Mercury has become a ubiquitous hazardous element even ending up in pristine areas such as the Arctic, where it biomagnifies and leaves especially top predators vulnerable to potential health effects. Here we investigate total mercury (THg) concentrations and dietary proxies for trophic position and habitat foraging (δ15N and δ13C, respectively) in fur of 30 Arctic wolves collected during 1869-1998 in the Canadian High Arctic and Greenland. Fur THg concentrations (mean ± SD) of 1.46 ± 1.39 µg g -1 dry weight are within the range of earlier reported values for other Arctic terrestrial species. Based on putative thresholds for Hg-mediated toxic health effects, the studied Arctic wolves have most likely not been at compromised health. Dietary proxies show high dietary plasticity among Arctic wolves deriving nutrition from both marine and terrestrial food sources at various trophic positions. Variability in THg concentrations seem to be related to the wolves' trophic position rather than to different carbon sources or regional differences (East Greenland, the Foxe Basin and Baffin Bay area, respectively). Although the present study remains limited due to the scarce, yet unique historic study material and small sample size, it provides novel information on temporal and spatial variation in Hg pollution of remote Arctic species.

Antimicrobials, such as fungicides and antibiotics, pose a risk for microbial decomposers (i.e., bacteria and aquatic fungi) and invertebrate detritivores (i.e., shredders) that play a pivotal role in the ecosystem function of leaf litter breakdown. Although waterborne toxicity and diet-related effects (i.e., dietary exposure and microorganism-mediated alterations in food quality for shredders) of fungicides and antibiotics on decomposer-detritivore systems have been increasingly documented, their joint effect is unknown. We therefore assessed waterborne and dietary effects of an antimicrobial mixture consisting of the fungicide azoxystrobin (AZO) and the antibiotic ciprofloxacin (CIP) on microbial decomposers and the shredder Gammarus fossarum using a tiered approach. We compared effect sizes measured in the present study with model predictions (i.e., independent action) based on published data. During a 7-day feeding activity assay quantifying waterborne toxicity in G. fossarum, the leaf consumption of gammarids was reduced by ∼60 % compared to the control when subjected to the mixture at concentrations of each component causing a 20 % reduction in the same response variable when applied individually. Moreover, the selective feeding of gammarids during the food choice assay indicated alterations in food quality induced by the antimicrobial mixture. The food selection and, in addition, the decrease in microbial leaf decomposition is likely linked to changes in leaf-associated bacteria and fungi. During a long-term assay, energy processing, growth and energy reserves of gammarids were increased in presence of 15 and 500 µg/L of AZO and CIP, respectively, through the dietary pathway. These physiological responses were probably driven by CIP-induced alterations in the gut microbiome or immune system of gammarids. In general, model predictions matched observed effects caused by waterborne exposure on the leaf consumption, energy processing and growth of gammarids during short- and long-term assays, respectively. However, when complex horizontal (bacteria and aquatic fungi) and vertical (leaf-associated microorganisms and shredders) interactions were involved, model predictions partly over- or underestimated mixture effects. Therefore, the present study identifies uncertainties of mixture effect predictions for complex biological systems calling for studies targeting the underlying processes and mechanisms.

Naturally occurring radionuclides, in particular, polonium-210 (210Po), have a greater contribution than anthropogenic radionuclides to the annual effective dose received by the general public due to consumption of seafood. Knowledge of potential trophic sources and transfer of 210Po to seabird species and subsequently to the Greenlandic people is, however, still poor. Here, we assess the transfer of 210Po and 210Pb to seabirds sampled during autumn and winter 2017 and 2018 in Greenland and provide a dose assessment. The activity concentrations of 210Po in muscle and liver, respectively, ranged from 0.2 ± 0.1 Bq kg-1 w.w. in glaucous gull (Larus hyperboreus) to 21.2 ± 22.6 Bq kg-1 w.w. in thick-billed murre (Uria lomvia) and from 32.0 ± 9.4 Bq kg-1 w.w. in common eider (Somateria mollissima) to 40.5 ± 49.0 Bq kg-1 w.w. in thick-billed murre. 210Po was non-uniformly distributed in the body of thick-billed murre. Kidneys and feathers showed higher 210Po activity concentrations than heart and bone. The 210Po/210Pb activity concentration ratios are higher than unity, indicating that 210Po is preferentially taken up by seabirds compared to its progenitor 210Pb. The derived annual absorbed dose from 210Po to the whole body of thick-billed murre was 6.4 × 102 ± 3.0 × 102 µGy. The annual effective dose to the average adult and representative person in Greenland due to ingestion of 210Po in seabirds was estimated to 13.0 µSv and 57.0 µSv, respectively. This derived dose is low and poses a slight risk, and risk communication is therefore deemed unnecessary.

Systemic neonicotinoid insecticides are increasingly used as a crop protection measure to suppress insect pests on trees. However, senescent foliage falling from treated trees represents a rarely studied pathway through which neonicotinoids may enter nontarget environments, e.g., surface waters. To estimate risk posed by this pathway, neonicotinoid residues were analyzed in foliage from black alder trees treated with one of three neonicotinoid insecticides (imidacloprid, thiacloprid, or acetamiprid) at five concentrations, each ranging from 0.0375-9.6 g active ingredient/cm trunk diameter at breast height (n = 3). Foliar residues measured at the time of leaf fall were used as input parameters for a model predicting imidacloprid water concentrations over a 100-m-long stream stretch as a consequence of remobilization from introduced foliage (input: 600 g foliage/m2 containing 80 µg imidacloprid/g). The water concentration (up to ∼250 ng/L) predicted by the model exceeded the recently proposed Maximum Permissible Concentration of 8.3 ng/L for ∼6.5 days. Moreover, dietary uptake was identified as an additional exposure route for aquatic organisms. The alternative pathway (i.e., introduction via leaf fall) and exposure route (i.e., dietary uptake) associated with the systemic nature of neonicotinoids should be accounted for during their registration process in order to safeguard ecosystem integrity.

We argued recently that the positive predictive value (PPV) and the negative predictive value (NPV) are valuable metrics to include during null hypothesis significance testing: They inform the researcher about the probability of statistically significant and non-significant test outcomes actually being true. Although commonly misunderstood, a reported p value estimates only the probability of obtaining the results or more extreme results if the null hypothesis of no effect was true. Calculations of the more informative PPV and NPV require a priori estimate of the probability (R). The present document discusses challenges of estimating R.

The level of protection provided by the present environmental risk assessment (ERA) of fungicides in the European Union for fungi is unknown. Therefore, we assessed the structural and functional implications of five fungicides with different modes of action (azoxystrobin, carbendazim, cyprodinil, quinoxyfen, and tebuconazole) individually and in mixture on communities of aquatic hyphomycetes. This is a polyphyletic group of fungi containing key drivers in the breakdown of leaf litter, governing both microbial leaf decomposition and the palatability of leaves for leaf-shredding macroinvertebrates. All fungicides impaired leaf palatability to the leaf-shredder Gammarus fossarum and caused structural changes in fungal communities. In addition, all compounds except for quinoxyfen altered microbial leaf decomposition. Our results suggest that the European Union's first-tier ERA provides sufficient protection for the tested fungicides, with the exception of tebuconazole and the mixture, while higher-tier ERA does not provide an adequate level of protection for fungicides in general. Therefore, our results show the need to incorporate aquatic fungi as well as their functions into ERA testing schemes to safeguard the integrity of aquatic ecosystems.

Although generally misunderstood, the p value is the probability of the test results or more extreme results given H0 is true: it is not the probability of H0 being true given the results. To obtain directly useful insight about H0, the positive predictive value (PPV) and the negative predictive value (NPV) may be useful extensions of null hypothesis significance testing (NHST). They provide information about the probability of statistically significant and non-significant test outcomes being true based on an a priori defined biologically meaningful effect size. The present study explores the utility of PPV and NPV in an ecotoxicological context by using the frequently applied Daphnia magna reproduction test (OECD guideline 211) and the chemical stressor lindane as a model system. The results indicate that especially the NPV deviates meaningfully between a test design strictly following the guideline and an experimental procedure controlling for α and ß at the level of 0.05. Consequently, PPV and NPV may be useful supplements to NHST that inform the researcher about the level of confidence warranted by both statistically significant and non-significant test results. This approach also reinforces the value of considering α, ß, and a biologically meaningful effect size a priori.
