RESUMO

Serotonin (5-HT) deficiency is a core biological pathology underlying depression and other psychiatric disorders whose key symptoms include decreased motivation. However, the exact role of 5-HT in motivation remains controversial and elusive. Here, we pharmacologically manipulated the 5-HT system in macaque monkeys and quantified the effects on motivation for goal-directed actions in terms of incentives and costs. Reversible inhibition of 5-HT synthesis increased errors and reaction times on goal-directed tasks, indicating reduced motivation. Analysis found incentive-dependent and cost-dependent components of this reduction. To identify the receptor subtypes that mediate cost and incentive, we systemically administered antagonists specific to 4 major 5-HT receptor subtypes: 5-HT1A, 5-HT1B, 5-HT2A, and 5-HT4. Positron emission tomography (PET) visualized the unique distribution of each subtype in limbic brain regions and determined the systemic dosage for antagonists that would achieve approximately 30% occupancy. Only blockade of 5-HT1A decreased motivation through changes in both expected cost and incentive; sensitivity to future workload and time delay to reward increased (cost) and reward value decreased (incentive). Blocking the 5-HT1B receptor also reduced motivation through decreased incentive, although it did not affect expected cost. These results suggest that 5-HT deficiency disrupts 2 processes, the subjective valuation of costs and rewards, via 5-HT1A and 5-HT1B receptors, thus leading to reduced motivation.

Assessing quality of life in animals is an art as much as a science. Despite the use of questionnaires and keeper reports which consider several aspects of well-being, the process often remains subjective. Keepers have unique insights, and anecdotal observations can be enhanced with objective data. We combined the art and science of assessments in this study on a geriatric macaque (1.0 lion-tailed/pig-tailed (Macaca silenus/macaca nemestrina) hybrid), using historic data to inform management decisions. Following the unexpected death of his cage mate, his activity and engagement with keepers decreased, and new concerning behaviors presented. While the zoo worked to identify new social opportunities, we used these data to develop a plan to improve his quality of life (e.g., increase training sessions, enrichment, social interactions). After intense implementation, we saw a significant increase in activity level and engagement with keepers; the frequency of unexpected behaviors suggesting a lower quality of life, however, increased over time. Our data allowed us to objectively compare changes in behavior, enabling the zoo to make the most informed animal management decision possible. [Figure: see text].

Cynomolgus macaques may undergo surgical procedures for scientific and veterinary purposes. Recognition and assessment of pain using validated tools is a necessary first step for adequately managing pain in these primates. Grimace scales are one means of assessing the occurance of acute pain using action units such as facial expressions and posture. The aim of this study was to create and validate a Cynomolgus Macaque Grimace Scale (CMGS). Cynomolgus macaques (n = 43) were video recorded before and after a surgical procedure. Images were extracted from videos at timepoints at which breakthrough pain might be expected based on analgesic pharmacokinetics. Using the CMGS images were scored by 12 observers blinded to animal identification, times, and conditions. To validate the tool, detailed behavioral analyses emphasizing changes to baseline activity ethograms were compared to grimace scores. Four action units were identified related to potential pain including orbital tightening, brow lowering, cheek tightening, and hunched posture. The CMGS tool was found to have moderate inter- (ICCaverage action unit mean ± SD: 0.67 ± 0.28) and good intra- (ICCsingle mean ± SD: 0.79 ± 0.14) observer reliability. Grimace scores increased significantly (p < 0.0001) in the first four post-operative timepoints compared to baseline, correlating with behavioral findings (rho range = 0.22-0.35, p < 0.001). An analgesic intervention threshold was determined and should be considered when providing additional pain relief. The CMGS was shown to be a reliable and valid tool; however, more research is needed to confirm external validity. This tool will be highly valuable for refining analgesic protocols and acute peri-procedural care for cynomolgus macaques.

Facial expressions are key to navigating social group life. The Power Asymmetry Hypothesis of Motivational Emancipation predicts that the type of social organization shapes the meaning of communicative displays in relation to an individual's dominance rank. The bared-teeth (BT) display represents one of the most widely observed communicative signals across primate species. Studies in macaques indicate that the BT display in despotic species is often performed unidirectionally, from low- to high-ranking individuals (signaling submission), whereas the BT display in egalitarian species is usually produced irrespective of dominance (mainly signaling affiliation and appeasement). Despite its widespread presence, research connecting BT displays to the power asymmetry hypothesis outside the Macaca genus remains scarce. To extend this knowledge, we investigated the production of BT in relation to social dominance in dyadic interactions (N = 11,377 events) of 11 captive bonobos (Pan paniscus). Although adult bonobos were more despotic than previously suggested in the literature, BT displays were produced irrespective of dominance rank. Moreover, while adults produced the BT exclusively during socio-sexual interactions, especially during periods of social tension, immature bonobos produced the BT in a wider number of contexts. As such, the results indicate that the communicative meaning of the BT display is consistent with signaling appeasement, especially in periods of social tension. Moreover, the BT display does not seem to signal social status, supporting the prediction for species with a high degree of social tolerance. These results advance our understanding of the origins of communicative signals and their relation to species' social systems.

In humans, risk attitude is highly context-dependent, varying with wealth levels or for different potential outcomes, such as gains or losses. These behavioral effects have been modelled using prospect theory, with the key assumption that humans represent the value of each available option asymmetrically as a gain or loss relative to a reference point. It remains unknown how these computations are implemented at the neuronal level. Here we show that macaques, like humans, change their risk attitude across wealth levels and gain/loss contexts using a token gambling task. Neurons in the anterior insular cortex (AIC) encode the 'reference point' (i.e., the current wealth level of the monkey) and reflect 'loss aversion' (i.e., option value signals are more sensitive to change in the loss than in the gain context) as postulated by prospect theory. In addition, changes in the activity of a subgroup of AIC neurons correlate with the inter-trial fluctuations in choice and risk attitude. Taken together, we show that the primate AIC in risky decision-making may be involved in monitoring contextual information used to guide the animal's willingness to accept risk.

Improving captive environments for primates has been an important tool to enhance animal welfare. One method has been to provide primates with naturalistic vegetated enclosures to promote species-specific behaviors, enhancing interactions with their social and natural environment, such as an increase in feeding, foraging, and positive social behaviors such as play. To investigate the benefits in which living in naturalistic environments promote general animal well-being, we observed immature and adult males living in two outdoor housed groups of Japanese macaques (Macaca fuscata) in the Kyoto University Primate Research Institute (KUPRI), Inuyama, Japan, from June of 2015 until June 2016. One enclosure was naturally vegetated and the other was not. We recorded male activity budgets, affiliative (groom, play) behaviors, and rates of agonistic interactions. To examine health status, we recorded and compared coat conditions for both groups. We found that males in the vegetated enclosure spent more time in social play than males in the non-vegetated enclosure, while males in the non-vegetated enclosure displayed more stereotypic behaviors and agonistic interactions. We recorded better coat conditions in the vegetated enclosure males while rates of social grooming or self-grooming were no different between males in the two enclosures. The males in the vegetated enclosure did not have activity budgets more similar to their wild counterparts; but they spent more time in feeding-related activities and less time resting, which was more similar to their wild counterparts than males in the non-vegetated enclosure. Our findings suggest that individuals housed in naturalistic environments have significantly greater behavioral and physical markers of wellbeing than those housed in unnatural, large outdoor enclosures. Although we found that males in both types of enclosures overall had similar time budgets to males in the wild, the detailed behavioral and health results suggest that the welfare benefits to males were greater in the vegetated enclosure, compared with non-vegetated enclosures. Efforts to mimic more natural environments should promote the well-being of primates.

Group living is a source of stress and an individuals' social environment has been shown to have a significant effect on its health and well-being. However, little is known about how different social organizations affect the stress levels of their members. Is living in a hierarchical society more or less stressful than living in a more tolerant structure? Here, we assess cortisol concentrations in the hair of two macaque species with radically different dominance styles: the egalitarian Tonkean macaque (Macaca Tonkeana) and the despotic long-tailed macaques (Macaca fascicularis). Hair was sampled in winter and again in late spring in two mixed-sex groups of 22 Tonkean macaques and 9 long-tailed macaques; Hair cortisol concentrations were significantly higher in the egalitarian Tonkean macaques than in the despotic long-tailed macaques, ranging from 161.13 to 938.8 pg/mg (mean ± SD 349.67 ± 126.22) and from 134.46 to 339.86 pg/mg (mean ± SD 231.2 ± 44.24), respectively. There was no difference between male and female cortisol concentrations, but hair cortisol increased with age in males. Dominance rank certainty was lower among female Tonkean macaques compared to long-tailed macaques. Our results suggest that species differences in dominance styles may translate into differences in long-term cortisol concentrations. We suggest that the higher cortisol concentrations in Tonkean macaques could be linked to the instability and lack of predictability and control around social relationships.


LAY SUMMARYBeing part of a social group can be very stressful, depending on the social structure of this group. We found that the more socially tolerant Tonkean macaques exhibited on average higher hair cortisol concentrations than more despotic long-tailed macaques. Males and females exhibited similar concentrations of hair cortisol in both species, but in male Tonkean macaques concentrations increased with age. The finding that overall cortisol levels were higher in the more tolerant species suggests that psychological arousal due to unpredictable social relations and mating competition may be an important driver of cortisol release in this species despite its overall tolerant social organization.

Nonhuman primates (NHP) are important translational models for cardiac aging. To assess progress in this research area and to provide a reference for other investigators, we identified papers indexed in PubMed to determine what species, ages, outcomes, treatments, and approaches have been studied. Since 1983, 33 studies of cardiac aging in NHP have been published. Of these, 27 used species of macaque, 6 baboon, 1 vervet, 1 orangutan, and 1 marmoset (some studies were multispecies). Common research approaches were echocardiography, ECG, and histology of the left ventricle. Only 10 studies performed sex-based analyses. The average age of the oldest macaque studied was 26 y. The reported mean lifespan of macaques in captivity is around 30 y. The age of the oldest baboon studied was 24 y. Baboons in captivity are reported to live on average to 21 y. Twelve studies took a "life course" approach, studying animals of a wide range of ages from less than or equal to 10 y through the late teens to thirties, and employing analyses designed to show change over time. Keeping NHP into old age is a major challenge for biomedical research. The ideal design is to start monitoring in early life and to track how cardiac structure and function change with age. Important issues for future research are an increased focus on life-course approaches, investment in existing life-course NHP cohorts, better reporting of study sample characteristics, more molecular studies to identify genetic risk factors and mechanisms, attention to sex as a biological variable, a move away from descriptive reports to mechanistic studies, development of biomarkers to predict disease risk, and exploration of interventions that are implemented early in life to prevent or delay age-related disease later in life. Reducing exposure to early life adversity, identifying early-life biomarkers of aging and age-related disease, and early treatment can contribute to longer health span.

In perceptual decisions, subjects infer hidden states of the environment based on noisy sensory information. Here we show that both choice and its associated confidence are explained by a Bayesian framework based on partially observable Markov decision processes (POMDPs). We test our model on monkeys performing a direction-discrimination task with post-decision wagering, demonstrating that the model explains objective accuracy and predicts subjective confidence. Further, we show that the model replicates well-known discrepancies of confidence and accuracy, including the hard-easy effect, opposing effects of stimulus variability on confidence and accuracy, dependence of confidence ratings on simultaneous or sequential reports of choice and confidence, apparent difference between choice and confidence sensitivity, and seemingly disproportionate influence of choice-congruent evidence on confidence. These effects may not be signatures of sub-optimal inference or discrepant computational processes for choice and confidence. Rather, they arise in Bayesian inference with incomplete knowledge of the environment.

Ketamine is an NMDA receptor antagonist commonly used to maintain general anesthesia. At anesthetic doses, ketamine causes high power gamma (25-50 Hz) oscillations alternating with slow-delta (0.1-4 Hz) oscillations. These dynamics are readily observed in local field potentials (LFPs) of non-human primates (NHPs) and electroencephalogram (EEG) recordings from human subjects. However, a detailed statistical analysis of these dynamics has not been reported. We characterize ketamine's neural dynamics using a hidden Markov model (HMM). The HMM observations are sequences of spectral power in seven canonical frequency bands between 0 to 50 Hz, where power is averaged within each band and scaled between 0 and 1. We model the observations as realizations of multivariate beta probability distributions that depend on a discrete-valued latent state process whose state transitions obey Markov dynamics. Using an expectation-maximization algorithm, we fit this beta-HMM to LFP recordings from 2 NHPs, and separately, to EEG recordings from 9 human subjects who received anesthetic doses of ketamine. Our beta-HMM framework provides a useful tool for experimental data analysis. Together, the estimated beta-HMM parameters and optimal state trajectory revealed an alternating pattern of states characterized primarily by gamma and slow-delta activities. The mean duration of the gamma activity was 2.2s([1.7,2.8]s) and 1.2s([0.9,1.5]s) for the two NHPs, and 2.5s([1.7,3.6]s) for the human subjects. The mean duration of the slow-delta activity was 1.6s([1.2,2.0]s) and 1.0s([0.8,1.2]s) for the two NHPs, and 1.8s([1.3,2.4]s) for the human subjects. Our characterizations of the alternating gamma slow-delta activities revealed five sub-states that show regular sequential transitions. These quantitative insights can inform the development of rhythm-generating neuronal circuit models that give mechanistic insights into this phenomenon and how ketamine produces altered states of arousal.

RESUMO

Motor actions in fMRI settings require specialized hardware to monitor, record, and control the subjects behavior. Commercially available options for such behavior tracking or control are very restricted and costly. We present a novel grasp manipulandum in a modular design, consisting of MRI-compatible, 3D printable buttons and a chassis for mounting. Button presses are detected by the interruption of an optical fiber path, which is digitized by a photodiode and subsequent signal amplification and thresholding. Two feedback devices (manipulanda) are constructed, one for macaques (Macaca mulatta) and one for human use. Both devices have been tested in their specific experimental setting and possible improvements are reported. Design files are shared under an open hardware license.

Regulation of energy allocation and metabolic rate plays an important role in determining behavior and fitness in wild animals, calling for the validation of non-invasive markers of energetic condition. Recently, the thyroid hormone triiodothyronine (T3) has emerged as a promising marker as concentrations decrease to lower the metabolic rate during energetically challenging periods. However, it remains largely unclear whether T3 merely represents an alternative or provides additional information compared to other compounds involved in the regulation of energy acquisition and allocation, like cortisol and C-peptide, as few joint measurements have been conducted to date in non-invasively collected samples. We aimed to validate the non-invasive measurement of immunoreactive urinary total T3 (uTT3), in comparison to urinary cortisol (uCort) and urinary C-peptide (uCP), as a marker of metabolic response to variation in food intake in macaques, and to address a number of issues regarding the collection, storage and processing of samples which are important for application of uTT3 measurements under field conditions. We used daily samples and body mass measures from a prior food restriction-refeeding experiment over 4 weeks with six captive macaques and analyzed concentrations of uTT3 and uCort in samples collected prior to (fasting) and after morning feeding (non-fasting). Concentrations of uTT3 decreased in response to restriction in food supply and were also lower during weeks of food restriction compared to weeks of refeeding. Variation in uTT3 also correlated positively with variation in body mass and concentrations of uCP. As expected, uCort showed the reverse pattern, increasing during food restriction and decreasing following refeeding, but was not associated with variation in body mass. Generally, compared to fasting samples, concentrations were higher in post-morning feeding, i.e. non-fasting, samples for uTT3 but not uCort. Contamination of urine samples with fecal matter, but not soil, and exposure to UV light led to a decrease in uTT3. uTT3 was largely unaffected by repeated freeze-thaw cycles and by refrigeration for medium-term storage (2 days) but degraded substantially when stored at ambient temperature for the same period. In conclusion, uTT3 measurements inform on the effect of food intake and its associated metabolic response to variation in energetic status. Since uTT3 is reasonably robust to many issues associated with collection and storage of urine samples under field conditions, it is a promising biomarker for studies of energetic condition and basal metabolic rate in wild macaques.

Rhesus macaque is one of the most widely used primate model animals for immunological research of infectious diseases including human immunodeficiency virus (HIV) infection. It is well known that major histocompatibility complex (MHC) class I genotypes affect the susceptibility and disease progression to simian immunodeficiency virus (SIV) in rhesus macaques, which is resembling to HIV in humans. It is required to convincingly determine the MHC genotypes in the immunological investigations, that is why several next-generation sequencing (NGS)-based methods have been established. In general, NGS-based genotyping methods using short amplicons are not often applied to MHC because of increasing number of alleles and inevitable ambiguity in allele detection, although there is an advantage of short read sequencing systems that are commonly used today. In this study, we developed a new high-throughput NGS-based genotyping method for MHC class I alleles in rhesus macaques and cynomolgus macaques. By using our method, 95% and 100% of alleles identified by PCR cloning-based method were detected in rhesus macaques and cynomolgus macaques, respectively, which were highly correlated with their expression levels. It was noted that the simulation of new-allele detection step using artificial alleles differing by a few nucleotide sequences from a known allele could be identified with high accuracy and that we could detect a real novel allele from a rhesus macaque sample. These findings supported that our method could be adapted for primate animal models such as macaques to reduce the cost and labor of previous NGS-based MHC genotyping.

The present study aimed to conduct a comparative assessment of the external morphology, relative tail length and colour variation (RGB additive colour model) of Macaca munzala, Tawang and Macaca assamensis, Goalpara (southern Brahmaputra population), using a non-invasive photogrammetry method. The study revealed that the relative tail length of M. munzala (0.43 ± 0.03) is similar to that of M. assamensis assamensis, a subspecies of M. assamensis. On the other hand, the relative tail length of M. assamensis, Goalpara (0.61 ± 0.04) is similar to M. assamensis pelops, the other subspecies of M. assamensis. A total of 12 external morphological traits were observed and similarities found between the two studied populations. The study also found that the species-specific traits of M. munzala such as "dark patch of the crown whorl", "dark patch of hair at the temporal side", "spectacle appearance around eyes" and "whip-like tail appearance" in the infant and "stocky tail" are present in M. assamensis, Goalpara as well. Likewise, the "chin and cheek whiskers" are found in adult female individuals of both species. The comparative assessment of the dorsal coat colour (RGB value) revealed a darker brown dorsal coat in M. munzala (R 123.14 ± 11.62; G 107.71 ± 10.37, B 89.43 ± 9.21) than in M. assamensis, Goalpara (R 136 ± 23.57, G 112 ± 15.63, B 97.83 ± 13.04). The comparative assessment of facial skin colour among the adult individuals showed that the male M. munzala has darker brown facial skin than that of M. assamensis, whereas the female M. munzala shows darker reddish facial skin compared to M. assamensis, Goalpara. The species-specific traits of M. leucogenys were also compared, and the traits "chin and cheek whiskers", "darker dorsal coat" and "round penile" appearance have a striking resemblance to those of the M. munzala population of Tawang. Thus, the external morphological traits that are being used to distinguish these macaque species are highly variable even within the same species, and there is an urgent need to identify more precise species-specific morphological traits.

Orbitofrontal cortex (OFC) is implicated in value-based decision-making, but it is unclear how downstream circuits read out complex OFC responses into separate representations of the relevant variables to support distinct functions at specific times. We recorded from single OFC neurons while macaque monkeys made cost-benefit decisions. Using a novel analysis, we find separable neural dimensions that selectively represent the value, choice, and expected reward of the present and previous offers. The representations are generally stable during periods of behavioral relevance, then transition abruptly at key task events and between trials. Applying new statistical methods, we show that the sensitivity, specificity and stability of the representations are greater than expected from the population's low-level features-dimensionality and temporal smoothness-alone. The separability and stability suggest a mechanism-linear summation over static synaptic weights-by which downstream circuits can select for specific variables at specific times.

OBJECTIVES: The cranium is generally considered more reliable than the postcranium for assessing primate taxonomy, although recent research suggests that pelvic shape may be equally reliable. However, little research has focused on intrageneric taxonomic discrimination. Here, we test the relative taxonomic efficacy of the cranium and os coxa for differentiating two macaque species, with and without considering sexual dimorphism. MATERIALS AND METHODS: Geometric morphometric analyses were performed on cranial and os coxa landmarks for 119 adult Macaca fascicularis, M. mulatta, and Chlorocebus pygerythrus. Among-group shape variation was examined using canonical variates analyses. Cross-validated discriminant function analysis provided rates of correct group classification. Additionally, average morphological distances were compared with neutral genetic distances. RESULTS: Macaque species were clearly differentiated, both cranially and pelvically, when sex was not considered. Males were more often correctly classified based on the os coxa, while female classification rates were high for both morphologies. Female crania and male os coxa were differentiated approximately the same as genetic distance, while male crania were more similar (convergent), and female os coxa were more divergent than expected based on genetic distance. DISCUSSION: The hypothesis that cranial and os coxal shape can be used to discriminate among macaque species was supported. The cranium was better at differentiating females, while the os coxa was better at differentiating male macaques. Hence, there is no a priori reason for preferring the cranium when assessing intragenetic taxonomic relationships, but the effects of high levels of sexual dimorphism must be corrected for to accurately assess taxonomic signatures.

The activity budget is important for understanding behavioural variability and adaptation in primates. Assamese macaques (Macaca assamensis) found in the limestone forest of Guangxi Nonggang National Nature Reserve, Southwest China, primarily feed on young leaves of Bonia saxatilis (a shrubby, karst-endemic bamboo). To understand how a specific bamboo leaf-based diet and ecological factors affect activity budget, one group of Assamese macaques was studied using instantaneous scan sampling for 1 year. The macaques spent most of their time feeding (32.7 ± 5.4%), followed by resting (28.6 ± 6.3%), moving (28.6 ± 5.3%), grooming (8.0 ± 3.0%), playing (1.7 ± 1.6%) and other activities (0.4 ± 0.2%). Their activity budget was similar to that of typical frugivorous primates and bamboo-dominated primates, which spent more time on active behaviours (feeding and moving, 61.3 ± 6.0%) than on inactive behaviours (resting and grooming, 36.6 ± 6.4%). The macaques spent significantly more time resting during the fruit-lean season and more time moving during the fruit-rich season. Their activity budget was significantly affected by diet. Resting time increased with decreased fruit consumption, whereas moving time decreased with the increasing mature leaf consumption. Playing time decreased when the macaques consumed more young bamboo leaves. The activity budget was also influenced by food availability and climatic factors. Resting time increased with decreasing temperature, whereas moving time increased with increasing day length and young leaf availability. Grooming time increased with decreasing day length and increasing temperature, and playing time increased with increasing day length. Our findings provide evidence of the importance of diet, food availability, temperature and day length in coping with seasonal variation in ecological factors, highlighting the need to increase knowledge of the behavioural ecology of the Assamese macaques living in the unique limestone forest and to understand the influence of a bamboo-dominated diet and ecological factors on their survival.

Primate behavior can be responsive to the different ecological pressures associated with different habitats, as well as to the effects of direct and indirect anthropogenic disturbance. The karst forest ecosystem of South Sulawesi (Indonesia) represents one of the few intact forests available for residual populations of the moor macaque, but our understanding of its habitat use is limited. In the present study, this gap in knowledge was addressed by observing the activity and habitat use of two groups of moor macaques and by assessing the suitability of different habitats in the karst forest. Through a fine-scale vegetation analysis of 1 ha of forest in Bantimurung Bulusaraung National Park, we identified the presence of two distinct habitats that differ in terms of forest structure and composition. The karst plain forest (KPF) provided a greater abundance and diversity of food resources than the karst tower forest (KTF). In addition, anthropogenic disturbance was high in the KPF but low in the KTF. Behavioral data collected via group scans indicate that the macaques devoted more time to feeding activities when in the KPF, suggesting an ability to adjust their feeding behavior to meet their nutritional needs. However, the larger of the two groups used the food-rich KPF more than expected, implying that the KTF may represent a valuable refuge for the smaller group, as it is a less risky portion of its home range. The results of this study therefore provide novel information on the ecology of moor macaques and their habitats that can inform conservation planning for remnant populations.

Blastocystis is an enteral eukaryote with an omnipresent existence in animals and humans globally. Animals have been proposed to be a major reservoir for the transmission of Blastocystis to individuals due to their high prevalence and large amount of zoonotic subtypes. However, limited data on Blastocystis infection in experimental macaques in China exists. The objective of the current study was to investigate the frequency and subtypes of Blastocystis infection in macaques in southwestern China. A total of 505 fecal samples were collected from experimental macaques in Yunnan province and were analyzed by nested PCR and phylogenetic analyses on the basis of small subunit rRNA (SSU rRNA) gene fragments. A total of 235 specimens were positive for Blastocystis sp., and the prevalence of Blastocystis sp. was 46.5% (235/505). Significant differences in prevalence were also observed among the various species of macaques (P < 0.0133, df = 2, χ2 = 8.64) and the different feed types (P < 0.0093, df = 1, χ2 = 6.77). Moreover, three zoonotic subtypes, ST1, ST3, and ST5, were identified by DNA sequence analysis. There were mainly single subtype infections with some mixed subtype infections, and the predominant subtype was ST3. The results suggested a high prevalence and diversified subtypes in macaques in Yunnan province, southwestern China. Macaques are likely to be potential reservoirs capable of zoonotic transmission of Blastocystis sp. to humans. To our knowledge, this study is the first large-scale systematic analysis of Blastocystis sp. colonization in Yunnan province in the subtropics of China; these results contribute to the in-depth study of genetic characteristics and the prevention, control, and treatment of Blastocystis sp. in macaques in Yunnan province and other regions.

In nonhuman animals, individuals of the same sex and age differ in their behavior patterns consistently across time, comparable with human personality differences. To draw conclusions about the adaptive value of behavior traits, it is essential to study them in the wild where animals are subject to the ecological pressures that promoted the evolution of behavior strategies. This study was conducted in the Phu Khieo Wildlife Sanctuary, Thailand, on 4 groups of habituated wild Assamese macaques by observers who had familiarized themselves with the subjects over the course of an ongoing long-term study. We used a multimethod approach enabling the most comprehensive understanding of variation in stable interindividual differences in a species-typical ecological setting. We combined trait ratings (TRs), assessed with observer-report questionnaires (54-item Hominoid Personality Questionnaire) of 107 individuals of diverse age-sex classes, with behavior codings (BCs) of 24 adult males. We found male and female personality constructs to be congruent and examined reliability and construct validity. Combining trait rating and behavioral coding, we found two solutions with five factors to best describe the personality structure of the males: one structure comprised the dimensions GregariousnessBC, AggressivenessBC, SociabilityBC and VigilanceBC, complemented by a ConfidenceTR domain and the other structure OpportunismTR, ConfidenceTR, FriendlinessTR, ActivityTR complemented with VigianceBC. We discuss our findings with regard to the importance of construct validity and reproducibility in the context of method development and standardization in nonhuman animal personality research.
