A major public health concern in Mexico is the natural contamination of groundwater with fluoride and arsenic. Therefore, this work aimed to evaluate the magnitude of human health risk after determining fluoride and arsenic concentrations in groundwater samples (n = 50) from the Metropolitan area of the city of San Luis Potosi, Mexico. Fluoride levels in water were determined via a potentiometric method using an ion-selective electrode. Arsenic concentrations in water samples were determined with an Atomic Absorption technique. Subsequently, a probabilistic health risk assessment was developed (Monte Carlo Analysis). Fluoride levels in water ranged from 0.20 to 3.50 mg/L. For arsenic, the mean level found in the assessed water samples was 15.5 ± 5.50 µg/L (range: 2.50-30.0 µg/L). In addition, when the probabilistic health risk assessment was completed, a mean HI (cumulative hazardous index) of higher than 1 was detected, indicating a high NCR (non-carcinogenic risk) for children and adults. According to the results found in this study, exposure protection campaigns are imperative in the Metropolitan area of the city of San Luis Potosí, Mexico, to successfully diminish exposure to arsenic and fluoride and, as a consequence, decrease the NCR in the population living in that region of Mexico.

Health complications have been associated with polycyclic aromatic hydrocarbons (PAHs) exposure, a widespread environmental pollutants family. Therefore, the objective of this investigation was to develop a probabilistic health risk evaluation (using Monte-Carlo simulation) in an infantile population living in areas with a high risk of pollution by PAHs (indoor wood combustion, brick kiln industry, municipal landfill, and low and high vehicular traffic) in Mexico. Urine samples were obtained from Mexican children (n = 135) and urinary 1-OHP concentrations (used as a PAHs biomarker) were quantified. Highest urinary 1-OHP concentrations were detected in children living in areas that use wood combustion as the principal indoor fuel (3.50 ± 0.95 µg/L). Nevertheless, estimated hazard quotients (HQ) lower than 1 were found in all assessed sites after Monte-Carlo analysis. Although HQ <1.0 (a toxic effect is not expected), more data are necessary to determine the real impact of PAHs exposure on children health status.

Recent studies have documented environmental contamination by PCBs in soil from different areas in Mexico (industrial, mining, and urban sites). However, the real significance of that soil contamination has not been established. Therefore, the aim of this study was to perform a human health risk assessment (Monte Carlos simulation) to evaluate the probable toxic effects of soils contaminated with PCBs on children in four sites in Mexico. A high non-carcinogenic risk (total nHQ = 1.1E+01; if nHQ ≥1, hazardous health effects cannot be ruled out) was found in Alpuyeca, Morelos, Mexico. Moreover, the total CR (cancer risk) found in Alpuyeca, Morelos is of concern (total CR = 5.1E-03), being that a cut-point of 1.0E-06 has been suggested as a safe level for cancer risk. Taking into consideration the data shown in this research, we conclude that a strategy to protect human health is necessary for the assessed sites.

It has been demonstrated that the human biomonitoring of susceptible populations is a valuable method for the identification of critical contaminants. Therefore, the purpose of this study was to assess the exposure profile for arsenic (As), lead (Pb), mercury (Hg), 1-hydroxypyrene (1-OHP), 1,1-bis(p-chlorophenyl)-2,2,2-trichloroethane (DDT), 1,1-dichloro-2,2-bis(p-chlorophenyl) ethylene (DDE), polybrominated diphenyl ethers (PBDEs), and polychlorinated biphenyls (PCBs) in children living in Ciudad Juarez, Chihuahua, Mexico (a major manufacturing center in Mexico). In 2012, we evaluated a total of 135 healthy children living in Ciudad Juarez since birth. The total PBDEs levels ranged from nondetectable (< LOD) to 215 ng/g lipid, with a mean total PBDEs level of 29.5 ± 53.0 ng/g lipid (geometric mean ± standard deviation). The mean total PCBs level in the study participants was 29.0 ± 10.5 ng/g lipid (range 4.50-50.0 ng/g lipid). Recently, in developing countries, polycyclic aromatic hydrocarbons (PAHs) have been considered contaminants of grave concern for women and children. Therefore, the aim of this study was twofold: (1) evaluate exposure assessment to PAHs using urinary 1-hydroxypyrene (1-OHP) as an exposure biomarker and (2) perform a health risk assessment in women from four different high risk scenarios in Mexico. From 2012 to 2013, in a cross-sectional study, we evaluated a total of 184 healthy women from the following scenarios: (A) indoor biomass combustion site (n = 50); (B) brick manufacturing site using different materials such as fuel sources (n = 70); (C) industrial site (n = 44); and (D) high vehicular traffic site (n = 20). 1-hydroxypyrene (1-OHP) was quantified using a high-performance liquid chromatography (HPLC) technique. Afterward, a probabilistic health risk assessment was performed (Monte Carlo analysis). Mean urinary 1-OHP levels found were 0.92 ± 0.92; 0.91 ± 0.83; 0.22 ± 0.19; and 0.14 ± 0.17 µg/L for scenario A, B, C, and D, respectively. Then, based on the measured urinary 1-OHP levels, the estimated median daily intake doses of pyrene were calculated: 659, 623, 162, and 77.4 ng/kg/day for the women participating in the study living in areas A, B, C, and D, respectively, and finally, the hazard quotient (HQ) was calculated (22 ± 21, 21 ± 20, 5.5 ± 5.5, and 2.6 ± 3.5; for areas A, B, C, and D, respectively), high health risk was noted for the women living in the studied communities. The data shown in this study (exposure levels to PAHs and health risk assessment) made it reasonable to conclude that the exposure levels found have a significant potential for generating adverse effects on human health in the studied scenarios.

The use of solid fuels for cooking and heating is likely to be the largest source of indoor air pollution on a global scale; these fuels emit substantial amounts of toxic pollutants such as polycyclic aromatic hydrocarbons (PAHs) when used in simple cooking stoves (such as open "three-stone" fires). Moreover, indoor air pollution from biomass fuels is considered an important risk factor for human health. The aim of this study was to evaluate the relationship between exposure to PAHs from wood smoke and vascular dysfunction; in a group of Mexican women that use biomass combustion as their main energy source inside their homes. We used 1-hydroxypyrene (1-OHP) as an exposure biomarker to PAHs and it was assessed using high performance liquid chromatography. The endothelium-dependent vasodilation was assessed through a vascular reactivity compression test performed with a pneumatic cuff under visualization of the brachial artery using high resolution ultrasonography (HRU). Assessment of the carotid intima-media thickness (CIMT) was used as an atherosclerosis biomarker (also assessed using HRU); and clinical parameters such as anthropometry, blood pressure, glucose, triglycerides, total cholesterol, HDL cholesterol, LDL cholesterol, among others were also evaluated. The mean concentration of urinary 1-OHP found in exposed women was 0.46±0.32µmol/mol Cr (range: 0.086-1.23µmol/mol Cr). Moreover, vascular dysfunction (diminished endothelium dependent vasodilation) was found in 45% of the women participating in the study. Association between vascular function and 1-OHP levels was found to be significant through a logistic regression analysis (p=0.034; r(2)=0.1329). Furthermore, no association between CIMT and clinical parameters, urinary 1-OHP levels or vascular dysfunction was found. Therefore, with the information obtained in this study, we advocate for the need to implement programs to reduce the risk of exposure to PAHs in communities that use biomass fuels as a main energy source.

In Mexico, polychlorinated biphenyls (PCBs) were principally used as heat transfer chemicals in electric transformers and capacitors as well as hydraulic fluids and lubricants in heavy electrical equipment since the early 1940s. However, although PCBs have been banned in Mexico, their past and present improper disposal has resulted in environmental contamination. Therefore, the aim of this study was to evaluate the PCBs levels in soil samples in the immediate area of an electric capacitor manufacturing industry, which was established several years ago in Alpuyeca, Morelos, Mexico. To confirm the presence of PCBs, surface soil samples (1-5 cm in depth) were collected from the vicinity of the industry. We determined the concentrations of 40 PCB congeners in soil samples using gas chromatography/mass spectrometry. The total PCBs levels in the soil samples ranged from 6.2 to 108460.6 µg kg(-1). Moreover, when we analyzed the results of the congeners (non-dioxin-like PCBs and dioxin-like PCBs), the levels of non-dioxin-like PCB congeners ranged from 5.7 to 103469 µg kg(-1) and the levels of dioxin-like PCB congeners ranged from 0.5 to 4992 µg kg(-1). Considering that soil is an important pathway of exposure in humans, analysis of PCBs levels in blood (as a biomarker of exposure) is necessary in individuals living in Alpuyeca, Morelos.

The purpose of this study was to measure levels of polybrominated diphenyl ethers (PBDEs) in the blood of children (50 individuals) living in Guadalajara, Jalisco, Mexico. We analyzed six PBDE congeners by gas chromatography-mass spectrometry. Total PBDE levels ranged from not detectable (nd) to 15.2 µg/L on a whole-weight basis and from nd to 6,435 ng/g lipid on a lipid-weight basis. The dominant congener in our study was BDE-153, followed by BDE-154, BDE-99, BDE-100, and BDE-47. Levels of BDE-209 were below the detection limit. Our data indicate that children living in the areas studied in this work are exposed to high levels of PBDEs.

The aim of this study was to evaluate hexachlorocyclohexane (HCH) exposure in children living in nine hot spots in four Mexican states. We analyzed HCH (α, ß, and γ-isomers) in blood using gas chromatography/mass spectrometry. HCH exposure level in 261 children was assessed and approximately 75 % of the children studied had detectable levels of HCH. These levels ranged from 188 to 40,096.7 ng/g lipid. The highest mean levels were found in Lacanja (5,446.9 ng/g lipid), an indigenous community in Chiapas, Mexico. Our data indicate high exposure to HCH in children living in these communities.

The aim of this study was to evaluate the exposure levels to persistent organic pollutants and 1-hydroxypyrene in children living in an endemic malaria zone in Mexico. The blood levels for 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT), 1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (DDE) and lindane ranged from 15.4 to 17,886.5 ng/g lipid, 6,624.3 to 100,119.0 ng/g lipid, and 351.1 to 6,153.8 ng/g lipid, respectively. For total polychlorinated biphenyls the blood levels ranged from 2,584.9 to 14,547.9 ng/g lipid. Regarding urinary 1-hydroxypyrene levels, the mean level was 2.9 ± 3.1 µmol/mol creatinine. In conclusion, the children in our study are exposed to levels higher than normal to mixtures of environmental contaminants.

PURPOSE: The aim of this study was assess co-exposure to DDT, DDE (main DDT metabolite), and PAHs (1-hydroxypyrene) in areas where biomass is used to cook and to heat homes and where DDT was used to combat malaria transmission. METHODS: During 2009, we analyzed a total of 190 blood and urine samples from children living in six communities in Mexico. Taking into account the environmental persistence and the toxicity of DDT, the Pan American Health Organization (PAHO) organized a surveillance program in Mesoamerica which included the detection of residual DDT in environmental (soil) and biological samples (fish tissue and children's blood). This program was carried out in communities from Mexico, Guatemala, El Salvador, Honduras, Nicaragua, Costa Rica and Panama. This paper presents the first report of that program. As expected, the results show that the levels for [summation operator] DDT in soil (outdoor or indoor) and fish samples in the majority of the locations studied are below guidelines. However, in some locations, we found children with high concentrations of DDT as in Mexico (mean level 50.2 ng/mL). Furthermore, in some communities and for some matrices, the DDT/DDE quotient is higher than one and this may reflect a recent DDT exposure. Therefore, more efforts are needed to avoid exposure and to prevent the reintroduction of DDT into the region. In this regard it is important to know that under the surveillance of PAHO and with the support of UNEP, a regional program in Mesoamerica for the collection and disposal of DDT and other POPs stockpiles is in progress.

PURPOSE: Biological monitoring of polycyclic aromatic hydrocarbons (PAHs) has expanded rapidly since urinary 1-hydroxypyrene (1-OHP) was suggested as a biological index for pyrene. Taking into account that pyrene is often present in PAHs mixtures, 1-OHP has also been considered an indirect indicator of exposure to these mixtures. Sources of PAHs in developing countries are numerous; however, exposure of children to PAHs has not been studied in detail. Therefore, the aim of this study was to assess exposure of children to PAHs in different scenarios: (a) children living next to highways with heavy traffic; (b) sanitary landfill; (c) brick kiln communities and (d) children exposed to biomass combustion. METHODS: A total of 258 children (aged 3-13) participated in the study. The analyses were performed by HPLC with fluorescence detector. Urinary 1-OHP concentrations were then adjusted by urinary creatinine. RESULTS: The highest levels of 1-OHP in this study were found in children exposed to biomass combustion (mean value 3.25 micromol/mol creatinine), but exposure was also detected in children living in communities with brick kiln industry (mean 0.35 micromol/mol creatinine), or in a community next to a sanitary landfill (with waste combustion) (0.30 micromol/mol creatinine) and in children exposed to traffic (mean value 0.2 micromol/mol creatinine and 0.08 micromol/mol creatinine). CONCLUSIONS: Considering our results and taking into account that millions of children in Mexico are living in scenarios similar to those studied in this work, the assessment of health effects in children exposed to PAHs is urgently needed; furthermore, PAHs have to be declared contaminants of concern at a national level.

Flame retardants (FRs) constitute a group of compounds that are added to materials in order to suppress, reduce, or delay fire. At present the most used FRs are the polybrominated diphenyl ethers (PBDEs), and diverse studies have found individuals exposed to them. However, few studies have reported data in children. The objective of this report was to assess PBDEs levels in children of six communities in México. During the year 2006 we analyzed a total of 173 healthy children (aged 6-13 years old). Plasma samples were taken and quantified (gas chromatography/mass spectrometer) for PBDEs. Six PBDEs congeners (BDE-47, BDE-99, BDE-100, BDE-153, BDE-154, and BDE-209) were quantified in blood serum. We detected exposure to PBDEs in all the communities. The total PBDEs levels ranged from no detectable (nd) to 43.4 ng g(-1) lipid, the dominant PBDE congener was BDE-47, followed by BDE-100, BDE-99 and BDE-153, whereas the levels of BDE-209 were below LOD. Children living in an industrial and urban area (Cd. Juarez, Chih) had the highest levels of PBDEs, approximately two times that of children living in El Refugio, S.L.P. (a rural area) or in Milpillas, S.L.P. (municipal landfill) and 4-5 times higher than levels found in children living in San Luis Potosi, S.L.P. (urban area), in Chihuahua, Chih. (urban area), and San Juan Tilapa, Edo. Mex. (municipal landfills). Results cannot be generalized since the communities selected are not representative of the Mexican population. However, they do indicate that Mexican children are exposed to PBDEs.

Environmental policies in Mexico have contributed to the reduction in the production or use of some persistent organic pollutants (POPs) and metals. However, monitoring of POPs concentrations in humans living in hot spots is lacking. Therefore, the objective of this study was to conduct a screening for POPs and metals first in Mexican children living in high-risk areas. During the year 2004, we analyzed a total of 229 healthy children (aged 6-12 years old) who resided in communities located in nine Mexican states. Organochlorine insecticides, PCBs and metals were quantified in plasma and urine samples. We detected p'p-DDE in all the children; moreover, p'p-DDT, lindane and hexachlorobenzene were detected respectively in 14%, 85% and 10% of the children studied. Measurable levels of PCBs were recorded in only one community, where six of 14 PCB congeners assayed were detected (numbers 52, 118, 138, 153, 170 and 180). All the children had detectable levels of lead in their blood (mean level, 4.6 microg dL(-1)); furthermore, 57% of the children studied had levels higher than 5.0 microg/dL. The mean level of urinary arsenic (UAs) for all the children was 22.35 microg g(-1) creatinine and 15% of those children had concentrations of UAs above 50 microg g(-1) creatinine. For cadmium, the mean urinary level was 0.78 microg g(-1) creatinine, and only one percent of the children had values above 2.0 microg g(-1) creatinine. The results cannot be generalized since the communities selected are not representative of the Mexican population; however, they indicate that Mexican children are exposed to chemicals and some at risk levels.

We reported previously that children are exposed to deltamethrin in malarious areas. In the present work we explored the levels of this insecticide in soil samples and also obtained relevant toxicokinetic data of deltamethrin in exposed children. Results show that, after spraying, indoor levels of deltamethrin in soil samples were higher than outdoor levels. The mean half-life estimated with these data was 15.5 days for outdoor samples and 15.4 days for indoor samples. Children's exposure to deltamethrin was assessed using as biomarkers the urinary concentrations of the metabolites 3-phenoxybenzoic acid (3-PBA) and cis-3-(2,2-dibromovinyl)-2,2-dimethylcyclopropane-1-carboxylic acid (Br2CA). The mean level of both biomarkers reached a peak within the first 24 hr postexposure; 6 months after the initial exposure, urinary levels of 3-PBA and Br2CA were found at levels observed before exposure. Approximately 91% of the total 3-PBA or Br2CA was excreted during the first 3 days after exposure. Therefore, we estimated a half-life for this period, the values for 3-PBA and Br2CA being almost identical (13.5 vs. 14.5 hr). Finally, considering reports about the genotoxicity of deltamethrin, we assessed DNA damage in children before and 24 hr after indoor spraying of deltamethrin; we found no differences in the comet assay end points. In conclusion, we observed exposure to deltamethrin in children, but we did not find any relationship between soil concentrations of deltamethrin and urinary levels of the metabolites. At least for genotoxicity, the exposed children appeared not to be at risk.
