RESUMO

Dynamic extraction of edible tissues of Jinjiang oyster (Crassostrea rivularis) and Flower clam (Paphia undulata) was conducted using a fully biomimetic digestion (in vitro) method. The impact of different cooking methods on the bioavailability of Zn and Cd in the edible shellfish tissues was analyzed, and the human health risk of Zn and Cd was evaluated. The results show that the gastric biomimetic extractions of Zn and Cd in unheated samples of C. rivularis and P. undulata were higher than those in the intestinal biomimetic extraction. The extraction patterns of cooked samples were consistent with those of raw samples. The bioavailability of Zn and Cd in cooked samples of C. rivularis was 94.9% and 82.5%, respectively, indicating increased Zn bioavailability but decreased Cd bioavailability compared to the raw samples. The bioavailability of Zn and Cd in cooked samples of P. undulata was 85.1% and 83.0%, respectively, both of which decreased compared to the raw samples. Consumption of C. rivularis can provide 21.0% to 34.2% of the daily required Zn intake, while consumption of P. undulata can provide 3.8% to 6.4%. The intake of Cd from both shellfish species is below the monthly tolerable intake recommended by FAO/WHO. Consuming cooked C. rivularis can increase the intake of Zn and decrease the intake of Cd.

Bioenrichment preference of arsenic and metals in wild marine organisms has been scarcely considered. Twenty species including fishes, cephalopods, crustaceans, and bivalve mollusks were collected from Dapeng (Mis) Bay and analyzed for arsenic and metals. Through this study, we had obtained the following four main conclusions: (1) average concentrations of arsenic and metals (µg/kg, wet weight) in the aquatic organism samples were 48.7 for Cr, 1762.0 for Mn, 20,632.8 for Fe, 33.0 for Co, 119.5 for Ni, 3184.7 for Cu, 12,040.5 for Zn, 389.0 for As, 189.1 for Se, 144.4 for Cd, 15.0 for Hg, and 55.3 for Pb; (2) factor analysis (FA) revealed that the studied twenty species exhibited three types of arsenic and metal bioenrichment preference;(3) non-carcinogenic health risk assessment indicated insignificant health effects from marine organism consumption; (4) carcinogenic health risk assessment revealed an unacceptable risk from consumption of nine species, seven of which were crustaceans.

BACKGROUND: Fibroblast growth factor (FGF) 14 is a member of the FGF family that is mainly expressed in the central nervous system. FGF14 has a close association with the occurrence of neurodegenerative conditions; however, its significance in Alzheimer's disease (AD) has yet to be evaluated. Therefore, we sought to obtain a large amount of exogenous FGF14 protein and explore its effect in a cellular model of AD. METHODS: FGF14 protein was expressed in an Escherichia coli system using gene recombination technology. Purified protein was obtained through washing and renaturation of inclusion bodies combined with nickel column affinity chromatography. The AD model was established via Aß25-35-induced injury in PC12 cells. Changes in the levels of lactate dehydrogenase and malondialdehyde were detected, and the neuroprotective effect of recombinant human FGF14 (rhFGF14) was evaluated through double-fluorescence staining and flow cytometry apoptosis detection. For further exploration of rhFGF14-mediated regulation of mitogen-activated protein kinase (MAPK) signaling, western blot was employed. RESULTS: We successfully induced large amounts of insoluble rhFGF14. Following solubilization and refolding of the rhFGF14 from inclusion bodies, high purity rhFGF14 was purified by Nickel affinity column chromatography. The results showed that rhFGF14 alleviated Aß25-3-induced PC12 cell injury by inhibiting the phosphorylation of p38, extracellular signal-regulated kinase 1/2, and c-Jun N-terminal kinase, thus suppressing the MAPK signaling pathway. CONCLUSIONS: FGF14 performed a neuroprotective role in our in vitro AD model via its inhibition of MAPK signaling, highlighting its potential as a therapeutic drug for neurodegenerative conditions.

Mucin-type O-glycosylation occurs on many proteins that transit the Golgi apparatus. These glycans impact structure and function of many proteins and have important roles in cellular biosynthetic processes, signaling and differentiation. Although recent technological advances have enhanced our ability to profile glycosylation of glycoproteins, limitations in the understanding of the biosynthesis of these glycan structures remain. Some of these limitations stem from the difficulty to track the biosynthetic process of mucin-type O-glycosylation, especially when glycans occur in dense clusters in repeat regions of proteins, such as the mucins or immunoglobulin A1 (IgA1). Here, we describe a series of nano-liquid chromatography (LC)-mass spectrometry (MS) analyses that demonstrate the range of glycosyltransferase enzymatic activities involved in the biosynthesis of clustered O-glycans on IgA1. By utilizing nano-LC-MS relative quantitation of in vitro reaction products, our results provide unique insights into the biosynthesis of clustered IgA1 O-glycans. We have developed a workflow to determine glycoform-specific apparent rates of a human UDP-N-acetylgalactosamine:polypeptide N-acetylgalactosaminyltrasnfersase (GalNAc-T EC 2.4.1.41) and demonstrated how pre-existing glycans affect subsequent activity of glycosyltransferases, such as core 1 galactosyltransferase and α2,3- and α2,6-specific sialyltransferases, in successive additions in the biosynthesis of clustered O-glycans. In the context of IgA1, these results have potential to provide insight into the molecular mechanisms implicated in the pathogenesis of IgA nephropathy, an autoimmune renal disease involving aberrant IgA1 O-glycosylation. In a broader sense, these methods and workflows are applicable to the studies of the concerted and competing functions of other glycosyltransferases that initiate and extend mucin-type core 1 clustered O-glycosylation.

Previous research has revealed associations between childhood maltreatment (CM) and adverse health behaviors. However, little is known about the relationship between CM and adolescent murderous ideation and behaviors. A total of 5726 middle and high school students completed the Childhood Trauma Questionnaire-Short Form and the Murderous Ideation and Behaviors Questionnaire. The findings revealed that the prevalence rates for murderous ideation, plans, preparation, and attempts were 9.9%, 2.8%, 1.3%, and 0.6%, respectively. The results of multinomial logistic regression models indicated that adolescents who experienced CM were more likely to exhibit murderous ideation and behaviors, with adjusted odds ratios (AORs) ranging from 2.55 to 22.31. Additionally, a significant dose-response relationship was found between the number of CM types experienced and murderous ideation and behaviors (AORs ranging from 1.52 to 2.45). The odds of participants who had experienced three or five types of CM were significantly associated with murderous ideation and behaviors, with AORs ranging from 4.55 to 28.30 and from 5.26 to 85.45, respectively. The findings highlighted that adolescents who engaged in murderous ideation and behaviors were more likely to have a personal history of CM and revealed a dose-response relationship between the number of CM types and murderous ideation and behaviors.

The concentrations of heavy metals (Cd, Pb, Cr, Ni, Cu, Zn, Hg and As) in surface sediments and bivalve mollusks in Kaozhouyang Bay were investigated. A biological risk analysis of the sediments indicated that ten sites (about 76.92% of the total number of sites) had a 21% incidence probability of toxicity. A health risk analysis of the bivalve mollusks indicated that Cu and As posed low risks to consumer health. On the basis of the target hazard quotient (THQ), adverse effects may occur based on total THQ (TTHQ). The highest TTHQ was found in the species, Ostrea rivularis, which had the highest capacity for the bioaccumulation (factor > 38) of Cd.

The fuzzy comprehensive assessment was undertaken to evaluate the marine sediment quality in Daya Bay, China based on the monitoring data of 2015 and China National Standard for Marine Sediment Quality (GB 18668-2002). The results demonstrated that the average metal concentrations (mg/kg) were 0.08 (Cd), 51.30 (Pb), 91.30 (Cr), 29.63 (Cu), 143.42 (Zn), 0.03 (Hg), and 7.31 (As), which were clearly higher with respect to their corresponding background values. Cr was the major pollutant based weight matrices calculated. Taken as whole, the membership degree of class I was a range from 0.65 to 1, suggesting that the marine sediment quality in Daya Bay belongs to class I. Pb stable isotopic analysis coupled with geographic information system (GIS) approach showed the sedimentary Pb mainly originated from petrochemical industry at northwest coast of Daya Bay.

BACKGROUND AND AIM: There is growing interest in how integrins play a role in cancer disease biology and what applications these may have in anti-cancer therapeutic development. This study investigates integrin αvß5 expression in non-small cell lung carcinoma (NSCLC) and its correlations with clinical information. METHODS: Tumor human tissue microarrays of 147 radically operated Chinese NSCLC patients were retrieved from the pathology archive, and sections were autoimmune stained along with positive and negative controls. Integrin αvß5 (P1F6) mouse monoclonal antibody were validated by both Western blotting and immunoreactivity on the same set of cell pellets, according to a precedent of expression levels in cell flow cytometry. The immunoreactivity of all patients' cases against integrin αvß5 antibody was graded in a semi-quantitative manner by two pathologists blind to any patient data. RESULTS: Overall survival significantly correlated to integrin αvß5 immunoreactivity by Cox regression analysis with P = 0.032. When applying Kaplan-Meier to analyze the comparison between positive and negative expression on lymph node metastasis patients, P = 0.082. Therefore, integrin αvß5 expression emerges as a significant prognostic factor for NSCLC. In total, 39 of 147 (26.5%) showed an integrin αvß5 immunoreactivity positive, as it ranged from 6.1% in squamous cell carcinoma to 19.7% in adenocarcinoma. This expression correlated significantly with histological subtypes (P = 0.007), tumor node metastasis classification of malignant tumors (P = 0.027) and lymph node metastases (P = 0.006). CONCLUSION: This study support the hypothesis that integrin αvß5 is a prognostic biomarker and correlates to metastasis with therapeutic development potential in NSCLC.
