RESUMO

Salmonella infections pose a significant threat to animal and human health. Phytochemicals present a potential alternative treatment. Galla chinensis tannic acid (GCTA), a hydrolyzable polyphenolic compound, inhibits bacterial growth and demonstrates potential as an alternative or supplement to antibiotics to prevent Salmonella infections. However, little is known about the antimicrobial mechanism of GCTA against Salmonella. Here, we revealed 456 differentially expressed proteins upon GCTA treatment, impacting pathways related to DNA replication, repair, genomic stability, cell wall biogenesis, and lipid metabolism using TMT-labeled proteomic analysis. TEM analysis suggested altered bacterial morphology and structure post-treatment. A Salmonella-infected-mouse model indicated that GCTA administration improved inflammatory markers, alleviated intestinal histopathological alterations, and reduced Salmonella enterica serovar Enteritidis (S. Enteritidis) colonization in the liver and spleen of Salmonella-infected mice. The LD50 of GCTA was 4100 mg/kg with an oral single dose, vastly exceeding the therapeutic dose. Thus, GCTA exhibited antibacterial and anti-infective activity against S. Enteritidis. Our results provided insight into the molecular mechanisms of these antibacterial effects, and highlights the potential of GCTA as an alternative to antibiotics.

Industrial transformation and high-quality urban development have become the core issues of urban-rural coordination and the leap forward in development in the new era. The research perspective of 'pattern-process-mechanism' is needed to reveal the spatiotemporal correlation characteristics of industrial transformation and urban economic efficiency evolution, and to expand its systematic, comprehensive and regional characteristics. Based on the geographical cognitive of local effects and spatial non-stationarity, we used a quantile regression model and a geographically weighted regression model to analyze the dynamic mechanism of industrial transformation and urban economic efficiency to explain the path characteristics of urban development and industrial transformation of the Yangtze River economic belt. The conclusions are as follows: (1) From 2000 to 2015, the average economic efficiency in the Yangtze River economic belt increased from 0.05 to 0.332, and the pattern gradually changed from spatial homogeneity to spatial mosaic; (2) From 2000 to 2015, the range and intensity of industrial transformation in the Yangtze River economic belt showed an increasing trend, while the speed of industrial transformation showed a downward trend, and the high-value unit of the three showed the characteristics of gradual homogenization; (3) From the perspective of the impact of industrial transformation on urban economic efficiency, the impact of the range and speed of industrial transformation on urban economic efficiency was gradually weakened, while the impact of the intensity of industrial transformation on urban economic efficiency was gradually strengthened, and the patterns of the three show the characteristics of a spatially inverted U-shaped distribution with high values in the middle reaches and low values in the upstream and downstream areas.

INTRODUCTION: Metagenomic Next-Generation Sequencing (mNGS) is an emerging technique for microbial identification and diagnosis of infectious diseases. The clinical utility of mNGS, especially its real-world impact on antimicrobial treatment and patient outcome has not been systematically evaluated. METHODS: We prospectively assessed the effectiveness of mNGS in 70 febrile inpatients with suspected infections at Hematology department of the Children's Hospital, National Clinical Research Center for Child Health. 69/70 patients were given empirical antibiotics prior to mNGS. A total of 104 samples (62 plasma, 34 throat swabs, 4 bone marrow, 4 bronchoalveolar lavage) were collected on day 1-28 (mean 6.9) following symptom onset and underwent mNGS testing. RESULTS: Traditional microbiological tests discovered causal microorganisms in 5/70 (7.14%) patients, which were also detected by mNGS. In addition, mNGS reported possible pathogens when routine tests were negative. Antibiotics were adjusted accordingly in 55/70 (78.6%) patients that led to improvement/relief of symptoms within 3 days. In contrast, mNGS results were considered irrelevant in 15/70 (21.4%) patients by a board of clinicians, based on biochemical, serological, imaging evidence, and experiences. CONCLUSION: mNGS expanded the capacity of pathogen detection and made a positive impact on clinical management of suspected infections through (a) differential diagnosis which may rule out infectious diseases and (b) adjustment or de-escalation of empirical antibiotics.
