RESUMO

Background: Previous research on the association between risk factors and gestational diabetes mellitus (GDM) primarily comprises observational studies with inconclusive results. The objective of this study is to investigate the causal relationship between 108 traits and GDM by employing a two-sample Mendelian randomization (MR) analysis to identify potential risk factors of GDM. Methods: We conducted MR analyses to explore the relationships between traits and GDM. The genome-wide association studies (GWAS) for traits were primarily based on data from the UK Biobank (UKBB), while the GWAS for GDM utilized data from FinnGen. We employed a false discovery rate (FDR) of 5% to account for multiple comparisons. Results: The inverse-variance weighted (IVW) method indicated that the genetically predicted 24 risk factors were significantly associated with GDM, such as "Forced expiratory volume in 1-second (FEV1)" (OR=0.76; 95% CI: 0.63, 0.92), "Forced vital capacity (FVC)" (OR=0.74; 95% CI: 0.64, 0.87), "Usual walking pace" (OR=0.19; 95% CI: 0.09, 0.39), "Sex hormone-binding globulin (SHBG)" (OR=0.86; 95% CI: 0.78, 0.94). The sensitivity analyses with MR-Egger and weighted median methods indicated consistent results for most of the trats. Conclusion: Our study has uncovered a significant causal relationship between 24 risk factors and GDM. These results offer a new theoretical foundation for preventing or mitigating the risks associated with GDM.

BACKGROUND: Laodelphax striatellus is one of the most destructive pests of rice and other cereal crops. Chemical control is still the most efficient way to control this pest, but insecticide resistance always threatens this approach. RESULTS: Monitoring data (2003-2020) showed that Chinese field populations of L. striatellus developed high-level buprofezin resistance within the first four years. This high-level resistance to buprofezin was stable for about ten years and persisted even when buprofezin selection pressure was absent. An established near-isogenic strain (YN-NIS) with 90.8-fold resistance to buprofezin had resistance inheritance of autosomal and incomplete dominance, and the resistance was controlled by multiple genes with no obvious fitness costs (relative fitness of 0.8707). Furthermore, the susceptibility of 29 field populations to another seven insecticides (2014-2020) showed that: (i) low-level resistance to pymetrozine, dinotefuran, sulfoxaflor and thiamethoxam was first detected in 2014 (eight years after introduction), 2016 (three years after), 2017 (four years after) and 2019 (19 years after), respectively, (ii) moderate resistance levels to chlorpyrifos were found for all populations across multiple years, and (iii) no resistance was detected for nitenpyram and triflumezopyrim. CONCLUSION: The fast buprofezin resistance development in L. striatellus would be caused by incomplete dominant resistance with almost no fitness cost in the resistant strain. Nitenpyram and triflumezopyrim showed no resistance and can be used as the main insecticide for the control of L. striatellus. These findings provide key fundamental information for controlling L. striatellus.

Yadong County located in the southern Himalayan mountains in Tibet, China, is an import frontier county. It was affected by landslides after the 2011 Sikkim earthquake (Mw = 6.8) and the 2015 Gorkha earthquake (Mw = 7.8). Casualties and property damage were caused by shallow landslides during subsequent rainfall on the earthquake-destabilized slopes. Existing researches have generally examined rainfall- and earthquake-triggered landslides independently, whereas few studies have considered the combined effects of both. Furthermore, there is no previous study reported on landslide hazards in the study area, although the area is strategically applicable for trade as it is close to Bhutan and India. This study developed a new approach that coupled the Newmark method with the hydrological model based on geomorphological, geological, geotechnical, seismological and rainfall data. A rainfall threshold distribution map was generated, indicating that the southeast part of Yadong is prone to rainfall-induced landslides, especially when daily rainfall is higher than 45 mm/day. Permanent displacement predictions were used to identify landslide hazard zones. The regression model used to calculate these permanent displacement values was 71% accurate. Finally, landslide probability distribution maps were generated separately for dry and wet conditions with rainfall of varying intensities. Results can serve as a basis for local governments to manage seismic landslide risks during rainy seasons.

Triflumezopyrim, a novel mesoionic chemical insecticide, is promoted as a powerful tool for control of susceptible and resistant hopper species in rice throughout Asia. For a newly commercialized insecticide it is important to establish susceptibility baseline, conduct susceptibility monitoring, and assess the risk of resistance via artificial selection to provide foundational information on designing resistance management strategy. The susceptibility baseline of triflumezopyrim was established for three rice planthopper species, Nilarpavata lugens (Stål), Sogatella furcifera (Horváth) and Laodelphax striatellus (Fallén). The LD50 of triflumezopyrim was 0.026, 0.032 and 0.094 ng/individual for the adults of the susceptible strains of S. furcifera, L. striatellus and N. lugens, respectively, determined by a topical application method. Using a rice stem (seedling) dipping method, the LC50 was determined as 0.042, 0.024 and 0.150 mg/L for the nymphs (3rd instar) of the three hopper species, respectively. In the meanwhile, the LC50 of Pyraxalt™ (triflumezopyrim 10% SC) was 0.064 mg/L for the N. lugens susceptible strain. Furthermore, the susceptibility of triflumezopyrim and other five neonicotinoid insecticides were monitored for N. lugens field populations collected from major rice production areas in China in 2015-2019. All monitored populations were susceptible to triflumezopyrim (0.5 to 3.9-fold resistance ratio), and showed no cross-resistance to the other five neonicotinoids. These results suggested that triflumezopyrim is a good option to control resistant N. lugens. In addition, a field-collected population of N. lugens was artificially selected with triflumezopyrim for 20 generations and resulted in 3.5-fold increase in LC50 from F0 and 6.0-fold increase from that of the susceptible strain. The realized heritability (h2) of resistance was estimated as 0.0451 by using threshold trait analysis. With this h2 value, the projected triflumezopyrim resistance development (a 10-fold increase in LC50) would be expected after 30.3 or 24.0 generations if 80% or 90% of the population was killed at each generation.

After pre-fatigue cycles at different strain amplitudes with different N/Nf values (33.3%, 50%, and 75%), specimens of HRB335 steel were subjected to uniaxial tension until failure. By this method the mechanical properties of the specimens after pre-fatigue testing were measured, and the fracture morphology and microscopic morphology in the vicinity of the specimen's neck surface near the fracture were observed. The verification of the characteristics to be used to estimate the damage caused during the loading cycles was conducted. By observing optical microscope images of the surface area near the neck of the specimens, it was found that the images of surface cracks were significantly different and strongly depended on the number of pre-fatigue cycles the specimen had undergone. In response to this phenomenon, both the microscopic images taken directly from the photos of the surface crack distribution and the binary images based on them were statistically analyzed, and then a parameter, S, denoted as the "unit crack area", characterizing the cumulative fatigue damage was suggested. Furthermore, the test procedure and the calculation formula for determining the image parameters were summarized, and a method for evaluating the remaining life of steel after low-cycles of reversed tension and compression was proposed.
