RESUMO

The human population will be approximately 9.7 billion by 2050, and food security has been identified as one of the key issues facing the global population. Agrochemicals are an important tool available to farmers that enable high crop yields and continued access to healthy foods, but the average new agrochemical active ingredient takes more than ten years, 350 million dollars, and 20,000 animals to develop and register. The time, monetary, and animal costs incentivize the use of New Approach Methodologies (NAMs) in early-stage screening to prioritize chemical candidates. This review outlines NAMs that are currently available or can be adapted for use in early-stage screening agrochemical programs. It covers new in vitro screens that are on the horizon in key areas of regulatory concern. Overall, early-stage screening with NAMs enables the prioritization of development for agrochemicals without human and environmental health concerns through a more directed, agile, and iterative development program before animal-based regulatory testing is even considered.

Nanotechnology is a rapidly developing discipline that has the potential to transform the way we approach problems in a variety of fields, including agriculture. The use of nanotechnology in sustainable agriculture has gained popularity in recent years. It has various applications in agriculture, such as the development of nanoscale materials and devices to boost agricultural productivity, enhance food quality and safety, improve the efficiency of water and nutrient usage, and reduce environmental pollution. Nanotechnology has proven to be very beneficial in this field, particularly in the development of nanoscale delivery systems for agrochemicals such as pesticides, fertilizers, and growth regulators. These nanoscale delivery technologies offer various benefits over conventional delivery systems, including better penetration and distribution, enhanced efficacy, and lower environmental impact. Encapsulating agrochemicals in nanoscale particles enables direct delivery to the targeted site in the plant, thereby reducing waste and minimizing off-target effects. Plants are fundamental building blocks of all ecosystems and evaluating the interaction between nanoparticles (NPs) and plants is a crucial aspect of risk assessment. This critical review therefore aims to provide an overview of the latest advances regarding the positive and negative effects of nanotechnology in agriculture. It also explores potential future research directions focused on ensuring the safe utilization of NPs in this field, which could lead to sustainable development. © 2024 Society of Chemical Industry.

In the modern fast-paced lifestyle, time-efficient and nutritionally rich foods like corn and oat have gained popularity for their amino acids and antioxidant contents. The increasing demand for these cereals necessitates higher production which leads to dependency on agrochemicals, which can pose health risks through residual present in the plant products. To first report the phytotoxicity for corn and oat, our study employs QSAR, quantitative Read-Across and quantitative RASAR (q-RASAR). All developed QSAR and q-RASAR models were equally robust (R2 = 0.680-0.762, Q2Loo = 0.593-0.693, Q2F1 = 0.680-0.860) and find their superiority in either oat or corn model, respectively, based on MAE criteria. AD and PRI had been performed which confirm the reliability and predictability of the models. The mechanistic interpretation reveals that the symmetrical arrangement of electronegative atoms and polar groups directly influences the toxicity of compounds. The final phytotoxicity and prioritization are performed by the consensus approach which results into selection of 15 most toxic compounds for both species.

The two-test in vitro battery for genotoxicity testing (Ames and micronucleus) has in the majority of cases replaced the three-test battery (as two-test plus mammalian cell gene mutation assay) for the routine testing of chemicals, pharmaceuticals, cosmetics, and agrochemical metabolites originating from food and feed as well as from water treatment. The guidance for testing agrochemical groundwater metabolites, however, still relies on the three-test battery. Data collated in this study from 18 plant protection and related materials highlights the disparity between the often negative Ames and in vitro chromosome aberration data and frequently positive in vitro mammalian cell gene mutation assays. Sixteen of the 18 collated materials with complete datasets were Ames negative, and overall had negative outcomes in in vitro chromosome damage tests (weight of evidence from multiple tests). Mammalian cell gene mutation assays (HPRT and/or mouse lymphoma assay (MLA)) were positive in at least one test for every material with this data. Where both MLA and HPRT tests were performed on the same material, the HPRT seemed to give fewer positive responses. In vivo follow-up tests included combinations of comet assays, unscheduled DNA synthesis, and transgenic rodent gene mutation assays, all gave negative outcomes. The inclusion of mammalian cell gene mutation assays in a three-test battery for groundwater metabolites is therefore not justified and leads to unnecessary in vivo follow-up testing.

A utilização de agrotóxicos ao redor do mundo é elevada, estimada em mais 2 milhões de toneladas e somente o continente americano emprega 1,2 milhão de toneladas de produtos. O Brasil possui na agricultura a sua principal atividade econômica e utilizou em 2021 aproximadamente 700 mil toneladas de agrotóxicos. O tomate é um dos vegetais mais cultivados e consumidos no mundo e o Brasil figura como o 10º maior produtor. O consumo anual médio de tomate do brasileiro é de 4,2 kg, é um vegetal nutritivo e com propriedades associadas à prevenção de câncer. O tomate é uma das culturas com maior uso de agrotóxicos e durante o período desse estudo, era autorizado o uso de 123 agrotóxicos. Os resíduos desses produtos nos alimentos podem acarretar em diversos problemas à saúde, mesmo em curta exposição (< 24h). Desse modo, uma das maneiras de garantir a segurança alimentar da população é a realização da avaliação de risco de contaminação dietética. No processo de validação 73 ingredientes ativos respeitaram os critérios do protocolo adotado. Para a realização da estimativa de risco de exposição dietética aguda, foram coletadas 30 amostras de tomate in natura e 11 de tomate pelado, entre setembro de 2021 e março de 2022. Para a extração dos compostos de interesse foi utilizado o método QuEChERS e para avaliação dos resíduos, a cromatografia líquida acoplada à espectrometria de massas. Das amostras de tomate in natura, somente seis (20 %) estavam isentas dos compostos pesquisados; 18 (60 %) apresentaram resíduo(s) abaixo do limite (LMR) estabelecido e seis amostras (20 %) foram consideradas impróprias ao consumo. Das amostras de tomate pelado, três estavam isentas dos agrotóxicos pesquisados e oito (72 %) apresentaram resíduo de carbendazim abaixo do LMR. Nenhuma das amostras mostrou potencial de contaminação aguda por agrotóxicos, apesar disso não é possível afirmar que não há risco, pois não há como estimar os potenciais efeitos adversos provenientes do consumo de um alimento com múltiplos compostos.


More than 2 million tons of pesticide products are used annually through the world and only the America continent was used 1,2 million tons of these products. The agriculture is the main economic activity in Brazil, this way in 2021 approximately 700 thousand tons of pesticide were applied in its crops. Tomato is one of the most cultivated and consumed vegetables in the world and Brazil is the 10th largest producer. The Brazilian people consumes an average of 4.2 kg of tomatoes by year, it is a nutritious vegetable with anti-cancer properties. Tomato crop is one of the which highest pesticide usages, during this study 123 compounds were permitted for this crop. Pesticide residues in food may causes several health problems, even in short-term exposition (< 24h), thus one of the ways to ensure the food safety to population is performing the dietary contamination risk assessment. In the validation process 73 active ingredients were within the established criteria. To perform the acute risk assessment of dietary exposure, were collected samples of: fresh tomato (30) and whole peeled tomato (11) in between September of 2021 and March of 2022. To extraction of interest substances was used the QuEChERS method and to residue evaluation the liquid chromatography coupled to mass spectrometry. Of the fresh tomato samples, only six (20 %) were free of searched analytes; 18 (60 %) showed residue(s) bellow the established limits (MRL) and six (20 %) were considered unfit to consumption. Of the whole peeled tomato samples, three were free of searched substances and eight (72 %) showed residue of carbendazim bellow the MRL. None of the samples showed potential for acute contamination by pesticide, however it is not possible to say that there is no risk, as there is no way to estimate the potential adverse effects on health arising from the consumption of a food with several compounds.

Toxicological assessments of newly developed agrochemical agents consider chemical modifications and their metabolic and biotransformation products. To carry out an in silico hazard assessment, understanding the type of chemical modification and its location on the original compound can greatly enhance the reliability of the evaluation. Here, we present and apply a method based on liquid chromatography-mass spectrometry (LC-MS) enhanced with infrared ion spectroscopy (IRIS) to better delineate the molecular structures of transformation products before in silico toxicology evaluation. IRIS facilitates the recording of IR spectra directly in the mass spectrometer for features selected by retention time and mass-to-charge ratio. By utilizing quantum-chemically predicted IR spectra for candidate molecular structures, one can either derive the actual structure or significantly reduce the number of (isomeric) candidate structures. This approach can assist in making informed decisions. We apply this method to a plant growth stimulant, digeraniol sinapoyl malate (DGSM), that is currently under development. Incubation of the compound in Caco-2 and HepaRG cell lines in multiwell plates and analysis by LC-MS reveals oxidation, glucuronidation, and sulfonation metabolic products, whose structures were elucidated by IRIS and used as input for an in silico toxicology assessment. The toxicity of isomeric metabolites predicted by in silico tools was also assessed, which revealed that assigning the right metabolite structure is an important step in the overall toxicity assessment of the agrochemical. We believe this identification approach can be advantageous when specific isomers are significantly more hazardous than others and can help better understand metabolic pathways.

The use of agrochemical and pharmaceutical active ingredients is essential in our modern society. Given the increased concern and awareness of the potential risks of some chemicals, there is a growing need to align with 'green chemistry' and 'safe and sustainable by design' principles and thus to evaluate the hazards of agrochemical and pharmaceutical active ingredients in early stages of R&D. We give an overview of the current challenges and opportunities to assess the principle of biodegradability in the environment. Development of new medium/high-throughput methodologies, combining predictive tools and wet-lab experimentation are essential to design biodegradable chemicals early in the active ingredient discovery and selection process.

In contrast to genotoxic carcinogens, there are currently no internationally agreed upon regulatory tools for identifying non-genotoxic carcinogens of human relevance. The rodent cancer bioassay is only used in certain regulatory sectors and is criticized for its limited predictive power for human cancer risk. Cancer is due to genetic errors occurring in single cells. The risk of cancer is higher when there is an increase in the number of errors per replication (genotoxic agents) or in the number of replications (cell proliferation-inducing agents). The default regulatory approach for genotoxic agents whereby no threshold is set is reasonably conservative. However, non-genotoxic carcinogens cannot be regulated in the same way since increased cell proliferation has a clear threshold. An integrated approach for the testing and assessment (IATA) of non-genotoxic carcinogens is under development at the OECD, considering learnings from the regulatory assessment of data-rich substances such as agrochemicals. The aim is to achieve an endorsed IATA that predicts human cancer better than the rodent cancer bioassay, using methodologies that equally or better protect human health and are superior from the view of animal welfare/efficiency. This paper describes the technical opportunities available to assess cell proliferation as the central gateway of an IATA for non-genotoxic carcinogenicity.

The increased use of agrochemicals raises concerns about environmental, animal, and mainly human toxicology. The development of New Approach Methodologies (NAMs) for toxicological risk assessment including new in vitro tests and in silico protocols is encouraged. Although agrochemical mutagenicity testing is well established, a complementary alternative approach may contribute to increasing reliability, with the consequent reduction of false-positive results that lead to unnecessary use of animals in follow-up in vivo testing. Additionally, it is unreasonable to underestimate the phototoxic effects of an accidental dermal exposure to agrochemicals during agricultural work or domestic application in the absence of adequate personal protection equipment, especially in terms of photomutagenicity. In this scenario, we addressed the integration of in vitro and in silico techniques as NAMs to assess the mutagenic and phototoxic potential of agrochemicals. In the present study we used the yno1 S. cerevisiae strain as a biomodel for in vitro assessment of agrochemical mutagenicity, both in the absence and in the presence of simulated sunlight. In parallel, in silico predictions were performed using a combination of expert rule-based and statistical-based models to assess gene mutations and phototoxicity. None of the tested agrochemicals showed mutagenic potential in the two proposed approaches. The Gly and 2,4D herbicides were photomutagenic in the in vitro yeast test despite the negative in silico prediction of phototoxicity. Herein, we demonstrated a novel experimental approach combining both in silico and in vitro experiments to address the complementary investigation of the phototoxicity and (photo)mutagenicity of agrochemicals. These findings shed light on the importance of investigating and reconsidering the photosafety assessment of these products, using not only photocytotoxicity assays but also photomutagenicity assays, which should be encouraged.

Periurban agriculture in Argentina is carried out by farmers without adequate control. The indiscriminate agrochemical application for productivity improvement negatively impacts the environment. The objective of this work was to test the quality of periurban agricultural soils by performing bioassays with Eisenia andrei as an indicator. Soils belonging to two orchards with intensive production (S: strawberry/broccoli crop plot and G: tomato/pepper crop greenhouse - Moreno District, Buenos Aires, Argentina) were sampled during 2015 and 2016. As subcellular biomarkers, cholinesterases (ChE), carboxylesterases (CaE), and glutathione-S-transferases (GST) activities were analysed in E. andrei (7-day exposure). While no effect on ChE activities was observed, CaE activities were significantly reduced 18% (S-2016 soil). GST activities were increased 35% and 30% by S-2016 and G-2016, respectively. CaE decrease together with GST increase could be indicative of a negative disturbance. Concerning whole organism biomarkers, reproduction (56-day exposure), avoidance (3-day exposure), and feeding activity (bait-lamina test, 3-day exposure) were analysed. A reduced cocoons' viability (50%), hatchability (55%), accompanied by a low number of juveniles (50%) were observed in all cases. Additionally, the earthworms exhibited significant avoidance responses to S-2015, S-2016 and G-2016 whereas G-2015 soil induced migration. No significant effect on the feeding activity was registered in any case. Most of the E. andrei biomarkers tested could constitute an early warning of harmful effects produced by polluted periurban soils, even if the agrochemical treatment applied remains unknown. The results reveal the need to develop an action plan to avoid further deterioration of the productive soil.

[RESUMO]. O objetivo deste estudo descritivo e exploratório, de base qualitativa, foi identificar as resoluções do MERCOSUL sobre resíduos de agrotóxicos em alimentos publicadas entre 1991 e 2022, analisando os processos de harmo- nização regional desses marcos e sua incorporação ao arcabouço regulatório dos Estados Partes fundadores do bloco (Argentina, Brasil, Paraguai e Uruguai). A análise identificou pontos importantes para a regulação e o monitoramento de resíduos de agrotóxicos em alimentos no MERCOSUL, como as sinonímias utilizadas na definição de agrotóxicos e a abrangência do sistema regulatório de cada país, as marcadas diferenças no alcance dos principais marcos regulatórios nacionais, a incorporação desigual de regulamentos internacionais e regionais pelos Estados Partes e os desafios para a harmonização da legislação sobre resíduos de agrotóxicos em alimentos no âmbito do MERCOSUL. Para além dos limitados avanços observados na tentativa de harmo- nizar a legislação pertinente dentro do bloco, observa-se a necessidade de avançar, nacional e regionalmente, nos processos regulatórios sobre resíduos de agrotóxicos em alimentos, garantindo a qualidade dos produtos e serviços ofertados à população e fortalecendo, assim, um comércio de alimentos mais seguros e produzidos a partir de processos menos prejudiciais ao ambiente no MERCOSUL.


[ABSTRACT]. The objective of this descriptive and exploratory study with a qualitative basis was to identify MERCOSUR resolutions on pesticide residues in food issued between 1991 and 2022, analyzing the regional harmoni- zation processes of these milestones and their incorporation into the regulatory framework of MERCOSUR founding Member States (Argentina, Brazil, Paraguay, and Uruguay). The analysis identified important points for the regulation and monitoring of pesticide residues in food in MERCOSUR, such as the synonyms used in the definition of pesticides and the scope of the regulatory system in each country, the marked differences in the scope of the main national regulatory frameworks, the unequal incorporation of international and regional regulations by the Member States and the challenges for harmonizing legislation on pesticide residues in food within MERCOSUR. In addition to the limited advances observed in the attempt to harmonize the relevant legislation within the bloc, there is a need to advance, nationally and regionally, in the processes to regulate pesticide residues in food, so as to ensure the quality of the products and services provided to the population and also to strengthen safer agro/food trade that relies on processes that are less harmful to the environment.

El objetivo de este estudio descriptivo y exploratorio, de base cualitativa, fue identificar las resoluciones del MERCOSUR sobre la gestión de residuos de productos agrotóxicos en los alimentos publicadas entre los años 1991 y 2022 y analizar los procesos de armonización regional de esos marcos y su incorporación al marco regulatorio de los Estados Partes fundadores del bloque (Argentina, Brasil, Paraguay y Uruguay). En el análisis se identificaron puntos importantes para la regulación y el seguimiento de la gestión de residuos de productos agrotóxicos en los alimentos en el MERCOSUR, tales como los sinónimos utilizados en la definición de productos agrotóxicos y la amplitud del sistema regulatorio de cada país, las diferencias más destacadas en el alcance de los principales marcos regulatorios nacionales, la desigualdad de la incorporación de los reglamentos regionales e internacionales por parte de los Estados Partes y los desafíos para la armonización de la legislación sobre residuos de productos agrotóxicos en los alimentos en el ámbito del MERCOSUR. Además de los limitados avances observados en el intento de armonizar la legislación pertinente dentro del bloque, existe la necesidad de avanzar, a nivel nacional y regional, en los procesos regulatorios sobre la gestión de residuos de productos agrotóxicos en los alimentos, asegurando la calidad de los productos y ser- vicios ofrecidos a la población y fortaleciendo así el comercio de alimentos más inocuos producidos a partir de procesos menos nocivos para el medioambiente en el MERCOSUR.

The rodent cancer bioassay has been the standard approach to fulfill regulatory requirements for assessing human carcinogenic potential of agrochemicals, food additives, industrial chemicals, and pharmaceuticals. Decades of research have described the limitations of the rodent cancer bioassay leading to international initiatives to seek alternatives and establish approaches that modernize carcinogenicity assessment. Biologically relevant approaches can provide mechanistic information and increased efficiency for evaluating hazard and risk of chemical carcinogenicity to humans. The application of human-relevant mechanistic understanding to support new approaches to carcinogenicity assessment will be invaluable for regulatory decision-making. The present work outlines the challenges and opportunities that authorities should consider as they come together to build a roadmap that leads to global acceptance and incorporation of fit-for-purpose, scientifically defensible new approaches for human-relevant carcinogenicity assessment of agrochemicals.

Este estudo propõe analisar as relações e os processos de subjetivação de mulheres quebradeiras de coco babaçu decorrentes das intervenções de políticas desenvolvimentistas em seus territórios de vida e reverberações no Movimento Interestadual de Quebradeiras de Coco Babaçu (MIQCB). Sob a perspectiva ético-estético-política da Cartografia, acompanhamos as narrativas das histórias de vida de 24 mulheres, suas atividades cotidianas e eventos do MIQCB, também analisamos os documentos das políticas. Entendemos que, ao passo que tais políticas de desenvolvimento rural contribuem para a melhoria das condições de vida, em termos materiais e simbólicos, elas também produzem ressonâncias relacionadas ao modo de subjetivação do tipo "empresário de si", que agenciam seus modos de viver, de produzir e de se relacionar consigo e com os outros na lógica capitalista neoliberal. A resistência às capturas neoliberais também estão presentes ao ampliarem as mobilizações coletivas do próprio movimento, articulando com outros na produção de um "comum".(AU)


This study proposes to analyze the relations and the processes of subjectivation of babassu coconut-breaker women arising from developmental policy interventions in their territories of life and reverberations in the Babassu Coconut-breaker Interstate Movement (MIQCB). From the ethical-aesthetic-political perspective of Cartography, we followed the narratives of the life stories of 24 women, their daily activities and promoted events by MIQCB, we also analyzed the policy documents. We understand that while these policies of rural development contribute to improve the living conditions, in material and symbolic terms, they also produce resonances related to the "self-entrepreneur" mode of subjectivation, which has been handling their ways of living, producing, and relating to themselves and others in the neoliberal capitalist logic. Resistance to neoliberal captures is also present as they expand the collective mobilizations of the movement itself, articulating with others, in the production of a "common."(AU)


Este estudio tiene como objetivo analizar los procesos de subjetivación de las mujeres que rompen coco babaçu que surgen de las intervenciones de las políticas de desarrollo en sus territorios de vida y las reverberaciones en el Movimiento Interestadual de las Mujeres que Rompen Coco Babaçu (MIQCB). Desde la perspectiva ético-estético-política de la Cartografía, seguimos las narraciones de las historias de vida de 24 mujeres, sus actividades diarias y eventos del MIQCB, y también analizamos los documentos de las políticas. Si bien estas políticas han contribuido a mejorar las condiciones de vida de las mujeres, en términos materiales y simbólicos, también han producido resonancias del modo de subjetivación "autoempresarial", que ha agenciado sus formas de vivir, producir y relacionarse consigo mismas y con los demás en la lógica capitalista neoliberal. La resistencia a las capturas neoliberales también está presente cuando amplían las movilizaciones colectivas del propio movimiento, articulándose con otros en la producción de un "común".(AU)

A luta pela terra e seu uso incide na forma como as subjetividades têm sido produzidas no Brasil. No contexto capitalista, patriarcal e machista, vive-se um processo de exploração da mão de obra de mulheres e de recursos naturais da Terra. Este estudo objetivou conhecer histórias de vida de mulheres agricultoras rurais que participam de movimentos sociais agroecológicos no Rio Grande do Sul. Participaram do estudo três mulheres agricultoras, com idades entre 21 e 53 anos, residentes e trabalhadoras em zonas rurais das cidades de Viamão e Rolante, escolhidas por conveniência. Os instrumentos utilizados foram: questionário sociodemográfico, entrevista de história de vida e observação participante, os quais foram, posteriormente, submetidos à análise temática. Os resultados demonstram que as histórias de vida das mulheres relatadas são marcadas por lutas e formas de resistência. Para além das situações de conflitos e falta de recursos, avistam-se questões próprias de gênero, como a invisibilidade feminina e a desigualdade no acesso à terra por mulheres. A construção de base e o fortalecimento do papel político das mulheres nos movimentos sociais permite inferir que existem melhorias significativas nas desigualdades e injustiças no meio rural. Contudo, pontua-se a necessidade de se promover o diálogo entre os movimentos sociais, as mulheres e a sociedade política sobre os modelos atuais de políticas públicas existentes, possibilitando, assim, avançar nas discussões a respeito da promoção da equidade de gênero nos espaços rurais, bem como potencializar o avanço das práticas agroecológicas em direção à superação do capitalismo.(AU)


The struggle for land and its use affects the way subjectivities have been produced in Brazil. In the capitalist, patriarchal, and male-chauvinist context, women's labor and the Earth's natural resources are strongly exploited. This study aimed to know the life stories of female farmers who participate in agroecological social movements in the state of Rio Grande do Sul. Three female farmers, aged from 21 to 53, residents and workers in rural areas of the municipalities of Viamão and Rolante, chosen by convenience, participated in the study. The instruments used were: sociodemographic questionnaire, a life story interview, and participant observation, which were later subjected to thematic analysis. The results show that the participants' life stories are marked by struggles and means of resistance. In addition to conflicts and lack of resources, there are specific gender issues, such as female invisibility and inequality in women's access to land. The grassroots construction and the strengthening of the political role of women in social movements allow us to infer that there are significant improvements in inequalities and injustices in rural areas. However, it is necessary to promote dialogue between social movements, women, and political society about the current models of existing public policies, thus making it possible to advance in discussions about gender equity in rural spaces, as well as to enhance the advancement of agroecological practices to overcoming capitalism.(AU)


La lucha por la tierra y su uso afecta a la forma en que se han producido las subjetividades en Brasil. En el contexto capitalista, patriarcal y machista, hay un proceso de explotación del trabajo de las mujeres y de los recursos naturales de la tierra. Este estudio tuvo como objetivo conocer las historias de vida de agricultoras rurales que participan en movimientos sociales agroecológicos en Rio Grande do Sul (Brasil). Participaron en el estudio tres agricultoras, de entre 21 y 53 años, residentes y trabajadoras en áreas rurales de las ciudades de Viamão y Rolante, elegidas por conveniencia. Los instrumentos utilizados fueron: cuestionario sociodemográfico, entrevista de historia de vida y observación participante, cuyos datos posteriormente se sometieron a análisis temático. Los resultados muestran que las historias de vida de las mujeres relatadas están marcadas por luchas y formas de resistencia. A las situaciones de conflicto y falta de recursos se suman cuestiones específicas de género, como la invisibilidad femenina y la desigualdad en el acceso de las mujeres a la tierra. La construcción popular y el fortalecimiento del papel político de las mujeres en los movimientos sociales permiten inferir que hay mejoras significativas en las desigualdades e injusticias en las zonas rurales. Sin embargo, es necesario promover el diálogo entre los movimientos sociales, las mujeres y la sociedad sobre los modelos actuales de las políticas públicas existentes, para posibilitar avances en las discusiones sobre la promoción de la equidad de género en los espacios rurales y potenciar el avance de las prácticas agroecológicas hacia la superación del capitalismo.(AU)

Conventional agricultural practices favoring the use of glyphosate-based herbicides (GBHs) increase the risk of GBH residues ending up in animal feed, feces, and, eventually, manure. The use of poultry manure as organic fertilizer in the circular food economy increases the unintentional introduction of GBH residues into horticultural and agricultural systems, with reportedly negative effects on the growth and reproduction of crop plants. To understand the potential lasting effects of exposure to GBH residues via organic manure fertilizers, we studied strawberry (Fragaria x vescana) plant performance, yield quantity, biochemistry, folivory, phytochemistry, and soil elemental composition the year after exposure to GBH. Although plants exposed to GBH residues via manure fertilizer were, on average, 23% smaller in the year of exposure, they were able to compensate for their growth during the following growing season. Interestingly, GBH residue exposure in the previous growing season led to a trend in altered plant size preferences of folivores during the following growing season. Furthermore, the plants that had been exposed to GBH residues in the previous growing season produced 20% heavier fruits with an altered composition of phenolic compounds compared to non-exposed plants. Our results indicate that GBHs introduced via manure fertilizer following circular economy practices in one year can have effects on perennial crop plants in the following year, although GBH residues in soil have largely vanished.

Trace and potentially toxic elements represent one class of food contaminants that has stimulated research. In markets, two main methods of growing vegetables are generally available: conventional and organic. Conventional farming has been the target of some concerns about the use of agrochemicals, especially the excessive use of pesticides, whereas organic agriculture minimizes the use of agrochemicals. As the main route for potentially toxic elements' absorption by humans is by food intake, it is important to evaluate if the method of cultivation influences their concentrations. This study evaluated the levels of potentially toxic elements and nutrients on four leafy vegetables: curly lettuce, collard greens, escarole, and rocket, cultivated by conventional and organic farming. We found that Al, Ba, Fe, and Sr levels were higher in conventional samples, whereas K, Pb, and Zn were higher in organic. Amongst the elements analysed, values of Fe, Al, and K were around 0.2, 0.3, and 70 g kg-1, respectively, except in collard greens, in which the values were lower. On the other hand, Ba, Sr, and Mn presented higher concentration in collard greens compared to the other vegetables in conventional cultivation (~ 35, 80, and 120 mg kg-1, respectively). The principal component analysis result shows that the samples were grouped according to the type of vegetable, regardless of the type of cultivation. Despite this, the evaluation of the cultivation by different types of farming is important in order to choose the healthiest option.

Phototoxicity testing is required by European regulations for agrochemicals with UV/visible molar extinction/absorption coefficient (MEC) higher than 10 L x mol-1 x cm-1 in the 290-700 nm wavelength range. Furthermore, regulations identify a need of considering human exposure in case of positive results. While in vitro OECD test guidelines are available for hazard characterisation, there is no guidance on how to utilise positive results in human exposure risk assessments. Our goal was to take a first step towards developing a NAM based tiered testing approach and a framework for non-dietary acute human dermal risk assessment for phototoxicity to agrochemicals. The proposed framework can be divided into a few steps: 1) use the OECD updated MEC values of 1000 L x mol-1 x cm-1 as trigger for phototoxicity testing; 2) establish a reference concentration (RfC) from in vitro phototoxicity studies using BMC approach, 3) estimate potential exposure to skin, target organ for phototoxicity, using EFSA exposure models, product specific labels and skin penetration values, and 4) phototoxicity risk assessment; 5) refinement to RfC and/or exposure estimates can be considered. Finally, case studies of a nematicide and an herbicide active substance are provided to illustrate the proposed framework.

The Central Andes of Peru are a region of great concern regarding pesticide risk to the health of local communities. Therefore, we conducted an observational study to assess the level of pesticide contamination among Andean people. Analytical chemistry methods were used to measure the concentrations of 170 pesticide-related compounds in hair samples from 50 adult Andean subjects living in rural and urban areas. As part of the study, a questionnaire was administered to the subjects to collect information regarding factors that increase the risk of pesticide exposure. Our results indicate that Andean people are strongly exposed to agrochemicals, being contaminated with a wide array of pesticide-related compounds at high concentration levels. Multivariate analyses and geostatistical modeling identified sociodemographic factors associated with rurality and food origin that increase pesticide exposure risk. The present study represents the first comprehensive investigation of pesticide-related compounds detected in body samples collected from people living in the Central Andes of Peru. Our findings pinpoint an alarming environmental situation that threatens human health in the region and provide a rationale for improving public policies to protect local communities.

European agricultural development in the 21st century will be affected by a host of global changes, including climate change, changes in agricultural technologies and practices, and a shift towards a circular economy. The type and quantity of chemicals used, emitted, and cycled through agricultural systems in Europe will change, driven by shifts in the use patterns of pesticides, veterinary pharmaceuticals, reclaimed wastewater used for irrigation, and biosolids. Climate change will also impact the chemical persistence, fate, and transport processes that dictate environmental exposure. Here, we review the literature to identify research that will enable scenario-based forecasting of environmental exposures to organic chemicals in European agriculture under global change. Enabling exposure forecasts requires understanding current and possible future 1.) emissions, 2.) persistence and transformation, and 3.) fate and transport of agricultural chemicals. We discuss current knowledge in these three areas, the impact global change drivers may have on them, and we identify knowledge and data gaps that must be overcome to enable predictive scenario-based forecasts of environmental exposure under global change. Key research gaps identified are: improved understanding of relationships between global change and chemical emissions in agricultural settings; better understanding of environment-microbe interactions in the context of chemical degradation under future conditions; and better methods for downscaling climate change-driven intense precipitation events for chemical fate and transport modelling. We introduce a set of narrative Agricultural Chemical Exposure (ACE) scenarios - augmenting the IPCC's Shared Socio-economic Pathways (SSPs) - as a framework for forecasting chemical exposure in European agriculture. The proposed ACE scenarios cover a plausible range of optimistic to pessimistic 21st century development pathways. Filling the knowledge and data gaps identified within this study and using the ACE scenario approach for chemical exposure forecasting will support stakeholder planning and regulatory intervention strategies to ensure European agricultural practices develop in a sustainable manner.

A field survey was conducted on five fish farms to trace glyphosate and malathion pollution with some physicochemical parameters. A precise half-life time, LC50-96h, of these agrochemicals on Oreochromis niloticus, as well as chronic exposure with organic selenium (OS) supplementation, were experimentally investigated. Oreochromis niloticus was subjected to the following: (negative control); (2 mg L-1 glyphosate); (0.5 mg L-1 malathion); (glyphosate 1.6 mg L-1 and 0.3 mg L-1 malathion); (glyphosate 2 mg L-1 and OS 0.8 g kg-1 diet); (malathion 0.5 mg L-1 and OS 0.8 g kg-1 diet) and (glyphosate 1.6 mg L-1; malathion 0.3 mg L-1 and OS 0.8 g kg-1 diet). Furthermore, data from the analyzed pond revealed a medium risk quotient (RQ) for both agrochemicals. The detected agrochemicals were related to their application, and vegetation type surrounding the farms, also their biodegradation was correlated to water pH, temperature, and salinity. Glyphosate and malathion had half-lives of 2.8 and 2.3 days and LC50-96h of 2.331 and 0.738 mg L-1, respectively. The severest nervous symptoms; increased oxidative stress markers, as well as high bacterial count in the livers and kidneys of fish challenged with Aeromonas hydrophila, were observed in the combined exposure, followed by a single exposure to malathion and then glyphosate. Organic selenium mitigated these impacts.
