China has experienced a boom expansion of non-grain production in recent years. While the non-grain production can increase the economic benefits of farmers, its expansion has significant impacts on the ecological environment and agricultural sustainability. This study attempted to assess the trade-offs between the economic benefits and environmental costs of non-grain production and to provide reference for future land use management. Focusing on the non-grain expansion in Tongxiang City, eastern China, empirical models and field surveys were used to evaluate its environmental impacts and monetary analysis was used to assess the trade-offs between the economic benefits and environmental costs. The results showed that the area of non-grain production increased by 2464.74 ha from 2005 to 2020, and pond fish farming accounted for the largest proportion. The economic benefits and environmental costs of non-grain production increased continuously during 2005-2020, and the net economic-environmental benefits gradually expanded after 2010. Trade-off analysis indicates that the economic benefits of duck rearing did not compensate for the environmental costs, while the other non-grain productions did. Nevertheless, the potential impact of non-grain conversion on the local environment is still underestimated. Some suggestions are proposed to achieve a win-win situation between cultivated land utilization and ecological protection.

Objectives: The purpose of this study is to describe the current situation and forecast the trends of co-infection between the human immunodeficiency virus (HIV) and drug-susceptible tuberculosis (DS-TB) in different countries, across various age groups and genders. Methods: We obtained data on the number of cases, age-standardized incidence rate, age-standardized prevalence rate, age-standardized rate of disability-adjusted life years (DALYs), and age-standardized death rate from the Global Burden of Disease (GBD) 2019 database. These data were used to describe the distribution and burden of co-infection between the human immunodeficiency virus (HIV) and DS-TB in different regions, genders, and age groups. We employed joinpoint regression analysis to analyze the temporal trends from 1990 to 2019. Additionally, an age-period-cohort model was established to forecast the future trends of co-infection up to 2040. Results: The prevalence and burden of co-infection varied across different age groups and genders. The territories with the higher disease burden were distributed in some Asian and African countries. In terms of temporal trends, the age-standardized incidence rate and age-standardized prevalence rate of HIV and DS-TB co-infection exhibited an overall increasing trend from 1990 to 2019, and the prediction indicated a slow downward trend from 2019 to 2040. Conclusions: The co-infection of HIV and DS-TB posed a grave threat to public health and economic development. What's more, there existed a significant disparity between the actual state of co-infection and the desired goals for prevention and control.

BACKGROUND: Patients with myasthenia gravis (MG) lose part of their working or living ability due to illness, and bring burden to caregivers. The purpose of this study was to explore the factors related to caregivers' disease family burden for MG patients in Northwest China. METHODS: The study utilized our Myasthenia Gravis database and distributed online questionnaires to both MG patients and their caregivers. The questionnaires included a general data collection form, the Patient Health Questionnaire-9 (PHQ-9) scale, and the Caregivers' Family Burden Scale of Disease (FBSD). Univariate analysis and multivariate linear regression analysis were run, with FBSD as the outcome variable for separate analyses. RESULTS: 178 MG patients were eligible for inclusion in the analysis, of whom 80 patients' caregivers had a positive family burden of MG. The daily activity burden of the family and the economic burden of the family were the heaviest among the six dimensions of the caregivers' family disease burdens. The factors independently associated with FBSD were depression symptom level, MG severity classification and family's monthly per capita income (p < 0.05). CONCLUSIONS: Depression symptom level, MG severity classification and family's monthly per capita income are independent factors related to the caregivers' disease family burden for MG patients.

The protection of farmland resources is related to world food security, as well as the smooth realization of the goals of UN 2030 Agenda for Sustainable Development and China's Rural Revitalization Plan. As the most active region in the global economic development and one of the main grain producing areas, the problem of farmland abandonment in the Yangtze River Delta gradually appears with the rapid development of urbanization. Therefore, based on the interpretation data of remote sensing images and field survey data of three periods in 2000, 2010, and 2018, this study used Moran's I and geographical barycenter model to reveal the spatiotemporal evolution law of farmland abandonment in Pingyang County of the Yangtze River Delta. And then, this study selected 10 indicators covering four categories: geography, proximity, distance, and policy, and used random forest model to reveal the main influencing factors of farmland abandonment in the study area. The results showed that the abandoned farmland increased from 441.58 hm2 in 2000 to 5797.40 hm2 in 2018. The hot spot and barycenter of land abandonment gradually shifted from the western mountainous areas to the eastern plain areas. Altitude and slope were the main factors affecting the farmland abandonment. The high the altitude and the great the slope, the farmland abandonment in mountainous areas were serious. The proximity factors had a greater impact on the expansion of farmland abandonment from 2000 to 2010, and then the impact weakened. In view of the above analysis, the countermeasures and suggestions for maintaining food security were finally put forward.

Global warming and intensified human activities have led to regional climate instability with increasing frequency and the persistence of high-temperature climate events. Eco-environmental protection and socio-economic development have been faced with rigorous threats. Taking the monthly maximum temperatures from 1950 to 2019 as the basic data source, the spatial-temporal evolution characteristics of seasonal average maximum temperature (AMT) were discerned using the Mann-Kendall test and unary linear regression method in China from 1950 to 2019. Combined with linear correlation, partial linear correlation, and wavelet analysis, the correlation between seasonal AMT characteristics and atmospheric circulations was analyzed quantitatively. The results showed that:① the AMT in all seasons had a significant upward trend, with an increase of 1.21, 0.08, 1.81, and 0.25℃ in spring, summer, autumn, and winter, respectively. The abrupt change times of the AMT were concentrated in the 1990s to the early 21st century. ② In terms of spatial distribution, except for in summer, the average trend rates of AMT in other seasons increased gradually from south to north, although the increasing degrees were different. Among them, the AMT change rate in spring-winter was the fastest in northeast and northwest China. ③ There were complex correlations between the AMT of every season and atmospheric circulation factors, and the distribution of the interrelation energy varied significantly in different frequency domains. Specifically, the Pacific Decadal Oscillation had a significant negative correlation with AMT in summer. The North Atlantic Oscillation had an active effect on AMT changes in summer, autumn, and winter. The Arctic Oscillation had a significant positive driving effect on AMT in all seasons, and there were significant positive or negative influences on the short-or long-term changes of AMT in spring and summer due to the different EI Niño-Southern Oscillation years. These results could provide a theoretical basis and technical reference for China to formulate scientific and effective response plans of climate change.

Fungicides containing active ingredients of boscalid and pyraclostrobin have been widely applied in watermelon disease control. To provide data for avoiding health hazards caused by fungicides, we investigated its terminal residues and evaluated the dietary risk. In this work, watermelon samples were collected from field sites in six provinces and analyzed with high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The average recoveries of boscalid and pyraclostrobin in the watermelon matrix were 97-108% and 93-103%, respectively, with the relative standard deviations (RSDs) ≤ 9.1%. The limits of quantifications (LOQs) were 0.01 and 0.005 mg/kg for boscalid and pyraclostrobin. Twenty-one days after applying the test pesticide with 270 g a.i./ha, the terminal residues of boscalid and pyraclostrobin were all below 0.05 mg/kg and below the maximum residue limits (MRLs) recommended by European Food Safety Authority (EFSA). According to the national estimated daily intake (NEDI), the risk quotients (RQs) of boscalid and pyraclostrobin were 48.4% and 62.6%, respectively. That indicated the pesticide evaluated in watermelon exhibited a low dietary risk to consumers. All data provide a reference for the MRL establishment of boscalid in watermelon for China.

To achieve urban sustainability, it is critical to enhance the environment, economy, and society simultaneously. This study adopted the revised genuine progress indicator (GPI) and ecological footprint (EF) to evaluate the ecological efficiency and economic sustainability of the Yangtze River Delta from 2000 to 2018. Spatial analysis was utilized to identify spatial autocorrelation. A total of 27 cities were then partitioned through k-means cluster analysis. The results showed that GPI and ecological efficiency improved rapidly, but economic sustainability showed a downward trend. GPI and GDP had a high degree of spatial correlation, especially in Suzhou-Wuxi-Changzhou Metropolitan Area. However, no spatial correlation existed between GPI and EF. The city with high GEE can reach 3000 $/gha, indicating the city consumed 1 global hectare to create $3000 of genuine economic growth. Shanghai, Hangzhou, and Taizhou were cities with the highest level of economic sustainability and ecological efficiency. The spatiotemporal characteristics of economic sustainability and ecological efficiency revealed in this study will provide theoretical guidance for alleviating ecological pressure and promoting economic sustainable development.

The control of non-grain production (NGP) has become a great challenge for cultivated land protection in China in recent years. A control method for NGP that can coordinate the conflicts between cultivated land protection and farmers' interest is urgently needed. Taking Tongxiang City as an example, this research proposed a solution for the control and management of NGP based on cultivated land multifunctional assessment. The GIS and AHP approach were used to assess production function via a comprehensive evaluation index. The InVEST and FMSPA models were applied to assess ecological function while, the Maxent model was applied to assess recreational function, then multifunctional comprehensive zoning was conducted through natural breakpoint method and spatial overlay analysis. Five development-oriented function zones were considered, including the core area of grain production plus areas for ecological agriculture, leisure agriculture, compound agriculture, and general farmland. Differentiated control measures for NGPs in each functional subarea are proposed considering the current NGP distribution of Tongxiang city. This research can provide a reference for subsequent improvement of land management policies and can aid the achievement of sustainable agricultural development and rural revitalization.

Nanoimprint technology is powerful for fabricating nanostructures in a large area. However, expensive equipment, high cost, and complex process conditions hinder the application of nano-imprinting technology. Therefore, double-layer self-priming nanoimprint technology was proposed to fabricate ordered metal nanostructures uniformly on 4-inch soft and hard substrates without the aid of expensive instruments. Different nanostructure (gratings, nanoholes and nanoparticles) and different materials (metal and MoS2) were patterned, which shows wide application of double-layer self-priming nanoimprint technology. Moreover, by a double-layer system, the width and the height of metal can be adjusted through the photoresist thickness and developing condition, which provide a programmable way to fabricate different nanostructures using a single mold. The double-layer self-priming nanoimprint method can be applied in poor condition without equipment and be programmable in nanostructure parameters using a single mold, which reduces the cost of instruments and molds.

The fungicides epoxiconazole and pyraclostrobin have been widely used to control wheat fusarium head blight. This study was designed to investigate the dissipation behaviors in different climate regions and provide data for the modification of maximum residue limits of the two fungicides. Wheat samples were collected from field sites in twelve different regions, China and analyzed with an HPLC-MS/MS method for simultaneous detection of epoxiconazole and pyraclostrobin in wheat. The average recoveries of epoxiconazole and pyraclostrobin in wheat matrix were 87-112% and 85-102%, respectively, with the relative standard deviations ≤8.1%. The limits of quantification of epoxiconazole and pyraclostrobin in grain and straw were both 0.01 mg/kg. The dissipations of epoxiconazole and pyraclostrobin followed first-order kinetics, with the half-lives of 10.3 days and 7.6 days, respectively. The terminal residues of epoxiconazole and pyraclostrobin in grain were below 0.034 and 0.028 mg/kg, separately, both lower than the maximum residue limits recommended by China. Based on Chinese dietary pattern and terminal residue distributions, the risk quotients of epoxiconazole and pyraclostrobin were 13.9% and 65.9%, respectively, revealing the evaluated wheat exhibited an acceptably low dietary risk to consumers.

The dissipation and residues of tembotrione in corn field application were investigated using liquid chromatography tandem mass spectrometry (LC-MS/MS) method. The average recoveries of tembotrione in maize, corncob, and straw were in the ranges of 98-107% with relative standard deviations (RSDs ≤9.3%), respectively. The recoveries of M5 was in the ranges of 90-108% in all three matrices of maize, with RSDs were 3.3-12.8%. The LODs for tembotrione and M5 in maize were 0.85 µg/L and 1.0 µg/L, 0.84 µg/L and 0.43 µg/L in corncob, 0.94 µg/L and 1.5 µg/L in straw, respectively. The LOQs of the method in maize grain, corncob and straw were 0.01, 0.01 and 0.05 mg/kg for both analytes, respectively. The dissipation of tembotrione in straw was in compliance with the first-order dynamic equation, with half-lives of 1.18-1.23 days at Beijing and Heilongjiang. Total residue of tembotrione in maize grain and corncob matrix were both below 0.02 mg/kg, lower than the max residue limit (MRL) recommended by european food safety authority (EFSA). Risk quotients (RQs) of this pesticide was assessed via comparing national estimated daily intake with acceptable daily intake. The dietary intake risk of tembotrione residue in maize was very low for all groups of Chinese residents. These data could provide scientific data and strategies and facilitate Chinese government to establish the MRLs of tembotrione.

Localized surface plasmon resonance-based plasmonic biosensors are interesting candidates for the design of portable optical biosensor platforms owing to their integration, miniaturization, multiparameter, real-time, and label-free detection characteristics. Plasmonic biosensor arrays that have been combined with microfluidics have been developed herein to detect exosomes label-free. Gold nano-ellipsoid arrays were fabricated with low-cost anodic aluminum oxide thin films that act as shadow masks for evaporation of Au. The nano-ellipsoid arrays were integrated with a microfluidic chip to achieve multiparameter detection. The anti-CD63 antibody that is specific to the exosome transmembrane protein CD63 is modified on the surface of the nano-ellipsoids. Exosome samples were injected into the biosensor platform at different concentrations and detected successfully. The detection limit was 1 ng/mL. The proposed plasmonic biosensor array can be universally applicable for the detection of other biomarkers by simply changing the antibody on the surface of the Au nano-ellipsoids. Moreover, this biosensor platform is envisaged to be potentially useful in the development of low-cost plasmonic-based biosensors for biomarker detection and for the investigation of exosomes for noninvasive disease diagnoses.

Dietary risk assessment generally combines food consumption data and the concentration of pesticide by using the risk quotient (RQ) method. Chlorothalonil is the second popular fungicide in the world, and its residue and risk assessment in water spinach remain unknown. In this paper, the field trials of chlorothalonil in water spinach were operated under good agricultural practice (GAP) in China to human health protective. The dissipation experiments demonstrated that chlorothalonil was rapidly degraded in water spinach, with the half-lives of 1.8-3.2 days, and the amount of its metabolite SDS-3701 (4-hydroxy-2,5,6-trichloroisophthalonitrile) taken up through the water spinach roots from the soil was minor. The terminal experiments disclosed that the average residues of chlorothalonil and SDS-3701 in water spinach were below 6.59 mg/kg and 0.01 mg/kg, respectively. The results suggested the chronic dietary risk probability of chlorothalonil was 51.95-59.15% in terms of all registered crops, and the acute dietary risk probability of chlorothalonil was 12.30%-63.01% in water spinach, highlighting that the dietary risk of chlorothalonil in water spinach under GAP was acceptable. MRL of chlorothalonil was proposed as 7 mg/kg for water spinach and 5 days was recommended as a safe pre-harvest interval (PHI) for chlorothalonil application in water spinach field.

OBJECTIVE: To evaluate whether the technique ofmicroarray construction can be applied to paraffin-embedded cell block specinmens. STUDY DESIGN: We used a manual microarray method for construction of a well-aligned cell microarray. First we evaluated the cellularity of the cell block and assigned the cases to the null, low, moderate or high cellularity category. Second, based on the different cellularity, we constructed 25 specimen cylinders into 1 block. We used routine hematoxylin-eosin (H-E) stain and immunocytochemistry (ICC) on the slide. RESULTS: All cell microarray paraffin block shaping and specimen cylinder arraying were finished in 1 step, so the specimen cylinders and the paraffin blocks of the cell microarray could very easily be incorporated. No sample was lost during the H-E staining process. However, a few samples fell off partially during the ICC process. Additionally, we observed that the higher cellularity groups yielded a better outcome as compared to the lower cellularity groups. CONCLUSION: This study introduced a very simple and economical technique for the creation of cell microarrays from cell blocks. This procedure should acquaint cytopathologists with microarray technology and encourage its use.

The characteristic pathological changes of Parkinson s disease (PD) include a severe loss of dopamine neurons in the substantia nigra and a severe decrease in dopamine in the striatum. Since the expression of tyrosine hydroxylase (TH) and aromatic L-amino acid decarboxylase (AADC) in the biosynthetic pathway for dopamine are low, a promising approach to the gene therapy of PD is to augment the gene expression of the enzymes in the biosynthetic pathway for dopamine. In the present study, human TH and AADC genes were reconstructed into retrovirous vectors pLHCX and pLNCX(2) respectively. Then pLHCX/TH and pLNCX(2)/AADC were transfected into packaging cell line PA317 with liposome. PA317/TH and PA317/AADC were selected by different antibiotics. Gene expression was examined by methods of immunohistochemistry and in situ hybridization. The catalytic activity of two cloned gene enzymes was assessed in vitro by HPLC-EC. Immunocytochemical staining showed that TH and AADC were expressed efficiently in vitro. Both TH and AADC mRNA were transcripted in PA317 cell lines by using in situ hybridazation. HPLC-EC experiments revealed that the transfected cells produced a significantly higher level of dopamine and L-dopa than the untransfected cells. The two genetically modified cells could improve the production of L-dopa and dopamine in response to suitable substrate. The present results suggest that not only recombinant TH and AADC genes are successfully expressed in vitro, but also the enzymes have respective functional activities. These results have set up a way for in vivo gene therapy of PD with TH and AADC genes.
