RESUMO

Nicotinamide adenine dinucleotide (NAD+) is an essential cofactor of critical enzymes including protein deacetylase sirtuins/SIRTs and its levels in mammalian cells rely on the nicotinamide phosphoribosyltransferase (NAMPT)-mediated salvage pathway. Intracellular NAMPT (iNAMPT) is secreted and found in the blood as extracellular NAMPT (eNAMPT). In the liver, the iNAMPT-NAD+ axis oscillates in a circadian manner and regulates the cellular clockwork. Here we show that the hypothalamic NAD+ levels show a distinct circadian fluctuation with a nocturnal rise in lean mice. This rhythm is in phase with that of plasma eNAMPT levels but not with that of hypothalamic iNAMPT levels. Chemical and genetic blockade of eNAMPT profoundly inhibit the nighttime elevations in hypothalamic NAD+ levels as well as those in locomotor activity (LMA) and energy expenditure (EE). Conversely, elevation of plasma eNAMPT by NAMPT administration increases hypothalamic NAD+ levels and stimulates LMA and EE via the hypothalamic NAD+-SIRT-FOXO1-melanocortin pathway. Notably, obese animals display a markedly blunted circadian oscillation in blood eNAMPT-hypothalamic NAD+-FOXO1 axis as well as LMA and EE. Our findings indicate that the eNAMPT regulation of hypothalamic NAD+ biosynthesis underlies circadian physiology and that this system can be significantly disrupted by obesity.

An environmental impact assessment of a landfarming process, which was performed at an actual petroleum-contaminated site, was conducted using a green and sustainable remediation (GSR) tool in this study. The landfarming process was divided into four stages: site preparation, installation, system operation, and system dismantling/waste disposal. The environmental footprints of greenhouse gas (GHG) emissions, water consumption, total energy usage, and air pollutants (SOx, NOx, and PM10) were analyzed. GHG emissions and water consumption were approximately 276 metric tons and 7.90E + 05 gal, respectively, in stage III, where they were the highest due to the consumables and equipment use in the system operation. Total energy usage had the highest value of 1.54E + 03 MMBTU in stage II due to material production. The SOx and NOx emissions primarily occurred in stages I and II due to energy usage. The PM10 was mostly emitted in stages I and III and was associated with heavy use of equipment. To reduce the environmental footprints, biodiesel and sunlight were suggested as alternatives in this study. The GHG and SOx emissions decreased to 1.7 and 4.4E-04 metric tons, respectively, on the basis of total emissions with a 1 % increase in biodiesel content, but the NOx emissions increased to 5.6E-03 metric tons. If sunlight was used instead of electricity, the GHG and NOx emissions could be reduced by as much as 79 and 84 %, respectively, and the SOx emissions could also be reduced.

ETHNOPHARMACOLOGICAL RELEVANCE: Tongue diagnosis is a significant procedure to examine the physiological and pathological changes of the human body in oriental medicine. However, the conventional method of tongue diagnosis including direct observation of tongue has limitations because of various external factors and subjective factors. Therefore, the current study investigated the usefulness of the tongue diagnosis system (TDS) as a diagnostic tool for evaluating tongue coating thickness (TCT) by assessing the agreement between the TDS and a gold standard established by assessors using the conventional method. MATERIALS AND METHODS: The present study was designed as a prospective clinical trial including 60 patients with functional dyspepsia. The TDS was used to capture tongue images twice within a 30-min interval to assess its reproducibility. Tongue coating percentage was measured by the TDS, and TCT was classified as either no coating, thin coating, or thick coating according to the existing diagnostic criteria. After both TDS examinations, the weight of tongue coating was quantitatively measured, and the correlation between the actual quantity of tongue coating and the percentage of the coating measured by the TDS was analyzed using Pearson׳s correlation. After collecting tongue images in all 60 patients, tongue coating was evaluated using a conventional method by 5 well-trained assessors to establish the gold standard for evaluating TCT, which allowed us to assess the diagnostic agreement between the TDS and the gold standard. After 2 weeks, TCT evaluation was repeated by the same assessors using the same images but in a random order. RESULTS: The agreement between the TDS and the gold standard for evaluating TCT was almost perfect (weighted kappa, 0.840), as was the reproducibility of the TDS (weighted kappa, 0.851). The percentage of tongue coating measured by the TDS was significantly correlated with the weight of tongue coating (r=0.442, p <0.001). The levels of intra-rater reliability ranged from substantial to almost perfect (range of weighted kappa, 0.777-0.923). The inter-rater reliability of 5 assessors was moderate (weighted kappa, 0.563). CONCLUSIONS: The present study demonstrated that the TDS can be used as a diagnostic tool for the objective and standardized evaluation of TCT in actual clinical practice.

A stabilization/solidification treatment scheme was devised to stabilize Pb and Cu contaminated soil from a firing range using renewable waste resources as additives, namely waste oyster shells (WOS) and fly ash (FA). The WOS, serving as the primary stabilizing agent, was pre-treated at a high temperature to activate quicklime from calcite. Class C FA was used as a secondary additive along with the calcined oyster shells (COS). The effectiveness of the treatment was evaluated by means of the toxicity characteristic leaching procedure (TCLP) and the 0.1 M HCl extraction tests following a curing period of 28 days. The combined treatment with 10 wt% COS and 5 wt% FA cause a significant reduction in Pb (>98 %) and Cu (>96 %) leachability which was indicated by the results from both extraction tests (TCLP and 0.1 M HCl). Scanning electron microscopy-energy dispersive X-ray spectroscopy (SEM-EDX) analyses are used to investigate the mechanism responsible for Pb and Cu stabilization. SEM-EDX results indicate that effective Pb and Cu immobilization using the combined COS-FA treatment is most probably associated with ettringite and pozzolanic reaction products. The treatment results suggest that the combined COS-FA treatment is a cost effective method for the stabilization of firing range soil.

Total petroleum hydrocarbon (TPH) is an important environmental contaminant that is toxic to human and environmental receptors. However, human health risk assessment for petroleum, oil, and lubricant (POL)-contaminated sites is especially challenging because TPH is not a single compound, but rather a mixture of numerous substances. To address this concern, this study recommends a new human health risk assessment strategy for POL-contaminated sites. The strategy is based on a newly modified TPH fractionation method and includes an improved analytical protocol. The proposed TPH fractionation method is composed of ten fractions (e.g., aliphatic and aromatic EC8-10, EC10-12, EC12-16, EC16-22 and EC22-40). Physicochemical properties and toxicity values of each fraction were newly defined in this study. The stepwise ultrasonication-based analytical process was established to measure TPH fractions. Analytical results were compared with those from the TPH Criteria Working Group (TPHCWG) Direct Method. Better analytical efficiencies in TPH, aliphatic, and aromatic fractions were achieved when contaminated soil samples were analyzed with the new analytical protocol. Finally, a human health risk assessment was performed based on the developed tiered risk assessment framework. Results showed that a detailed quantitative risk assessment should be conducted to determine scientifically and economically appropriate cleanup target levels, although the phase II process is useful for determining the potency of human health risks posed by POL-contamination.

Human health risk assessments for petroleum, oil, and lubricant (POL)-contaminated sites are more complicated than for sites contaminated by single compounds due to the complex composition and various analytical methods associated with total petroleum hydrocarbons (TPH). Although several TPH fractionation methods are commonly used, including that of the TPH Criteria Working Group (TPHCWG), an efficient and economical human health risk assessment method is not yet available. To address this concern, a new modified fractionation strategy is recommended in this study, which resolves the problems of the current TPH fractionation methods while retaining reliability in the results. For the purpose of this study, the distribution characteristics of the 13 TPHCWG fractions were examined, and human health risk assessments for the POL-contaminated sites were performed. The results show that aliphatic EC8-16 and aromatic EC10-21 among the 13 TPH fractions are major contributors to human health risks along all exposure routes, making up approximately 96% of the hazard index (HI) of the TPH fractions, on average. Therefore, it is reasonable to concentrate on aliphatic EC8-16 and aromatic EC10-21 fractions, rather than to study all of the TPH fractions, in evaluating human health risk for TPH-contaminated sites.
