Tectonic lakes are among the most geologically fascinating and environmentally versatile hydrobiological systems found on the earth's surface. We conducted a study on the limnology of Tasek Lake, a tectonic lake located in the Indo-Burma Province of the South Asian region. Physico-chemical parameters of the lake's water along with its plankton were considered for the study. Their relationship was analysed by understanding their seasonal variations and through linear regression models. The water quality index (WQI), plankton diversity indices and canonical correspondence analysis (CCA) were computed. The ichthyofaunal diversity was also studied to get an insight into the lake's fishery potential. A preliminary assessment on the economic feasibility of converting Tasek Lake into a fishery was also completed. Results indicate moderate eutrophication in the lake and the plankton population is observed to be rich and abundant. The WQI value confirms the water to be of "very poor" quality. The CCA was done to analyze the relationships of physico-chemical parameters with months and seasons, and the relation between seasons and plankton assemblages. Results corroborate the results of WQI. Identified fish population suggest ample fishery potential of the lake. The economic assessment reveals that in order to maintain the ecological sustainability of the lake, it should be transformed into a recreational fishery, following a catch-and-release model. The study calls for urgent restoration of the lake so that not only its pristine ecology is survived but also its fishery potential is sustainably harnessed and local livelihood is improved.

BACKGROUND: Resveratrol (3,5,4'-trihydroxy-trans-stilbene) is a natural phytoalexin synthesized by plants in response to stress. This compound has several beneficial documented properties, namely anti-inflammatory, antioxidant, neuroprotective and antimicrobial activities. In this study the antimicrobial activity of resveratrol against Listeria monocytogenes and Listeria innocua was investigated. RESULTS: Resveratrol had a minimum inhibitory concentration of 200 µg mL(-1) for the tested strains, with time-kill curves demonstrating bacteriostatic activity. Inhibition of biofilm formation was also assessed, with resveratrol strongly inhibiting biofilm formation by both species even at subinhibitory concentrations. Overall, resveratrol showed antimicrobial properties on planktonic cells and on biofilm formation ability. Considering the potential use of resveratrol as a food preservative, the antimicrobial efficacy of resveratrol in food was studied in milk, lettuce leaf model and chicken juice. Resveratrol retained greater efficacy in both lettuce leaf model and chicken juice, but milk had a negative impact on its antilisterial activity, indicating a possible reduction of resveratrol availability in milk. CONCLUSION: This study reinforces resveratrol as an antimicrobial agent, pointing out its antibiofilm activity and its potential use as preservative in some food matrices. © 2016 Society of Chemical Industry.

The unwanted impacts of non-indigenous species have become one of the major ecological and economic threats to aquatic ecosystems worldwide. Assessing the potential dispersal and colonization of non-indigenous species is necessary to prevent or reduce deleterious effects that may lead to ecosystem degradation and a range of economic impacts. A three dimensional (3D) numerical model has been developed to evaluate the local dispersal of the planktonic larvae of an invasive bivalve, Asian clam (Corbicula fluminea), by passive hydraulic transport in Lake Tahoe, USA. The probability of dispersal of Asian clam larvae from the existing high density populations to novel habitats is determined by the magnitude and timing of strong wind events. The probability of colonization of new near-shore areas outside the existing beds is low, but sensitive to the larvae settling velocity ws. High larvae mortality was observed due to settling in unsuitable deep habitats. The impact of UV-radiation during the pelagic stages, on the Asian clam mortality was low. This work provides a quantification of the number of propagules that may be successfully transported as a result of natural processes and in function of population size. The knowledge and understanding of the relative contribution of different dispersal pathways, may directly inform decision-making and resource allocation associated with invasive species management.

Studies of the Slovakian Danube have indicated profound changes in the structures of several invertebrate assemblages in the section of the river bypassed by the Gabcíkovo hydroelectric project in 1992 and in adjacent floodplain water bodies. The present study investigates the relationship between hydrological connectivity and species diversity in the old main channel and adjacent left-bank water bodies, using resident cladocerans as indicators of the ecological integrity of the river-floodplain system. The work aimed to quantify cladoceran habitat preferences using habitat values (HV) and indication weights (IW) calculated from data accumulated during long-term monitoring of planktonic communities of the Danube floodplain (1,840.5-1,804 rkm) and supplemented with data from the literature. Although not listed as an indicator group in the European Union Water Framework Directive, cladocerans are shown to be highly apposite in the ecological assessment of river-floodplain systems. Of 71 recorded cladoceran species, 19 taxa showed a preference for eupotamal habitats, 24 species preferred eupotamal B/parapotamal waters, and 28 cladocerans were found to prefer the plesiopotamal/paleopotamal habitat type. Identified habitat types follow a gradient of hydrological connectivity with the main river channel, ranging from the eupotamal to more or less isolated floodplain water bodies. A secondary aim was to use the calculated values to assess the ecological integrity of the river-floodplain system. The key element of this procedure is the calculation of the Floodplain Index (FI) (Chovanec et al. Large Rivers, 15(1-4), 169-185 2005) from the summation of the habitat values and indication weights of all species present at the sampling sites. Calculated index values indicate the extent of disruption to lateral connectivity in the floodplain area.

This study was designed to evaluate the lytic activity of bacteriophage P22 against Salmonella Typhimurium ATCC 19585 (Salmonella Typhimurium P22(-)) at various multiplicities of infections (MOIs), the susceptibility of preattached Salmonella cells against bacteriophage P22, and the effect of P22-mediated bacterial lysates (extracellular DNA) on the attachment ability of Listeria monocytogenes ATCC 7644 and enterohemorrhagic Escherichia coli ATCC 700927 to surfaces. The numbers of attached Salmonella Typhimurium P22(-) cells were effectively reduced to below the detection limit (1 log CFU/ml) at the fixed inoculum levels of 3 × 10(-) CFU/ml (MOI = 3.12) and 3 × 10(3) CFU/ml (MOI = 4.12) by bacteriophage P22. The attached Salmonella Typhimurium P22(-) cells remained more than 2 log CFU/ml, with increasing inoculum levels from 3 × 10(4) to 3 × 10(7) CFU/ml infected with 4 × 10(8) PFU/ml of P22. The number of preattached Salmonella Typhimurium P22(-) cells was noticeably reduced by 2.72 log in the presence of P22. The highest specific attachment ability values for Salmonella Typhimurium P22(-), Salmonella Typhimurium ATCC 23555 carrying P22 prophage (Salmonella Typhimurium P22(+)), L. monocytogenes, and enterohemorrhagic E. coli were 2.09, 1.06, 1.86, and 1.08, respectively, in the bacteriophage-mediated cell-free supernatants (CFS) containing high amounts of extracellular DNA. These results suggest that bacteriophages could potentially be used to effectively eliminate planktonic and preattached Salmonella Typhimurium P22(-) cells with increasing MOI. However, further research is needed to understand the role of bacteriophage-induced lysates in bacterial attachment, which can provide useful information for the therapeutic use of bacteriophage in the food system.

The present study deals with the limnobiotic status of three selected lakes of Himachal Pradesh using physicochemical and biological parameters (especially phytoplankton and zooplankton) over a period of 2 years. One hundred forty-eight species belonging to nine groups of phytoplankton and 79 species belonging to five groups of zooplankton were identified from the lakes. Trophic level and the pollution status of the lakes were assessed upon the basis of Shannon diversity index (H'), species richness index (S), and physicochemical parameters. Plankton population size was correlated with biotic and abiotic parameters (pH, alkalinity, temperature, dissolved oxygen, transparency, phosphate, chloride, and nitrate). The present investigation revealed that the distribution of plankton species depended upon the physicochemical parameters of the environment. Based on water quality standards given by the Central Pollution Control Board, the water quality was between "A-B" at Prashar wetland, "C-D" at Kuntbhyog Lake, and "D-E" at Rewalsar Lake. The results from the present study indicated that the potential of planktons as bioindicators of trophic status is very high.

World consumption of seafood continues to rise, but the seas and oceans are already over-exploited. Land-based (saline) aquaculture may offer a sustainable way to meet the growing demand for fish and shellfish. A major problem of aquaculture is nutrient waste, as most of the nutrients added through feed are released into the environment in dissolved form. Wetlands are nature's water purifiers. Constructed wetlands are commonly used to treat contaminated freshwater effluent. Experience with saline systems is more limited. This paper explores the potential of constructed saline wetlands for treating the nutrient-rich discharge from land-based saline aquaculture systems. The primary function of constructed wetlands is water purification, but other ancillary benefits can also be incorporated into treatment wetland designs. Marsh vegetation enhances landscape beauty and plant diversity, and wetlands may offer habitat for fauna and recreational areas. Various approaches can be taken in utilizing plants (halophytes, macro-algae, micro-algae) in the treatment of saline aquaculture effluent. Their strengths and weaknesses are reviewed here, and a conceptual framework is presented that takes into account economic and ecological benefits as well as spatial constraints. Use of the framework is demonstrated for assessing various saline aquaculture systems in the southwestern delta region of the Netherlands.

AIMS: To evaluate the antimicrobial activities of aspirin, EDTA and an aspirin-EDTA (A-EDTA) combination against Pseudomonas aeruginosa, Escherichia coli and Candida albicans in planktonic and biofilm cultures. METHODS AND RESULTS: Minimal inhibitory concentrations (MIC) and minimal biocidal concentrations (MBC) were determined using twofold broth microdilution and viable counting methods, respectively. Aspirin's recorded MIC values ranged from 1.2 to 2.7 mg ml(-1). Checkerboard assay demonstrated a synergism in antimicrobial activity upon combination. Aspirin's minimal biofilm eradication concentration values (MBEC) against the established biofilms ranged between 1.35 and 3.83 mg ml(-1). A complete eradication of bacterial biofilms was achieved after a 4-h treatment with the A-EDTA combination. CONCLUSION: Both aspirin and EDTA possess broad-spectrum antimicrobial activity for both planktonic and biofilm cultures. Aspirin used at the MBEC for 24 h was successful in eradicating P. aeruginosa, E. coli and C. albicans biofilms established on abiotic surfaces. Moreover, the exposure to the A-EDTA combination (4 h) effected complete bacterial biofilm eradication. SIGNIFICANCE AND IMPACT OF THE STUDY: There is a continuous need for the discovery of new antimicrobial agents. Aspirin and EDTA are 'nonantibiotic drugs', the combination of which can be used successfully to treat and eradicate biofilms established on abiotic surfaces.

Monitoring of biofilms subjected to different operating conditions was performed using a flow cell system. The system was fed by chlorine-free tap water, with and without added nutrients (0.5 mg l(-1) carbon, 0.1 mg l(-1) nitrogen and 0.01 mg l(-1) phosphorus), and biofilms were grown on polyvinyl chloride (PVC) and stainless steel (SS) coupons, both in laminar and turbulent flow. The parameters analysed were culturable cells, using R2A, and total bacteria, which was assessed using the 4,6-diamino-2-phenylindole (DAPI) staining method. The impact of the different operating conditions in the studied parameters was established using Multivariate Analysis of Variance (MANOVA). From the most relevant to the least relevant factor, the total and culturable bacteria in biofilms increased due to the addition of nutrients to water (F = 20.005; p < 0.001); the use of turbulent (Re = 11000) instead of laminar (Re = 2000) hydrodynamic flows (F = 9.173; p < 0.001); and the use of PVC instead of SS as the support material (F = 2.848; p = 0.060). Interactions between these conditions, namely between surface and flow (F = 8.235; p < 0.001) and also flow and nutrients (F = 5.498; p < 0.05) have also proved to significantly influence biofilm formation. This work highlights the need for a deeper understanding of how the large spectrum of conditions interact and affect biofilm formation potential and accumulation with the final purpose of predicting the total and culturable bacteria attached to real drinking water distribution pipes based on the system characteristics.

OBJECTIVES: To quantitatively compare the antibiotic susceptibility of biofilms formed by the coagulase-negative staphylococci (CoNS) Staphylococcus epidermidis and Staphylococcus haemolyticus with the susceptibility of planktonic cultures. METHODS: Several CoNS strains were grown planktonically or as biofilms to determine the effect of the mode of growth on the level of susceptibility to antibiotics with different mechanisms of action. The utility of a new, rapid colorimetric method that is based on the reduction of a tetrazolium salt (XTT) to measure cell viability was tested by comparison with standard bacterial enumeration techniques. A 6 h kinetic study was performed using dicloxacillin, cefazolin, vancomycin, tetracycline and rifampicin at the peak serum concentration of each antibiotic. RESULTS: In planktonic cells, inhibitors of cell wall synthesis were highly effective over a 3 h period. Biofilms were much less susceptible than planktonic cultures to all antibiotics tested, particularly inhibitors of cell wall synthesis. The susceptibility to inhibitors of protein and RNA synthesis was affected by the biofilm phenotype to a lesser degree. Standard bacterial enumeration techniques and the XTT method produced equivalent results both in biofilms and planktonic assays. CONCLUSIONS: This study provides a more accurate comparison between the antibiotic susceptibilities of planktonic versus biofilm populations, because the cell densities in the two populations were similar and because we measured the concentration required to inhibit bacterial metabolism rather than to eradicate the entire bacterial population. While the biofilm phenotype is highly resistant to antibiotics that target cell wall synthesis, it is fairly susceptible to antibiotics that target RNA and protein synthesis.

Flow cytometry was used to characterize bacterioplankton from two tropical environments in Brazil: the eutrophic Guanabara Bay and the oligotrophic southwest Atlantic Ocean. Bacterial abundance was evaluated by flow cytometry, and cells were stained with SYTO 13, allowing demonstration of differences in nucleic acid content. Bacterial production was also evaluated by means of 3H-leucine incorporation. Bacterial numbers were different for both sites. In Atlantic Ocean samples, we found a maximum of 5.50 x 10(5) cells ml(-1), and low nucleic acid content organisms predominated. In Guanabara Bay, bacterial numbers were one order of magnitude higher than in the ocean, and they varied from outer bay (1.01 x 10(6) cells ml(-1)) to inner bay (6.90 x 10(6) cells ml(-1)). Bacterial activity in ocean samples varied from 4.6 to 126 ng C l(-1) h(-1), while in the bay, mean values ranged from 1.95 microg C l(-1) h(-1) (outer bay) to 7.35 microg C l(-1) h(-1) (inner bay). Values found for both parameters are characteristic of different trophic situations. These results illustrate the utility of cytometric analyses of bacterioplankton populations in characterizing their large spatial and temporal scales of distribution in aquatic ecosystems.

During the autumn of 1991, numerous seabird fatalities in Monterey Bay, California, led to the discovery of a new domoic acid-producing diatom, Pseudonitzschia australis. Since this initial event, sizable populations of P. australis, as well as other likely toxin producers, P. pungens f. multiseries and P. pseudodelicatissima, have occurred biannually in Monterey Bay. Using the highly sensitive FMOC-HPLC method, we detected domoic acid whenever Pseudonitzschia australis was found in the plankton, even at densities as low as 4.0 x 10(3) cells/L. Based on correlations of domoic acid and P. australis abundances and the overwhelming biovolume dominance of P. australis, we conclude that P. australis has been the major domoic acid producer during the period of our study. Our study suggests that P. australis cells may always be toxic in natural populations and that toxin concentrations on a per cell basis have no statistically significant relationship to population density or to nutrient concentrations other than silicate. Cellular levels of domoic acid were positively correlated with silicate concentrations, which is at variance with reports from prior culture experiments. These conclusions must be tentative because of the limited extent of our sampling. Nevertheless, these preliminary data indicate that further investigations of environmental conditions affiliated with cell growth and toxin production in P. australis are warranted. As a practical matter, domoic acid in the pelagic environment cannot be reliably or consistently detected by monitoring domoic acid levels in intertidal mussels. Direct measurement of domoic acid using sensitive HPLC methods is probably the most cost-effective and accurate approach for an ongoing phycotoxin monitoring program.
