INTRODUCTION: Citri Sarcodactylis Fructus (CSF), a common fruit and traditional Chinese medicine (TCM), has been hindered in its further development and research owing to the lack of comprehensive and specific quality evaluation standards. OBJECTIVE: This study aimed to establish clear TCM quality standards related to the therapeutic mechanisms of CSF and to provide a basis for subsequent research and development. METHODS: Ultra-high performance liquid chromatography coupled with hybrid quadrupole-orbitrap high-resolution mass spectrometry (UPLC-Q-orbitrap HRMS) technology was used to comprehensively identify CSF components and explore their absorbance levels in rat serum. Network pharmacology research methods were employed to investigate the potential mechanisms of action of the identified components in the treatment of major clinical diseases. Subsequently, a combination of HPLC chromatographic fingerprinting for qualitative analysis and multi-index content determination was used to evaluate the detectability of the identified quality markers (Q-markers). RESULTS: Twenty-six prototype components were tentatively characterized in rat serum. Network pharmacology analysis showed six effective components, namely 7-hydroxycoumarin, isoscopoletin, diosmin, hesperidin, 5,7-dimethoxycoumarin, and bergapten, which played important roles in the treatment of chronic gastritis, functional dyspepsia, peptic ulcer, and depression and were preliminarily identified as Q-markers. The results of content determination in 15 batches of CSF indicated significant differences in the content of medicinal materials from different origins. However, compared with the preliminarily determined Q-markers, all six components could be measured and were determined as Q-markers of CSF. CONCLUSION: The chemical Q-markers obtained in this study could be used for effective quality control of CSF.

A novel phosphate-accumulating organism (PAO), Arthrobacter sp. HHEP5 was isolated from mariculture effluents. It produced no hemolysin and was susceptible to most antibiotics. It had removal efficiencies of above 99% for 1-10 mg/L phosphorus at 18-28 °C, pH 5.5-8.5, salinities 0-3%, C/N ratios 5-20, P/N ratios 0.1-0.2 and 20-260 rpm. It exhibited simultaneous aerobic phosphorus removal, nitrification and denitrification with the highest ammonium, nitrite, nitrate removal efficiencies of 99.87%, 100%, 99.37%. Phosphorus removal was accomplished by assimilating phosphate with the existence of polyphosphate kinase completely under aerobic condition. Genes involved in nitrogen removal were amplified. 99% of phosphorus and 95% of nitrogen in both mariculture and domestic wastewater were removed by HHEP5. This study provided sound methods for future screening of PAOs and new perspectives for renewed cognition of phosphorus removal process. Wide adaptation and remarkably aerobic phosphorus, nitrogen removal performances would make HHEP5 a promising candidate in wastewater treatment.

A study was conducted to determine the genetic diversity of 39 determinate and indeterminate tomato inbred lines collected from China, Japan, S. Korea, and USA. Using 35 SSR polymorphic markers, a total of 150 alleles were found with moderate levels of diversity, and a high number of unique alleles existing in these tomato lines. The mean number of alleles per locus was 4.3 and the average polymorphism information content (PIC) was 0.31. Unweighted Pair Group Method with Arithmetic Mean (UPGMA) clustering at genetic similarity value of 0.85 grouped the inbred lines into four groups, where one USA cultivar formed a separate and more distant cluster. The most similar inbred lines are from USA, both with determinate type, whereas the most different lines are from USA (Us-16) and Japan (Ja-2) with determinate and indeterminate growth habit, respectively. Clustering was consistent with the known information regarding geographical location and growth habit. The genetic distance information reported in this study might be used by breeders when planning future crosses among these inbred lines.
