In recent years, strategic plans for poultry production have emphasized quantitative traits, particularly body weight and carcass traits (meat yield), in response to overpopulation challenges. Candidate genes such as adenylosuccinate lyase (ADSL), melanocortin-4-receptor (MC4R), and calpain 1 (CAPN1) have played vital roles in this context due to their associations with muscle growth and body composition. This study aims to investigate the influence of polymorphisms and gene expressions of the aforementioned genes on body weight (BW), growth rate (GR), breast weight (BrW), and thigh weight (TW) across four distinct chicken breeds: Fayoumi, Matrouh, Mamourah, and Leghorn. The use of PCR-SSCP analysis revealed genetic polymorphisms through the identification of various patterns (genotypes) within the three examined genes. The ADSL, MC4R, and CAPN1 genes exhibited five, three, and two different genotypes, respectively. These polymorphisms displayed promising connections with enhancing economically significant production traits, particularly BW, BrW and TW. Furthermore, gene expression analyses were conducted on breast and thigh tissues obtained from the chicken breeds at 60 days of age, where ADSL and MC4R exhibited a noteworthy up-regulation in Fayoumi and Matrouh breeds, and down-regulation in Mamourah and Leghorn. In contrast, CAPN1 expression decreased across most breeds with a slight increase noted in Fayoumi breed. In conclusion, this investigation underscores the substantial impact of ADSL, MC4R, and CAPN1 genes on economically important production traits within Egyptian domestic chicken breeds. Consequently, these genes emerge as significant molecular markers, holding potential utility in avian selection and breeding programs aimed at enhancing productive performance.

Understanding the structural properties of keratin is of great importance to managing their potential application in keratin-inspired biomaterials and its management of wastes. In this work, the molecular structure of chicken feather keratin 1 was characterized by AlphaFold2 and quantum chemistry calculation. The predicted IR spectrum of the N-terminal region of feather keratin 1, consisting of 28 amino acid residues, was used to assign the Raman frequencies of the extracted keratin. The MW of experimental samples were 6 & 1 kDa while the predicted MW (∼10 kDa) of ß-keratin. Experimental analysis shows the magnetic field treatment could affect the functional and surface structural properties of keratin. The particle size distribution curve illustrates the dispersion of particle size concentration, while TEM analysis demonstrates the reduction of particle diameter to 23.71 ± 1.1 nm following treatment. High-resolution XPS analysis confirmed the displacement of molecular elements from their orbital.

1. The time-energy budget method estimates the energy used for physical activity (PA) by integrating behaviour PA patterns with energy cost for specific PAs. Nevertheless, information about individual energy cost by type of PA are not available and so this study estimated the energy cost of PA for growing broilers.2. An indirect calorimetry system for single birds was constructed to measure the variation in the rate of O2 consumption (V˙O2, L/min) and rate of CO2 production (V˙CO2, L/min) produced by these PAs.3. A total of five birds were used in a replicated trial where their body weight (BW) ranged from 1.5 to 2.5 kg to measure the increase in heat production (HP) above resting levels as a result of PA. The procedure in the chamber was divided into five steps: (1) initial baselining, (2) resting metabolic rate, (3) PA such as feeding, drinking and other standing activities, (4) removal of gas exchange produced in step 3, and (5) final baselining. The PA was recorded using a video camera fixed at the chamber's top (and outside).4. The area under V˙CO2 and V˙O2 curves was used to calculate the CO2 production (vCO2, L) and O2 consumption (vO2, L). Then, the HP (cal/kg-0.75) was calculated according to the Brouwer equation. Two observers analysed the video records to estimate the time spent for each PA (seconds and frequency).5. To calculate the energetic coefficients, the HP was regressed with the function of time spent to perform each PA allowing to estimate the energy cost for eating, drinking and stand activities, which were 0.607, 0.352 and 0.938 cal/kg-0.75/s, respectively.

Modern commercial broiler is growing very rapidly and its amino acid requirement is not fulfilling. An experimental trial was conducted to study the effect of super-dosing of lysine in fish meal-based diets (50% fish meal and 50% SBM) on production performance, protein digestibility and economic efficiency in male and female broiler chickens. Four hundred and eighty (480) one-day-old male and female broiler chicks were divided into forty experimental units of 12 birds each. Five levels of dietary lysine i.e. 90, 100, 110, 120 and 130% of Ross-308 recommendation in male and female birds were separately used. Weight gain and feed intake were higher (p < 0.05) in birds received 100 and 110% recommended lysine than other levels. Feed conversion ratio and EPEF were improved (p < 0.05) in birds received 100% recommended lysine than other levels. Higher (p < 0.05) CP digestibility and lower production cost per kg live weight were noted for birds fed diet containing lysine 100, 110 and 120% than 90 and 130%. Male birds had higher (p < 0.05) WG, FI, EPEF, protein digestibility and lower production cost per kg live weight than female birds. In conclusion, lysine levels below 100% and above 110% of Ross recommended levels had poor production performance, protein digestibility and economic efficiency.

Poultry production is linked to veterinary drug use to treat diseases. Few ectoparasitic compounds are approved for poultry. Fipronil is a pesticide widely used in agriculture. It is also a drug authorized to control ectoparasites in small animals and, in some countries, in cattle. There has been evidence of fipronil extra-label use in laying hens, mainly to control the red mite Dermanyssus gallinae. Fipronil's popularity is due to its high toxicity to invertebrates. It could be metabolized to more toxic metabolites that potentially damage human health. In the present study, we carry out a quantitative exposure assessment and risk characterization for fipronil residues in laying hen eggs for local consumption in five cities of Buenos Aires province in Argentina, namely, Azul, Balcarce, Juarez, Chaves, and Tandil. Consumption surveys and egg sampling were conducted for three summer periods. Eggs were analyzed by UFLC-MS-MS. Fipronil prevalence, residue concentrations, residue stability to cooking methods, egg consumption, among the most important variables were modeled. The results indicated that 20.7% of samples contained fipronil residues. The highest residue was fipronil sulfone metabolite. Fipronil concentrations quantified ranged between 10 and 2510 ppb (median value = 150 ppb). When eggs were cooked, fipronil residues were stable. The exposure assessment and risk characterization revealed that the highest probability of consuming eggs with fipronil residues above the admissible limits was for young adults (20.8%), followed by babies (16.9%), young children (16.4%), children (13.4%), teenagers (10.3%), older adults (9.41%), and adults (8.65%). These results suggest an unacceptable risk associated with egg consumption with fipronil residues for all age groups. PRACTICAL APPLICATION: Fipronil is widely used as an extra-label way on laying hens since its use is prohibited in poultry production both in Argentina and in most countries. This molecule has been classified as Class II, a moderately hazardous pesticide because it could damage various human organs. Fipronil residues in eggs could be one of the exposure pathways for consumers. Monitoring residual levels and carrying out the health risk assessment in eggs are thus in an urge.

Meat quality traits are an important economic trait and remain a major argument, from the producer to the consumer. However, there are a few candidate genes and pathways of chicken meat quality traits that were reported for chicken molecular breeding. The purpose of the present study is to identify the candidate genes and pathways associated with meat quality underlying variations in meat quality. Hence, transcriptome profiles of breast tissue in commercial Digao (DG, 5 male) and Chahua (CH, 5 male) native chicken breeds were analyzed at the age of 100 days. The results found 3525 differentially expressed genes (DEGs) in CH compared to DG with adjusted p-values of ≤0.05 and log2FC ≥ 0.1 FDR ≤ 0.05. Functional analysis of GO showed that the DEGs are mainly involved in the two types of processes of meat quality, such as positive regulation of the metabolic process, extracellular structure organization, collagen trimer, cellular amino acid metabolic process, cellular amino acid catabolic process, and heme binding. Functional analysis of KEGG showed that the DEGs are mainly involved in the two types of processes of meat quality, such as oxidative phosphorylation, carbon metabolism, valine, leucine, and isoleucine degradation, and fatty acid degradation. Many of the DEGs are well known to be related to meat quality, such as COL28A1, COL1A2, MB, HBAD, HBA1, ACACA, ACADL, ACSL1, ATP8A1, CAV1, FADS2, FASN, DCN, CHCHD10, AGXT2, ALDH3A2, and MORN4. Therefore, the current study detected multiple pathways and genes that could be involved in the control of the meat quality traits of chickens. These findings should be used as an essential resource to improve the accuracy of selection for meat traits in chickens using marker-assisted selection based on differentially expressed genes.

1. A field assessment was performed to map the extent of crop usage and thus retention time in broiler chickens. In addition, a broiler experiment was carried out to study the short-term effect of acid addition on phytase efficacy in the crop.2. In the field assessment, the crop content of 40 ad libitum fed broiler chickens from 4 different farms were sampled at 10, 20 and 30 d of age. The dry matter (DM) content varied from 0 to 32 g.3. From 11 d of age, 120 individually caged chickens were intermittently fed a high phytate-P diet with either no addition or 500 FYT C. braakii-derived phytase added or both phytase and 1.4% formic acid added. Excreta were collected for assessment of phosphorus (P) retention. At 20 and 21 d of age, starved birds were fed for 1 h, and thereafter crop and gizzard contents were collected every 20 min until 140 min after start of the feeding. At 60 and 140 min, the contents from the jejunum and ileum were collected.4. All diets reduced the concentration of phytate in the crop, however the combination of acid and phytase resulted in a higher degradation (P < 0.05) than the other diets from 20 min after the start of feeding. Simultaneously, the concentration of the smaller inositol phosphate isomers, such as inositol-5-phosphate, increased (P < 0.05). Phytase increased (P < 0.05) P retention, and the combination of acid and phytase increased jejunal P digestibility (P < 0.05) compared to the other diets.5. The results indicated that lower pH in the crop due to acid addition improved phytase efficacy and increased P digestibility in the anterior digestive tract, even with short retention times.

BACKGROUND: Poultry feed consists mainly of conventional grains and protein supplements, however, using treated unconventional agro-industrial by-products as replacements of corn soybean-based diet can minimize production costs and improve productivity. Therefore, in this study, the effects of fermented or enzymatically treated dried brewer grains (DBG) on growth, expression of digestive enzymes and nutrient transporters genes and the profitability of the rations were evaluated. A total of 1600 one-day-old Ross 308 broiler chicks were randomly distributed in 2 × 4 factorial arrangement (eight treatments with ten replicates, 20 birds/replicate). Experimental diets included two controls; negative control (basal corn-soybean diet; NC) and positive control (basal corn-soybean diet with exogenous enzymes; PC), and six diets in which basal diet was replaced by three levels of fermented DBG (FDBG; 5, 10 or 15%), or enzyme-treated DBG (DBG 5, 10 or 15%+Enz), for 38 days. RESULTS: The results described that feeding FDBG (three levels) or DBG5%+Enz improved (P < 0.05) BW gain and feed efficiency of broilers. Also, feeding FDBG10% yielded the best improvement in weight gain (10%), compared to NC group. Increasing the inclusion levels of DBG either fermented or enzymatically treated up-regulated (p < 0.01) expression of digestive-genes in proventriculus (PGC and PGA5, range 1.4-1.8 fold), pancreas (AMY2A, PNLIP, CELA1, and CCK; range 1.2-2.3 fold) and duodenum (CAT1, CAT2, GLUT1, GLUT2, LAT1, Pep1; range 1.3-3 fold) when compared to NC group. Feeding treated DBG significantly increased (p < 0.05, range 4.5-13.6%) gizzard relative weight compared to NC and PC groups. An additional benefit was lower (p < 0.01) cholesterol content from 66.9 mg/100 mg (NC) to 62.8 mg/100 mg (FDBG5 or 10%) in thigh meat. Furthermore, the least cost feed/kg body gain was achieved in FDBG10% and DBG5%+Enz groups, with approx. 16% reduction compared to NC cost, leading to increasing the income gross margin by 47% and 40% in FDBG10% and DBG5%+Enz groups, respectively. CONCLUSIONS: Substitution of corn-soybean based diet with 10% FDBG or 5% DBG+Enz resulted in better growth and higher economic efficiency of broilers chickens.

Genetic selection in broilers has resulted in improved growth performance, meat yield, and feed conversion efficiency. However, consumers have become increasingly concerned about modern broiler welfare that is related to their rapid growth rate, which may be alleviated by nutrient dilution. This study was conducted to investigate the effects of dietary amino acid (AA) reduction on the growth performance and internal organ development of different genetic strains of broilers. A randomized completed block design with a factorial arrangement of 10 treatments (5 strains × 2 AA levels) was used. The 5 different strains of broilers were fed either a control diet, with digestible AA (lysine, total sulfur AA, and threonine) at the highest recommended levels for the 5 strains, or an AA-reduced diet, with the digestible AA being 20% lower than the control diet. Feed conversion ratio was increased by AA reduction in all 5 strains during day 0-14, 14-28, and 28-41 but was not affected from day 41-55. Body weight and feed intake responses to AA reduction varied in the different strains and ages of birds. Liver weight relative to BW on day 40, and weights of the duodenum and jejunum relative to BW on day 60 were increased by decreasing the dietary AA concentration. These results indicate that the birds had adjusted their organ growth and metabolism in response to increases in digestion, absorption, and utilization efficiency to accommodate a decrease in dietary AA content. Surprisingly, the cost of feed required to produce the same BW was decreased in 4 of 5 strains on both day 41 and 55, which was largely because of the lower price of the diets containing reduced AA levels and the later compensatory growth experienced by the birds fed AA-reduced diets. In the future, when dietary AA levels need to be adjusted to control growth rate and improve welfare status, the genetic strain, age of the birds, and targeted goals need to be taken into consideration.

Most of the cereal-based ingredients used in poultry feed are contaminated with ochratoxin-A (OTA). We have investigated H-ß zeolite (HBZ) as a new OTA binder for poultry, along with widely used clay mineral-based product (CM), using in vitro and in vivo methods. In vitro binding experiment was carried out using a biphasic assay, consisting of adsorption at pH 3.2 and desorption at pH 6.8. High adsorption (>98%) with less desorption (<5%) was observed for HBZ, whereas CM showed high binding (>98%) and moderate desorption (48%). In the in vitro experiments with the different simulated gastro-intestinal pH buffers, HBZ did not desorb OTA at any of the pH. Desorption of OTA was observed with CM, as the pH increases. From the in vitro kinetic and chemisorption studies, faster, stronger, and higher adsorption was observed for HBZ. Thermodynamic studies showed positive entropy (22.76 KJ/mol K) for HBZ, signifying predominant hydrophobic interactions towards OTA, whereas CM exhibited negative entropy (-3.67 KJ/mol K). The in vivo binding efficacy of HBZ and CM was tested in 5-wk-old broiler chickens. The study consisted of 4 experimental groups, each with 6 replicates having 2 birds per replicate. The groups were control, negative control (no toxin binder), T1 (HBZ at 1 kg/ton of feed), and T2(CM at 1 kg/ton of feed). Except control, all the replicates received 20 µg of OTA in the feed. Excreta samples of T1, T2, and NC contained 11.57, 7.16, and 2.78 µg of OTA respectively, which was significantly different from each other (P < 0.05). A growth performance trial was conducted in broiler chickens for 35 D. A total of 288 one-day-old birds were randomly segregated to 3 treatment groups, each with 8 replicates of 12 birds each. Treatment groups consisted of control, T1, and T2, treated with no toxin binder, HBZ, and CM at 1 kg/ton of feed, respectively. None of the treatment groups including control, affected BW gain, and feed conversion ratio (P > 0.05).

Nuclear magnetic resonance (NMR) metabolomics profiling was evaluated as a new tool in sensory assessment of protein hydrolysates. Hydrolysates were produced on the basis of different raw materials (cod, salmon, and chicken), enzymes (Food Pro PNL and Bromelain), and hydrolysis time (10 and 50 min). The influence of raw material and hydrolysis parameters on sensory attributes was determined by traditional descriptive sensory analysis and 1H NMR spectroscopy. The raw material had a major influence on the attribute intensity and metabolite variation, followed by enzyme and hydrolysis time. However, the formation of bitter taste was not affected by the raw material. Partial least-squares regression (PLSR) on 1H NMR and sensory data provided good models (Q2 = 0.55-0.89) for 11 of the 17 evaluated attributes, including bitterness. Significant metabolite-attribute associations were identified. The study confirms the potential prediction of the sensory properties of protein hydrolysates from cod, salmon, and chicken based on 1H NMR metabolomics profiling.

A study was carried out to evaluate the effect of dietary inclusion of Zymomonas mobilis degraded cassava sifting (ZDCS) on growth response, apparent nutrient digestibility, and ileal digesta viscosity of broiler chickens. Five diets containing undegraded and degraded cassava sifting were formulated to replace wheat offal at 0, 50, and 100% levels. Two hundred and forty (240) one-day-old Marshall broiler chickens were randomly allotted to the five dietary treatments in a completely randomized design (CRD); significant means were separated using Duncan's multiple range test at p < 0.05. The biodegradation of cassava sifting with Zymomonas mobilis significantly increased crude protein content by 44.59% while crude fiber and neutral detergent fiber significantly decreased by 23.08% and 6.38%, respectively. The results showed that birds fed 50% ZDCS had the best (p > 0.05) feed conversion ratio (FCR) at the starter phase. The replacement of wheat offal with 100% ZDCS improved (p < 0.05) the crude fiber digestibility at both starter and finisher phases. Also, the birds fed 100% ZDCS had the lowest (p < 0.05) value of ileal digesta viscosity. The birds fed 50% ZDCS had the highest (p < 0.05) values of gross revenue, gross profit, rate of return on investment, and economic efficiency while the least values for gross profit, rate of return on investment, and economic efficiency were obtained in 100% ZDCS. The study concluded that replacement of wheat offal with 50% ZDCS in the ration of broiler chickens improved FCR, crude fiber digestibility (CFD), and rate of return on investment and economic efficiency.

Protein sources are known to be the second largest component in the poultry sector. Traditionally, fish and soya-bean meals are known to supply very good protein; however, these are restricted in supply and more expensive than energy sources. The prices of soya-bean meal are currently high and tend to fluctuate with changes in climatic conditions and social situations in the countries where it is produced. Developing countries like South Africa have made enormous investments in soya-bean production, despite that the country still imports considerable volumes of this crop and is not self-sufficient. This then means that there is an urgent need to seek for alternative and cost-effective protein sources that can provide the same nutrients as soya-bean and fish meal for poultry production. Tenebrio molitor L. which is commonly known as yellow mealworm has a huge potential to substitute commonly used protein sources in poultry diets. Mealworms are easy to breed and do not require large area for production. Moreover, they have high nutritional value comparable to that of soya-bean and fishmeal. However, the only limiting nutrient for mealworms is calcium which can be easily supplemented in the diets. Therefore, this review sets out to explore the importance of replacing soya bean with mealworms in poultry diets. Furthermore, the life cycle of meal worms will also be discussed.

Theory predicts that males will strategically invest in ejaculates according to the value of mating opportunities. While strategic sperm allocation has been studied extensively, little is known about concomitant changes in seminal fluid (SF) and its molecular composition, despite increasing evidence that SF proteins (SFPs) are fundamental in fertility and sperm competition. Here, we show that in male red junglefowl, Gallus gallus, along with changes in sperm numbers and SF investment, SF composition changed dynamically over successive matings with a first female, immediately followed by mating with a second, sexually novel female. The SF proteome exhibited a pattern of both protein depletion and enrichment over successive matings, including progressive increases in immunity and plasma proteins. Ejaculates allocated to the second female had distinct proteomic profiles, where depletion of many SFPs was compensated by increased investment in others. This response was partly modulated by male social status: when mating with the second, novel female, subdominants (but not dominants) preferentially invested in SFPs associated with sperm composition, which may reflect status-specific differences in mating rates, sperm maturation and sperm competition. Global proteomic SF analysis thus reveals that successive matings trigger rapid, dynamic SFP changes driven by a combination of depletion and strategic allocation.

The global popularity of chicken in human diets make it an obvious choice for enrichment with DHA and LC-PUFA. There is presently a need for a robust method for the analysis of chicken tissues and where the fitness for purpose of the method has been demonstrated. The purpose of this paper is to present the validation of the AOAC method 996.06 on five different chicken tissues; breast, thigh, skin, kidney and liver. The parameters investigated as part of the validation study included; linearity and range, the limit of detection and limit of quantification, accuracy, repeatability, inter-analyst reproducibility, and specificity. The method was further applied to assess the stability of DHA and other specific LC-PUFA in chicken tissues over short and long timepoints. The disclosure of this information is relevant for researchers concerned with the analysis of LC-PUFA in regulatory and efficacy studies.

BACKGROUND: The global market for protein drugs has the highest compound annual growth rate of any pharmaceutical class but their availability, especially outside of the US market, is compromised by the high cost of manufacture and validation compared to traditional chemical drugs. Improvements in transgenic technologies allow valuable proteins to be produced by genetically-modified animals; several therapeutic proteins from such animal bioreactors are already on the market after successful clinical trials and regulatory approval. Chickens have lagged behind mammals in bioreactor development, despite a number of potential advantages, due to the historic difficulty in producing transgenic birds, but the production of therapeutic proteins in egg white of transgenic chickens would substantially lower costs across the entire production cycle compared to traditional cell culture-based production systems. This could lead to more affordable treatments and wider markets, including in developing countries and for animal health applications. RESULTS: Here we report the efficient generation of new transgenic chicken lines to optimize protein production in eggs. As proof-of-concept, we describe the expression, purification and functional characterization of three pharmaceutical proteins, the human cytokine interferon α2a and two species-specific Fc fusions of the cytokine CSF1. CONCLUSION: Our work optimizes and validates a transgenic chicken system for the cost-effective production of pure, high quality, biologically active protein for therapeutics and other applications.

Tetracyclines, sulfonamides and amphenicols are broad spectrum antimicrobial drugs that are widely used in poultry farming. However, a high proportion of these drugs can be excreted at high concentrations in droppings, even after the end of a therapy course. This work intended to assess and compare concentrations of florfenicol (FF), florfenicol amine (FFa), chlortetracycline (CTC), 4-epi-chlortetracycline (4-epi-CTC), and sulfachloropyridazine (SCP) in broiler chicken droppings. To this end, 70 chickens were housed under controlled environmental conditions, and assigned to experimental groups that were treated with therapeutic doses of either 10% FF, 20% CTC, or 10% SCP. Consequently, we implemented and designed an in-house validation for three analytical methodologies, which allowed us to quantify the concentrations of these three antimicrobial drugs using liquid chromatography coupled to mass spectrometry (LC-MS/MS). Our results showed that FF and FFa concentrations were detected in chicken droppings up to day 10 after ceasing treatment, while CTC and 4-epi-CTC were detected up to day 25. As for SCP residues, these were detected up to day 21. Noticeably, CTC showed the longest excretion period, as well as the highest concentrations detected after the end of its administration using therapeutic doses.

An increasing amount of evidence suggests that phthalic acid esters (PAE), polychlorinated biphenyls (PCB), polybrominated diphenyl ethers (PBDE), and organochlorine pesticides (OCP) are related to mutagenic, carcinogenic, and endocrine disruptor effects (EDCs). These lipophilic compounds are highly resistant to breakdown processes, and consequently remain in the environment, followed by uptake into the food chain. Human exposure to lipophilic compounds results from the consumption of food containing EDCs, mainly foodstuffs of animal origin with a high fat content, since these contaminants accumulate in fatty tissues. Foodstuffs in which EDCs can accumulate include meat, fish, eggs, and milk. We investigated the contamination in edible eggs to determine whether relative differences in the contaminants' residue levels appeared in three types of egg production (i.e., battery, free-range, and organic). The results showed that PAEs, especially dimethyl phthalate contamination, was the most abundant in the battery eggs, and the PCBs, PBDEs, and OCPs were the most abundant in the free-range eggs. The eggs were contaminated by more than one chemical, and as many as five contaminants (PCB180, PBDE47, dimethyl phthalate, diethyl phthalate, and di-n-butyl phthalate in battery eggs, and PCB138, PCB153, PCB180, diethyl phthalate, and di-2-ethylhexyl phthalate in organic eggs) were detected in the same egg. However, none of the chemicals detected were at the maximum limit of acceptable risk.

Dietary phosphorus (P) oversupply is costly to the poultry industry and represents a point source of excessive P discharge to the environment via manure application to soils. The current study was conducted to investigate the effects of dietary available P (AP) level on growth performance, plasma biochemistry, bone characteristics, and P flow in order to assess minimal AP needs of pullets during the pre-lay period. A total of 192 Lohmann pullet chicks were randomly allocated to one of 4 treatments with 8 replicate cages of 6 birds per cage. The pullets of Low (L; the lowest AP regimen) were fed diets in a sequence of 0.200, 0.175, and 0.150% AP for the age period of 0 to 4 wk, 4 to 8 wk, and 8 to 16 wk, respectively. The AP level was increased by an increment of 0.1% in each phase in Low-Medium (LM), Medium-High (MH) and High (H), thus making H (control) consisting of 0.500, 0.475, and 0.450% AP. Overall, there were no main effects of treatment on body weight gain (BWG), feed intake (FI) and feed conversion ratio (FCR). However, compared with the H treatment, BWG or FI of the L treatment was lower during the first 8 wk post-hatch, but became comparable or higher over the last 8 wk (P < 0.05); FCR of L birds was greater over wk 0 to 4 but became smaller over wk 8 to 16 (P < 0.05). Plasma P was lower in L than H treatments at wk 8 but turned comparable at wk 16 (P < 0.001). Bone characteristics were not different for pullets fed various AP regimens. Total P intake and excretion were reduced with lowering dietary AP (P < 0.001), but its retention percent was not affected. The results indicated that the lowest AP regimen (0.200-0.175-0.150%) in the current study was adequate to support healthy growth and development of pre-lay pullets, while reducing P excretion in manure.

Solute carriers (SLCs) are vital as they are responsible for a major part of the molecular transport over lipid bilayers. At present, there are 430 identified SLCs, of which 28 are called atypical SLCs of major facilitator superfamily (MFS) type. These are MFSD1, 2A, 2B, 3, 4A, 4B, 5, 6, 6 L, 7, 8, 9, 10, 11, 12, 13A, 14A and 14B; SV2A, SV2B and SV2C; SVOP and SVOPL; SPNS1, SPNS2 and SPNS3; and UNC93A and UNC93B1. We studied their fundamental properties, and we also included CLN3, an atypical SLC not yet belonging to any protein family (Pfam) clan, because its involvement in the same neuronal degenerative disorders as MFSD8. With phylogenetic analyses and bioinformatic sequence comparisons, the proteins were divided into 15 families, denoted atypical MFS transporter families (AMTF1-15). Hidden Markov models were used to identify orthologues from human to Drosophila melanogaster and Caenorhabditis elegans Topology predictions revealed 12 transmembrane segments (for all except CLN3), corresponding to the common MFS structure. With single-cell RNA sequencing and in situ proximity ligation assay on brain cells, co-expressions of several atypical SLCs were identified. Finally, the transcription levels of all genes were analysed in the hypothalamic N25/2 cell line after complete amino acid starvation, showing altered expression levels for several atypical SLCs.
