Fish from the pike (Esox) genus are valued in gastronomy for their superior meat quality. However, they can cause allergic reactions in sensitive consumers. This work aimed to fill the gap in the detection of pike allergens using molecular-biological techniques. New, fast, and accurate loop-mediated isothermal amplification (LAMP) and real-time PCR (qPCR) assays were designed to detect pike DNA using the parvalbumin gene as a marker. LAMP was assessed by electrophoresis, SYBR green optical detection, and real-time fluorescence detection. The latter was the most sensitive, detecting as little as 0.78 ng of pike DNA; the qPCR detection limit was 0.1 ng. The LAMP analysis took 20-70 min, which is significantly faster than qPCR. The study provides reliable detection and quantification of the parvalbumin gene in both fresh and processed samples and further highlights the versatility of the use of the parvalbumin gene for the authentication of food products and consumer protection via refined allergen risk assessment that is independent of the type of tissue or food processing method used.

There is a pressing need for more-holistic approaches to fisheries assessments along with growing demand to reduce the health impacts of sample collections. Metabolomic tools enable the use of sample matrices that can be collected with minimal impact on the organism (e.g., blood, urine, and mucus) and provide high-throughput, untargeted biochemical information without the requirement of a sequenced genome. These qualities make metabolomics ideal for monitoring a wide range of fish species, particularly those under protected status. In the current study, we surveyed the relative abundances of 120 endogenous metabolites in epidermal mucus across eight freshwater fish species belonging to seven phylogenetic orders. Principal component analysis was used to provide an overview of the data set, revealing strong interspecies relationships in the epidermal mucous metabolome. Normalized relative abundances of individual endogenous metabolites were then used to identify commonalities across multiple species, as well as those metabolites that showed notable species specificity. For example, taurine was measured in high relative abundance in the epidermal mucus of common carp (Cyprinus carpio), northern pike (Esox lucius), golden shiner (Notemigonus crysoleucas), rainbow trout (Oncorhynchus mykiss), and rainbow smelt (Osmerus mordax), whereas γ-amino butyric acid (GABA) exhibited a uniquely high relative abundance in flathead catfish (Pylodictis olivaris). Finally, hierarchical cluster analysis was used to evaluate species relatedness as characterized by both the epidermal mucous metabolome (phenotype) and genetic phylogeny (genotype). This comparison revealed species for which relatedness in the epidermal mucous metabolome composition closely aligns with phylogenetic relatedness (e.g., N. crysoleucas and C. carpio), as well as species for which these two measures are not well aligned (e.g., P. olivaris and Polyodon spathula). These, and other findings reported here, highlight novel areas for future research with fish, including development of epidermal mucous-based markers for non-invasive health monitoring, sex determination, and hypoxia tolerance.

Viral hemorrhagic septicemia virus (VHSV) is an ongoing cause of disease and mortality in freshwater fishes across the Great Lakes region of the Midwestern United States. Antibody detection assays such as enzyme-linked immunosorbent assay (ELISA) are nonlethal serological methods that can have significantly shorter turnaround times than the current validated viral detection diagnostic methodology for VHSV: cell culture with confirmation by polymerase chain reaction (PCR). This study evaluated an ELISA that detects nonneutralizing antinucleocapsid antibodies to VHSV in Northern Pike Esox lucius. Juvenile Northern Pike were experimentally infected with VHSV by intraperitoneal injection. The infected fish were monitored for 12 weeks for signs of disease, and weekly serum samples were obtained. An analysis of the survival data showed that mortality occurred significantly more quickly in inoculated fish than in control fish. Fish that were infected by injection showed a significant increase in antibody response by 2 weeks postinfection. However, variation in the rate and pattern of antibody response among the infected fish was high at any given point. The optimum window for detecting antibodies in Northern Pike is 2-12 weeks postinfection, which generally follows the median time to appearance of clinical signs (21 d postinfection). The receiver-operating characteristic curve analysis showed the ELISA to have a sensitivity of 80.5% and a specificity of 63.2% in Northern Pike, but these values can be adjusted by choosing different percent inhibition cutoffs, which may facilitate the use of the test for specific management goals. The results of this study offer insights into the disease progression and immune kinetics of VHSV, including interindividual variation, which will aid in the management of this economically important virus.

Industrial activities conducted in Northern Canada have raised concerns among Indigenous communities regarding wildlife contamination and potential consequences for human health. Therefore, an investigation on the chemical (metals/metalloids) and biological (parasite) burden of adult walleye (Sandervitreus) and northern pike (Esoxlucius) from Montreal Lake, Saskatchewan, was conducted to assess health risks related to fish consumption. Dissection revealed that both fishes displayed typical parasite communities, with Eubothrium sp. (Cestoda) and Raphidascarisacus (Nematoda) occurring the most frequently. None of the identified parasite species were infectious to humans. Concentrations of most inorganic contaminants in fish muscle were low and both walleye and pike can be considered healthy components of a balanced diet. However, due to slightly elevated mercury concentrations, excessive daily consumption of these fishes is not recommended, as mercury exposure over time may lead to adverse health effects.

Lakes receiving effluent from the Key Lake uranium mill in northern Saskatchewan contain elevated trace metals, some of which are associated with increased reactive oxygen species (ROS) in cells and tissues causing oxidative stress. The potential for oxidative stress was assessed in juvenile (age 1+) northern pike (Esox lucius) collected from two exposure (high and low) and one reference lake near the Key Lake operation. The concentrations of total, reduced and oxidized glutathione and the ratio of oxidized to reduced glutathione in liver and kidney did not differ significantly among pike collected from exposure and reference lakes, with the exception of low exposure pike kidney that had significantly greater oxidized glutathione and ratio of oxidized to reduced glutathione. The concentrations of by-products of lipid peroxidation (malondialdehyde and 4-hydroxyalkenal) were significantly greater in kidney of pike collected from the reference lake compared to both exposure lakes. The activity of the antioxidant enzyme glutathione peroxidase in liver was greater in pike collected from the high exposure lake compared to the reference lake. Histopathological evaluations revealed greater pathology in reference lake pike as indicated by a greater number of pyknotic and fragmented nuclei and dilated tubules as well as a thickening of Bowman's capsule in kidney, and as a thickening of the primary filament epithelial padding in gills. In liver, hepatocyte morphology, including transsectional area and degree of vacuolation, differed among lakes without any clear signs of pathology. Trace metal analyses of muscle showed that eight elements (arsenic, cobalt, copper, iron, molybdenum, selenium, thallium, and uranium) were significantly elevated in pike collected from both exposure lakes compared to reference. These results provide only limited evidence of oxidative stress in exposure pike tissues and no evidence of histopathology despite indications that trace metals, most notably arsenic and selenium, were bioaccumulating in tissue.

Uranium mining and milling operations in northern Saskatchewan (Canada) release effluents with elevated levels of certain trace metals and metalloids, including selenium. The goal of the present study was to evaluate the presence of selenium-induced deformities in northern pike (Esox lucius) and white sucker (Catostomus commersoni) larvae originating from adults collected downstream of a uranium mine. Eggs were fertilized in the field and incubated in the laboratory following a two-way (crossover) analysis-of-variance experimental design to discriminate effects from maternal transfer versus those from exposure to site water in the developing embryos. Selenium concentrations in northern pike and white sucker eggs (8.02 and 4.89 microg/g dry wt, respectively; mean +/- standard error throughout) from the exposure site were approximately two- to threefold higher than reference (2.35 +/- 0.20 and 1.94 +/- 0.25 microg/g dry wt, respectively). Among all evaluated deformities (skeletal curvatures, craniofacial deformities, fin deformities, and edema), only edema in white sucker fry from the exposure site was slightly elevated ( approximately 3%) compared to reference. The occurrence of edema, however, can be associated with factors other than selenium (e.g., other metals and organic compounds). Both fish species displayed strong linear relationships between the selenium concentrations in eggs and other tissues (muscle, liver, kidney, and bone), suggesting that selenium concentrations in eggs could be predicted from selenium concentrations in adult tissues. The lack of a clear, toxic response in the present study is in agreement with selenium thresholds for early life-stage deformities reported in other studies, with egg selenium concentrations in northern pike and white sucker collected at the exposure site being less than the 10 microg/g (dry wt) threshold associated with the presence of deformities.

Sediments contaminated by various sources of mercury (Hg) were studied at 8 sites in Sweden covering wide ranges of climate, salinity, and sediment types. At all sites, biota (plankton, sediment living organisms, and fish) showed enhanced concentrations of Hg relative to corresponding organisms at nearby reference sites. The key process determining the risk at these sites is the net transformation of inorganic Hg to the highly toxic and bioavailable methylmercury (MeHg). Accordingly, Hg concentrations in Perca fluviatilis were more strongly correlated to MeHg (p < 0.05) than to inorganic Hg concentrations in the sediments. At all sites, except one, concentrations of inorganic Hg (2-55 microg g(-1)) in sediments were significantly, positively correlated to the concentration of MeHg (4-90 ng g(-1)). The MeHg/Hg ratio (which is assumed to reflect the net production of MeHg normalized to the Hg concentration) varied widely among sites. The highest MeHg/Hg ratios were encountered in loose-fiber sediments situated in southern freshwaters, and the lowest ratios were found in brackish-water sediments and firm, minerogenic sediments at the northernmost freshwater site. This pattern may be explained by an increased MeHg production by methylating bacteria with increasing temperature, availability of energy-rich organic matter (which is correlated with primary production), and availability of neutral Hg sulfides in the sediment pore waters. These factors therefore need to be considered when the risk associated with Hg-contaminated sediments is assessed.

This article introduces a new general method for genealogical inference that samples independent genealogical histories using importance sampling (IS) and then samples other parameters with Markov chain Monte Carlo (MCMC). It is then possible to more easily utilize the advantages of importance sampling in a fully Bayesian framework. The method is applied to the problem of estimating recent changes in effective population size from temporally spaced gene frequency data. The method gives the posterior distribution of effective population size at the time of the oldest sample and at the time of the most recent sample, assuming a model of exponential growth or decline during the interval. The effect of changes in number of alleles, number of loci, and sample size on the accuracy of the method is described using test simulations, and it is concluded that these have an approximately equivalent effect. The method is used on three example data sets and problems in interpreting the posterior densities are highlighted and discussed.

We developed an empirical Bayes procedure to estimate genetic distances between populations using allele frequencies. This procedure makes it possible to describe the skewness of the genetic distance while taking full account of the uncertainty of the sample allele frequencies. Dirichlet priors of the allele frequencies are specified, and the posterior distributions of the various composite parameters are obtained by Monte Carlo simulation. To avoid overdependence on subjective priors, we adopt a hierarchical model and estimate hyperparameters by maximizing the joint marginal-likelihood function. Taking advantage of the empirical Bayesian procedure, we extend the method to estimate the effective population size using temporal changes in allele frequencies. The method is applied to data sets on red sea bream, herring, northern pike, and ayu broodstock. It is shown that overdispersion overestimates the genetic distance and underestimates the effective population size, if it is not taken into account during the analysis. The joint marginal-likelihood function also estimates the rate of gene flow into island populations.
