RESUMO

Liquid crystal monomers (LCMs) are widely used in liquid crystal displays (LCDs) and are proposed to be a new generation of environmentally persistent, bioaccumulative and toxic (PBT) substances that are increasingly detected in rivers and seas. However, there is a lack of in vivo data that characterize adverse responses and toxic mechanisms of LCMs on aquatic organisms. The aim of this study was to comprehensively investigate the effect of four typical LCMs on the lethality, growth, molting, and reproductive capacity of Daphnia magna (D. magna), a highly studied aquatic species in environmental toxicology. Whole body and enzymatic biomarkers (i.e., body length, chitobiase, acetylcholinesterase, antioxidant defense) were measured to assess the toxicity of LCMs. The 48 h mortality rate and observations of disrupted thorax development and inhibition of ecdysis indicate that D. magna are sensitive to LCMs exposure. Oxidative stress, impaired neurotransmission, and disruptions in molting were observed in short-term biomarker tests using LCMs. A 21 day exposure of D. magna to LCMs resulted in reduced growth, reproduction, and population intrinsic growth rate. In addition, chitobiase and 20-hydroxyecdysone, enzymes important for the molting process, were altered at 7, 14 and 21 d. This is hypothesized to be related to endocrine imbalance resulting from LCM exposure. Based on molecular docking simulations, there is evidence that LCMs bind directly to ecdysteroid receptors; this may explain the observed endocrine disrupting effects of LCMs. These data support the hypothesis that LCMs are endocrine disrupting chemicals in aquatic species, impacting the process of molting. This may subsequently lead to lower reproduction and unbalanced population dynamics.

Sous vide can keep the nutritional properties and improve taste of food compared with other conventional methods. In addition, this method may reduce the risk of recontamination after cooking and during storage. The purpose of this paper was to study the effects of four cooking methods (steaming, microwaving, baking, and frying) on the sensory and digestibility on sturgeon steak pretreated by sous vide during the cold storage (0-25 days). The results showed that the digestibility of steaming and microwaving groups (range from 80.34% to 90.12%) significantly higher than that of the other treatment groups (p < .05); however, the overall acceptability of the two groups was lower. What more, the frying group has the highest acceptability and the lowest digestibility (range from 65.12% to 70.89%). The springiness (4.12-6.56 mm) and chewiness (1.75-3.12 mm) of the frying group were significantly higher than those of the other treatment groups, which was consistent with the results of scanning electron microscopy (SEM) that frying treatment group has a denser structure and smaller pores. With the prolonged refrigeration time, especially between 15 and 25 days, the volatile flavor components (nitrogen oxide, methane, and alcohol) and stagnant water (T21) were significantly decreased. Principal component analysis showed that the moisture content was the main factor affecting the overall acceptability and best consumption time of the sturgeon was within 15 days. Simulating the effects of home cooking conditions and refrigeration storage time on the quality of sturgeon steak provided a reference for consumers using similar products.

Acetochlor is a high-volume herbicide used on a global scale and toxicity assessments are needed to define its potential for adverse effects in wildlife and humans. This study was conducted to determine the effects of acetochlor on human liver carcinoma cells (HepG2), a cell model widely used to assess the potential for chemical hepatotoxicity. Experiments were conducted at concentrations ranging 0-800 µM acetochlor over a 12 to 48h period to quantify underlying mechanisms of toxicity. Our data indicate that acetochlor suppressed HepG2 cell proliferation in both a concentration- and time-dependent manner. Acetochlor induced reactive oxygen species (ROS) generation more than 700% with exposure to 400 µM acetochlor, and acetochlor decreased the activities and levels of anti-oxidant responses (superoxide dismutase, glutathione) following exposure to 100 µM, 200 µM and 400 µM acetochlor. Acetochlor also (1) induced HepG2 cell damage through apoptotic-signaling pathways; (2) enhanced intracellular free Ca2+ concentration (>400%); (3) decreased mitochondrial transmembrane potential (∼77%), and reduced ATP levels (∼65%) following exposure to 400 µM acetochlor compared to untreated cells. Notably, cell cycle progression was blocked at G0/G1 phase in HepG2 cells when treated for 24 h with 400 µM acetochlor. Taken together, acetochlor induced significant cytotoxicity toward HepG2 cells, and the underlying toxicity mechanisms appear to be related to ROS generation, mitochondrial dysfunction and disruption in the cell cycle regulation. These data contribute to toxicity assessments for acetochlor, a high-use herbicide, to quantify risk to wildlife and human health.

The objective of this study was to determine whether toxicity in mouse fibroblast cells (3T3 cells) could predict toxicity in mice. Synthesized data on toxicity was subjected to regression analysis and it was observed that relationship of toxicities between mice and 3T3 cells was not strong (R2 = 0.41). Inclusion of molecular descriptors (e.g. ionization, pKa) improved the regression to R2 = 0.56, indicating that this relationship is influenced by kinetic processes of chemicals or specific toxic mechanisms associated to the compounds. However, to determine if we were able to discriminate modes of action (MOAs) in mice using the toxicities generated from 3T3 cells, compounds were first classified into "baseline" and "reactive" guided by the toxic ratio (TR) for each compound in mice. Sequence, binomial and recursive partitioning analyses provided strong predictions of MOAs in mice based upon toxicities in 3T3 cells. The correct classification of MOAs based on these methods was 86%. Nearly all the baseline compounds predicted from toxicities in 3T3 cells were identified as baseline compounds from the TR in mice. The incorrect assignment of MOAs for some compounds is hypothesized to be due to experimental uncertainty that exists in toxicity assays for both mice and 3T3 cells. Conversely, lack of assignment can also arise because some reactive compounds have MOAs that are different in mice compared to 3T3 cells. The methods developed here are novel and contribute to efforts to reduce animal numbers in toxicity tests that are used to evaluate risks associated with organic pollutants in the environment.

PURPOSE: The environmental issues caused by the municipal solid waste disposal are becoming a worldwide concern. METHODS: We studied the situations both domestically and abroad by the strategic environmental assessment (SEA) approach and also conducted comprehensive evaluations of garbage disposal in Changchun City. RESULTS: On the basis of this study, we found that SEA is of great importance in the municipal solid waste disposal. Moreover, with the rapid socioeconomic development of Changchun City, municipal solid waste production increases on an annual basis, and thus, good waste management planning is of great significance. CONCLUSIONS: Considering the situation of the economic development of Changchun City, garbage disposal was handled mainly in the major sanitary landfills with appropriate use of incineration technology. This plan is environmentally friendly at a relatively high degree and has met the requirements of minimum investment. It also takes into account the requirements of the development of incineration technology. Regarding environmental pollution in terms of groundwater pollution and atmospheric pollution, this plan is a feasible one by meeting various requirements with low environmental impact among the three plans discussed in this study.

The concentrations of eight heavy metals (Cu, Zn, Pb, Cd, Cr, Ni, Hg, and As) in the intertidal surface sediments from Quanzhou Bay were determined to evaluate their levels and spatial distribution due to urbanization and economic development of Quanzhou region, southeast China. The ranges of the measured concentrations in the sediments are as follows: 24.8-119.7 mg/kg for Cu, 105.5-241.9 mg/kg for Zn, 34.3-100.9 mg/kg for Pb, 0.28-0.89 mg/kg for Cd, 51.1-121.7 mg/kg for Cr, 16.1-45.7 mg/kg for Ni, 0.17-0.74 mg/kg for Hg, and 17.7-30.2 mg/kg for As. The overall average concentrations of above metals exceed the primary standard criteria but meet the secondary standard criteria of the Chinese National Standard of Marine Sediment Quality. Several contents of Cu and Hg exceed the secondary standard criteria at some stations. The results of geoaccumulation index (Igeo) show that Cd causes strong pollution in most of the study area. There are no significant correlations among most of these heavy metals, indicating they have different anthropogenic and natural sources. Some locations present severe pollution by heavy metals depending on the sources, of which sewage outlets, aquatic breeding, and commercial ports are the main sources of contaminants to the area.
