Taxus. A total of 72 key informants (60 men, 12 women), recommended by the majority of people in informal group discussions at each study site, were interviewed to collect information on the importance of yews. RESULTS: This study reports multidimensional uses of yews commonly practiced by different indigenous communities of Nepal and compared those with published uses along the HKH region. The key informants cited a total 45 uses under 21 categories. A greater use diversity and high consensus value for use types were recorded for medicinal uses (gastro-intestinal ailments, cough and cold, skeleto-muscular system problem and others medicinal importance) followed by fruit consumption, household tools, agriculture implements and timber. A decline of yew populations and associated traditional knowledge among the younger generations of indigenous people was found. CONCLUSION: The present study shows a strong agreement of ethnobotanical knowledge on yews between communities of Mongols and Caucasian origins. Our findings further revealed the potential for additional therapeutic applications in yews of the HKH region, besides cancer treatment. To compensate the low yield of 'Taxol', and the fact that three yew species are involved, the reported species-specific curative properties need to be validated scientifically and evaluated clinically. Moreover, initiatives should be taken immediately to stop further degradation of yew populations and the associated indigenous knowledge in the HKH region.

Ex situ seed storage underpins global agriculture and food supplies and enables the conservation of thousands of wild species of plants within national and international facilities. As an insurance policy against extinction, ex situ seed conservation is estimated to cost as little as 1% of in situ conservation. The assumptions, costs, risks and scientific challenges associated with ex situ plant conservation depend on the species, the methods employed and the desired storage time. Recent, relatively widespread evidence of less than expected longevity at conventional seed bank temperatures, innovations in the cryopreservation of recalcitrant-seeded species and economic comparators provide compelling evidence that ultra-cold storage should be adopted for the long-term conservation of plants. Policy instruments, such as the Global Strategy for Plant Conservation (2011-2020), should respond to the evidence base and promote the implementation of cryopreservation for both tropical and temperate plants.
