RESUMO

The water balance budget in remote plateau lakes provides a fundamental information on the local climate-hydrological pattern. However, integrated investigation on the waters entering the lake, especially groundwater, was limited. To assess the current climate stress on Yunnan-Guizhou Plateau lakes, we collected rivers, groundwater, lake, and precipitation with varying isotopic compositions in the Chenghai Lake basin over four separate campaigns during a hydrological year. The wide and enriched variation of isotope composition in rivers, groundwater, and lake indicate that they have undergone distinct evaporations, which further reveal the recharging and mixing processes. Based on the similar isotopic signals between rivers and precipitation, rivers can serve as proxies for precipitation. Groundwater was primarily replenished by high mountain precipitation duo to the stable isotopic values in aquifers. Even through mass water in lake was able to smooth out some variability, the considerable isotopic variation of lake during the four collections suggested the influence of meteorological conditions. According to the assessment of isotope balance model, lake evaporation accounts for almost 65 % of the total inflow for one year, which partially explains the climate stress on the lake level. As the most sensitive variables, changes in relative humidity (h) and isotope composition of atmospheric moisture (δA) resulted in remarkable variations in E/I ratios and the constructed water isotope framework. These results shed light on the capacity of evaporation relative to lake input and provide interpretations on local paleoclimate and predicted-climate construction.

Tungsten carbide (WC) is an alternative to the costly and resource-constrained Pt-based catalysts. Herein, a one-step facile and easily scalable approach is reported to synthesize ultrafine WC nanocrystals encapsulated in porous N-doped carbon nanospheres (NC) by simple self-polymerization, drying, and annealing. It is worth mentioning that this developed method has four novel features: (1) the synthesis process, without any hard template or hydrocarbon gas feeding, is, notably, very facile and efficient with low cost; (2) the carbon coating on WC nanocrystals not only restrains coarsening of particles but also creates strong coupling interactions between the nanocrystallines and the conductive carbonaceous matrix; (3) uniform grape-like WC@NC nanospheres with high specific surface area can be obtained in a large scale; and (4) single-phase WC can be achieved. As a result, WC@NC demonstrates remarkable hydrogen evolution reaction (HER) electrocatalytic performance with overpotentials of 127 and 141 mV at a current density of 10 mA cm-2 and Tafel slopes of 56.3 and 78.7 mV dec-1 in acid and alkaline media, respectively. Our density functional theory calculations manifest that the strong synergistic electronic effect between WC and its intimately bonded carbon shell vastly boosts the HER electrocatalytic activity. WC@NC catalysts as a cathode are further tested in a home-made electrolyzer with 0.78 A cm-2 achieved at a cell voltage of 2 V at 80 °C and operated stably at 200 mA cm-2 for more than 20 h.

To evaluate the toxicity of arsenic trioxide (As2O3) in the muscular tissues (wing, thigh and pectoral) of birds, 72 one-day-old Hy-line cocks were selected and randomly divided into four groups. They were fed either a commercial diet or an arsenic-supplemented diet containing 7.5, 15 or 30 mg/kg As2O3. The experiment lasted for 90 days and the samples of muscular tissues were collected at 30, 60 and 90 days. The results showed that As2O3 exposure significantly lowered the activities of antioxidant enzymes (catalase (CAT), glutathione peroxidase (GSH-Px)) and inhibition ability of hydroxyl radicals (OH) and increased the malondialdehyde (MDA) contents. Furthermore, the mRNA levels of inflammatory cytokines (tumor necrosis factor-α (TNF-α), nuclear factor-kappa B (NF-κB), cyclooxygenase-2 (COX-2), inducible NO synthase (iNOS), prostaglandin E synthase (PTGEs)) and heat shock proteins (HSPs) in muscular tissue were significantly upregulated in the As2O3 exposure groups. The results indicated that As2O3 exposure resulted in oxidative damage, induced the inflammatory response, and influenced the mRNA levels of HSPs in muscular tissue of cocks. Additionally, the results suggested that HSPs possibly resisted due to the As2O3 exposure-induced oxidative stress and inflammatory response, which provided a favorable environment and played protective roles in the muscular tissues of cocks. The information presented in this study is helpful to understand the mechanism of As2O3 toxicity in bird muscular tissues.

A rapid immunochromatographic strip (ICS) test for specific detection of lily symptomless virus (LSV) was developed. The test is based on a double-antibody sandwich format and employs two distinct anti-LSV polyclonal antibodies (IgG1 and IgG2). The first antibody, IgG1 was used as the detection antibody conjugated to colloidal gold and the second antibody, IgG2 was used to as the capture antibody at the test line. The performance of the ICS test was evaluated and the results obtained were compared with a quadruplex RT-PCR assay. When serial dilutions of purified LSV were tested, the LSV detection limit of the ICS test was 6.0 × 10(-8)mg/mL, which was the same as the quadruplex RT-PCR assay. Relative to quadruplex RT-PCR, the specificity and sensitivity of the ICS were 98.6% and 100%, respectively for field leaf samples. There was significant agreement between the results of the ICS and quadruplex RT-PCR tests (κ = 0.983). Compared with conventional lily virus detection methods, our ICS test has many advantages: simple, fast, low cost, high sensitivity and specificity, and has applications in the laboratory and in the field to detect and control LSV.

BACKGROUND: Carbon nanotubes have shown broad potential in biomedical applications, given their unique mechanical, optical, and chemical properties. In this pilot study, carbon nanotubes have been explored as multimodal drug delivery vectors that facilitate antiangiogenic therapy in zebrafish embryos. METHODS: Three different agents, ie, an antiangiogenic binding site (cyclic arginine-glycin-easpartic acid), an antiangiogenic drug (thalidomide), and a tracking dye (rhodamine), were conjugated onto single-walled carbon nanotubes (SWCNT). The biodistribution, efficacy, and biocompatibility of these triple functionalized SWCNT were tested in mammalian cells and validated in transparent zebrafish embryos. RESULTS: Accumulation of SWCNT-associated nanoconjugates in blastoderm cells facilitated drug delivery applications. Mammalian cell xenograft assays demonstrated that these antiangiogenic SWCNT nanoconjugates specifically inhibited ectopic angiogenesis in the engrafted zebrafish embryos. CONCLUSION: This study highlights the potential of using SWCNT for generating efficient nanotherapeutics.

OBJECTIVE: To develop an alternative method for assessment of gene delivery systems in vivo. METHODS: Mouse primary spleen lymphocytes were genetically modified in vitro by a retroviral vector harboring a Gaussia luciferase (Gluc) expression cassette. After implantation of these cells into recipient mice, the expression of Gluc was detected in whole blood or plasma collected. RESULTS: As little as 10 muL whole blood drawn from the recipient mice could guarantee prompt reading of Gluc activity with a luminometer. And the reading was found in good correlation with the number of genetically modified spleen lymphocytes implanted to the mice. CONCLUSIONS: Gluc may be useful as an in vivo reporter for gene therapy researches, and Gluc blood assay could provide an alternative method for assessment of gene delivery systems in vivo.
