BACKGROUND: Research suggests that effort-cost decision-making (ECDM), the estimation of work required to obtain reward, may be a relevant framework for understanding motivational impairment in psychotic and mood pathology. Specifically, research has suggested that people with psychotic and mood pathology experience effort as more costly than controls, and thus pursue effortful goals less frequently. This study examined ECDM across psychotic and mood pathology. HYPOTHESIS: We hypothesized that patient groups would show reduced willingness to expend effort compared to controls. STUDY DESIGN: People with schizophrenia (N = 33), schizoaffective disorder (N = 28), bipolar disorder (N = 39), major depressive disorder (N = 40), and controls (N = 70) completed a physical ECDM task. Participants decided between completing a low-effort or high-effort option for small or larger rewards, respectively. Reward magnitude, reward probability, and effort magnitude varied trial-by-trial. Data were analyzed using standard and hierarchical logistic regression analyses to assess the subject-specific contribution of various factors to choice. Negative symptoms were measured with a clinician-rated interview. STUDY RESULTS: There was a significant effect of group, driven by reduced choice of high-effort options in schizophrenia. Hierarchical logistic regression revealed that reduced choice of high-effort options in schizophrenia was driven by weaker contributions of probability information. Use of reward information was inversely associated with motivational impairment in schizophrenia. Surprisingly, individuals with major depressive disorder and bipolar disorder did not differ from controls. CONCLUSIONS: Our results provide support for ECDM deficits in schizophrenia. Additionally, differences between groups in ECDM suggest a seemingly similar behavioral phenotype, reduced motivation, could arise from disparate mechanisms.

BACKGROUND: Working memory (WM) deficits are seen as a core deficit in schizophrenia, implicated in the broad cognitive impairment seen in the illness. Here we examine the impact of WM storage of a single item on the operation of other cognitive systems. METHODS: We studied 37 healthy controls (HCS) and 43 people with schizophrenia (PSZ). Each trial consisted of a sequence of two potential target stimuli, T1 and T2. T1 was a letter presented for 100 ms. After delays of 100-800 ms, T2 was presented. T2 was a 1 or a 2 and required a speeded response. In one condition, subjects were instructed to ignore T1 but respond to T2. In another condition, they were required to report T1 after making their speeded response to T2 (i.e. to make a speeded T2 response while holding T1 in WM). RESULTS: PSZ were dramatically slowed at responding to T2 when T1 was held in WM. A repeated measures ANOVA yielded main effects of group, delay, and condition with a group by condition interaction (p's < 0.001). Across delays, the slowing of the T2 response when required to hold T1 in memory, relative to ignoring T1, was nearly 3 times higher in PSZ than HCS (633 v. 219 ms). CONCLUSIONS: Whereas previous studies have focused on reduced storage capacity, the present study found that PSZ are impaired at performing tasks while they are successfully maintaining a single item in WM. This may play a role in the broad cognitive impairment seen in PSZ.

Event-related potentials (ERPs) are a powerful tool in understanding and evaluating cognitive, affective, motor, and sensory processing in both healthy and pathological samples. A typical ERP recording session takes considerable time but is designed to isolate only 1-2 components. Although this is appropriate for most basic science purposes, it is an inefficient approach for measuring the broad set of neurocognitive functions that may be disrupted in a neurological or psychiatric disease. The present study provides a framework for more efficiently evaluating multiple neural processes in a single experimental paradigm through the manipulation of functionally orthogonal dimensions. We describe the general MONSTER (Manipulation of Orthogonal Neural Systems Together in Electrophysiological Recordings) approach and explain how it can be adapted to investigate a variety of neurocognitive domains, ERP components, and neural processes of interest. We also demonstrate how this approach can be used to assess group differences by providing data from an implementation of the MONSTER approach in younger (18-30 y of age) and older (65-85 y of age) adult samples. This specific implementation of the MONSTER framework assesses 4 separate neural processes in the visual domain: (1) early sensory processing, using the C1 wave; (2) shifts of covert attention, with the N2pc component; (3) categorization, with the P3 component; and (4) self-monitoring, with the error-related negativity. Although the MONSTER approach is primarily described in the context of ERP experiments, it could also be adapted easily for use with functional magnetic resonance imaging.

To determine whether data quality is meaningfully reduced by high electrode impedance, EEG was recorded simultaneously from low- and high-impedance electrode sites during an oddball task. Low-frequency noise was found to be increased at high-impedance sites relative to low-impedance sites, especially when the recording environment was warm and humid. The increased noise at the high-impedance sites caused an increase in the number of trials needed to obtain statistical significance in analyses of P3 amplitude, but this could be partially mitigated by high-pass filtering and artifact rejection. High electrode impedance did not reduce statistical power for the N1 wave unless the recording environment was warm and humid. Thus, high electrode impedance may increase noise and decrease statistical power under some conditions, but these effects can be reduced by using a cool and dry recording environment and appropriate signal processing methods.
