RESUMO

Using Saccharomyces cerevisiae as an experimental model, the potential toxicological effects of Fe3O4 nanoparticles (Fe3O4-NPs) were investigated following exposure to 0-600 mg/L for 24 h. Results revealed that cell proliferation was significantly inhibited by Fe3O4-NPs with an IC50 value of 326.66 mg/L. Mortality showed a concentration-dependent increase, and the highest concentration in this study (600 mg/L) resulted in 22.30% mortality. In addition, Effects on proliferation and mortality were accounted for Fe3O4-NPs rather than iron ion released from Fe3O4-NPs. Scanning and transmission electron microscope observation showed that Fe3O4-NPs extensively attached on the cell surfaces, causing cells to deform and shrink. Moreover, Fe3O4-NPs could be internalized in S. cerevisiae cells via endocytosis and then be distributed in cytoplasm and vesicles. The data of uptake kinetics demonstrated that the maximal accumulation (4.898 mg/g) was reached at 15 h. Besides, percentage of late apoptosis/necrosis was observably increased (p < 0.01) at 600 mg/L (15.80%), and the expression levels of apoptosis-related genes (SOD, Yca1 and Nuc1) were dramatically increased following exposure to Fe3O4-NPs for 24 h. As expected, mitochondrial transmembrane potential was significantly decreased (p < 0.01) at 50-600 mg/L, and biomarkers of oxidative stress (ROS, CAT and SOD) were also markedly changed following exposure. Altogether, the combined results so far indicated Fe3O4-NPs could induce S. cerevisiae cell apoptosis that mediated by mitochondrial impairment and oxidative stress.

BACKGROUND: We examined the association of housing affordability with physical and mental health in Hong Kong, where there is a lack of related research despite having the worst housing affordability problem in the world, considering potential mediating effect of deprivation. METHODS: A stratified random sample of 1978 Hong Kong adults were surveyed. Housing affordability was defined using the residual-income (after housing costs) approach. Health-related quality of life was assessed by the Short-Form Health Survey version 2 (SF-12v2), from which the physical component summary (PCS) and mental component summary (MCS) measures were derived. Multivariable linear regressions were performed to assess associations of housing affordability with PCS and MCS scores, adjusting for sociodemographic, socioeconomic and lifestyle factors. Mediation analyses were also conducted to assess the mediating role of deprivation on the effect of housing affordability on PCS or MCS. RESULTS: Dose-response relationships were observed between housing affordability and mean PCS score (ß (95% CI) compared with the highest affordable fourth quartile: -2.53 (-4.05 to -1.01), -2.23 (-3.54 to -0.92), -0.64 (-1.80 to 0.51) for the first, second and third quartiles, respectively) and mean MCS score (ß (95% CI): -3.87 (-5.30 to -2.45), -2.35 (-3.59 to -1.11), -1.28 (-2.40 to -0.17) for the first, second and third quartiles, respectively). Deprivation mediated 34.3% of the impact of housing unaffordability on PCS and 15.8% of that on MCS. CONCLUSIONS: Housing affordability affects physical and mental health, partially through deprivation, suggesting that housing policies targeting deprived individuals may help reduce health inequality in addition to targeting the housing affordability problem.

Ichthyophthirius multifiliis, an external fish parasite, often causes significant economic damage to the aquaculture industry. Since the use of malachite green was banned, the search of alternative substance to control I. multifiliis infections becomes stringent. In present study, in vitro and in vivo anti-ich efficacies of isopsoralen and psoralidin, two active compounds isolated from methanol extract of Psoralea corylifolia by bioassay-guided fractionation based on the efficacy of anti-ich encysted tomonts, were evaluated. In vitro antiprotozoal efficacy of psoralidin is much better than that of isopsoralen. Psoralidin can kill all theronts at concentrations of 0.8 mg/L or more during 4 h exposure; and terminate reproduction of I. multifiliis post 6 h exposure of protomonts to 0.9 mg/L and encysted tomonts to 1.2 mg/L. In vivo trials showed that 5 h exposure of infected fish to 2.5 mg/L of psoralidin significantly reduced the number of theronts released from tomonts. Furthermore, we observed that a part of protomonts, collected from infected fish post treatment, presented characteristic morphological changes of apoptosis after staining with Annexin V-EGFP/propidium iodide, indicating the possible mechanism of psoralidin against I. multifiliis trophont in situ. On the basis of these results, psoralidin can be used as a potential lead compound for the development of commercial drug against I. multifiliis.

Trifloxystrobin (TFS) is the widely used strobilurin fungicide. However, little information is so far available regarding the uptake kinetics and developmental toxicity of TFS to fish. The present study was conducted to investigate the uptake kinetics, potential environment risk and toxicity of TFS on Gobiocypris rarus embryos. Results revealed that increased malformation, decreased body length and heart rate, affected spontaneous movement and swimming speed provide a gradual concentration-dependent manner; values of 144 h LC50 (median lethal concentration) and EC50 (median effective concentration) were 1.11 and 0.86 µg L(-1). Continuous exposure to TFS resulted in a steady accumulation with no evidence of elimination. Enzyme activities were significantly changed; reactive oxygen species and DNA damage were significantly induced after TFS treatment. Certain genes related to cell apoptosis (p53), metabolism (cyp1a), stress response (hsp70) and blood vessels (vezf1) development were all significantly up-regulated. This is the first study to define uptake kinetics and to focus on behavioral consequences, physiological changes and mRNA expression following TFS exposure in the early life stages of fish. Our results suggest that TFS is highly toxic to fish embryos.

Cyanobacteria are a diverse group of Gram-negative bacteria that produce an array of secondary compounds with selective bioactivity against vertebrates, invertebrates, fungi, bacteria and cell lines. Recently the main methods of controlling cyanobacteria are using chemicals, medicinal plants and microorganism but fewer involved the safety research in hydrophytic ecosystems. In search of an environmentally safe compound, 53 chemicals were screened against the developed heavy cyanobacteria bloom Microcystis aeruginosa using coexistence culture system assay. The results of the coexistence assay showed that 9 chemicals inhibited M. aeruginosa effectively at 20 mg L(-1) after 7 days of exposure. Among them dimethomorph, propineb, and paraquat were identified that they are safe for Chlorella vulgaris, Scenedesmus obliquus, Carassius auratus (Goldfish) and Bacillus subtilis within half maximal effective concentration (EC50) values 5.2, 4.2 and 0.06 mg L(-1) after 7 days, respectively. Paraquat as the positive control observed to be more efficient than the other compounds with the inhibitory rate (IR) of 92% at 0.5 mg L(-1). For the potential inhibition mechanism, the chemicals could destroy the cell ultrastructure in different speed. The safety assay proved dimethomorph, propineb and paraquat as harmless formulations or products having potential value in M. aeruginosa controlling, with the advantage of its cell morphology degrading ability.

Toxicity assessment of Zn-Co mixtures involves multiple ions interactions. The negative potential (ψ0) at the cell membrane surface (CMs) concentrated cationic toxicants (denoted {M(2+)}0) and influenced the rhizotoxicity of Co(2+) or Zn(2+). The single and joint rhizotoxicity of Co(2+) and Zn(2+) to wheat (Triticum aestivum L.) were examined, coupled with different Ca(2+) levels. Joint effects of Zn(2+), Co(2+) and Ca(2+) were estimated by the linearly extended concentration addition (CA) and response addition (RA) models. Incorporation of Ca(2+) in single metal toxicity assessment significantly enhanced the prediction accuracy (r(2) increased from 0.948 to 0.550 for Zn(2+) and from 0.903 to 0.611 for Co(2+), respectively). ψ0 affected the multiple metals toxicity in both conventional mixture models (r(2)=0.814 for CA model and 0.820 for RA model). Concretely, {Zn(2+)}0 alleviated the toxicity of {Co(2+)}0, while {Co(2+)}0 had non-significant effect on {Zn(2+)}0 toxicity. Growth responses to {Ca(2+)}0 were substantially affected by {Zn(2+)}0 and {Co(2+)}0. Ca addition in medium decreased the {M(2+)}0 by reducing the ψ0 negativity, moreover this addition alleviated Ca deficiency at CMs induced by Zn(2+) (or Co(2+)). These consistent results from both extended CA and RA models indicated that ψ0 provided a novel sight for understanding the rhizotoxicity of multiple metals.

Lantang pig (16 sows), which is one of the south China type pig breeds, was crossed with Landrace pig (8 boars) to construct the resource population. According to the pig linkage map of USDA-MARC2.0, 31 microsatellite DNA markers on pig chromosomes 1, 4, 7, and 8 were used to genotype the parents F1 and F2. The distance between adjacent markers was about 10 to 20 cM. The gene frequency, heterozygosity (h), and polymorphism information content (PIC) were calculated. The marker genotype of parents, F1, and F2 were obtained with WAVEO nucleotide fragment analysis system (DHPLC) and ABI 377 DNA sequencer. Twenty-one microsatellite markers on chromosomes 1, 4, and 8 were genotyped with ABI 377. The length of the DNA fragments of 18 alleles on 13 microsatellite markers were beyond the range reported on the web site. The loci of new alleles were 62% of total markers. The heterozygosities of the 31 microsatellite ranged from 0.043 to 0.7855, the heterozygosities of 70% loci were over 0.6, the average heterozygosity was 0.6460. The average polymorphism information content (PIC) of 31 microsatellite markers was 0.5949, the PIC of 77.4% loci were over 0.5. The values of h and PIC suggested that polymorphism information of these markers in the resource population was plentiful. These markers could be used to map quantitative traits loci of important economic traits in this population.
