RESUMO

Chlorinated paraffins (CPs), which were conventionally classified into short- (SCCPs), medium- (MCCPs) and long- (LCCPs) chain CPs, have received growing attention due to their wide usage and extensive detection in environmental samples and biota. The number of studies regarding the biomonitoring of CPs in human beings increased rapidly and their health risk gained great concern. This review summarized their occurrence and homologue patterns in human matrices including blood/serum, placenta, cord serum and breast milk. As the production and usage of SCCPs was progressively banned after being listed in Annex A of the Stockholm Convention, the production of MCCPs and LCCPs was stimulated. Accordingly, the ratio of MCCPs/SCCPs in human samples has increased rapidly in the last 5 years. The current understanding of exposure routes and risk assessments of CPs was also reviewed. Oral dietary intake is the most predominant source of daily CP intake, but dust ingestion, inhalation and dermal exposure is also nonnegligible, especially for MCCPs and LCCPs. Furthermore, the reported upper bound of the estimated daily intakes (EDIs) in various risk assessment studies was close to or exceeded the tolerable daily intakes (TDIs). Considering the bioaccumulation and long-lasting exposure of CPs, their health impacts on humans and the ecosystem required continuous monitoring and evaluation.

The present study investigated the temporal and spatial distributions, partition behaviors, sources, and risks of 14 perfluoroalkyl acids (PFAAs) in the aquatic environment of a fluorine industry-impacted region. The total concentrations of 14 PFAAs (ΣPFAAs) were 118.10-2235.4 ng/L, 40.00-2316.1 ng/g dw, and 6.90-180.5 ng/g dw in dissolved, suspended particle matter (SPM), and sedimentary phases, respectively. The predominant pollutants in the dissolved and SPM phases were perfluoroalkyl carboxylic acids (PFCAs) with carbon chain lengths <9, whereas C13 and C14 PFCAs accounted for a large proportion in the sedimentary phase. The dry season exhibited the highest concentration of ΣPFAAs in the dissolved phase (500.9 ± 350.2 ng/L), while the wet season showed the highest concentrations of ΣPFAAs in the SPM and sedimentary phases (591.6 ± 469.1 ng/g dw and 59.7 ± 35.5 ng/g dw, respectively). Significantly higher concentrations of PFAAs have been found in sewage plant and industrial areas. The concentration of PFAAs in the Xupu water source area (XPS) was slightly higher than that in other water source areas of the Yangtze River, which were either not affected or were less affected by the fluorine industry. The log KD-SPM (distribution coefficient between SPM and water), log KD-SED (distribution coefficient between sediment and water), and log KOC-SED (the organic carbon normalized distribution coefficient) of PFAAs showed significant differences between the wet season and dry season, which may also be affected by carbon chain length. Source identification results showed that industries, wastewater discharge, and nonpoint sources were the main sources of PFAAs in this region. The ecological risk posed by long-chain PFAAs in aquatic organisms cannot be ignored, especially in areas with intensive industrial and agricultural activities. Health risks may exist for local toddlers with long-term exposure to perfluorooctanoic acid (PFOA) through drinking water intake and dermal contact.

Oral leukoplakia is the most common type of oral potentially malignant disorders and considered a precursor lesion to oral squamous cell carcinoma. However, a predictor of oral leukoplakia prognosis has not yet been identified. We investigated whether copy number alteration patterns may effectively predict the prognostic outcomes of oral leukoplakia using routinely processed paraffin sections. Comparison of copy number alteration patterns between oral leukoplakia with hyperplasia (HOL, n=22) and dysplasia (DOL, n=21) showed that oral leukoplakia with dysplasia had a higher copy number alteration rate (86%) than oral leukoplakia with hyperplasia (46%). Oral leukoplakia with dysplasia exhibited a wider range of genomic variations across all chromosomes compared with oral leukoplakia with hyperplasia. We also examined a retrospective cohort of 477 patients with oral leukoplakia with hyperplasia with detailed follow-up information. The malignant transformation (MT, n=19) and leukoplakia recurrence (LR, n=253) groups had higher frequencies of aneuploidy events and copy number loss rate than the free of disease (FD, n=205) group. Together, our results revealed the association between the degree of copy number alterations and the histological grade of oral leukoplakia and demonstrated that copy number alteration may be effective for prognosis prediction in oral leukoplakia patients with hyperplasia.

Polychlorinated biphenyls (PCBs) may transfer into the neonates through the placental transfer and via breastfeeding after the delivery, thus might be harmful to the infant. Sixty colostrum samples in the Yangtze River Region were collected to investigate the concentration, distribution pattern, and enantiomer characteristic of the PCB exposure. Among all samples, over 90% of pollutants were tetra-to hepta-chlorinated PCBs. The sum concentration of the PCB was 512 (IQR: 322-856) ng g-1 lipid weight. Enantiomer fraction (EF) of PCB 95 and PCB 149 was found lower than the racemic value, while EFs of PCB 45 and PCB 136 were found higher and near-racemic state, respectively. The concentration pattern and enantiomeric properties of the PCBs indicated that the mothers from Mianyang had a recent exposure to PCBs. Among all samples, similar exposure and metabolic pathways of the PCB congeners were observed. PCB exposure showed no significant correlation with the birth outcome of the infants, but 43.3% of the infants have potential health risks via breastfeeding.

Based on the existing comprehensive ecological risk assessment methods of PAHs, this paper proposed an improved hierarchical Archimedean copula integral assessment (HACIA) model with the optimization in the model selection mechanism and accelerating the calculation speed, and according to which performed the sensitivity analysis of the integrated risk relative to the underlying grouped risk probability. Taihu Lake in China and the Bay of Santander in Spain were taken as study areas, whose samples were obtained and extracted concentrations of 16 priority polycyclic aromatic hydrocarbons (PAHs). After briefly analyzing their concentration characteristics and source, their comprehensive ecological risks were evaluated by the improve HACIA model and their sensitivity was also analyzed. The results proved that, for Taihu Lake, pyrogenic sources occupied the dominance, especially grass, coal and wood combustion, while the risk proportion of 5-rings PAHs was the lowest indeed based on the improved HAICA model. For the Bay of Santander, source apportionment indicated both petrogenic and pyrogenic sources, mainly from vehicle emissions including gasoline and diesel engines, and 4-ring PAHs were urgently needed to be managed. However, the sensitivity analysis results of two study areas showed that the most effective control target for reducing integral risk has no obvious relationship with the maximum grouped risk. And a clear linear relationship between the maximum sensitivity range and the logarithm of the initial overall risk only presented in one of study areas, which required further research to clarify. In brief, the improved HACIA model is helpful to evaluate the comprehensive ecological risk of 16 PAHs, and formulate risk management strategies based on grouped risk assessment and sensitivity analysis, with the former points out the admonitory risk and the latter helps to find the most effective mitigation measures.

Information regarding the enantioselective endocrine disruption of chiral herbicides is scarce. This study assessed the disrupting effects of eight typical chiral herbicides on corticosteroids (including glucocorticoids and mineralocorticoids). Enantioselectivity of eight chiral herbicides were evaluated for their agonistic/antagonistic effects on glucocorticoid receptor (GR) and mineralocorticoid receptor (MR) with CHOK1 cell line using reporter gene assay. Their influence on the production of corticosteroids were further investigated in H295R cell line using enzyme-linked immunosorbent assay (ELISA). None of the racemates or enantiomers of eight chiral herbicides exhibited GR or MR agonistic activity at non-cytotoxic concentrations. However, rac-propisochlor and S-imazamox antagonized cortisol-induced transactivation of GR by 21.79% and 38.73% at the concentration of 1.0 × 10-7 M and 1.0 × 10-6 M, respectively, and R-napropamide remarkably attenuated aldosterone-induced MR transactivation by 68.78% at 1.0 × 10-6 M. The secretion of cortisol was significantly restrained after treated with 1.0 × 10-6 M rac-propisochlor and rac-/R-napropamide at the concentration of 1.0 × 10-6 M by 26.49%, 30.10% and 35.27%, respectively, while this glucocorticoid was remarkably induced by 1.0 × 10-5 M rac-diclofop-methyl and its two enantiomers at the concentration of 1.0 × 10-5 M by 75.60%, 100.1% and 68.78%, respectively. Exposure to rac-propisochlor (1.0 × 10-6 M), S-diclofop-methyl (1.0 × 10-5 M) or rac-/S-/R- acetochlor (1.0 × 10-6 M) and rac-/S-/R-lactofen (1.0 × 10-6 M) inhibited the secretion of aldosterone by approximately 40%. Our findings suggested that chiral herbicides disrupted corticosteroid homeostasis in an enantioselective way. Therefore, more comprehensive screening is required to better understand the ecological and health risks of chiral pesticides.

Hydropower development is the key strategy in many developing countries for energy supply, climate-change mitigation and economic development. However, it is commonly assumed that river dams retain nutrients and therefore reduce downstream primary productivity and fishery catches, compromising food security and causing trans-boundary disputes. Contrary to expectation, here we found that a cascade of reservoirs along the upper Mekong River increased downstream bioavailability of nitrogen and phosphorus. The dams caused phytoplankton density to increase with hydraulic residence time and stratification of the stagnant reservoirs caused hypoxia at depth. This allowed the release of bioavailable phosphorus from the sediment and an increase in dissolved inorganic nitrogen as well as a shift in nitrogen species from nitrate to ammonium, which were transported downstream by the discharge of water from the base of the dam. Our findings provide a new perspective on the environmental impacts of river dams on nutrient cycling and ecosystem functioning, with potential implications for sustainable development of hydropower worldwide.

Short-chain chlorinated paraffins (SCCPs), which are candidate persistent organic pollutants (POPs) according to the Stockholm Convention, are of great concern because of their persistent bioaccumulation, long-range transport and potential adverse health effects. However, data on the endocrine-disrupting effects of SCCPs remain scarce. In this study, we first adopted two in vitro models (reporter gene assays and H295R cell line) to investigate the endocrine-disrupting effects of three SCCPs (C10-40.40%, C10-66.10% and C11-43.20%) via receptor mediated and non-receptor mediated pathway. The dual-luciferase reporter gene assay revealed that all test chemicals significantly induced estrogenic effects, which were mediated by estrogen receptor α (ERα), in the following order: C11-43.20%>C10-66.10%>C10-40.40%. Notably, C10-40.40% and C10-66.10% also demonstrated remarkable anti-estrogenic activities. Only C11-43.20% showed glucocorticoid receptor-mediated (GR) antagonistic activity, with a RIC20 value of 2.6×10(-8)mol/L. None of the SCCPs showed any agonistic or antagonistic activities against thyroid receptor ß (TRß). Meanwhile, all test SCCPs stimulated the secretion of 17ß-estradiol (E2). Both C10-66.10% and C11-43.20% increased the production of cortisol at a high level in H295R cell lines. In order to explore the possible mechanism underlying the endocrine-disrupting effects of SCCPs through the non-receptor pathway, the mRNA levels of 9 steroidogenic genes were measured by real-time polymerase chain reaction (RT-PCR). StAR, 17ßHSD, CYP11A1, CYP11B1, CYP19 and CYP21 were upregulated in a concentration-dependent manner by all chemicals. The data provided here emphasized that comprehensive assessments of the health and ecological risks of emerging contaminants, such as SCCPs, are of great concern and should be investigated further.
