RESUMO

Alcohol-to-jet (ATJ) Synthetic Kerosene with Aromatics (SKA) fuels are produced by dehydration and refining of alcohol feed stocks. ATJ SKA fuel known as SB-8 was developed by Swedish Biofuels as a cooperative agreement between Sweden and AFRL/RQTF. SB-8 including standard additives was tested in a 90-day toxicity study with male and female Fischer 344 rats exposed to 0, 200, 700, or 2000 mg/m3 fuel in an aerosol/vapor mixture for 6 hr/day, 5 days/week. Aerosols represented 0.04 and 0.84% average fuel concentration in 700 or 2000 mg/m3 exposure groups. Examination of vaginal cytology and sperm parameters found no marked changes in reproductive health. Neurobehavioral effects were increased rearing activity (motor activity) and significantly decreased grooming (functional observational battery) in 2000 mg/m3 female rats. Hematological changes were limited to elevated platelet counts in 2000 mg/m3 exposed males. Minimal focal alveolar epithelial hyperplasia with increased number of alveolar macrophages was noted in some 2000 mg/m3 males and one female rat. Additional rats tested for genotoxicity by micronucleus (MN) formation did not detect bone marrow cell toxicity or alterations in number of MN; SB-8 was not clastogenic. Inhalation results were similar to effects reported for JP-8. Both JP-8 and SB fuels were moderately irritating under occlusive wrapped conditions but slightly irritating under semi-occlusion. Exposure to SB-8, alone or as 50:50 blend with petroleum-derived JP-8, is not likely to enhance adverse human health risks in the military workplace.

Diluted bitumen (dilbit) is an unconventional oil produced by the oil sands industry in Canada. Despite the knowledge available on hydrocarbon toxicity, the effects of diluted bitumen on benthic organisms are still largely unknown. Moreover, in Quebec there are only provisional threshold values of 164 mg/kg C10-C50 for chronic effects and 832 mg/kg for acute effects. The protectiveness of these values for benthic invertebrates has not been tested for heavy unconventional oils such as dilbit. Two benthic organisms, the larvae of Chironomus riparius and Hyalella azteca, were exposed to these two concentrations and to an intermediate concentration (416 mg/kg) of two dilbits (DB1 and DB2) and a heavy conventional oil (CO). The aim of the study was to assess the sublethal and lethal effects of spiked sediment by dilbit. The oil was rapidly degraded in the sediment, especially in the presence of C. riparius. Amphipods were much more sensitive to oil than chironomids. LC50-14d values for H. azteca were 199 mg/kg C10-C50 for DB1, 299 mg/kg for DB2 and 8.42 mg/kg for CO compared to LC50-7d values for C. riparius of 492 mg/kg for DB1, 563 mg/kg for DB2 and 514 mg/kg for CO. The size of the organisms was reduced compared to controls for both species. The defense enzymes (GST, GPx, SOD and CAT) were not good biomarkers in these two organisms for this type of contamination. The current provisional sediment quality criteria seem too permissive for heavy oils and should be lowered.

Legal restrictions on vehicle engine exhaust gas emission control do not always go hand in hand with an actual reduction in the emissions of toxins into the atmosphere. Moreover, the methods currently used to measure exhaust gas emissions do not give unambiguous results on the impact of the tested gases on living organisms. The method used to assess the actual toxicity of gases, BAT-CELL Bio-Ambient-Tests using in vitro tests, takes into account synergistic interactions of individual components of a mixture of gases without the need to know its qualitative and quantitative composition and allows for determination of the actual toxicity of the gas composition. Using the BAT-CELL method, exhaust gases from passenger vehicles equipped with spark-ignition engines complying with the Euro 3 and Euro 6 emission standards were tested. The results of toxicological tests were correlated with the results of chromatographic analysis. It was shown that diverse qualitative composition of the mixture of hydrocarbons determining the exhaust gases toxicity may decrease the percentage value of cell survival. Additionally, it was proven that the average survival of cells after exposure to exhaust gases from tested vehicles meeting the more restrictive Euro 6 standard was lower than for vehicles meeting the Euro 3 standard thus indicating the higher toxicity of exhaust gases from newer vehicles.

Remedial guidelines for petroleum hydrocarbons (PHCs) in soil aid in the mitigation of risks to human health and the environmental. However, some remediation guidelines may overestimate the potential for adverse effects to native plant species, contributing to unnecessary remedial efforts in attempts to meet the guidelines. At sites where PHC-contaminated soils undergo weathering, some PHCs may persist but with decreased bioavailability to organisms. In this study, the toxicity of both coarse and fine-grained subarctic soils, contaminated with weathered PHCs were assessed using five native plant species (Picea mariana, Achillea millefolium, Alnus viridis, Elymus trachycaulus and Salix bebbiana). Soil toxicity tests were conducted in a growth chamber with parameters set to simulate the site's subarctic climate conditions. Reference toxicant tests using boric acid were conducted to provide confidence in the interpretation of the results for the PHC-contaminated soils, and also provide new information on the sensitivities of the four boreal species to boric acid. All plants exhibited reduced growth and germination rates as boric acid concentrations increased. Despite exceeding the Canada-wide standard guidelines for Fraction 3 PHCs, field-collected contaminated soils had no significant negative impacts on the growth (i.e., length, dry weight and emergence) of any of the plant species tested.

Paraffin waxes and white mineral oils are distinct petroleum products separated from a common feedstock by crystallization, where only n-alkanes, iso- and cyclo-alkanes with a linear backbone of ∼ 20 carbon atoms long, selectively crystalize out from the oil to form the wax, which is solid at room temperature, whereas oils remain liquid. Up until the 90's, these differences were reflected in separated regulatory assessments. A paradigm shift occurred when Fischer 344 rats (F-344) developed liver epithelioid granuloma following exposure to low and medium viscosity oils or waxes. This lesion was used as common denominator between these products to be jointly assessed under the common term "mineral hydrocarbons - MHC", obviating compositional differences. This regulatory paradigm dominated for the next 30 years, exacerbated by the EFSA 2012 evaluation using the analytical term "MOSH" (mineral oil saturated hydrocarbons) which encompassed these products under single chromatography fraction. The reconstruction of historical developments, together with recent EFSA-sponsored studies of toxicity and accumulation and supporting literature, has allowed us to understand the etiology of the F-344 rat hepatic epithelioid granuloma, which is presented in an adverse outcome pathway (AOP). Considering chemical composition, it clearly demonstrates that the hepatic effects in F-344 rats caused by linear alkanes of waxes are irrelevant for humans. Waxes are thus not MOSH and should thus be evaluated on their own merit. The term MOSH should not include n-alkanes and be exclusively used to mineral oil fractions when considering their chemical makeup for a relevant human hazard assessment.

The ever-increasing anthropogenic activities have adversely impacted coral reef ecosystems and their ecological functions. This calls for an urgent assessment of the health state of these valuable ecosystems to justify the need for mitigation and proper management efforts. In this contribution, we used multiple indicators to assess the impact of intense oil-related activities on coral reefs in two near-by impacted and non-impacted islands in the northwestern Persian Gulf. The efficacy of indices was assessed using estimations of the effect size (omega-squared), precision, and decision trees (Classification and Regression Tree (CART)). The results demonstrated that the combination of bioaccumulation of Æ©PAH in coral tissues, the percent of live coral cover, and the Sediment Constituent (SEDCON) Index were the most robust proxies reflecting the influence of human activities on reef's health. Based on sedimentary PAH concentration, the CART classified most of the indicators into two classes consisting of those in impacted and those in non-impacted locations, further supporting the feasibility of the employed indices. The findings of this study provided a warning of degradation in coral reefs of the island subjected to PAH pollution. This encourages decision-makers to execute routine monitoring and mitigation practices to maintain healthy reefs in the study areas.

Toxicity monitoring of environmental pollutants especially petroleum hydrocarbons as priority pollutants is an important environmental issue. This study addresses a rapid, sensitive and cost effective method for the detection of total petroleum hydrocarbons (TPHs) using Aliivibrio fischeri bioluminescence inhibition bioassay. At the first step, the optimum conditions including time, pH and temperature for growth of A. fischeri were determined. Then, two methods were used to evaluate the toxicity of petroleum compounds. In the first method, short-term (15 min) and long-term (16 h) toxicity assays were performed. In the second method luminescence kinetics of A. fischeri was investigated during 24 h. The results demonstrated the most appropriate time for the bacterial growth occurred 16 h after inoculation and optimum temperature and pH were found 25 °C and 7, respectively. Short-term and long-term toxicity did not indicate any toxicity for various concentrations of TPHs (30, 50, 110, 160, 220 mg/L). Considering the luminescence kinetics of A. fischeri the long-term assay was introduced as 6 h. The half maximal effective concentration (EC50) was achieved 1.77 mg/L of TPHs. It is concluded that the luminescence kinetics of A. fischeri can be a valuable approach for assessing toxicity of TPHs in aquatic environments.

A toxicological investigation was conducted for alcohol-to-jet (ATJ) fuels intended as a 50:50 blend with petroleum-derived fuel Jet Propulsion (JP)-8. The ATJ synthetic paraffinic kerosene (SPK) fuel was produced by Gevo (Englewood CO) and derived either from biomass (bio) or non-biomass sources. All toxicity tests were performed with one or both ATJ fuels following addition of a standard additive package required for JP-8. The primary fuel, Gevo (bio) ATJ SPK produced from biomass-derived iso-butanol, exhibited the same dermal irritation potential in rabbits as JP-8; the non-biomass-derived fuel was less irritating. The Gevo (bio) fuel was non-clastogenic in micronucleus testing with rats and neither version was mutagenic in the bacterial reverse mutation assay. A 90-day study was performed with Gevo (bio) ATJ SPK by exposing male and female Fischer 344 rats to target concentrations of 0, 200, 700 or 2000 mg/m3 of fuel, 6 hr per day, 5 days a week for 69 exposure days and included neurobehavioral assays and reproductive health evaluations in the study design. Results were negative or limited to irritant effects in the respiratory system due to exposure to a vapor and aerosol mixture in the 2000 mg/m3 exposure group. Occupational exposure limits for JP-8 were proposed for these ATJ fuels since these fuels display similar or somewhat lower toxicity than JP-8. As both versions of the Gevo ATJ jet fuel were similar, handling of either fuel alone or in a blend with petroleum-derived JP-8 appears unlikely to increase human health risks for workers.

RESUMO

Soil contamination with total petroleum hydrocarbons (TPH) is widespread throughout the globe due to the massive production of TPH anthropogenically and its occurrence in the soil. TPH is toxic to beneficial soil organisms and humans and thus has become a serious concern among the public. Traditionally TPH toxicity in the soil is estimated based on chemical fractions and a range of bioassays including plants, invertebrates and microorganisms. There is a large inconsistency among ecotoxicology data using these assays due to the nature of TPH and their weathering. Therefore, in this article, we critically reviewed the weathered conditions of TPH, the potential fate in soil and the bioindicators for the assessment of the ecotoxicity. Based on the current research and the state-of-the-art problem, we also highlighted key recommendations for future research scope for the real-world solution of the ecotoxicological studies of hydrocarbons.

This paper presents the results of two different studies investigating the acute and chronic toxicity of two blends of diluted bitumens ("dilbit") and weathered dilbit on freshwater fish and invertebrates after exposure to different concentrations of physically-dispersed (water accommodated fraction; WAF) and chemically-dispersed (chemically-enhanced WAF; CEWAF). The first study evaluated the acute and chronic toxicity of weathered, unweathered and dispersed Access Western Blend (AWB) dilbit on Fathead minnow (Pimephales promelas). In the second study, acute and chronic toxicity of weathered and unweathered Cold Lake Blend (CLB) dilbit was assessed on Rainbow trout (Oncorhynchus mykiss), and two invertebrate species, Daphnia (Daphnia magna) and Ceriodaphnia (Ceriodaphnia dubia). For Fathead minnow, unweathered AWB WAF demonstrated a significantly higher acute toxicity (LC50-96 h = 0.628 g/L) compared to the weathered AWB WAF (LC50-96 h = 2.06 g/L). Chronic toxicity tests showed that Fathead minnow lethality was also higher for unweathered AWB (LC50-7 d = 0.593 g/L) compared to the weathered AWB (LC50-7 d = 1.31 g/L) whereas larval growth toxicity was lower for unweathered AWB (IC25-7 d=0.312 g/L) compared to the weathered dilbit (IC25-7 d = 0.096 g/L). Rainbow trout exposed to unweathered CLB demonstrated a significantly higher toxicity (LC50-96 h = 5.66 g/L) compared to the weathered CLB (LC50 > 18 g/L). Lethality (LC50 = 6.43 g/L) was observed in Ceriodaphnia exposed to the CLB WAF while no mortality was observed with the weathered CLB. The reproductive effects on Ceriodaphnia were greater with the CLB (IC25 <1.0) than with the weathered CLB (IC25 = 3.99 g/L). Volatile organic compounds (VOC), polycyclic aromatic hydrocarbons (PAH) and total petroleum hydrocarbons (TPH) increased as the dilbit CLB and AWB WAF concentrations increased. The total VOC and the total PAHs (including the alkylated PAHs) presented similar concentrations in the weathered and unweathered AWB WAF. These compounds seem therefore not be affected by the AWB WAF weathering process. However, VOC and PAH concentrations decreased significantly in the Rainbow trout and Ceriodaphnia toxicity tests using the weathered CLB WAF. VOC and PAH concentrations were also lower in the weathered AWB CEWAF used for Fathead minnow chronic study.

Fischer-Tropsch (FT) Synthetic Paraffinic Kerosene (SPK) jet fuel is a synthetic organic mixture intended to augment petroleum-derived JP-8 jet fuel use by the U.S. armed forces. The FT SPK testing program goal was to develop a comparative toxicity database with petroleum-derived jet fuels that may be used to calculate an occupational exposure limit (OEL). Toxicity investigations included the dermal irritation test (FT vs. JP-8 vs. 50:50 blend), 2 in vitro genotoxicity tests, acute inhalation study, short-term (2-week) inhalation range finder study with measurement of bone marrow micronuclei, 90-day inhalation toxicity, and sensory irritation assay. Dermal irritation was slight to moderate. All genotoxicity studies were negative. An acute inhalation study with F344 rats exposed at 2000 mg/m3 for 4 hr resulted in no abnormal clinical observations. Based on a 2-week range-finder, F344 rats were exposed for 6 hr per day, 5 days per week, for 90 days to an aerosol-vapor mixture of FT SPK jet fuel (0, 200, 700 or 2000 mg/m3). Effects on the nasal cavities were minimal (700 mg/m3) to mild (2000 mg/m3); only high exposure produced multifocal inflammatory cell infiltration in rat lungs (both genders). The RD50 (50% respiratory rate depression) value for the sensory irritation assay, calculated to be 10,939 mg/m3, indicated the FT SPK fuel is less irritating than JP-8. Based upon the proposed use as a 50:50 blend with JP-8, a FT SPK jet fuel OEL is recommended at 200 mg/m3 vapor and 5 mg/m3 aerosol, in concurrence with the current JP-8 OEL.

The evaluation of mineral oils by the Joint FAO/WHO Expert Committee on Food Additives (JECFA) provided high acceptable daily intakes for classes largely falling into the mass range strongly accumulated by humans. Because they are roughly 2 orders of magnitude above the present exposure, they authorize strongly increased exposure. An approach based on accumulation seems more adequate. Increased organ weights might be more critical than granulomas. Aromatic hydrocarbons with 1-2 aromatic rings should be distinguished from those with at least 3 aromatic rings. If mineral oil saturated hydrocarbon limits were low, no limit might be needed for the 1-2 ring aromatics. It should be considered to phase out substantial use of mineral oils in food application.

The European chemical control regulation (REACH) requires that data on physical/chemical, toxicological and environmental hazards be compiled. Additionally, REACH requires formal assessments to ensure that substances can be safely used for their intended purposes. For health hazard assessments, reference values (Derived No Effect levels, DNELs) are calculated from toxicology data and compared to estimated exposure levels. If the ratio of the predicted exposure level to the DNEL, i.e. the Risk Characterization Ratio (RCR), is less than 1, the risk is considered controlled; otherwise, additional Risk Management Measures (RMM) must be applied. These requirements pose particular challenges for complex substances. Herein, "white spirit", a complex hydrocarbon solvent, is used as an example to illustrate how these procedures were applied. Hydrocarbon solvents were divided into categories of similar substances. Representative substances were identified for DNEL determinations. Adjustment factors were applied to the no effect levels to calculate the DNELs. Exposure assessments utilized a standardized set of generic exposure scenarios (GES) which incorporated exposure predictions for solvent handling activities. Computer-based tools were developed to automate RCR calculations and identify appropriate RMMs, allowing consistent communications to users via safety data sheets.

Female Fischer 344 rats were exposed to three MOSH mixtures: oils largely below and above C25 (S-C25 and L-C25) and a 1:1 mixture of L-C25 with a wax; doses of 400, 1000 and 4000mg/kg feed were administered during 120days. MOSH were determined by on-line HPLC-GC-FID in liver, spleen, adipose tissue and the carcass. The composition of the hydrocarbons accumulated in the tissues was further analyzed by comprehensive two-dimensional GC (GC×GC). MOSH in the mass range of C26-30 were more strongly accumulated than those between C20-25, which does not support the present classification of MOSH differentiating at n-C25 for risk assessment. Compared to the total of the MOSH, n-alkanes and n-alkyl monocyclic naphthenes were generally enriched in adipose tissue. In liver and spleen, n-alkanes up to C25 were eliminated, but strongly accumulated at around C30. Based on this profile, poor solubility and the melting points, it is hypothesized that crystallization protects these wax components against metabolism and elimination. In the liver, relative retention of n-alkanes decreased again beyond C30, accentuated at high exposure, suggesting reduced absorption. Compared to the animal data, accumulation of n-alkanes from food sources, such as apples, into human tissues seems low, perhaps because of low absorption due to their presence in crystalline form. A series of dominant isoalkanes, accumulated in all tissues analyzed, was characterized, though without proposing a structure. Implications on present regulation of white mineral oil products are discussed.

CONTEXT: EpiAirway™ 3-D constructs are human-derived cell cultures of differentiated airway epithelial cells that may represent a more biologically relevant model of the human lung. However, limited information is available on their utility for exposures to air pollutants at the air-liquid interface (ALI). OBJECTIVE: To assess the biological responses of EpiAirway™ cells in comparison to the responses of A549 human alveolar epithelial cells after exposure to air pollutants at ALI. METHODS: Cells were exposed to filtered air, 400 ppb of ozone (O3) or a photochemically aged Synthetic Urban Mixture (SynUrb54) consisting of hydrocarbons, nitrogen oxides, O3 and other secondary oxidation products for 4 h. Basolateral supernatants and apical washes were collected at 9 and 24 h post-exposure. We assessed cytotoxicity by measuring lactate dehydrogenase (LDH) release into the culture medium and apical surface. Interleukin 6 (IL-6) and interleukin 8 (IL-8) proteins were measured in the culture medium and in the apical washes to determine the inflammatory response after exposure. RESULTS: Both O3 and SynUrb54 significantly increased basolateral levels of LDH and IL-8 in A549 cells. No significant changes in LDH and IL-8 levels were observed in the EpiAirway™ cells, however, IL-6 in the apical surface was significantly elevated at 24 h after O3 exposure. CONCLUSION: LDH and IL-8 are robust endpoints for assessing toxicity in A549 cells. The EpiAirway™ cells show minimal adverse effects after exposure suggesting that they are more toxicologically resistant compared to A549 cells. Higher concentrations or longer exposure times are needed to induce effects on EpiAirway™ cells.

Toxicological evaluation of Clarias gariepinus from bitumen-polluted River Oluwa, Nigeria, was carried out in furtherance of studies on the environmental impacts of the bitumen exploration in Ondo State, Nigeria. Samples were taken from three different (two polluted and one as control) sites. The effect of changes in monthly seasonal flow rate was assessed for the sites of study. Blood plasma clinical-chemical parameters (BCCPs) and histological changes/lesions in various organs were evaluated as markers of pollution in the fish blood using standard methods. The result of the physicochemical properties of water from the sampling points revealed some of the values conforming to approved standards while others showed deviation. Significant differences were found in the blood and histological endpoints between the control and the polluted sites as well as between the two seasons evaluated across the sites. The public health implications of consuming this fish are fully discussed.
