RESUMO

The major lipids and antioxidant activities of Asterias rolleston gonad lipids were evaluated systematically. Major lipids of A. Rolleston gonad lipids were triacylglycerols (TAGs) and phospholipids (PLs). Total lipids were composed of 15.62% of polyunsaturated fatty acids (PUFAs), and 40.81% of monounsaturated fatty acids (MUFAs). The most abundant PUFA were C20:5n-3 (EPA) (6.28%) and C22:6n-3 (DHA) (5.80%). Predominantly composed of phosphatidylcholine (PC) and phosphatidylethanolamine (PE), polar lipids were rich in PUFAs and could contain up to 34.59% EPA and DHA, and PE and PI (phosphatidylinositol) were also found to be the main carriers of EPA and ARA (arachidonic acid) in polar lipids. The MUFA and PUFA of Sn-2 in TAG are 39.72% and 30.37%, respectively. A total of 64 TAG species were identified, with Eo-P-M, Eo-Eo-M, and M-M-Eo being the main TAGs components. Moreover, A. rollestoni gonad lipids exhibited potent radical scavenging activities and reducing power in a dose-dependent manner.

There is limited information on fluoride toxicity and risk overview on ecotoxicological risks to aquatic invertebrate populations particularly molluscan taxa. This necessitated the assessment of toxicity responses in the freshwater snail, Bellamya bengalensis exposed to environmentally relevant concentrations of sodium fluoride. Under lethal exposures (150, 200, 250, 300, 400 and 450 mg/l), the median lethal concentrations (LC50) were determined to be 422.36, 347.10, 333.33 and 273.24 mg/l for B. bengalensis at 24, 48, 72 and 96 h respectively. The rate of mortality of the snails was increased significantly with elevated concentrations of the toxicant. The magnitude of toxicity i.e., toxicity factor at different time scale was also higher with increased exposure duration. Altered behavioural changes i.e., crawling movement, tentacle movement, clumping tendency, touch reflex and mucous secretion in exposed snail with elevated concentrations and exposure duration. Similarly, oxygen consumption rate of the treated snail also lowered significantly during 72 and 96 h of exposure. Under 30-day chronic exposures (Control-0.00 mg/L; T1-27.324 mg/L; T2-54.648 mg/L), protein concentrations in gonad and hepatopancreas of exposure groups was significantly lowered. Chronic exposures also revealed lowered haemocytes counts in exposure groups. The potential for loss of coordination, respiratory distress and physiological disruption in organisms exposed to environmentally relevant concentrations of fluoride was demonstrated by this study. The estimation and magnitude of toxicity responses are necessary for a more accurate estimation of ecological risks to molluscan taxa and invertebrate populations under acute and chronic fluoride exposures in the wild.

RESUMO

HIGHLIGHTS: All heavy metals, except Hg, were well accumulated in liver compared with muscle and gonads. Estimated daily intakes of heavy metals were within 2% of the provisional maximum tolerable daily intakes, Hazard quotients of heavy metals were less than 1.0. Appropriate intake control of G. chalcogrammus is necessary to protect human health in the future.

The aim of the present work was to extend our knowledge on the variability of trace metals in sea urchin tissues, focusing on seasonal fluctuations (2010 February for "winter," May for "spring," August for "summer," November for "autumn") in the three different sampling sites of Algerian west coast (Oran Harbor (S1), Ain Defla (S2), and Hadjaj (belonging to Mostaganem City S3)). For this purpose, the bioavailability (metal indices) and bioaccumulation (metal concentrations in soft tissues) of heavy metals (Zn, Cu, Pb, and Cd), the physiological characteristics (e.g., biological indices such as condition index (CI), repletion index (RI), gonad index (GI)), and the biometric parameters (diameter (D) and the height (H)) of sea urchins Paracentrotus lividus were assessed and related to seasons and sites. To investigate the metal bioavailability to sea urchins more precisely, the metal indices were used as a reliable tool in the present work, instead of the metal concentrations only. The interest to standardize metal concentrations with the weight of the urchin test is to overcome the metal burden variations in the soft tissues of urchin related to the seasonal weight changes of the soft body of animal. We evidence that the most contaminated sites were S1 and S2. Furthermore, it should be noted that the bioavailability of metals, corresponding to the values of metal indices, is also more pronounced in S1 and S2 compared to that measured in S3. Thus, a correlation is observed between seasonal metal content in urchin tissues from the three sites and the corresponding metal indices. The high metal concentrations were obtained during the period when RI and CI were highest. So, it appears that the bioaccumulation of metals in sea urchins of the three sites studied is significantly influenced by the reproductive cycle and diet, feeding activity, and physiological state of these organisms. We noticed that the sea urchins from the sites S1 and S2 were small in size. It is probable that these animals, whose internal tissues contained high concentrations of metals, have been exposed to metal pollution, which might have affected both their growth and altered their physiological capacity. This approach is very original and might be used in the monitoring of the quality of coastal waters, and the present work provided a useful data set for Mediterranean monitoring network.

In the present study, concentrations of arsenic (As), cadmium (Cd), cobalt (Co), chromium (Cr), copper (Cu), manganese (Mn), nickel (Ni), lead (Pb), and zinc (Zn) were measured in the muscle, gill, and gonads of the pelagic fish species Trachurus mediterraneus, Engraulis encrasicolus ponticus, and Sprattus sprattus that are important both commercially and for the ecosystems in the Black Sea. The samples were collected during 2011. The metals were determined using inductively coupled plasma mass spectrometry (ICP-MS) following an acid digestion. The highest concentrations of As, Cd, Co, Cr, Cu, Pb, and Zn were found in E. encrasicolus ponticus, whereas the greatest concentrations of Ni were found in T. mediterraneus and Mn in S. sprattus. Results showed that average metal concentrations in the tissues of T. mediterraneus, E. encrasicolus ponticus, and S. sprattus decreased in the order gill > gonad > muscle, gonad > gill > muscle, and gill > gonad > muscle, respectively, for the three species. When metal concentrations of fish tissues were compared between fish gender, there were only statistical differences in the gonads of the studied fish species (p < 0.05). The present study demonstrated that the metals have different correlations with condition factor (CF) and gonadosomatic index (GSI) of the fish species. Cr showed statistically important positive correlation to the GSI in male T. mediterraneus. Co showed statistically important positive correlation to CF in female E. encrasicolus ponticus, and also Co and Cd showed correlation to CF in male T. mediterraneus. Cd concentrations in the muscle tissues of the fish species were above the maximum acceptable concentration for human consumption.

The objective of this research was to utilize the QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe) method to extract a broad range of persistent organic pollutants from sturgeon organs (liver and gonad) as indicators of exposure. The analyte list was prioritized to include carcinogenic polyaromatic hydrocarbons (PAHs), the most commonly occurring polybrominated diphenyl ethers (PBDEs) and polychlorinated biphenyls (PCBs), persistent bioaccumulative and toxic chemicals (PBTs), and emergent contaminants of concern (ECCs) as indicators of human sewage exposure. White sturgeon (Acipenser transmontanus) were selected for this study to support a larger ecotoxicological study to monitor contaminants as an assessment of fish health. Organ tissues contained high lipid content with percentages of 15% and 34% for liver and gonad, respectively. Overall recoveries from fortified sturgeon tissues were high, 71-98% for PAHs, 60-107% for PBDEs and PCBs, 86-107% for PBT chemicals, and 88-107% for ECCs with the exception of octinoxate (28%) from liver tissues. Analyte recovery trends decreased as analyte lipophilicity and molecular weight increased. These recoveries demonstrate that extraction using QuEChERS can be used for screening of the most common bioaccumulating organic compounds in high lipid fish tissue using a single extraction and analysis.

Concentrations of Mn, Fe, Co, Ni, As, Se, Cd, Ag and Pb in scale, skin, muscle, gills, liver and the gonads of Gilthead seabream (Sparus aurata), European seabass (Dicentrarchus labrax), Black Sea salmon (Salmo trutta labrax), Horse mackerel (Trachurus trachurus), Red mullet (Mullus barbatus) and Whiting (Merlangius merlangius euxinus) from the Black Sea, in Turkey, were investigated. Elemental analyses were performed with inductively coupled plasma-mass spectrometry after sample preparation by microwave digestion. Mean metal concentrations in different tissues were in the following ranges: Mn 0.09-23.1, Fe 0.58-326, Co 0.01-0.22, Ni 0.03-1.34, As 0.13-3.40, Se 0.13-4.42, Ag 0.01-0.18, Cd 0.32-6.25, Pb 0.02-0.38 mg kg⁻¹ wet weight. Metal concentrations in the muscles of the examined species were generally lower than those in scale, skin, gills, liver and the gonads. The described method was validated by analysis of Dogfish Liver-certified reference material, DOLT-4.

Current scientific advances in metal and metalloid risk assessment were applied to evaluate risk to aquatic and riparian wildlife species potentially impacted by residual coal fly ash after cleanup of an unprecedented large ash release into an aquatic environment-the first assessment of its kind. Risk was evaluated using multiple lines of evidence (LOE), including 1) tissue-based risk assessment of inorganic concentrations in piscivorous and insectivorous bird eggs and raccoon organs, 2) deterministic and probabilistic diet-based risk estimates for 10 receptors species, 3) raccoon health metrics, and 4) tree swallow nest productivity measures. Innovative approaches included use of tissue-based toxicity reference values (TRVs), adjustment of bioavailability in the dietary uptake models (using sequential metal extractions in sediment), partitioning chemical species into uptake compartments (e.g., prey gut, nongut, sediment), incorporating uncertainty in both modeled dose and dietary TRVs, matching TRVs to chemical forms of constituents, and pairing these LOEs with reproductive success or health status of sensitive receptor species. The weight of evidence revealed that risk to wildlife from residual ash was low and that risk, though low, was most pronounced for insectivorous birds from exposure to Se and As. This information contributes to the debate surrounding coal combustion residue regulations prompted by this ash release. Because of the responsible party's proactive approach of applying state-of-the-art methods to assess risk using several LOEs that produced consistent results, and because of their inclusion of the regulating agencies in decisions at every step of the process, the risk assessment results were accepted, and an effective approach toward cleanup protective of the environment was quickly implemented. This study highlights the value of using multiple LOEs and the latest scientific advances to assist in timely decision making to obtain an effective remedy for an emergency spill.

Persistent organic pollutants (POPs) in streamwater can sometimes exceed the guidelines values reported for biota and human protection in watersheds with intensive agriculture. Oxidative stress and cytotoxicity are some of the markers of exposure to POPs in fish. Accumulation of organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) as well as lipid peroxidation (LPO) was assessed in wild silverside (Odontesthes bonariensis) from maturation and pre-spawning stages sampled in a typical soybean growing area. Pollutants were quantified by gas chromatography with electron capture detection and LPO by the method of thiobarbituric acid reactive substances. Concentrations of POPs were in the following order: OCPs>PCBs>PBDEs in all organs and stages. Liver, gills and gonads had the highest OCP concentrations in both sexes and stages with a predominance of endosulfan in all samples. Matured individuals, sampled after endosulfan application period, showed higher endosulfan concentrations than pre-spawning individuals. The predominance of endosulfan sulfate could be due to direct uptake from diet and water column, as well as to the metabolism of the parent compounds in fish. The prevalence of p,p'-DDE in liver would also reflect both the direct uptake and the metabolic transformation of p,p'-DDT to p,p'-DDE by fish. The highest levels of PBDEs and PCBs were found in gills and brain of both stages of growth. The pattern BDE-47>BDE-100 in all samples corresponds to pentaBDE exposure. In the case of PCBs, penta (#101 and 110) and hexa-CB congeners (#153 and 138) dominated in the maturation stages and tri (#18) and tetra-CB (#44 and 52) in pre-spawning stages, suggesting biotransformation or preferential accumulation of heavier congeners during gonadal development. Differences in LPO levels in ovaries were associated with growth dilution and reproductive stage. Differences in LPO levels in gills were related with pesticide application periods. As a whole, endosulfan, a current-use pesticide, constituted the main pollutant found in wild silverside reflecting the intense agriculture activity in the study area. Moreover endosulfan was positively correlated with LPO.

BACKGROUND: The razor clam Sinonovacula constricta is a benthic intertidal bivalve species with important commercial value. Despite its economic importance, knowledge of its transcriptome is scarce. Next generation sequencing technologies offer rapid and efficient tools for generating large numbers of sequences, which can be used to characterize the transcriptome, to develop effective molecular markers and to identify genes associated with growth, a key breeding trait. RESULTS: Total RNA was isolated from the mantle, gill, liver, siphon, gonad and muscular foot tissues. High-throughput deep sequencing of S. constricta using 454 pyrosequencing technology yielded 859,313 high-quality reads with an average read length of 489 bp. Clustering and assembly of these reads produced 16,323 contigs and 131,346 singletons with average lengths of 1,376 bp and 458 bp, respectively. Based on transcriptome sequencing, 14,615 sequences had significant matches with known genes encoding 147,669 predicted proteins. Subsequently, previously unknown growth-related genes were identified. A total of 13,563 microsatellites (SSRs) and 13,634 high-confidence single nucleotide polymorphism loci (SNPs) were discovered, of which almost half were validated. CONCLUSION: De novo sequencing of the razor clam S. constricta transcriptome on the 454 GS FLX platform generated a large number of ESTs. Candidate growth factors and a large number of SSRs and SNPs were identified. These results will impact genetic studies of S. constricta.

Proopiomelanocortin (POMC) cDNA was cloned from sea bass (Dicentrarchus labrax) pituitary gland. A 743 nucleotide sequence was obtained coding for the following sequences flanked by sets of proteolytic cleavage sites: ACTH (Ser(88)-Met(127)), alpha-MSH (Ser(88)-Gly(102)), CLIP (Pro(106)-Met(127)), beta-LPH (Glu(131)-Gln(208)), gamma-LPH (Glu(131)-Ser(175)), beta-MSH (Asp(159)-Ser(175)), and beta-endorphin (Tyr(178)-Gln(208)). No region homologous to gamma-MSH/joining peptide (a tetrapod POMC feature) was found. Amino acid sequence identity was high with other teleostean species considered (tilapia: 73%) and lower with elasmobranchs (dogfish: 42%). However, the presumed biologically active peptides were highly conserved within all species considered: alpha-MSH (93-100%), ACTH (80-95%) and beta-endorphin (54-90%). Real-time PCR allowed us to quantify the expression of the POMC in different tIssues of the sea bass: pituitary gland, liver, gonad and head kidney. No significant POMC expression was found in the integument. In pituitary gland, gonads, head kidney and liver, POMC expression was respectively, 1.26x10(10), 2.67x10(5), 2.06x10(4) and 1.67x10(4) copies/ micro g mRNA.
