RESUMO

BACKGROUND: Attachment material is one of the contributing factors to the degree of wear of the attachment components in mandibular implant-retained overdentures. The purpose of this in vitro study was to compare the wear behavior of 2 different attachment systems of dissimilar materials in mandibular implant-retained overdentures by qualitative and quantitative methods. METHODS: Two attachment systems of different materials were utilized (n = 16); Titach (Dental Evolutions Inc, Beverly Hills, CA, USA) with a titanium-to-titanium interface and Locator R-Tx (Zest Anchors Inc, Escondido, CA, USA) with a titanium-to-nylon interface. One thousand cycles of overdenture insertion and removal simulating 1-year clinical use were performed. All matrices were removed from the overdentures and all patrices were unscrewed from the implants for wear assessment quantitively using a stereomicroscope and qualitatively using a scanning electron microscope. Data were analyzed by using an independent sample t test. RESULTS: After cyclic loading, stereomicroscopic findings showed that the Titach group had statistically significant higher wear value than the Locator R-Tx group (p < 0.001). Moreover, scanning electron microscopy images showed noticeable abrasion in Titach patrix at the area of highest convexity. However, the Locator R-Tx matrix displayed an apparent tear of rubber inserts. CONCLUSIONS: Titach attachment with the titanium-to-titanium interface revealed more wear than Locator R-Tx attachment with the titanium-to-nylon interface. Thus, the type of attachment material influences the degree of wear of the attachment components.

RESUMO

This research studied the occurrence, risk assessment and metals associated with microplastic (MPs) in soil of different land-use types in the south west of Iran. One hundred samples were collected from topsoil and MPs were extracted using the floatation method. In total, 9258 MPs particles with mean of 92.85 ± 119.24 particles kg-1 were counted. The mean MPs abundance in urban soils was 2.8 and 3.2 times higher than in industrial and agricultural soils, respectively. Fragment (43%) and small MPs (100-250 µm; 41%), were the dominant shape and size, respectively. Four main polymer types including Poly Ethylene (High Density Poly Ethylene and Low Density Poly Ethylene), Nylon (PA), Poly Propylene (PP), and Poly Styrene (PS) were identified. Nylon (29%) and PE (29%) were dominant polymer types. MPs particles in soil contained different levels of metals such as Cd, Cr, Cu, Fe, Hg, Pb, Zn, and Y, except La and Yb. The mean concentrations of metals were higher in MPs than in soil. The indices of MPs-induced risk (Hstudyarea=16.8) showed a hazard level (III) in the study area. Pollution load index (PLILandusetype) showed hazard level (II) for urban soils and hazard level (I) for industrial and agriculture soils, respectively. Overall, risk index indicated high to extreme danger for MPs pollution in the study area. This is the comprehensive study on the occurrence of soil MPs and associated metals, which provides basic information for a further study concerning ecosystem health in Shiraz.

RESUMO

Microplastic pollution on bathing beaches threatens the health of human beings and coastal organisms. There is a lack of assessment on the level of microplastic pollution and the health risk associated with plastics. As one of the earliest open bathing beaches in China, Liandao is well known as the two high-quality beaches. However, little is known about the extent of microplastic pollution on these bathing beaches. Based on the analysis of microplastic pollution abundance, distribution, shape, size, color, and composition at the Liandao bathing beaches, this study puts forward a novel approach to comprehensively evaluate the microplastic pollution level and risk level by using the Nemerow pollution index (NPI) and polymer hazard index (PHI). The results show that the average abundance of microplastics on the Liandao bathing beaches is 135.42 ± 49.58 items/kg; the main shapes are fibers, fragments, and granules. Most of the microplastics are transparent, brown, and black, accounting for 71.54%, and they have an average particle size of 0.63 ± 0.43 mm. The main components are PE, PP, PS, PET, and nylon, of which nylon appears in the highest proportion (54.77%). The microplastic NPI and PHI values are 0.38 and 74.81, respectively, indicating that the pollution level and health risk index of microplastics on the Liandao bathing beaches are both low. With the increase in population and per capita consumption, plastic waste generated on land will continue to increase. Finally, this study puts forward some suggestions regarding microplastic monitoring, plastic waste management, and environmental attitudes and behavior.

RESUMO

The microfibers generated from Polyester and Nylon based materials during washing cause serious environmental pollution by both contaminating the aquatic environment and the livelihood of the underwater creatures as well. This study aims at investigating the microfiber-pollution in wash effluents collected from different regions of Kolkata which is believed to be one of the microfiber-polluted cities in the South-east Asia in recent times. In this work, packed bed microfiltration (PBMF) was adopted in an economic and eco-friendly manner to arrest adequate amounts of microfibers and non-biodegradable matters present in the water samples collected from different regions of Kolkata and its surrounding areas. Moreover, effective parameters such as packed bed height to diameter ratio (H/D), mess size of the adopted filtration unit were varied from 0.71 to 2.85 and 60 to 100, respectively to understand the efficacy of the approach and to justify the potential of such alternative in order to alleviate the concern as well. The present study reveals that the microfiltration efficiency of the proposed PBMF unit was achieved maximum 93.5% for sample A and 92.2% for sample D respectively to reduce the microfiber count from 7614 to 543 in an hour operation at a flow rate of 60 L h-1. Besides, the cost of such system was found to be promising as much as 5 US$ on a yearly basis.

RESUMO

In this study, a novel anthropomorphic head phantom for quantitative image quality assessment in cone beam computed tomography (CBCT) is proposed. The phantom is composed of tissue equivalent materials (TEMs) which are suitable for cost-efficient fabrication methods such as silicone casting and 3D printing. A monocalcium phosphate/gypsum mixture (MCPHG), nylon and a silyl modified polymer gel (SMP) are proposed as bone, muscle and brain equivalent materials respectively. The TEMs were evaluated for their radiodensity in terms of Hounsfield Units (HU) and their x-ray scatter characteristics. The median radiodensity and inter quartile range (IQR) of the MCPHG and SMP were found to be within the range of the theoretical radiodensity for bone and brain tissue: 922 (IQR = 156) and 47 (IQR = 7) HU respectively. The median radiodensity of nylon was slightly outside of the HU range of muscle tissue, but within the HU range of a combination of muscle and adipose tissue: -18 (IQR = 40) HU. The median ratios between the measured scatter characteristics and simulated tissues were between 0.84 and 1.13 (IQR between 0.05 and 0.14). The preliminary results of this study show that the proposed design and TEMs are potentially suitable for the fabrication of a cost-efficient anthropomorphic head phantom for quantitative image quality assessment in CT or CBCT.

RESUMO

Charcoal is a popular form of biofuel embraced for domestic and industrial purposes. However, the use of Charcoal has some associated challenges, such as the required charcoal pot and setting it into the fire at first by using Charcoal-Ignition-Aiders (CIA) (e.g. discarded paper, nylon, rubber, plastics, petrol, the residue of processed palm oil, maise cob, wood, and kerosene). Coupled with the chemical properties of Charcoal, the resulting gases from CIA are capable of polluting the environment with perceived Adverse-Health-Implications (AHI) on the ecosystem. Therefore, this study conducted a safety assessment of charcoal biofuel usage and the effects of common CIA on combustion indices. This study followed standard methods and the use of peculiar equipment. This study established that Charcoal is commonly used in the studied area because it is cheap, readily available and requires less technical know-how. Considering the combustion indices, using paper as a CIA generated the lowest carbon monoxide (CO) value, 28.1 ppm, with 3,434.54 ppm volatile organic compound, VOC. Compared with the ACGIH standard permissible exposure level of ≤ 30 ppm, the paper gave a lesser CO value of 28.10 ppm among all the CIA. At the same time, all the CIA recorded higher VOC compared with EPA standard permissible exposure level of ≤ 15 ppm. ANOVA analysis conducted on the socio-demographic profile of the respondents, cooking attributes of the respondents, and use of charcoal pot types by the respondents in Zone 1, Zone 2, and Zone 3 gave p-values of 0.032, 0.028, and 0.039, respectively. These imply significant differences within the zones in each of the indices. The average energy content reported for charcoals sourced from oak trees, afara, obeche, mahogany, and iroko woods is 3,2149 kJ/kg compared to the lower ones. Therefore, this study recommended using these charcoals alongside discarded paper as CIA because they are a better combination to reduce AHI.

RESUMO

Laboratories and their criminal justice systems are confronted with challenges for implementing new technologies, practices, and policies even when there appears to be demonstrative benefits to operational performance. Impacting decisions are the often higher costs associated with, for example, new technologies, limited current budgets, and making hard decisions on what to sacrifice to take on the seemingly better approach. A prospective cost-benefit analysis (CBA) could help an agency better formulate its strategies and plans and more importantly delineate how a relatively small increase to take on, for example, a new technology can have large impact on the system (e.g., the agency, other agencies, victims and families, and taxpayers). To demonstrate the process and potential value a CBA was performed on the use of an alternate and more expensive swab with reported better DNA yield and being certified human DNA free (i.e., nylon 4N6FLOQSwabs®), versus the traditional less costly swab (i.e., cotton swab). Assumptions are described, potential underestimates and overestimates noted, different values applied (for low and modest to high), and potential benefits (monetary and qualitative) presented. The overall outcome is that the cost of using the more expensive technology pales compared with the potential tangible and intangible benefits. This approach could be a guide for laboratories (and associated criminal justice systems) worldwide to support increased funding, although the costs and benefits may vary locally and for different technologies, practices, and policies. With well-developed CBAs, goals of providing the best services to support the criminal justice system and society can be attained.

RESUMO

Microplastic pollution and its potential impacts on humans become a global concern. This study is the first study examining the microplastic ingestion in the commercially important species Oncorhynchus mykiss Rainbow trout (Walbaum, 1792), Sparus aurata Gilthead seabream Linnaeus, 1758, and Dicentrarchus labrax European seabass (Linnaeus, 1758) from Turkey. The occurrence frequency of microplastic (MP)in the gastrointestinal tract (GIT) was varied between 50 and 63 %. The highest MP abundance in the GIT was estimated in Rainbow trout (1.2 MPs individual-1) followed by European seabass (0.95 MPs individual-1) and Gilthead seabream (0.8 MPs individual-1). Most of the extracted microplastic particles were black (61 %) and blue (27 %) in color and fiber (80 %) in shape. Major identified polymers were polyethylene (25 %), polyester (20 %), polyamide (10 %). Since fish consumption is an important route for MPs, results light up the danger potential for humans. This study will fill the information gap in Turkey and show the necessity of protection measures in aquaculture industry.

RESUMO

In aquatic environments, microplastics (MPs) are pervasive which could have a considerable negative impact on the environment, organisms and pose a risk to human health. However, knowledge about the exposure and ecological risk of MPs in the coastal ecosystems of developing countries is limited. In this study, we analyzed salt samples from five commonly consumed processed and unprocessed sea salts of different commercial brands originated from 15 salt pans in Bangladesh to assess the abundance, characteristics and potential risks of MPs. The quantities of MPs in unprocessed salts (average 195 ± 56 item/kg) were higher than those in the processed salts (average 157 ± 34 item/kg). One-way analysis of variance (ANOVA) showed significant (p < 0.05) differences among the average numbers of MPs in both processed and unprocessed salts. MP levels in this study were 2-3 times higher than those reported from some other countries. Fiber-shaped and transparent MPs were dominant in both cases. MPs less than 0.5 mm in size were the most abundant in both unprocessed (58.2%) and processed (62.2%) salts. Fourier-transform infrared spectroscopy (FTIR) analysis confirmed five types of polymers, including polyethylene terephthalate (PET-35%), polypropylene (PP-27.5%), polyethylene (PE-25%), polystyrene (PS-10%), and Nylon (2.5%) in the studied salts. The sea salts were classified as potential hazard index (PHI) levels IV to V, indicating serious MP contamination, whereas potential ecological risk factor (Ei), potential ecological risk index (RI), and pollutant load index (PLI) indicated moderate levels of pollution of MPs. Domestic and municipal wastewater effluents to Bay of Bengal and fishing activities may attributed to presence of MPs in the sea salt. These findings can be used by consumers, salt industries and policy makers to reduce MPs levels during consumption, production and policymaking.

RESUMO

This international scale study measured the prevalence of indoor microplastics (MPs) in deposited dust in 108 homes from 29 countries over a 1-month period. Dust borne MPs shape, colour, and length were determined using microscopy and the composition measured using µFTIR. Human health exposure and risk was assessed along with residential factors associated with MPs via a participant questionnaire. Samples were categorised according to each country's gross national income (GNI). Synthetic polymers dominated in low income (LI) (39%) and high income (HI) (46%) while natural fibres were the most prevalent in medium income (MI) (43%) countries. Composition and statistical analysis showed that the main sources of MPs and dust were predominantly from indoor sources. Across all GNI countries, greater vacuuming frequency was associated with lower MPs loading. High income country samples returned higher proportions of polyamides and polyester fibres, whereas in LI countries polyurethane was the most prominent MPs fibre. Exposure modelling showed infants (0-2 years) were exposed to the highest MPs dose through inhalation (4.5 × 10-5 ± 3 × 10-5) and ingestion (3.24 × 10-2 ± 3.14 × 10-2) mg/kg-Bw/day. Health risk analysis of constituent monomers of polymers indicates cancer incidence was estimated at 4.1-8.7 per million persons across age groups. This study's analysis showed socio-economic factors and age were dominant variables in determining dose and associated health outcomes of MPs in household dust.

RESUMO

This study aimed to introduce new composites, containing polyamide-6 (PA6) and lead monoxide (PbO), to protect against ionizing photon sources used for diagnostic and therapeutic purposes. Five composites, containing various weight percentages of PbO filler (0, 5, 10, 20, and 50%), were developed in this study. Initially, the numerical attenuation value was estimated using the XMuDat program by calculating the mass attenuation coefficients at different energy levels. Next, the samples were synthesized based on the melt-mixing method in a laboratory mixing extruder. Their characteristics were also determined by scanning electron microscopy (SEM), energy dispersive X-ray (EDX) analysis, X-ray diffraction (XRD), and thermogravimetric analysis (TGA). Finally, experimental radiation attenuation tests were carried out. Based on the SEM results, the acceptable filler weight percentage was up to 20%. However, substantial aggregates were formed at the highest weight percentage. The results of XRD analysis showed a higher tendency for crystallization by decreasing the amorphous area while increasing the filler weight percentage. Moreover, the mass loss rate was monitored at different temperatures, revealing that the filler incorporation improved the thermal durability of the samples. The radiation results showed a good agreement between the experimental and computational data, except when aggregates formation was substantial. The experimental data revealed that when the lead weight percentage increased from 0% (crude PA6) to 50%, the half-value layer decreased from 3.13 to 0.17 cm at an energy level of 59 keV and from 7.28 to 4.97 cm at an energy level of 662 keV. Following the studied mechanism, the superiority of lead/polyamide composites can be found in the high adsorption of photon radiation at low energies (E < 0.20 MeV) and significant attenuation at medium and higher energies. Considering these promising results, the shielding properties of these composites can be further analyzed via more practical investigations.

RESUMO

BACKGROUND: Ischemic preconditioning of the forehead flap prior to pedicle division helps to improve angiogenesis. Clamping the pedicle by a clamped rubber band with vessel forceps is often clinically applied. However, the severe pain and unstable blocking effect influenced the preconditioning process. In this study, we described an innovative device designed for ischemic preconditioning and compared its efficacy with the clamped rubber band. METHODS: The device consists of a self-locking nylon cable tie with a buckle and a rubber tube. The rubber tube is fed over the cable tie to act as a soft outer lining and the cable tie is tightened across the pedicle to block the perfusion for ischemic preconditioning. This device and the standard clamped rubber band were applied respectively before division surgery. The constriction effect, reliability, reproducibility, and the patients' pain tolerance were compared. RESULTS: A total of 20 forehead flaps were included. The cable tie had less incidence of loosening (7.7% vs. 16.6%, p < 0.05) and maintained the pressure more effectively. The pain score for the nylon cable tie was significantly lower than the clamped rubber band (4.25 ± 1.02 vs. 6.75 ± 1.12, p < 0.05), especially for 10 pediatric patients (4.50 ± 0.85 vs. 8.10 ± 1.20, p < 0.01). All 20 pedicles were successfully divided at 19 to 22 days with no surgical complications. CONCLUSION: Compared with the clamped rubber band, the cable tie produces a more reliable and reproducible ischemic preconditioning effect. It is also better tolerated by the patients. Therefore, we recommend using the nylon cable tie as the preferred device for ischemic preconditioning of the forehead flap.

RESUMO

BACKGROUND: In order to mitigate the challenges in microsurgical skill acquisition and training, especially in the COVID-19 era, we devised a novel microsurgical telementoring protocol for imparting microsurgical skill training in a socially distanced setting. We objectively analyzed its feasibility among neurosurgical trainees. METHODS: In a controlled experimental design, 8 residents at different stages of their tenure participated in a lazy glass microsurgical simulator-based telementoring exercise. Microsuturing with 4-0 silk, 10-0 nylon on silastic sheets, and eggshell peeling tasks were performed by the residents prior to and after a telementoring session by a panel of 4 neurosurgical experts. Impact of telementoring was assessed in terms of surgical accuracy, efficiency, and dexterity by providing objective (Performance score [PS]), subjective (Neurosurgery Education and Training School [NETS] score), and cumulative scores (CS). Subgroup analysis was performed to assess the impact at different stages of residency. RESULTS: PS, NETS score, and CS were significantly improved by telementoring sessions for 10-0 nylon micro-suturing (P < 0.001), and egg-hell peeling tasks (P < 0.01). PS and CS improved significantly (P = 0.01) after telementoring sessions for 4-0 silk microsuturing. Both pre- and post-training CS were similar across the 2 subgroups PGY 1-4 and PGY 5-6 (P > 0.05). CONCLUSIONS: Telementoring is a viable alternative for neurosurgical resident training in the COVID-19 era, where reduction in elective surgeries and social distancing norms preclude conventional teaching. Lazy glass microsurgical simulator-based structured telementoring protocol is a cost-effective tool to augment surgical proficiency and finesse, irrespective of stage of residency.

RESUMO

OBJECTIVE: To report the use of nylon cable ties (NCT) for omentectomy in the horse. STUDY DESIGN: Experimental study. ANIMALS: Eight healthy adult horses. METHODS: Horses underwent nylon cable tie (NCT) ligation of the greater omentum after ventral midline celiotomy. The time required to complete the omentectomy was recorded. Horses were recovered for 14 days before repeat celiotomy, adhesions assessment, and histological examination of the omentectomy site using a proposed histologic grading scheme. The total time for omentectomy procedure and histologic score was assessed for normality. Data are expressed as mean ± standard deviation. RESULTS: NCT ligation provided sufficient hemostasis to complete the omentectomy (28 ± 15 s), without rescue ligation. No gross evidence of intra-abdominal adhesion or morbidity was associated with the omentectomy site 14 days after surgery. NCT were intact at the site of application, covered with smooth fibrous connective tissue. Adiponecrosis with minimal inflammation and fibrovascular occlusion of omental vessels was present at the surgical site. Mild inflammation was present at the NCT-tissue interface. CONCLUSION: The use of NCT resulted in fast and effective omentectomy in healthy horses without short-term evidence of inflammatory reaction or intra-abdominal adhesion. CLINICAL SIGNIFICANCE: The described technique provides an alternative for omentectomy in healthy adult horses.

RESUMO

Small molecules in food contact materials may migrate into food during their contact. To extensively analyze the migrants, non-targeted screening is needed to detect the migrants. The migrants' detection is difficult because of the complexity and the trace amount of the migrants. In this work, the dissolution precipitation method was used to extract small molecules in Polyamide (PA) kitchenware. The extract solutions were screened by ultra-performance liquid chromatography-quadrupole time-of-flight-mass spectrometry (UPLC-QTOF-MS) for non-targeted analysis and 64 different small molecules in materials were identified through the screening of a self-built database. Then, migration tests were performed to analyze migrants in food simulants. It suggests that the abundance of PA oligomers was the highest in migrants. The risk assessment of migrants revealed that the exposure of most migrants was at a safer level unlike the exposure of PA oligomers that exceeded their threshold of toxicological concern (TTC).

RESUMO

Textiles damage analysis is a very valuable tool in forensic investigations. However, to date, very little research has been carried out to understand the impact of bullet causing damages to clothing. According to the review of the most recent scientific papers, the frictional heating and crushing action of a bullet passing through synthetic fibres cause a unique transformation in their ends called mushroom-shaped morphology. In this study, the textile remains of six individuals executed during the first decade of the Chilean military dictatorship period (1973-1990) were analysed. The purpose was to examine their clothing in order to describe the fibre defects in the bullet holes. The fibres were directly observed using two different models of stereomicroscopy (MZ16A and EZ4D, Leica Microsystem Ltd., Wetzlar, Germany) and through a combination of transmitted, oblique and co-axial illumination (with Leica DFC500 Digital Camera), at × 230 and at a resolution of up to 840 Lp/mm. The mushroom-shaped morphology, along with rupturing of yarns, fibrillation or splitting of fibres, was observed in the bullet holes. Although the mushroom-shaped is a useful pattern for bullet hole identification in synthetic fibres, further research needs to be performed for developing a sounder interpretational framework of this type of forensic evidence.

RESUMO

BACKGROUND: Surgeons use absorbable and nonabsorbable sutures for epidermal wound closure. No large, randomized studies have compared the effect of these suture types on facial scar appearance. OBJECTIVE: To assess postsurgical facial scar appearance using either rapidly absorbable polyglactin 910 or nylon for epidermal closure. METHODS: Randomized, blinded, split-scar clinical trial. A total of 105 patients with facial wounds resulting from Mohs micrographic surgery excisions were randomly assigned for epidermal closure with rapidly absorbable 5-0 polyglactin 910 (Vicryl Rapide) on one half of the repair and 5-0 nylon (Ethilon) on the other half. Two physicians (1 dermatologist and 1 plastic surgeon), unaware of the original suture location, examined photographs of each healed wound at 6 months after surgery and graded the appearance of each half of the scar using the visual analog scale, wound evaluation scale, and Stony Brook Scar Evaluation Scale. RESULTS: At 6 months, there was no significant difference in the combined mean (standard deviation) visual analog scale scores (83.1 [14.2] and 83.0 [13.7]), Stony Brook Scar Evaluation Scale scores (4.3 [0.9] and 4.4 [0.9]), or wound evaluation scale scores (5.3 [1.1] and 5.2 [1.1]) for rapidly absorbable polyglactin 910 versus nylon (P = .72, .57, and .21, respectively). LIMITATIONS: Single institution. CONCLUSIONS: Both rapidly absorbable polyglactin 910 and nylon sutures placed through the epidermis resulted in an equivalent photographic appearance of facial scars at 6 months after surgery.

RESUMO

Transplantation of pancreatic islets is becoming a promising therapy for people with type I diabetes. In this study, we present a compact fluidic system that enables assessment of islet functionality ex vivo for efficient islet transplantation. The fluidic system includes a micromesh sheet-embedded chip. Islets can be loaded easily on the micromesh sheet and observed clearly by microscopy. Islets on the mesh sheet mainly remained in place during perfusion and did not get damaged by hydraulic pressure because of high porosity of the micromesh sheet. The fluidic system was assembled with a sample fraction chip of polydimethylsiloxane. The chip includes a channel and columns, both having surfaces that were super-hydrophilized so that solutions could flow smoothly within the chip by gravity. Using mouse pancreatic islets, a dynamic glucose-stimulated insulin secretion test was performed to examine the performance of the fluidic system. The system successfully analyzed levels and patterns of insulin secretion upon exposure of the islets to low- and high-glucose solutions in turns, thus demonstrating its capacity to assess islet functions more easily and cost-effectively.

RESUMO

In the last few years, several studies have investigated microplastics (MPs) in marine ecosystems, but data monitoring and assessing the occurrence in freshwater environments are still scarce. The present study aims to investigate the occurrence, distribution, and chemical composition of MP pollution in Vesijärvi lake and Pikku Vesijärvi pond close to the city of Lahti (Finland) in winter. Sediment, snow, and ice core samples were collected near the shore of these two aquatic systems. MPs were analysed and identified by a non-destructive method using Fourier transform infrared spectroscopy (FTIR) 2D imaging. The mean concentrations of MPs detected in sediment, snow, and ice samples were 395.5 ± 90.7 MPs/kg, 117.1 ± 18.4 MPs/L, and 7.8 ± 1.2 MPs/L, respectively. FTIR results showed the predominant abundance of microplastics, such as polyamides (up to 53.3%), polyethylene and polypropylene (up to 17.1%), and natural fragments such as cellulose (up to 45.8%) and wool (up 18.8%) in the same size range. The potential release of MPs arising from stormwaters and sport and recreational activities was evidenced.

RESUMO

Developing a mechanistic understanding of protein dynamics and conformational changes at polymer interfaces is critical for a range of processes including industrial protein separations. Salting out is one example of a procedure that is ubiquitous in protein separations yet is optimized empirically because there is no mechanistic description of the underlying interactions that would allow predictive modeling. Here, we investigate peak narrowing in a model transferrin-nylon system under salting out conditions using a combination of single-molecule tracking and ensemble separations. Distinct surface transport modes and protein conformational changes at the negatively charged nylon interface are quantified as a function of salt concentration. Single-molecule kinetics relate macroscale improvements in chromatographic peak broadening with microscale distributions of surface interaction mechanisms such as continuous-time random walks and simple adsorption-desorption. Monte Carlo simulations underpinned by the stochastic theory of chromatography are performed using kinetic data extracted from single-molecule observations. Simulations agree with experiment, revealing a decrease in peak broadening as the salt concentration increases. The results suggest that chemical modifications to membranes that decrease the probability of surface random walks could reduce peak broadening in full-scale protein separations. More broadly, this work represents a proof of concept for combining single-molecule experiments and a mechanistic theory to improve costly and time-consuming empirical methods of optimization.
