The Valparaiso region in Chile was decreed a zone affected by catastrophe in 2019 as a consequence of one of the driest seasons of the last 50 years. In this study, three varieties ('Alfa-INIA', 'California-INIA', and one landrace, 'Local Navidad') of kabuli-type chickpea seeds produced in 2018 (control) and 2019 (climate-related catastrophe, hereafter named water stress) were evaluated for their grain yield. Furthermore, the flavonoid profile of both free and esterified phenolic extracts was determined using liquid chromatography-mass spectrometry, and the concentration of the main flavonoid, biochanin A, was determined using liquid chromatography with diode array detection. The grain yield was decreased by up to 25 times in 2019. The concentration of biochanin A was up to 3.2 times higher in samples from the second season (water stress). This study demonstrates that water stress induces biosynthesis of biochanin A. However, positive changes in the biochanin A concentration are overshadowed by negative changes in the grain yield. Therefore, water stress, which may be worsened by climate change in the upcoming years, may jeopardize both the production of chickpeas and the supply of biochanin A, a bioactive compound that can be used to produce dietary supplements and/or nutraceuticals.

Chickpea (CP), a legume of the family Fabaceae, is an important nutrient-rich food providing protein, essential amino acids, vitamins, dietary fibre, and minerals. Unfortunately, several IgE-binding proteins in CP have been detected that are responsible for allergic manifestations in sensitized population. Therefore, the prevalence of CP induced allergy prompted us towards purification, characterization and allergenicity assessment of a major ∼26kDa protein from chickpea crude protein extract (CP-CPE). Purification of CP 26kDa protein was done using a combination of fractionation and anion exchange chromatography. This protein was further characterized as "Chain A, crystal structure of a plant albumin" from Cicer arietinum with Mol wt 25.8kDa by Liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. Further, allergenic potential of purified 25.8kDa protein was assessed using in vivo and in vitro model. Purified protein showed IgE-binding capacity with sensitized BALB/c mice and CP allergic patient's sera. Enhanced levels of specific and total IgE, MCP-1, MCPT-1, myeloperoxidase, histamine, prostaglandin D2, and cysteinyl leukotriene were found in sera of mice treated with CP ∼26kDa protein. Further, expressions of Th2 cytokines (i.e. IL-4, IL-5, IL-13), transcription factors (i.e. GATA-3, STAT-6, SOCS-3) and mast cell signaling proteins (Lyn, cFgr, Syk, PLC-γ2, PI-3K, PKC) were also found increased at mRNA and protein levels in the intestines of mice treated with CP ∼26kDa protein. In addition, enhanced release of ß-hexosaminidase, histamine, cysteinyl leukotriene and prostaglandin D2 were observed in RBL2H3 cell line when treated (125µg) with CP 26kDa protein. Conclusively, in vivo and in vitro studies revealed the allergenic potential of purified CP 26kDa protein. Being a potential allergen, plant albumin may play a pivotal role in CP induced allergenicity. Current study will be helpful for better development of therapeutic approaches to prevent the allergenicity in CP sensitized individuals.

Chemical investigation of Cicer microphyllum resulted in the isolation and characterization of eight natural products viz. Stigmasterol, Oleanolic acid-3-acetate, Oleanolic acid, Biochanin A, Genistein, Pratensein, Chrysoeriol, and Luteolin. Herein, we report a novel, accurate, and cost-effective high-performance thin-layer chromatography method for the simultaneous quantification of the isolated natural products on silica-gel 60F254 plates using the solvent system n-hexane/ethyl acetate/formic acid (9.0:6.5:0.8, v/v/v). Natural products were quantified after postchromatographic derivatization with ceric ammonium sulfate. The method was validated as per the International Conference on Harmonization guidelines. All calibration curves showed a good linear relationship (r > 0.9943) within the test range. Precision was assessed by intra- and interday tests with relative standard deviations <1.82%, accuracy validation recovery 98.38-99.57% with relative standard deviations <1.00%. On quantification, Pratensein was a major constituent (0.921%). The screening for cytotoxic activity using a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay resulted into identification of Luteolin as potent molecule with IC50 3.5 and 25.6 µg/mL against murine melanoma and human epidermoid carcinoma cell lines, respectively.

Chickpea is grain legumes grown mainly in areas with temperate and semiarid climate. It is characterized by a high content of protein, fat, vitamins, fiber, and a lower content of carbohydrates than flour of wheat. Chickpeas may contain antinutritional compounds that can impair utilization of the nutrients by people. Heat treatment is an effective method to increase the amount of protein available for intestinal digestibility. Adding chickpeas to a foodstuff can increase their nutritional value and reduce the acrylamide content. Acrylamide is an antinutritional substance present in foods, such as bread, snacks, and chips. Chickpea flour and protein may be new way to a reduce the content of acrylamide in products of this type. The addition of chickpea flour affects the sensory and textural properties.

Dissipation of ß-cyfluthrin and imidacloprid in chickpea pods and leaves was measured following three applications of Solomon 300 OD (ß-cyfluthrin 9 percent +imidacloprid 21 percent) at 200 and 400mLha(-1). Residues of ß-cyfluthrin on chickpea pods and leaves were found to be below its limit of quantification (LOQ) of 0.01mgkg(-1) after 7 days at both the dosages. Similarly, imidacloprid residues were found to be below its LOQ of 0.01mgkg(-1) at 10 days. Half-life periods on chickpea pods and leaves for ß-cyfluthrin were found to be 1.06 and 0.58 days, whereas for imidacloprid these values were observed to be 2.07 and 1.75 days at recommended dose. As the theoretical maximum residue contributions on chickpea pods and leaves are found to be less than the maximum permissible intake values even on 0 day, therefore consumer health risks are minimal at both the dosages on chickpea.

AIM: Chickpea (Cicer arietinum L) is a traditional Uighur herb. In this study we investigated the estrogenic activities of the isoflavones extracted from chickpea sprouts (ICS) in ovariectomized rats. METHODS: Ten-week-old virgin Sprague-Dawley female rats were ovariectomized (OVX). The rats were administered via intragastric gavage 3 different doses of ICS (20, 50, or 100 mg·kg(-1)·d(-1)) for 5 weeks. Their uterine weight and serum levels of 17ß-estradiol (E2), follicle stimulating hormone (FSH) and luteinizing hormone (LH) were measured. The epithelial height, number of glands in the uterus, and number of osteoclasts in the femur were histologically quantified, and the expression of proliferating cell nuclear antigen (PCNA) was assessed immunohistochemically. Bone structural parameters, including bone mineral density (BMD), bone volume/tissue volume (BV/TV), trabecular thickness (Tb.Th) and trabecular separation (Tb.Sp) were measured using Micro-CT scanning. RESULTS: Treatments of OVX rats with ICS (50 or 100 mg·kg(-1)·d(-1)) produced significant estrogenic effects on the uteruses, including the increases in uterine weight, epithelial height and gland number, as well as in the expression of the cell proliferation marker PCNA. The treatments changed the secretory profile of ovarian hormones and pituitary gonadotropins: serum E2 level was significantly increased, while serum LH and FSH levels were decreased compared with the vehicle-treated OVX rats. Furthermore, the treatments significantly attenuated the bone loss, increased BMD, BV/TV and Tb.Th and decreased Tb.Sp and the number of osteoclasts. Treatment of OVX rats with the positive control drug E2 (0.25 mg·kg(-1)·d(-1)) produced similar, but more prominent effects. CONCLUSION: ICS exhibits moderate estrogenic activities as compared to E2 in ovariectomized rats, suggesting the potential use of ICS for the treatment of menopausal symptoms and osteoporosis caused by estrogen deficiency.

Ochratoxin A content in 100 grain and derived products were determined by high-performance liquid chromatography with immunoaffinity column clean-up and fluorometric detection. Ochratoxin A was detected in 32% of green gram, 13.3% of chickpea, 10% of lentil and 17.5% of wheat flour. Ochratoxin contamination was below the regulatory limits of the European Union and of Iran. Recovery was 97% and the limit of detection was 0.12 ng g⁻¹.

Fusarium diseases cause major economic losses in wheat-based crop rotations. Volatile organic compounds (VOC) in wheat and rotation crops, such as chickpea, may negatively impact pathogenic Fusarium. Using the headspace GC-MS method, 16 VOC were found in greenhouse-grown wheat leaves: dimethylamine, 2-methyl-1-propanol, octanoic acid-ethyl ester, acetic acid, 2-ethyl-1-hexanol, nonanoic acid-ethyl ester, nonanol, N-ethyl-benzenamine, naphthalene, butylated hydroxytoluene, dimethoxy methane, phenol, 3-methyl-phenol, 3,4-dimethoxy-phenol, 2,4-bis (1,1-dimethyethyl)-phenol, and 1,4,7,10,13,16-hexaoxacyclooctadecane; and 10 VOC in field-grown chickpea leaves: ethanol, 1-penten-3-ol, 1-hexanol, cis-3-hexen-1-ol, trans-2-hexen-1-ol, trans-2-hexenal, 3-methyl-1-butanol, 3-hydroxy-2-butanone, 3-methyl-benzaldehyde and naphthalene. Also found was 1-penten-3-ol in chickpea roots and in the root nodules of two of the three cultivars tested. Chickpea VOC production pattern was related (P=0.023) to Ascochyta blight severity, suggesting that 1-penten-3-ol and cis-3-hexen-1-ol were induced by Ascochyta rabiei. Bioassays conducted in Petri plates established that chickpea-produced VOC used in isolation were generally more potent against Fusarium graminearum and Fusarium avenaceum than wheat-produced VOC, except for 2-ethyl-1-hexanol, which was rare in wheat and toxic to both Fusarium and tetraploid wheat. Whereas exposure to 1-penten-3-ol and 2-methyl-1-propanol could suppress radial growth by over 50% and octanoic acid-ethyl ester, nonanol, and nonanoic acid-ethyl ester had only weak effects, F. graminearum and F. avenaceum growth was completely inhibited by exposure to trans-2-hexenal, trans-2-hexen-1-ol, cis-3-hexen-1-ol, and 1-hexanol. Among these VOC, trans-2-hexenal and 1-hexanol protected wheat seedlings against F. avenaceum and F. graminearum, respectively, in a controlled condition experiment. Genetic variation in the production of 2-ethyl-1-hexanol, a potent VOC produced in low amount by wheat, suggests the possibility of selecting Fusarium resistance in wheat on the basis of leaf VOC concentration. Results also suggests that the level of Fusarium inoculum in chickpea-wheat rotation systems may be reduced by growing chickpea genotypes with high root and shoot levels of trans-2-hexen-1-ol and 1-hexanol.
