BACKGROUND: The changes in demographic and family structures have weakened the traditional norms of filial piety and intergenerational relationships dramatically. This study aims to examine the dynamic association between financial support of adult children to their parents and informal care provision in China and its differences in household registration, residence arrangement and community-based care services. METHODS: Data was derived from the 2008-2018 Chinese Longitudinal Healthy Longevity Survey (CLHLS), which is a longitudinal survey of a nationally representative sample of individuals aged 60 and over. Random effects model was used to assess the association between financial support and informal care provision of adult children to their parents. RESULTS: It was found that financial support showed an upward trend while informal care provision showed a download trend from 2008 to 2018. The result indicated a significant and negative association between financial support and informal care provision of adult children to their parents (B = -0.500, 95% confidence interval (CI) = -0.761 to -0.239). And the association was significant among elderly people who were from urban areas (B = -0.628, 95% CI = -0.970 to -0.287), co-resided with adult children (B = -0.596, 95% CI = -0.939 to -0.253), and had community-based services (B = -0.659, 95% CI = -1.004 to -0.315). CONCLUSION: Financial support was negatively associated with informal care provision of adult children to their parents in China, and the association has differences in household registration, residence arrangement and community-based care services. It is suggested that policymakers should prioritize planning interventions for elderly care services and establish a family caregiver support system.

China is promoting the coordinated development of the Beijing-Tianjin-Hebei region as a national strategy project; however, water scarcity and water quality problems will become a bottleneck restricting high-quality development. This study aimed to explore a feasible industrial synergy optimisation pathway to realise the collaborative development of economic growth and water environment improvement, combined with incentives for environmental efficiency improvement and reclaimed water utilisation. Research methods integrate input-output modelling, system dynamics, and multi-objective programming to construct a complex multi-region model. A dynamic simulation measure was adopted to simulate the economic and environmental impacts of different approaches that mix from 2020 to 2030 under water resource environment constraints. According to the simulation results, the annual economic growth rate of the entire region can exceed 6.1%, and the emission intensities of water pollutants decrease by more than 60.0%. In addition, traditional manufacturing industries that achieve cross-regional synergy can still release location advantages without negative environmental impacts. Furthermore, regional collaborative development optimises the allocation of water resources and alleviates water stress. Moreover, the pollutant emission reduction effect of source control in Hebei was more effective than in other cities. Finally, reclaimed water, as the end treatment measure, has the largest marginal effect on improving the trade-off between economic and environmental improvement in the long run. This study provides a new approach for multi-regional industrial synergy development and optimal allocation of resources and contributes to the high-quality development of the watershed.

Cervical and endometrial cancers are common gynecologic cancers. Protein phosphatase 1 regulatory subunit 14B (PPP1R14B) is aberrantly expressed in several tumors, while its functions in cervical and endometrial cancers remain largely uncertain. The differentially expression of PPP1R14B in cervical and endometrial cancers was predicted by GEPIA2 and Human Protein Atlas databases. The diagnostic value was analyzed by AUC curve. The association between PPP1R14B expression and overall survival was predicted using Kaplan-Meier Plotter database. The function of PPP1R14B was investigated according to in vitro assessment. PPP1R14B and phosphorylation level of Akt were analyzed through western blotting. Cell proliferation was investigated by CCK-8 and EdU staining assays. Cell apoptosis was evaluated via TUNEL staining and caspase-3 activity assays. PPP1R14B level was upregulated in cervical and endometrial cancers, and it was associated with diagnosis and worse prognosis. PPP1R14B silencing constrained cell proliferation and promoted cell death in cervical and endometrial cancers cells. PPP1R14B knockdown suppressed activation of the Akt pathway. Re-activation of the Akt signaling reversed the anti-proliferative and cell death-promoting roles of PP1R14B knockdown in cervical and endometrial cancers cells. In conclusion, PPP1R14B knockdown represses cell proliferation and facilitates cell death by inhibiting the activation of the Akt signaling in cervical and endometrial cancers.

Single-cell impedance analysis can provide valuable information for characterizing and discriminating cells. In this paper, a cost-effective portable microfluidic impedance cytometer (MIC) was proposed to realize the broadband impedance analysis of cells by using maximum length sequence (MLS) and viscoelastic focusing. The MIC comprised a microfluidic chip with a straight microchannel, two indium tin oxide (ITO) electrodes, and a home-made platform performing maximum length sequence technique. The viscoelastic focusing enabled cells to focus into a single train to eliminate the influence of cell position variation on acquired electrical signals and allow the cells to pass through the detection region one by one. The MLS technique realized the fast broadband impedance detection of single cells at a low hardware cost. The impedance data under multiplex frequencies was obtained to uncover the dielectric properties of white blood cells (WBCs) and MCF-7 cancer cells. The machine learning was used to train the impedance data and to identify cell types. The results indicated that 98.98% of MCF-7 cells and 98.65% of WBCs were correctly identified. Our MIC showed a potential to be developed as a cost-effective and portable device for point-of-care testing of circulating tumor cells from patients' peripheral blood.

BACKGROUND: Elderly individuals who experience falls suffer from higher levels of anxiety because of physical or mental injury. This study examined the association between falls and anxiety among elderly Chinese individuals. It also explored the mediating roles of functional ability and social participation in the link between falls and anxiety. METHODS: The analytical sample included 8233 elderly people aged 60 and above, and prospective data were obtained from the Chinese Longitudinal Healthy Longevity Survey (CLHLS). Anxiety was evaluated by a 7-item Generalized Anxiety Disorder (GAD-7) scale, and falls were determined by self-report. The association between falls and anxiety was assessed by linear regression. Mediation analysis was used to explore the potential mediating roles of functional ability and social participation on the association between falls and anxiety. RESULTS: Suffering falls predicted higher anxiety levels among elderly individuals (B = 0.608, 95% CI: 0.471, 0.746). Functional ability and social participation play partial mediating roles in the association between falls and anxiety, and the mediating effects were 0.036 (95% CI: 0.020, 0.058) and 0.005 (95% CI: 0.003, 0.014), respectively. The serial mediating effect of functional ability and social participation on the association between falls and anxiety was 0.003 (95% CI: 0.002, 0.005). LIMITATIONS: This study is based upon cross-sectional data, which limit inferring causality. CONCLUSIONS: This study suggests that policy-makers should explore how to encourage elderly individuals who experience falls to restore functional ability and participate in appropriate social activities to alleviate anxiety.

We developed a low-cost polymer-film spiral inertial microfluidic device for the effective size-dependent separation of malignant tumor cells. The device was fabricated in polymer films by rapid laser cutting and chemical bonding. After fabricating the prototype device, the separation performance of our device was evaluated using particles and cells. The effects of operational flow rate, cell diameter, and cell concentration on the separation performance were explored. Our device successfully separated tumor cells from polydisperse white blood cells according to their different migration modes and lateral positions. Then, the separation of rare cells was carried out using the high-concentration lysed blood spiked with 200 tumor cells. Experimental results showed that 83.90% of the tumor cells could be recovered, while 99.87% of white blood cells could be removed. We successfully employed our device for processing clinical pleural effusion samples from patients with advanced metastatic breast cancer. Malignant tumor cells with an average purity of 2.37% could be effectively enriched, improving downstream diagnostic accuracy. Our device offers the advantages of label-free operation, low cost, and fast fabrication, thus being a potential tool for effective cell separation.

The issue of trans-boundary water conflict has become an important factor affecting the relations between basin countries. The key to solve the current conflict problem is to realize the fair and reasonable allocation of trans-boundary water resources. Based on the satisfaction perspective, we developed an asymmetric Nash negotiation model to obtain an optimal and feasible allocation scheme for the trans-boundary water resources. Firstly, based on the two international water laws, we analyzed the influencing factors including water demand differences, resource endowment differences, and water efficiency differences, and by combing with the flexible weight constraint, we built the fair and reasonable allocation pattern for trans-boundary water resources. Secondly, under the constraint of the allocation pattern, we determined the ideal negotiation scheme of each basin country by considering their selection preference. Thirdly, we built a satisfaction degree function and established an asymmetric Nash negotiation model. This is used to build a fair negotiation mechanism among basin countries, and the allocation scheme after negotiation is regarded as the optimal allocation scheme. Lastly, we took the Lancang-Mekong river basin as an example. For this example, the results indicate the following: (1) after considering multiple factors comprehensively, China and Thailand obtained a higher proportion of trans-boundary water resource quota under different preference scenarios, while Myanmar obtained a lower proportion of trans-boundary water resource quota; (2) taking each basin country as the negotiation agent, the optimal allocation scheme with the introduction of fair negotiation mechanism has a higher degree of satisfaction, with an average of over 87.19%, which is more stable and easy to be accepted by all basin countries; (3) from the perspective of the change rate and the average satisfaction of the basin countries, the optimal allocation scheme under the resource endowment preference scenario obtained the highest satisfaction among basin countries. This study aims to improve the practicability and acceptability of trans-boundary water resources allocation, thus providing technical support for reducing trans-boundary water resources conflicts.

In this work, we demonstrate a single-view field filter (SVFF) device for the efficient filtration and enumeration of rare tumor cells in the blood. In our device, the track-etched membrane is integrated within a low-cost polymer-film microfluidic chip, and multiplex microfiltration chambers are designed. Our device permits the performing of multiple sample tests on a single membrane and the dynamical observation of the entire filtration process in a single field of view. To characterize the device performance, our device is first tested using tumor cells, and three different cell behaviors are observed during the filtration process. Finally, we successfully apply our device for the separation of rare tumor cells from the lysed blood samples at various flow rates. The recovery rates of 93.3, 87.6, and 84.1% can be respectively achieved at the throughputs of 50, 100, and 150 µL/min. Our single-view field filter (SVFF) device offers the advantages of label-free filtration, efficient enumeration, easy integration, and low cost, and holds the potential to be used as an efficient tool for the filtration and enumeration of rare cells.

We developed a low-cost multi-core inertial microfluidic centrifuge (IM-centrifuge) to achieve a continuous-flow cell/particle concentration at a throughput of up to 20 mL/min. To lower the cost of our IM-centrifuge, we clamped a disposable multilayer film-based inertial microfluidic (MFIM) chip with two reusable plastic housings. The key MFIM chip was fabricated in low-cost materials by stacking different polymer-film channel layers and double-sided tape. To increase processing throughput, multiplexing spiral inertial microfluidic channels were integrated within an all-in-one MFIM chip, and a novel sample distribution strategy was employed to equally distribute the sample into each channel layer. Then, we characterized the focusing performance in the MFIM chip over a wide flow-rate range. The experimental results showed that our IM-centrifuge was able to focus various-sized particles/cells to achieve volume reduction. The sample distribution strategy also effectively ensured identical focusing and concentration performances in different cores. Finally, our IM-centrifuge was successfully applied to concentrate microalgae cells with irregular shapes and highly polydisperse sizes. Thus, our IM-centrifuge holds the potential to be employed as a low-cost, high-throughput centrifuge for disposable use in low-resource settings.

Microfluidic technologies for cell separation were reported frequently in recent years. However, a compact microfluidic instrument enabling thoroughly automated cell separation is still rarely reported until today due to the difficult hybrid between the macrosized fluidic control system and the microsized microfluidic device. In this work, we propose a novel and automated microfluidic instrument to realize size-based separation of cancer cells in a label-free and high-throughput manner. Briefly, the instrument is equipped with a fully integrated microfluidic device and a set of robust fluid-driven and control units, and the instrument functions of precise fluid infusion and high-throughput cell separation are guaranteed by a flow regulatory chip and two cell separation chips which are the key components of the microfluidic device. With optimized control programs, the instrument is successfully applied to automatically sort human breast adenocarcinoma cell line MCF-7 from 5 mL of diluted human blood with a high recovery ratio of ∼85% within a rapid processing time of ∼23 min. We envision that our microfluidic instrument will be potentially useful in many biomedical applications, especially cell separation, enrichment, and concentration for the purpose of cell culture and analysis.

This work presents the fabrication of a microfluidic autoregulatory valve which is composed of several layers of thin polymer films (i.e., polyvinyl chloride (PVC), polyethylene terephthalate (PET) double-sided tape, and polydimethylsiloxane (PDMS)). Briefly, pulsed UV laser is employed to cut the microstructures of through grooves or holes in the thermoplastic polymer films, and then the polymer-film valves are precisely assembled through laminating the PDMS membranes to the thermoplastic polymer films through the roll-lamination method. The effective bonding between the PVC film and the PDMS membrane is realized using the planar seal method, and the valve is sandwiched and compressed by a home-made housing to achieve the good seal effect. Then, the flow performances of the prototype valve are examined, and constant flow autoregulation is realized under the static or dynamic test pressures. The long-term response of the valve is also studied and minimum flow-rate decrements are found over a long actuation time. The fabrication method proposed in this work is successful for the low-cost and fast prototyping of the polymer-film valve. We believe our method will also be broadly applicable for fabrication of other low-cost and disposable polymer-film microfluidic devices.

BACKGROUND: All versions of the Montreal Cognitive Assessment (MoCA) lack population-based data of 80-plus individuals. The norms and cut-off scores for mild cognitive impairment (MCI) and dementia of the MoCA are different among five Chinese versions. OBJECTIVE: To provide the cut-off scores in detecting MCI and dementia of the Peking Medical Union College Hospital version of the MoCA (MoCA-P). METHODS: In a cross-sectional survey, Chinese veterans aged ≥60 years completed the MoCA-P and the Mini-Mental State Examination (MMSE). RESULTS: Among 7,445 elderly veterans, 5,085 (68.30%) were aged ≥80 years old, 2,621 (35.20%) had 6 years of education or less, 6,847 (91.97%) were male, and 2,311 (31.04%) and 984 (13.22%) veterans were diagnosed as having MCI and dementia, respectively. Adding two points and one point to the MoCA scores for the primary and middle school groups, respectively, can fully adjust for the notable impact of education but cannot compensate for the effect of age. In the three age groups (60-79, 80-89, and ≥90 years old), the optimal MoCA-P cut-off scores for detecting MCI were ≤25, ≤24, and ≤23, respectively, and for detecting dementia were ≤24, ≤21, and ≤19, respectively, which demonstrated relatively high sensitivities and specificities. The areas under the curves for the MoCA-P for detecting MCI and dementia (0.937 and 0.908, respectively) were greater than those for the MMSE (0.848 and 0.892, respectively). CONCLUSION: Compared with the MMSE, the MoCA-P is significantly better for detecting MCI in the elderly, particularly in the oldest old population, and it also displays more effectiveness in detecting dementia.

The wide production and use of phthalate esters (PAEs) in both industry and commercial products lead to their ubiquitous existence in the environment. However, understanding flow and pathway of human exposure to PAEs from sources to receptors is necessary and challenging. In this study, we selected final sewage sludge, an inevitable byproduct of wastewater treatment plants (WWTP), as one type of important carrier/sources of PAEs to clarify the flow of PAEs between human and the environment, e.g. the release by human to sludge and in turn ingestion by human after these sludges were disposed as soil amendment. Twenty-five sludge samples were collected from 25 wastewater treatment plants (WWTPs) in Shanghai, East China. Of all 16 PAE congeners, di(2-ethylhexyl) phthalate (DEHP) and dibutyl phthalate (DnBP) were predominant with mean concentrations of 97.4 and 22.4 µg/g dw, respectively, both locating at the high end of the global range. WWTP treating industrial waster generally contained higher PAEs compared to those treating domestic wastewater, but no positive relationship was found between PAE levels with the percentage of industrial wastewater. Principal component analysis (PCA) showed that similar PAE sources for all WWTPs in Shanghai with three exceptions, in which specific PAE products were made. The annual mass loadings via sludge of DEHP, DnBP, and Σ16PAEs were 31.4, 7.44, and 39.6 tons in Shanghai and 1042, 247, and 1314 tons in China, respectively, only accounting for 0.09% of the total consumption of PAEs in China. If this sludge is applied in soil, human will take 16.4 and 3.8 µg/kg bw for DEHP and DnBP every day, respectively, via dietary and soil ingestion, which were lower than the toxicological safety parameters. To the best of our knowledge, this is the first report to analyze the flow of sludge-borne PAEs from human release to human intake.

OBJECTIVE: Multiple linkage and association studies have suggested chromosome 8q24 as a promising candidate region for bipolar disorder (BP). We performed a detailed association analysis assessing the contribution of common genetic variation in this region to the risk of BP. METHODS: We analyzed 2,756 single nucleotide polymorphism (SNP) markers in the chromosome 8q24 region of 3,512 individuals from 737 families. In addition, we extended genotype imputation methods to family-based data and imputed 22,725 HapMap SNPs in the same region on 8q24. We applied a family-based method to test 15,552 high-quality genotyped or imputed SNPs for association with BP. RESULTS: Our association analysis identified the most significant marker (p=4.80 × 10(-5) ), near the gene encoding potassium voltage-gated channel KQT-like protein (KCNQ3). Other marginally significant markers were located near adenylate cyclase 8 (ADCY8) and ST3 beta-galactoside alpha-2,3-sialyltransferase 1 (ST3GAL1). CONCLUSIONS: We developed an approach to apply MACH imputation to family-based data, which can increase the power to detect association signals. Our association results showed suggestive evidence of association of BP with loci near KCNQ3, ADCY8, and ST3GAL1. Consistent with genes identified by genome-wide association studies for BP, our results suggest the involvement of ion channelopathy in BP pathogenesis. However, common variants are insufficient to explain linkage findings in 8q24; other genetic variation should be explored.
