RESUMO

The use of contaminated raw materials can lead to the transfer of mycotoxins into the final product, including beer. This study describes the use of the commercially available immunoaffinity column 11+Myco MS-PREP® and UPLC-MS/MS for the determination of mycotoxins in pale lager-type beers brewed in Czech Republic and other European countries. The additional aim of the work was to develop, optimize and validate this analytical method. Validation parameters such as linearity, limit of detection (LOD), limit of quantification (LOQ), precision and accuracy were tested. The calibration curves were linear with correlation coefficients (R2 > 0.99) for all mycotoxins under investigation. The LOD ranged from 0.1 to 50 ng/L and LOQ from 0.4 to 167 ng/L. Recoveries of the selected analytes ranged from 72.2 to 101.1%, and the relative standard deviation under conditions repeatability (RSDr) did not exceed 16.3% for any mycotoxin. The validated procedure was successfully applied for the analysis of mycotoxins in a total of 89 beers from the retail network. The results were also processed using advanced chemometric techniques and compared with similar published studies. The toxicological impact was taken into account.

For quantifying muscle strength in clinical and research practice, establishing the reliability of measurements, specifically to the procedures used, is essential for credible findings. The objective was to establish the reliability of isokinetic measurement of ankle plantar and dorsal flexors (PF/DF) and invertors and evertors (INV/EV) on an IsoMed 2000 dynamometer. Twenty healthy subjects (10 males, 10 females, mean age: 23.1 ± 3.1 years) completed an isokinetic measurement session. The intraclass correlation coefficient (ICC) and standard error of measurement were assessed for peak torque and work of ankle PF/DF (concentric and eccentric) and INV/EV (concentric) for the preferred and nonpreferred limb. Standardized isokinetic measurements of reciprocal PF/DF and INV/EV muscle actions were associated with ICC ranging from 0.77 to 0.98 for the majority of observed parameters. The exception was work in the eccentric mode in the ankle DF and peak torque in the concentric mode in the ankle INV on the preferred limb, where ICC ranged from 0.64 to 0.71. The IsoMed 2000 isokinetic dynamometer can be reliably employed in future studies for reciprocal ankle PF/DF and INV/EV assessment in healthy adult subjects after implementation of a familiarization session.

PURPOSE: The aim of the study was to determine whether it is possible to assess static balance on an unstable surface using center of pressure velocities obtained with a force platform when standing on a wobble board. METHODS: The center of pressure velocities were recorded with a force platform within three days (four trials per day) in thirty young adults in three conditions: standing on a rigid surface, compliant surface, and on a wobble board. Reliability of mean velocities of the center of pressure was examined using intraclass correlation coefficients. Relationships between the three conditions were assessed with Pearson correlation coefficients. RESULTS: Intra-session reliability was excellent for standing on a rigid surface and on a compliant surface and good for standing on a wobble board. Inter-session reliability was good for all parameters in all conditions, except for poor reliability in the anterior-posterior direction in standing on a wobble board. All correlations between the same parameter in different conditions were statistically significant (P < 0.05), except for velocity of the center of pressure in the anterior-posterior direction between stance on a rigid surface and stance on a wobble board. CONCLUSIONS: Centre of pressure velocity parameters obtained with a force plate when standing on a wobble board can provide valuable information about postural stability in unstable conditions.

This study examines a relatively new method of studying and quantifying human postural stability in patients with degenerative cerebellar disease. Trunk sway and feet sway were measured during quiet stance. To test the method, ten patients and eleven healthy subjects performed two different stance tasks: standing with eyes open on a firm surface and standing with eyes closed on a foam support surface. Data were recorded using three body-worn gyroscopes (Xsens Technologies B.V.) to measure roll and pitch angular movements of the lower trunk, and left and right foot. The pitch versus roll plots of the trunk and feet were created, and the areas of the convex hull shapes were calculated. It was found that the area of the convex hull of the pitch versus roll plots is suitable for the identification of postural instability disorders caused by degenerative cerebellar disease.

AIMS: The objectives of this pilot study were to assess the potential use of 3D videography for analyzing the motion of the body center of mass (COM) in newborns and to determine differences in spontaneous movements between preterm and full-term infants. METHODS: The group comprised 10 preterm newborns (gestational age at birth between 26 and 37 weeks; birth weight 800 to 2960 g; gestational age at the time of examination 34 to 39 weeks) and 10 full-term infants (gestational week 38 to 41; birth weight 2810 to 4360 g). To determine the range of motion of the COM, 3D videography was used (2 cameras, 25 Hz). When recording their movements, the infants were in the supine position, calm and awake. The recordings were processed using the APAS software. Selected points on the body were marked to obtain data for calculating the basic parameters of COM trajectories. RESULTS: The range of motion of the COM in both craniocaudal and anteroposterior directions was significantly greater in premature infants (P < 0.05 and P < 0.01, respectively) than in full-term babies. The variability of motion of the COM was significantly greater in the craniocaudal (P < 0.01) and anteroposterior (P < 0.05) directions in preterm babies. This was also valid for the velocity of motion of the COM in the craniocaudal direction (P < 0.05). CONCLUSIONS: 3D videography can be used for experimental assessment of motor behavior in preterm infants. Basic kinematic characteristics of the motion of the COM (range, variability, velocity) are greater in preterm infants.

This study assessed the measurement error of a monitoring system, the Video Manual Motion Tracker 1.0 (VMMT1.0), during time-motion analysis of basketball players. In this study, four reliability parameters were used to assess the measurement error of the system: the systematic bias, the inter-observer reliability, the intra-observer reliability and the absolute reliability. A basketball game video was used for the analysis. To assess the inter-observer reliability, two observers analysed a player's covered distance for 50 different periods of the game. To assess the relative and absolute reliability of the covered distance, the chosen players were monitored three times by 41 qualified observers. The findings did not indicate a significant systematic bias in the measurement error using the VMMT1.0 (one-way ANOVA, P > 0.05). The intra-observer reliability of the monitoring system was rated as very high (intraclass correlation, ICC = 0.999), similar to its inter-observer reliability (Pearson product-moment correlation, r = 0.994). The absolute reliability does not appear to be significant (standard error of measurement, SEM = 0.34 m). The results showed that the measurement error of the VMMT1.0 is acceptable and comparable with that of other time-motion analysis techniques.
