RESUMO

Hydrogels are considered an ideal platform for personalized healthcare due to their unique characteristics, such as their outstanding softness, appealing biocompatibility, excellent mechanical properties, etc. Owing to the high similarity between hydrogels and biological tissues, hydrogels have emerged as a promising material candidate for next generation bioelectronic interfaces. In this review, we discuss (i) the introduction of hydrogel and its traditional applications, (ii) the work principles of hydrogel in bioelectronics, (iii) the recent advances in hydrogel bioelectronics for health monitoring, and (iv) the outlook for future hydrogel bioelectronics' development.

RESUMO

Urban expansion, especially the construction of megacities, increases carbon emissions and adversely affects the carbon storage of terrestrial ecosystems. However, scientific land-use management policies can increase carbon storage. This study takes two megacities at different stages of development, Beijing and Tianjin, as examples to explore the impact of different ecological conservation scenarios on both urban land use and carbon storage to provide recommendations for the construction planning of large cities with low-carbon development as the goal. Furthermore, we coupled the patch-generating land use simulation (PLUS) model with the integrated valuation of ecosystem services and tradeoffs (InVEST) model to simulate land use and carbon storage under a natural development scenario, a planned ecological protection scenario (PEPS), and a policy-based ecological restoration scenario (PERS). From 2000 to 2020, both cities had different degrees of construction land expansion and carbon loss, and Tianjin's dynamic degree of construction land was 0.94% higher than Beijing's, with a carbon loss 183,536.19 Mg higher than Beijing's; this trend of reducing carbon reserves will continue under the natural development scenario (NDS). Under the PEPS and PERS, the carbon stock of both cities increases, and the impact on Tianjin is greater, with an increase of 4.51% and 8.04%, respectively. Under PERS, the carbon stock increases the most, but the dynamic degree of construction land use is negative for both cities. Beijing's carbon stock is 0.40% lower than Tianjin's, which deviates slightly from the trend of urban economic development. Megacities in the rapid development stage can refer to Tianjin, strictly following the ecological protection land planning scope and vigorously implementing ecological restoration policies to effectively increase regional carbon stock. Megacities in the mature stage of development can refer to Beijing, and flexibly implement ecological restoration policies to increase regional carbon stock without affecting the city's economic development.

Root nitrogen (N)-uptake rate and uptake preference, and their association with root morphological and chemical traits are important to characterize root N-acquisition strategies of trees. However, how the root N-acquisition strategy varies with tree age, especially for those species that coexist at a common site, remains unknown. In this study, a field isotopic hydroponic method was used to determine the uptake rate and contribution of NH4+, NO3- and glycine, for three coexisting ectomycorrhizal coniferous species [Pinus koraiensis (Korean pine), Picea koraiensis (Korean spruce) and Abies nephrolepis (smelly fir)] at three age classes (young, middle-aged and mature) in a temperate forest. Concurrently, root morphological and chemical traits, as well as mycorrhizal colonization rate were determined. Our results show that the root uptake rate of total N and NH4+ gradually decreased across all three species with increasing tree age. The three species at all age classes preferred NH4+, except for middle-aged Korean spruce and mature smelly fir, which preferred glycine. In contrast, all three species showed the lowest acquisition of NO3-. According to the conceptual framework of 'root economics space', only a 'collaboration' gradient (i.e. dimension of root diameter vs specific root length or area) was identified for each species, in which root N-uptake rate loaded heavily on the side of 'do-it-yourself' (i.e. foraging N more by roots). Young trees of all species tended to exhibit the 'do-it-yourself' strategy for N uptake, and mature trees had an 'outsourcing' strategy (i.e. foraging N by a mycorrhizal partner), whereas middle-aged trees showed a balanced strategy. These findings suggest that shifts of root N-acquisition strategy with tree age in these species are mainly mediated by root traits along the 'collaboration' gradient, which advances our understanding of belowground competition, species coexistence and N cycling in temperate forests.

Introduction: Despite the widespread use of the unilateral striatal 6-hydroxydopamine (6-OHDA) lesion model in mice in recent years, the stability of behavioral deficits in the 6-OHDA striatal mouse model over time is not yet clear, raising concerns about using this model to evaluate a compound's long-term therapeutic effects. Materials and methods: In the current study, mice were tested at regular intervals in the cylinder test and gait analysis beginning 3 days after 6-OHDA injection of 4 and 8 µg and lasting until 56 days post-lesion. Apomorphine-induced rotational test and rotarod test were also performed on Day 23 and 43 post-lesion, respectively. Immunohistochemistry for dopaminergic neurons stained by tyrosine hydroxylase (TH) was also performed. Results: Our results showed that both the 4 and 8 µg 6-OHDA lesion groups exhibited forelimb use asymmetry with a preference for the ipsilateral (injection) side on Day 3 and until Day 21 post-lesion, but did not show forelimb asymmetry on Day 28 to 56 post-lesion. The 8 µg 6-OHDA lesion group still exhibited forelimb asymmetry on Day 28 and 42 post-lesion, but not on Day 56. The gait analysis showed that the contralateral front and hind step cycles increased from Day 3 to 42 post-lesion and recovered on Day 56 post-lesion. In addition, our results displayed a dose-dependent reduction in TH+ cells and TH+ fibers, as well as dose-dependent apomorphine-induced rotations. In the rotarod test, the 8 µg 6-OHDA lesion group, but not the 4 µg group, decreased the latency to fall on the rotarod on Day 43 post-lesion. Conclusion: In summary, unilateral striatal 6-OHDA injections of 4 and 8 µg induced spontaneous motor impairment in mice, which partially recovered starting on Day 28 post-lesion. Forced motor deficits were observed in the 8 g 6-OHDA lesion group, which remained stable on Day 43 post-lesion. In addition, the rotarod test and apomorphine-induced rotational test can distinguish between lesions of different extents and are useful tools for the assessment of functional recovery in studies screening novel potential therapies.

Objective: This study estimates the economic burden imposed on families by comparing the hospitalization costs of T1DM children with and without medical insurance in Shandong province. Methods: Our data comprised 1,348 T1DM inpatient records of patients aged 18 years or younger from the hospitalization information system of 297 general hospitals in 6 urban districts of Shandong Province. Descriptive statistics are presented and regression analyses were conducted to explore the factors associated with hospitalization costs. Results: Children with medical insurance had on average total hospitalization expenditures of RMB5,833.48 (US$824.02) and a hospitalization stay of 7.49 days, compared with the children without medical insurance who had lower hospitalization expenditures of RMB4,021.45 (US$568.06) and an average stay of 6.05 days. Out-of-pocket expenses for insured children were RMB3,036.22 (US$428.89), which is significantly lower than that of the uninsured children (P < 0.01). Out-of-pocket (OOP) expenditures accounted for 6% of the annual household income of insured middle-income families, but rose to a significant 25% of the annual income for low-income families. These OOP expenditures imposed a heavy economic burden on families, with some families experiencing long-term financial distress. Both insured and uninsured families, especially low-income families, could be tipped into poverty by hospitalization costs. Conclusion: Hospitalization costs imposed a significant economic burden on families with children with T1DM, especially low-income insured and uninsured families. The significantly higher hospitalization expenses of insured T1DM children, such as longer hospitalization stays, more expensive treatments and more drugs, may reflect both excess treatment demands by parents and over-servicing by hospitals; lower OOP expenses for uninsured children may reflect uninsured children from low-income families forgoing appropriate medical treatment. Hospital insurance reform is recommended.

The purpose of this study was to explore the temporal and spatial distribution characteristics and potential sources of ozone (O3) in the Shandong Peninsula over a long period of time based on the analysis of the temporal and spatial changes in O3 concentration in Shandong Peninsula from 2005 to 2020. We used wavelet analysis, the entropy weight method, and correlation analysis to discuss O3 and its influencing factors and researched the potential sources of O3 in Shandong Peninsula. The results showed that:① in terms of the time pattern, the near-surface O3 in Shandong Peninsula showed a "triple peak" trend from 2005 to 2020, reaching the maximum value of[(40.48±7.64) µg·m-3] in 2010 and a minimum value of[(36.63±5.61) µg·m-3] in 2013. The season was expressed as:summer[(42.49±1.7) µg·m-3]>spring[(40.65±0.6) µg·m-3]>autumn[(36.47±0.7) µg·m-3]>winter[(36.46±0.3) µg·m-3]. ② In terms of the spatial pattern, the O3 concentration of Shandong Peninsula gradually increased with the increase in latitude from 2005 to 2020, showing the characteristics of high concentrations in the east and west and low in the middle region. During the 16-year evolution of the O3 concentration, there was a 1.5 a main oscillation period. ③The analysis of meteorological conditions revealed that O3 concentration was positively correlated with temperature, precipitation, relative humidity, and sunshine hours, whereas pressure and wind speed were negatively correlated. In the analysis of social factors, soot (dust) emissions were the most obvious factor affecting the third indicator, with a weight of 0.25. ④ Through simulating the trajectory of airflow from different regions (Ji'nan and Qingdao), it was found that the ocean airflow contributed 10.69% to Jinan and 48.94% to Qingdao. There was 64.04% of the long-distance air mass transmission path coming from the northwest, and 43.69% of the short-distance air mass transmission path was from the Bohai Sea and the Yellow Sea, followed by Shandong Province with 21.01%. ⑤ The analysis of potential sources of O3 showed that the potential sources of Ji'nan were mainly distributed in Jinzhou, Liaoning Province, northern Jiangsu Province, Hubei Province, and Anhui Province, with a WPSCF value >0.6, and Qingdao's WPSCF value of >0.6 was mainly distributed in the Yellow Sea area. The O3 contribution of Jining City, Linyi City, Xuzhou City, Huaibei City, and Lianyungang City was >40 µg·m-3. The area with >45 µg·m-3 in Qingdao was mainly in the Yellow Sea. Through the analysis of potential sources in the Shandong Peninsula, particular attention should be paid to the supply of industrial sources in the surrounding areas and the marine sources provided by marine air pollution.

This study was to assess the prognosis stratification of the clinical-pathologic staging system incorporating estrogen receptor (ER)-negative disease, the nuclear grade 3 tumor pathology (CPS + EG), Neo-Bioscore, and a modified Neo-Bioscore system in breast cancer patients after preoperative systemic therapy (PST). A retrospective multicenter cohort study was conducted from 12 participating hospitals' databases from 2006 to 2015. Five-year disease free survival (DFS), disease specific survival (DSS), and overall survival (OS) were calculated using Kaplan-Meier Method. Area under the curve (AUC) of the three staging systems was compared. Wald test and maximum likelihood estimates in Cox proportional hazards model were used for multivariate analysis. A total of 1,077 patients were enrolled. The CPS + EG, Neo-Bioscore, and modified Neo-Bioscore could all stratify the DFS, DSS, and OS (all P < 0.001). While in the same stratum of Neo-Bioscore scores 2 and 3, the HER2-positive patients without trastuzumab therapy had much poorer DSS (P = 0.013 and P values < 0.01, respectively) as compared to HER2-positive patients with trastuzumab therapy and HER2-negative patients. Only the modified Neo-Bioscore had a significantly higher stratification of 5-year DSS than PS (AUC 0.79 vs. 0.65, P = 0.03). So, the modified Neo-Bioscore could circumvent the limitation of CPS + EG or Neo-Bioscore. CLINICAL TRIAL REGISTRATION: ClinicalTrials.gov, identifier NCT03437837.

The agricultural wastes disposal and polluted water purification are always the key issues of environmental restoration. In this work, a magnetic biogas residue-based biochar (mBR-C) by direct pyrolysis and sonochemical method was prepared from biogas residue (BR). Response design methodology based on Box-Behnken design was used for the preparation parameters optimization. The characterization results identified that mBR-C had well-developed pore structure and surface area, which was beneficial to diffuse and capture heavy metal ions. Traces of toxic heavy metal in mBR-C was leached (˂0.04 mg/L) through TCLP method, indicating the environmental safety of the magnetic biochar. Meanwhile, the mBR-C exhibited excellent solid-liquid separation efficiency because of its strong magnetism. The series of adsorption experiments indicated that mBR-C could capture Cu2+ and Pb2+ rapidly, and the maximum adsorption capacity for Cu2+ and Pb2+ was 75.76 and 181.82 mg/g, respectively, which was higher than some other biochars previously reported. mBR-C was further applied in the synthetic wastewater treatment, which could effectively purify at least 600 mL (150 BV) to meet emission standards. After several column adsorption-desorption cycles, the adsorption capacity could still reach 85%, implying that mBR-C has good reusability and stability. Overall, the mBR-C can be used as an eco-friendly, desirable, economic and recyclable biosorbent in heavy metal polluted water treatment, providing a new idea for a combination of biogas residue recycle and wastewater treatment.

Infectious bursal disease virus (IBDV) is still a vital etiological agent in poultry farms. IBDV outbreaks occasionally occur due to the presence of very virulent, reassortment or variant strains. Vaccine immunization has played crucial roles in IBD control for decades. However, survival pressure of IBDV from the vaccine immunization also increases the reassortments of circulating viruses. In this study, an IBDV strain was isolated from several broiler farms in Henan Province, central part of China, and named IBDV HN strain. Based on the results of RT-PCR, sequencing and phylogenic analyses of VP1 and VP2 genes, the IBDV HN strain is a novel reassortment strain in the Henan region. Segment A of this strain appears to originate from the very virulent IBDV strain, while segment B comes from the other field reassortment strains. This may be the result of natural reassortant of virus circulating in the field. About 60% (6/10) of experimentally infected specific pathogen-free chickens died after 3 to 5 d post-infection with typical symptom and pathological lesions. The IBDV HN strain was prone to horizontal transmission, which poses a serious threat to the chicken industry. Further investigation on the prevalence, virulence, and evolution of HN strain IBDV will provide a foundation for the prevention and control of the disease in this region.

Ozonation has been identified effective technique to degrade phenolic compounds, and production of intermediate dimers are major threat. In this study, we systematically investigated the degradation of Pentabromophenol (PBP) in an aqueous medium by using two different ozone generators (sources: air and water). We studied various factors that influenced the degradation kinetics of PBP, including the pH (7.0, 8.0, and 9.0), humic acid (HA) and anions (Cl-, SO42-, NO3-, and HCO3-). PBP was efficiently degraded within 5 min (O3 source: water) and 45 min (O3 source: air) at pH 8.0 maintained by phosphate buffer. Reaction kinetics revealed 17 b y-products with five possible pathways, including dimers with their isomers and lower bromophenols. Furthermore, the frontier molecular orbital theory was employed to confirm the proposed ozonation pathways, including the breakage of the CO bond at C5 and C4 positions, and the cleavage of the CC bond at C3 and C6 position. Product P5, P14 (hydroxyl-nonabromophenyl ether) and P15 (dihydroxyl-octabromophenyl ether) were identified with isomers. Ecological Structure Activity Relationships toxicity assessment resulted into the conversion of highly toxic PBP (acute toxicity: LC50 = 0.11 mg L-1 for fish, LC50 = 0.124 mg L-1 for daphnia, and EC50 = 0.118 mg L-1 for green algae) to less harmful products aside from dimers. P14 (acute toxicity: LC50 = 1.04 × 105) found to be more toxic as compare to PBP. From these findings, we concluded that ozonation is an effective and ideal process for PBP degradation.

RESUMO

A novel, simple, one-step and one-tube detection method was developed for ultrasensitive detection of polynucleotide kinase (PNK) activity on the basis of dual enzyme-synergistic signal amplification. This method was also demonstrated to work well for PNK inhibitor screening and endogenous PNK detection in cell lysates at a single-cell level.

Fabrication of a high quality quantum dot (QD) film is essentially important for a high-performance QD light emitting diode display (QLED) device. It is normally a high-cost and multiple-step solution-transfer process where large amounts of QDs were needed but with only limited usefulness. Thus, developing a simple, efficient, and low-cost approach to fabricate high-quality micropatterned QD film is urgently needed. Here, we proposed that the Chinese brush enables the controllable transfer of a QD solution directly onto a homogeneous and ultrasmooth micropatterned film in one step. It is proposed that the dynamic balance of QDs was enabled during the entire solution transfer process under the cooperative effect of Marangoni flow aroused by the asymmetric solvent evaporation and the Laplace pressure different by conical fibers. By this approach, QD nanoparticles were homogeneously transferred onto the desired area on the substrate. The as-prepared QLED devices show rather high performances with the current efficiencies of 72.38, 26.03, and 4.26 cd/A and external quantum efficiencies of 17.40, 18.96, and 6.20% for the green, red, and blue QLED devices, respectively. We envision that the result offers a low-cost, facile, and practically applicable solution-processing approach that works even in air for fabricating high-performance QLED devices.

PURPOSE: Depression and anxiety have been considered as the most common comorbidities in maintenance hemodialysis (MHD) patients. This study aimed to assess mental disorder in hemodialysis patients and to study the indicators and correlates of psychological disturbance among patients receiving MHD in China. METHODS: A cross-sectional study was conducted in 187 outpatients undergoing regular hemodialysis for at least 3 months. All the participants completed the Hospital Anxiety and Depression Scale (HADS), Functional Assessment of Chronic Illness Therapy-Fatigue (FACIT-Fatigue, Chinese mainland version), Pittsburgh Sleep Quality Index (PSQI), Family APGAR Index (PAGAR), Family Adaptability and Cohesion Evaluation Scale (FACES II, Chinese version) and the Social Functioning Questionnaire (SFQ). RESULTS: Based on the cutoff point of HADS-D/A ≥ 8, 51 (27.3%) and 29 (15.5%) patients reported depression and anxiety, respectively; 26 (13.9%) of all reported both depression and anxiety. Depressive patients had lower economic status, less subsidies, less than 3 years duration on hemodialysis and comorbidities when compared to patients without depression (all P < 0.05). HADS-D showed positive correlations with HADS-A, PSQI and SFQ and negative correlations with FACIT-Fatigue, PAGAR, family cohesion and adaptability (all P < 0.001). Patients with better family cohesion showed higher level of psychosocial well-being no matter their economic status. Binary logistic regression analysis demonstrated that anxiety (OR 1.80, P < 0.001) and bad social functioning (OR 1.31, P < 0.001) were independently associated with depression. CONCLUSION: More attention should be paid on assessment and management psychological disturbance, and development family/social/medical supporting system for Chinese patients receiving MHD.

CONTEXT: Fatigue is a common symptom reported by patients with end-stage renal disease, and it can significantly decrease patients' quality of life. A brief and convenient fatigue assessment tool is needed for Chinese patients on maintenance dialysis. OBJECTIVES: To determine the psychometric characteristics of the Chinese version of the Functional Assessment of Chronic Illness Therapy-Fatigue (FACIT-Fatigue) in patients receiving maintenance dialysis. METHODS: The Chinese version of the FACIT-Fatigue was obtained from the FACIT system. Test-retest reliability of this scale was examined using intraclass correlation coefficients, and the internal consistency was calculated by Cronbach's alpha. Content validity was examined using the content validity index (CVI), scale-level CVI/universal agreement, and scale-level CVI/average agreement. Construct validity was tested using Pearson product-moment correlations of the FACIT-Fatigue scores with the Revised Piper Fatigue Scale, Hospital Anxiety and Depression Scale, Pittsburgh Sleep Quality Index, and disease characteristics during the same period. RESULTS: A total of 172 patients (111 males and 61 females, mean age 52.6 ± 12.5 years) completed this study, with a median FACIT-Fatigue score of 41 (first and third quartiles 34.3-46). The Chinese version of the FACIT-Fatigue had excellent test-retest reliability (intraclass correlation coefficient = 0.98) and internal consistency (Cronbach's alpha = 0.92); the validity of the scale was supported by CVI, scale-level CVI/universal agreement, and scale-level CVI/average agreement values of at 0.67-1, 0.85, and 0.96, respectively. The standard error of measurement of the FACIT-Fatigue was 1.2. The significant correlations between the FACIT-Fatigue score and the Revised Piper Fatigue Scale (r = -0.658), Hospital Anxiety and Depression Scale (-0.566), and Pittsburgh Sleep Quality Index (-0.489) were supported by the FACIT-Fatigue, with good construct validity (all P < 0.01). CONCLUSION: The FACIT-Fatigue had acceptable validity and reliability for maintenance dialysis patients and can be used as a valid tool for the measurement of fatigue among these Chinese patients.

We report a study of DNA deformations using a coarse-grained mechanical model and quantitatively interpret the allosteric effects in protein-DNA binding affinity. A recent single-molecule study (Kim et al. Science 2013, 339, 816) showed that when a DNA molecule is deformed by specific binding of a protein, the binding affinity of a second protein separated from the first protein is altered. Experimental observations together with molecular dynamics simulations suggested that the origin of the DNA allostery is related to the observed deformation of DNA's structure, in particular, the major groove width. To unveil and quantify the underlying mechanism for the observed major groove deformation behavior related to the DNA allostery, here we provide a simple but effective analytical model where DNA deformations upon protein binding are analyzed and spatial correlations of local deformations along the DNA are examined. The deformation of the DNA base orientations, which directly affect the major groove width, is found in both an analytical derivation and coarse-grained Monte Carlo simulations. This deformation oscillates with a period of 10 base pairs with an amplitude decaying exponentially from the binding site with a decay length lD ≈10 base pairs as a result of the balance between two competing terms in DNA base-stacking energy. This length scale is in agreement with that reported from the single-molecule experiment. Our model can be reduced to the worm-like chain form at length scales larger than lP but is able to explain DNA's mechanical properties on shorter length scales, in particular, the DNA allostery of protein-DNA interactions.
