RESUMO

Cadmium (Cd) pollution induced by smelting process is of great concern worldwide. However, the comprehensive risk assessment of Cd exposures in smelting areas with farming coexist is lacking. In this study, atmospheric deposition, soil, surface and drinking water, rice, wheat, vegetable, fish, pork, and human hair samples were collected in rice-wheat rotation area near nonferrous smelter to investigate smelting effect on environmental Cd pollution and human health. Results showed high Cd deposition (0.88-2.61 mg m-2 year-1) combined with high bioavailability (37-42% totality) in study area. Moreover, 90%, 83%, 57%, and 3% of sampled soil, wheat, rice, and vegetable of Cd were higher than national allowable limits of China, respectively, indicating smelting induced serious environmental Cd pollution. Especially, higher Cd accumulation occurred in wheat compared to rice by factors of 1.5-2.0. However, as for Cd exposure to local residents, due to rice as staple food, rice intake ranked as main route and accounted for 49-53% of total intake, followed by wheat and vegetable. Cd exposure showed high potential noncarcinogenic risks with hazard quotient (HQ) of 0.63-4.99 using Monte Carlo probabilistic simulation, mainly from crop food consumption (mean 94% totality). Further, residents' hair Cd was significant correlated with HQ of wheat and rice ingestion, highlighting negative impact of cereal pollution to resident health. Therefore, smelting process should not coexist with cereal cultivating.

The risk assessment of heavy metals (HMs) in sewage sludge (SS) is essential before land application. Six HMs in nineteen SS collected in the Yangtze River Delta were analyzed to assess risks to environment, ecosystem, and human health. HMs concentrations were ranked in the order of Zn > Cu > Cr > Ni > Pb > Cd, with Cu, Zn, and Ni in a total of 16% of samples exceeding the legal standard. Zn showed greatest extractability according to EDTA-extractable concentrations. HMs in 16% of SS samples posed heavy contamination to the environment with Zn as the major pollutant. HMs in 26% of samples posed ecological risk to the ecosystem and Cd was the highest risky HM. The probabilistic health risk assessment revealed that HMs posed carcinogenic risks to all populations, but non-carcinogenic risks only to children. This work will provide fundamental information for land application of SS in this area.

The Japanese encephalitis virus (JEV) is a mosquito-borne flavivirus that causes viral encephalitis in humans, pigs and other mammals across Asia and the Western Pacific. Genetic screening tools such as CRISPR screening, DNA sequencing and RNA interference have greatly improved our understanding of JEV replication and its potential antiviral approaches. However, information on exon and intron mutations associated with JEV replication is still scanty. CRISPR-Cas9-mediated cytosine base editing can efficiently generate C: G-to-T: A conversion in the genome of living cells. One intriguing application of base editing is to screen pivotal variants for gene function that is yet to be achieved in pigs. Here, we illustrate that CRISPR-Cas9-mediated cytosine base editor, known as AncBE4max, can be used for the functional analysis of calreticulin (CALR) variants. We conducted a CRISPR-Cas9-mediated cytosine base editing screen using 457 single guide RNAs (sgRNAs) against all exons and introns of CALR to identify loss-of-function variants involved in JEV replication. We unexpectedly uncovered that two enriched sgRNAs targeted the same site in intron-2 of the CALR gene. We found that mutating four consecutive G bases in the intron-2 of the CALR gene to four A bases significantly inhibited JEV replication. Thus, we established a CRISPR-Cas9-mediated cytosine-base-editing point mutation screening technique in pigs. Our results suggest that CRISPR-mediated base editing is a powerful tool for identifying the antiviral functions of variants in the coding and noncoding regions of the CALR gene.

Soil salinization and water deficits are considered the primary factors limiting economic development and environmental improvement in arid areas. However, there remains limited knowledge of the adaptability of typical shrubs to salinization of desert areas in arid zones. This study was conducted in a desert oasis transition zone (Tarim River, China), aiming to investigate: i) the spatial-temporal changes in soil salinity; ii) the interactions between the pedoenvironment vs typical shrub (Calligonum mongolicum). The van Genuchten soil salinity retention ensemble model (TVGSSREM-3D) was developed to simulate variations in soil water-salt transport in the desert-oasis zone and to accurately explain the main factors influencing Calligonum mongolicum desert-oases transition areas. The results showed that monthly average salinity ranged from 2.0 to 8.0 g kg-1, with a peak in August (9.17 g kg-1). The presence of human activities (Salt Drainage Canal) and the distribution of Calligonum mongolicum resulted in a clear spatial salinity zonation. Moreover, analysis of environmental indicators using the TVGSSREM-3D model revealed strong correlations between the distribution of salinity in Calligonum mongolicum desert-oases transition areas and groundwater depth (GD), minimum relative humidity (MRH), and water vapor pressure (WVP). These findings provide a scientific basis for stabilizing, restoring, and reconstructing the ecosystem of the oasis-desert transition zone.

To address the challenge of no-reference image quality assessment (NR-IQA) for authentically and synthetically distorted images, we propose a novel network called the Combining Convolution and Self-Attention for Image Quality Assessment network (Conv-Former). Our model uses a multi-stage transformer architecture similar to that of ResNet-50 to represent appropriate perceptual mechanisms in image quality assessment (IQA) to build an accurate IQA model. We employ adaptive learnable position embedding to handle images with arbitrary resolution. We propose a new transformer block (TB) by taking advantage of transformers to capture long-range dependencies, and of local information perception (LIP) to model local features for enhanced representation learning. The module increases the model's understanding of the image content. Dual path pooling (DPP) is used to keep more contextual image quality information in feature downsampling. Experimental results verify that Conv-Former not only outperforms the state-of-the-art methods on authentic image databases, but also achieves competing performances on synthetic image databases which demonstrate the strong fitting performance and generalization capability of our proposed model.

Water resources in Central Asia are very scarce due to natural and anthropogenic impacts. Water shortages have been a major factor hampering the socio-economic development of Central Asia. Exploring internal interactions among climate change, human activities and terrestrial hydrological cycles will help to improve the management of water resources in Central Asia. In this paper, hydro-climatic and anthropogenic data for the period 2003-2016 from the Gravity Recovery and Climate Experiment (GRACE), the Global Land Data Assimilation System (GLDAS), the Climatic Research Unit (CRU) and the Moderate Resolution Imaging Spectroradiometer (MODIS) were used to analyze the influence of natural factors and human activities on changes of available water (AWC). The terrestrial water storage derived from GRACE and GLDAS remarkably declined in 2008, due to a serious drought, but increased thereafter. The AWC positively responded to the vegetation index, evapotranspiration, potential evapotranspiration and air temperature at a lag of 0-1 month, but to precipitation at a lag of 2-3 months. Results of correlation analysis with a spatial square moving window indicated that forests, grasses, croplands and water areas presented significantly positive correlations with AWC, while barren areas and urban areas were negatively correlated with AWC. According to the Boruta algorithm and the Random Forest model, natural factors, namely precipitation, evapotranspiration and potential evapotranspiration, were major factors for AWC in the whole Central Asia. Human activities had direct and indirect impacts on AWC. With the development of society and economy, croplands and urban areas gradually increased, resulting in a rising demand for water withdrawals for agriculture irrigation and industry. The unreasonable utilization and exploitation of water resources led to vegetation degradation and ecosystem deterioration, which would worsen the shortage of water resources in arid regions of Central Asia.

There is an increasing evidence linking protective effect of selenium (Se) against Pb toxicology; however, Pb exposure risk assessments usually consider only the environmental Pb contamination and dietary intake. Based on the current understanding of mechanisms of SePb interactions, the physiological function/toxicology of Se and the toxicology of Pb, a new criterion for Se and Pb exposure assessment is developed. Additionally, seven existing criteria were also used to assess the resident health risks around a smelter in China. The Pb concentrations in locally-produced foods exceeded the national tolerance limits of China and the Se in the foods were similar to those in areas with adequate Se levels. In accordance with the illustrated assessments of the new criterion and seven existing criteria, we found a large knowledge gap between the new and traditional assessments of exposure to Pb and/or Se. The new assessment criteria suggested that almost all the residents were facing the Se deficiency and 58% of the residents not only had the adverse health of Se deficiency, but also had the health risks of Pb toxicity. The Pb and Se in the hair and urine may partly support the new criterion. This study suggested that the process of Se counteracting the Pb toxicity may result in Se deficiency. Pb exposure combined Se intake should be considered in future assessments of Pb exposure (or Se intake).

The relationship between economic development and environment and the evolution characteristics of spatial pattern in Hexi Corridor of Northwest China were analyzed based on Landsat images in 1985, 1995, 2000 and 2011 with twenty counties in Hexi Corridor chosen as the basic research units. The ecological economic harmony during 1985-2011 was estimated according to ESV (ecosystem services value) and EEH (ecological and economic harmony) index with the ecosystem services value estimation methods. The results showed that the land type of the study area dramatically changed during the study period, the grassland decreased badly, and the construction land and cultivated land increased quickly. The ESV showed an overall downward trend, especially in the Shiyang River basin and the middle of Heihe River. The ESV in the Shule River basin in this period. After 2000, the economic growth speeded up visibly in the study area. The economic development concentrated in the resource-based cities and regional central cities, and declined from the center of corridor to the both sides. The ecological-economic relation in Hexi Corridor experienced a transformation of "preliminary deterioration--further deterioration--low grade coordination". The EEH had large changes in the Shiyang River basin and the middle of Heihe River, which experienced a transformation of "conflict--more conflicts--less conflicts", however, there was little change in Shule River basin. The development mode and the comprehensive reclamation of Shiyang River basin and Heihe River basin had a significant influence on the regional ecological and economic harmony.

PCR-RFLP was used to analyze the polymorphisms of MC4R, LEP, H-FABP genes in a swine breed composite (DIV2) and 4 swine breeds (Yorkshire, Landrace, Meishan, Bamei). The association study of these polymorphisms with several economic traits was carried out on a DIV2 population. The results obtained showed that MC4R/TaqI genotype had an effect for average backfat thickness (P < 0.05) and lean meat percentage (P < 0.05). At locus LEP/HinfI animals of AA genotype had lower test daily gain than that of BB (P < 0.01) or AB genotype (P < 0.05). At the H-FABP/HaeIII locus lean meat percentage of the individuals with genotype DD were higher than that with genotype dd (P < 0.05). Linkage disequilibrium analysis among MC4R, LEP and H-FABP revealed that these genes were independent. This represented two or more genes that could be combined together within one genotype in order to facilitate breeding for objective traits. In addition, a method allowing simultaneous detection of fragments of MC4R and LEP gene was developed.
