BACKGROUND: Growing population demands more animal protein products. Pork remains one of the traditional and relatively sustainable types of meats for human consumption. In this paper, life-cycle assessment was performed using data from 12 pig farms. In parallel, a survey on the consumption of pork meat products was conducted analyzing responses from 806 pork meat consumers. The study aims to provide a quantitative calculation of six environmental footprints associated with the consumption of pork meat products in Serbia by analyzing data from pig farms and a pork meat consumption survey. RESULTS: Results revealed that pork meat production is responsible for the emission of 3.50 kg CO2e kg-1 live weight, 16.1 MJe kg-1 , 0.151 mg R11e kg-1 , 31.257 g SO2e kg-1 , 55.030 g PO4e kg-1 and 3.641 kg 1.4 dBe kg-1 . Further calculations reveal that weekly emissions of various environmental potentials associated with an average consumer of pork meat products in Serbia are estimated at values of 4.032 kg CO2e week-1 , 18.504 MJe week-1 , 0.17435 mg R11e week-1 , 35.972 g SO2e week-1 and 63.466 g PO4e week-1 . CONCLUSIONS: Results show that, on the one hand, pork products are responsible for environmental production impacts that mainly occur on farms while, on the other hand, consumption is characterized with high meat inclusion rates. As a leverage strategy it is recommended for producers to concentrate on lowering the production impacts rather than trying to reach consumers for sustainability conciseness. © 2020 Society of Chemical Industry.

Data from three experiments conducted to evaluate the effects of increasing available P in swine diets were used to compare two different bone processing methods. Our objective was to determine if the procedures influenced treatment differences and the ability to detect changes in the percentage bone ash. In each experiment, pigs (nursery pigs in experiments 1 and 2, and finishing pigs in experiment 3) were fed a wide range of available phosphorus levels provided from either increasing monocalcium P or added phytase. At the completion of each experiment, a subset of pigs was euthanized, and either fibulas (experiments 1 and 2) or metacarpals (experiment 3) were collected to determine the percentage bone ash. Bones were processed by cleaning away all soft tissues followed by ether extraction for 7 d (defatted), or no lipid extraction (non-defatted), and then ashed. In nursery and finishing pigs, defatted bones had increased (P < 0.001) percentage bone ash compared with non-defatted bones. No evidence of a method × treatment interaction or linear and quadratic interactions were observed in bone ash weight and percentage bone ash (P > 0.10) for nursery pigs; however, a linear interaction was detected (P < 0.05) in percentage bone ash for grow-finish pigs. This response was minimal and likely due to increased variation observed in grow-finish pigs when bones were not defatted. The processing method did not affect the ability to detect differences among treatments as a result of changing dietary P concentrations in the nursery or grow-finish pigs. In summary, either non-defatted or defatted bone processing methods can be used to determine bone ash weight and percentage bone ash as a way to assess bone mineralization and dietary treatment differences in nursery pigs; however, the increased variation observed in mature pigs suggests that defatted bone processing is the preferred method for grow-finish pigs.

The objective of this study was to evaluate the effects of brown rice particle size on the apparent total tract digestibility (ATTD) of energy and nutrients in diets fed to pigs at four different stages and determine the optimal particle size (OPS) of brown rice for young pigs and adult sows. Eighteen weanling piglets (initial body weight (BW): 10.2 ± 0.4 kg), 18 growing barrows (initial BW: 35.6 ± 1.5 kg), 24 gestating sows (initial BW: 220 ± 2.8 kg), and 24 lactating sows (initial BW: 208 ± 3.8 kg) were allotted to 1 of 3 or 4 diets based on completely randomized design with six replicates per diet. Within each stage, brown rice-soybean meal diets were formulated, and the only difference among diets was the brown rice used was ground to the specified particle size. Each stage lasted 19 days, including 7 days for cage adaptation, 7 days for diet adaptation, and 5 days for total feces and urine collection. For weanling and growing pigs, the results showed that pigs fed brown rice milled to 600 µm had a greater ATTD of dry matter (DM), gross energy (GE), and crude protein (CP) than pigs fed brown rice ground to 800 µm. However, there was no improvement in the ATTD of energy and nutrients for pigs fed brown rice milled to 600 µm versus 400 µm. The concentration of nitrogen (N) in feces significantly reduced (p < 0.01) as brown rice particle size decreased from 800 to 400 µm. However, there were no differences in phosphorus (P) output and absorbed P among diets. For gestating and lactating sows, a reduction in particle size from 1,000 to 800 µm significantly improved (p < 0.01) the ATTD of DM, GE, and CP in diets. However, there was also no improvement in the ATTD of energy and nutrients for pigs fed brown rice milled from 800 to 400 µm. In conclusion, considering the energy required for milling and nutrient digestibility, milling brown rice to 600 and 800 µm are recommended in diets for young pigs and adult sows, respectively. The OPS of brown rice for pigs at different physiological stages should be considered to economically and accurately formulate diets.

Diet treatments were arranged in a split-plot design with the whole-plot consisting of 1 of 6 concentrations of dietary Cu (22 to 134 mg/kg total Cu) and the subplot using 1 of 2 sampling techniques (probe vs. hand grab). A total of 6 feeders per treatment were sampled using a brass open handle probe. The probe was inserted into the feeder 4 times to obtain a 900 g of sample. The hand-collected samples were obtained by inserting a bare hand into the feeder approximately 8 times to obtain a 900 g of sample. Within a feeder and sampling technique, subsamples (200 g) were created by using a sample splitting device. In addition to the 6 individual feeder samples, a subsample (33 g) from each individual feeder was pooled within dietary treatment and sampling technique to form a single composite sample (200 g). This process was repeated until 4 individual composite samples were created for each diet and sampling technique. Next, all samples were ground through a centrifugal mill and submitted for mineral analysis in duplicate for Cu, Zn, Ca, and P analysis. Results indicated variability when sampling feeders with a probe were reduced (P = 0.013) for Cu and marginally reduced (P = 0.058) for Ca when compared with hand-sampling. However, no evidence for differences was detected among sampling techniques for Zn and P for the individual feeder analysis. When samples were pooled from 6 feeders to form a single composite sample, there was no evidence for differences detected among sampling techniques for Cu, Zn, Ca, and P analysis. From these results, sampling frequency calculations were determined to assess sampling accuracy within a 95% confidence interval. Results indicated that the number of feeders or composite samples required to analyze was less for Cu, Zn, Ca, and P analysis when using a probe compared with a hand sampling. In summary, sampling with a probe is associated with less variability on an individual sample basis, but when individual samples are pooled to form a composite sample, there was no evidence for difference among sampling techniques. Our results suggest samples collected for these analyses with a probe and composited would be the best option to minimize variation and analytical costs.

Bayesian population pharmacokinetic models of florfenicol in healthy pigs were developed based on retrospective data in pigs either via intravenous (i.v.) or intramuscular (i.m.) administration. Following i.v. administration, the disposition of florfenicol was best described by a two-compartment open model with the typical values of half-life at α phase (t1/2α ), half-life at ß phase (t1/2ß ), total body clearance (Cl), and volume of distribution (Vd ) were 0.132 ± 0.0289, 2.78 ± 0.166 hr, 0.215 ± 0.0102, and 0.841 ± 0.0289 L kg-1 , respectively. The disposition of florfenicol after i.m. administration was best described by a one-compartment open model. The typical values of maximum concentration of drug in serum (Cmax ), elimination half-life (t1/2Kel ), Cl, and Volume (V) were 5.52 ± 0.605 µg/ml, 9.96 ± 1.12 hr, 0.228 ± 0.0154 L hr-1  kg-1 , and 3.28 ± 0.402 L/kg, respectively. The between-subject variabilities of all the parameters after i.m. administration were between 25.1%-92.1%. Florfenicol was well absorbed (94.1%) after i.m. administration. According to Monte Carlo simulation, 8.5 and 6 mg/kg were adequate to exert 90% bactericidal effect against Actinobacillus pleuropneumoniae after i.v. and i.m. administration.

Respiratory and enteric pathogens such as Mycoplasma hyopneumoniae (Mh) and Lawsonia intracellularis (LI) reduce lean accretion and feed efficiency (FE) in growing pigs. However, the metabolic mechanism by which this occurs is still unknown. Therefore, the primary aim of this study was to examine the metabolic adaptation of pigs presented with a dual Mh and LI challenge (MhLI). A secondary objective was to examine if selection for high FE, modeled by selection for low residual feed intake (RFI), alters molecular response to disease. Using a 2 × 2 factorial design, 6 littermate pairs from a high RFI (HRFI) and 6 littermate pairs from a low RFI (LRFI) line (barrows, 66 ± 2 kg BW) were selected, with 1 pig from each pair assigned to individual pens in either the challenge or the nonchallenge (control) rooms (n = 6 barrows per line/challenge). On days post inoculation (dpi) 0, MhLI pigs were inoculated intragastrically with LI and intratracheally with Mh. Pig and feeder weights were recorded at dpi 0, 7, 14, and 21. On dpi 21, pigs were euthanized and tissues and blood were collected. Markers of oxidative stress, skeletal muscle metabolism and proteolysis, and liver gluconeogenesis were evaluated to determine the effects of MhLI, RFI line, and their interaction. The interaction of line and challenge was not significant (P > 0.05) for any measure. Overall, MhLI pigs had lower ADG (38%, P < 0.001), ADFI (25%, P < 0.001), and G:F (19%, P = 0.012) compared with controls. As expected, LRFI pigs had lower ADFI (P = 0.028) for the same ADG, giving them greater G:F (P = 0.021) than HRFI pigs. Challenged pigs had greater reactive oxygen species (ROS) production in the LM and liver (P < 0.10) but did not have greater skeletal muscle proteolysis. Liver gluconeogenesis was also not upregulated (P > 0.05) due to MhLI. These results provide further evidence that selection for LRFI does not negatively affect response to disease. In addition, these results suggest that postabsorptive metabolic functions are altered due to MhLI challenge. The MhLI challenge induced mitochondrial dysfunction, evident by greater ROS production, and caused pigs to favor glycolytic energy generation. However, skeletal muscle proteolysis and liver gluconeogenesis were not upregulated during MhLI challenge. These data suggest that during mild disease stress, pigs can meet energy demands without reliance on nutrient mobilization and gluconeogenesis.

Practical evidence suggests possible beneficial effects with the combined use of prebiotics and probiotics which can improve production parameters. The objective of the study was to investigate the addition of Saccharomyces cerevisiae (SC) as prebiotic and the combination of Lactobacillus spp. (L), Bacillus spp. (B) as probiotics on productive parameters and economic feasibility. Four hundred male pigs, hybrids of commercial genetic lines (Pietrain), were used: T1 = control group, T2 = 4 kg/tonne SC, T3 = 0.8 kg/tonne feed L and B, T4 = 4 kg/tonne SC + 0.8 kg/tonne L and B. Productive parameters were recorded in the treatment groups for four periods. Then, the viscera of five pigs per treatment were collected after slaughter to evaluate the histological changes and cytokine concentrations in the ileum. The weight gains of groups at 70-100, 100-125 and 125-150 days in the T4 group showed statistically increases (p < .05). Feed intake had a significant difference (p < .05) in T3 versus T1. The feed-conversion ratio improved for all periods in the T4 group (p < .05). The eosinophil, mononuclear infiltration and cytokines (tumor necrosis factor-α and interleukin-6) in the mucosa were lower for treatments with probiotics. In conclusion, there was an economic benefit when using both prebiotics and probiotics in the diet of pigs from weaning to finishing.

Physiologically based pharmacokinetic (PBPK) models are scientific methods used to predict veterinary drug residues that may occur in food-producing animals, and which have powerful extrapolation ability. Quinocetone (QCT) and mequindox (MEQ) are widely used in China for the prevention of bacterial infections and promoting animal growth, but their abuse causes a potential threat to human health. In this study, a flow-limited PBPK model was developed to simulate simultaneously residue depletion of QCT and its marker residue dideoxyquinocetone (DQCT) in pigs. The model included compartments for blood, liver, kidney, muscle and fat and an extra compartment representing the other tissues. Physiological parameters were obtained from the literature. Plasma protein binding rates, renal clearances and tissue/plasma partition coefficients were determined by in vitro and in vivo experiments. The model was calibrated and validated with several pharmacokinetic and residue-depletion datasets from the literature. Sensitivity analysis and Monte Carlo simulations were incorporated into the PBPK model to estimate individual variation of residual concentrations. The PBPK model for MEQ, the congener compound of QCT, was built through cross-compound extrapolation based on the model for QCT. The QCT model accurately predicted the concentrations of QCT and DQCT in various tissues at most time points, especially the later time points. Correlation coefficients between predicted and measured values for all tissues were greater than 0.9. Monte Carlo simulations showed excellent consistency between estimated concentration distributions and measured data points. The extrapolation model also showed good predictive power. The present models contribute to improve the residue monitoring systems of QCT and MEQ, and provide evidence of the usefulness of PBPK model extrapolation for the same kinds of compounds.

The efficacy of four agricultural byproducts (ABPs) and two commercial binders (CBs) to reduce the gastrointestinal absorption of a mixture of mycotoxins was tested in piglets using urinary mycotoxin biomarkers as indicator of the absorbed mycotoxins. Twenty-eight piglets were administered a bolus contaminated with the mycotoxin mixture containing or not ABP or CB. Twenty-four hour urine was collected and analyzed for mycotoxin biomarkers by using a multiantibody immunoaffinity-based LC-MS/MS method. Each bolus contained 769 µg of fumonisin B1 (FB1), 275 µg of deoxynivalenol (DON), 29 µg of zearalenone (ZEN), 6.5 µg of aflatoxin B1 (AFB1) and 6.6 µg of ochratoxin A (OTA) corresponding to 2.2, 0.8, 0.08, 0.02, and 0.02 µg/g in the daily diet, respectively. The percentage of ABP in each bolus was 50%, whereas for the two CBs the percentages were 5.2 and 17%, corresponding to 2.8, 0.3, and 0.9% in the daily diet, respectively. The reduction of mycotoxin absorption was up to 69 and 54% for ABPs and CBs, respectively. White grape pomace of Malvasia was the most effective material as it reduced significantly (p < 0.05) urinary mycotoxin biomarker of AFB1 (67%) and ZEN (69%), whereas reductions statistically not significant were observed for FB1 (57%), DON (40%), and OTA (27%). This study demonstrates that grape pomace reduces the gastrointestinal absorption of mycotoxins. This agricultural byproduct can be considered an alternative to commercial products and used in the feed industries as an effective, cheap, and natural binder for multiple mycotoxins.

An experiment was conducted to determine protein quality in processed protein sources using the content of AA, -methylisourea (OMIU)-reactive Lys, Maillard reaction products (MRP), and cross-link products; the standardized ileal digestibility (SID) of CP and AA; and growth performance in growing pigs as criteria. Differences in protein quality were created by secondary toasting (at 95°C for 30 min) of soybean meal (SBM) and rapeseed meal (RSM) in the presence of lignosulfonate resulting in processed SBM (pSBM) and processed RSM (pRSM). The processing treatment was used as a model for overprocessed protein sources. Ten growing pigs were each fed 1 of the 4 diets containing SBM, pSBM, RSM, or pRSM in each of 3 periods. Ileal chyme was collected at the end of each period and analyzed for CP, AA, and OMIU-reactive Lys. Diets were analyzed for furosine and carboxymethyllysine (CML) as an indicator for MRP and lysinoalanine (LAL), which is a cross-link product. The SBM and RSM diets contained furosine, CML, and LAL, indicating that the Maillard reaction and cross-linking had taken place in SBM and RSM, presumably during the oil extraction/desolventizing process. The amounts of furosine, CML, and LAL were elevated in pSBM and pRSM due to further processing. Processing resulted in a reduction in total and OMIU-reactive Lys contents and a decrease in G:F from 0.52 to 0.42 for SBM and 0.46 to 0.39 for RSM ( = 0.006), SID of CP from 83.9 to 71.6% for SBM and 74.9 to 64.6% for RSM ( < 0.001), and SID of AA ( < 0.001), with the largest effects for total and OMIU-reactive Lys. The effects of processing could be substantial and should be taken into account when using processed protein sources in diets for growing pigs. The extent of protein damage may be assessed by additional analyses of MRP and cross-link products.

Three iso-energetic and iso-nitrogenous diets were fed to finishing pigs for 28 days to investigate the mammalian target of rapamycin (mTOR) and ubiquitin-proteasome signaling pathways of skeletal muscle by altering compositions of dietary energy sources. Diet A, diet B, and diet C contained 44.1%, 37.6%, and 30.9% starch; 5.9%, 9.5%, and 14.3% ether extract (EE); and 12.6%, 15.4%, and 17.8% neutral detergent fiber (NDF), respectively. An increase of mRNA expression of MuRF1 (1.09 ± 0.10 vs 1.00 ± 0.08) and MAFbx (1.10 ± 0.06 vs 1.00 ± 0.11) and a decrease of concentrations of plasma insulin (8.2 ± 0.8 vs 10.8 ± 1.2) and glucose (5.76 ± 0.12 vs 6.43 ± 0.33) together with mRNA expression of IRS (0.78 ± 0.19 vs 1.01 ± 0.05) and Akt (0.92 ± 0.01 vs 1.00 ± 0.05) were observed in pigs fed diet C compared with diet A. Protein levels of total and phosphorylated mTOR (0.31 ± 0.04 vs 0.48 ± 0.03 and 0.39 ± 0.01 vs 0.56 ± 0.02), 4EBP1 (0.66 ± 0.06 vs 0.90 ± 0.09 and 0.60 ± 0.12 vs 0.84 ± 0.09), and S6K1 (0.66 ± 0.01 vs 0.89 ± 0.01 and 0.48 ± 0.03 vs 0.79 ± 0.02) were decreased; however, total and phosphorylated AMPK (0.96 ± 0.06 vs 0.64 ± 0.04 and 0.97 ± 0.09 vs 0.61 ± 0.09) were increased in pigs fed diet C compared with diet A. In conclusion, diet C suppressed the mTOR pathway and accelerated the ubiquitin-proteasome pathway in skeletal muscle of finishing pigs.

This study evaluated the theoretical clinical outcome of three marbofloxacin posology regimens in two groups of pigs (weaners and fatteners) for the treatment of Actinobacillus pleuropneumoniae (App) and Haemophilus parasuis (Hp) infection and the appearance of resistant bacteria due to the antibiotic treatment. The probability of target attainment (PTA) for pharmacokinetic/pharmacodynamics (PK/PD) ratios associated with clinical efficacy and with the appearance of antimicrobial resistance for fluoroquinolones at each minimum inhibitory concentration (MIC) or mutant prevention concentration (MPC) were calculated, respectively. The cumulative fraction of response (CFR) was calculated for the three posology regimens against App and they ranged from 91.12% to 96.37% in weaners and from 93% to 97.43% in fatteners, respectively. In the case of Hp, they ranged from 80.52% to 85.14% in weaners and from 82.01% to 88.49% in fatteners, respectively. Regarding the PTA of the PK/PD threshold associated with the appearance of antimicrobial resistance, results showed that marbofloxacin would prevent resistances in most of the animals up to the MPC value of 1 µg/mL.

The aim of this study was to investigate the presence of arsenic, lead and cadmium residues in samples of liver, kidney and muscle of poultry and swine during the years from 2002 to 2008. A total of 1978 samples were analyzed: 1031 of poultry and 947 of swine from Brazilian slaughterhouses. The samples were analyzed at the National Agricultural Laboratory using the atomic absorption spectrometry technique. In poultry, the arsenic residues were detected in 53.6% of liver samples, although no results have exceeded the MRL. In kidneys, 39.7% of the samples showed measurable residues and there was no violation of the limits. Regarding lead, there were 5 contaminated liver samples and 24 contaminated kidney samples (1.5 and 3.6% respectively). In muscle tissue there was only one sample with residues...


Objetivou-se investigar a presença de resíduos de arsênico, chumbo e cádmio em amostras de fígado, rins e músculo de aves e suínos, durante os anos de 2002 a 2008. Um total de 1.978 amostras foi analisado: 1031 de aves e 947 de suínos provenientes de matadouros brasileiros. As amostras foram analisadas no Laboratório Nacional Agropecuário (Lanagro), sendo utilizada a técnica de espectrometria de absorção atômica. Em aves, os resíduos de arsênico foram detectados em 53,6% das amostras de fígado, embora sem exceder o LMR. Nos rins, 39,7% das amostras mostraram resíduos mensuráveis e não houve violação dos limites. Em relação ao chumbo, cinco amostras de fígado e 24 de amostras de rins estavam contaminadas (1,5 e 3,6%, respectivamente). No tecido muscular, houve apenas uma amostra com resíduos. Para o cádmio, 3,8% das amostras apresentavam valores abaixo do LMR. Resíduos de cádmio foram encontrados em 110 amostras de rins (16,3% testados), mas apenas uma ultrapassou o limite máximo permitido pela legislação nacional. Em suínos, resíduos de arsênico foram detectados em 15,3% das amostras de fígado, sem exceder o LMR. Nos rins, 14,2% das amostras apresentaram resíduos mensuráveis...

Porcine ß-defensin 2 (pBD2), a recently discovered porcine defensin that is produced by the intestine, exerts antimicrobial activities and innate immune effects that are linked to intestinal diseases in pigs. Here, we report a codon-optimised protein corresponding to mature pBD2 cDNA that was expressed and purified in Pichia pastoris yeast. The highest amount of secreted protein (3,694.0 mg/L) was reached 144 h into a 150-h induction during high-density cultivation. Precipitation followed by gel exclusion chromatography yielded 383.7 mg/L purified recombinant pBD2 (rpBD2) with a purity of ~93.7 %. Two recombinant proteins of 5,458.5 and 5,258.4 Da were detected in the mass spectrum due to variation in the amino-terminus. The rpBD2 exhibited high antimicrobial activity against a broad range of pig pathogenic bacteria (minimal inhibitory concentration [MIC] 32-128 µg/mL); the highest activity was observed against Salmonella choleraesuis, Staphylococcus aureus and Streptococcus suis (MIC 32-64 µg/mL). However, rpBD2 also inhibited the growth of probiotics such as Lactobacillus plantarum, Bacillus subtilis and Saccharomyces cerevisiae, but at lower efficacies than the pathogens. Purified or unpurified rpBD2 also maintained high activity over a wide range of pH values (2.0-10.0), a high thermal stability at 100 °C for 40 min and significant resistance to papain, pepsin and trypsin. In addition, the activity of rpBD2 towards S. aureus was unaffected by 10 mM dithiothreitol (DTT) and 20 % dimethyl sulphoxide (DMSO). Our results suggest that pBD2 could be produced efficiently in large quantities in P. pastoris and be a substitute for traditional antibiotics for growth promotion in the porcine industry.

To estimate the consumer exposure to olaquindox (OLA) residues in porcine edible tissues, a physiologically based pharmacokinetic (PBPK) model for methyl-3-quinoxaline-2-carboxylic acid (MQCA), the marker residue of OLA, was developed in pigs based on the assumptions of the flow-limited distribution, hepatic metabolism, and renal excretion. The model included separate compartments corresponding to blood, muscle, liver, kidney, adipose, and an extra compartment representing the remaining carcass. Physiological parameters were determined from literatures. Plasma protein binding, partition coefficients, and renal clearance for MQCA were determined in in vitro and in vivo studies. The metabolic conversion of OLA to MQCA was assumed as a simple, one-step process, and an apparent first-order rate constant (k) was employed to describe this metabolic process. The PBPK model was optimized and validated with plasma and tissue data from literatures and our study. Sensitivity analysis and Monte Carlo simulation were also implemented to estimate the influence of model parameters on the goodness of fit. When compared with the observed data, the PBPK model underestimated the MQCA level in all compartments at the early time points, whereas gave excellent predictions of MQCA concentration in porcine edible tissues at later time points. The correlation coefficients between the predicted and observed values were over 0.88. The consistency between the model predictions and the real residues of OLA in pigs proved the good applicability of our model in food safety risk assessment.

Currently, the determination of health risks to pesticide applicators from dermal exposure to these chemicals is assessed using either a concentrate of the compound or a relevant aqueous dilution. Neither of these conditions reflects a normal exposure of an individual when re-entering an area after pesticide application, that is, contact with dried residue of the diluted product on foliage. Methodology has therefore been developed to determine a relevant estimate of this potential dermal re-entry exposure from pesticide residues. Potential delivery platforms have been characterized for the transfer of pesticide residue to skin. Spin coating has been used to deposit uniform pesticide layers on to each platform. Five pesticides have been chosen to encompass a wide range of physicochemical properties: atrazine, 2,4-dichlorophenoxyacetic acid (2,4-D), chlorpyrifos, monocrotophos, and acetochlor. In vitro (Franz diffusion cell) experiments have been performed to monitor the transfer of these pesticides from the delivery platforms onto and through excised porcine skin. Parallel experiments were also conducted with aqueous pesticide dilutions for comparison, and a final in vivo measurement using ibuprofen (as a model compound) complemented the in vitro data. The results demonstrate that transfer of chemical residue onto and subsequently through the skin is dependent on the physical attributes of the residue formed. Thus, assessing dermal exposure to pesticides based on skin contact with either the chemical concentrate or a relevant aqueous dilution may incorrectly estimate the risk for re-entry scenarios.

In vitro digestors can be used to provide bioaccessibility values to help assess the risk from incidental human ingestion of contaminated soils. It has been suggested that these digestors may need to include a lipid sink to mimic human uptake processes. We compare the correspondence between in vivo polycyclic aromatic hydrocarbon (PAH) uptake for eight different PAH contaminated soils with PAH release in in vitro digestors in the presence and absence of a lipid sink. Lipid sinks were essential to the success of the in vitro digestors in predicting juvenile swine PAH uptake. In the presence of the lipid sink, results of the In Vitro Digestion model (IVD) closely corresponded with a slope of 0.85 (r(2) = 0.45, P < 0.07) to the in vivo results. The Relative Bioaccessibility Leaching Procedure (RBALP) results did not correspond to the in vivo study but did tightly reflect total soil PAH concentration. We conclude that the basis of this difference between digestors is that the RBALP used an aggressive extraction technique that maximized PAH release from soil. Systemic uptake in juvenile swine was not linked to soil PAH concentration but rather to the thermodynamic properties of the soil.

The present study aimed to establish hyperinsulinemic euglycemic step clamping with tracer glucose infusion and labeled glucose infusate (step hot-GINF HEC) for assessment of acute insulin resistance in anesthetized pigs and to arrange for combination with invasive investigative methods. Tracer enrichment was measured during D-[6,6-(2)H(2)]glucose infusion before and after surgical instrumentation (n = 8). Insulin dose-response characteristics were determined by two step hot-GINF HEC procedures, with accordingly labeled glucose infusates performed at a total of six insulin infusion rates ranging from 0.2 to 2.0 mU kg(-1) min(-1) (n = 8). Finally, three-step hot-GINF HEC (0.4, 1.2, and 2.0 mU kg(-1) min(-1)) was performed subsequent to major surgical trauma (n = 8). Tracer enrichment, basal glucose kinetics, and circulating levels of C-peptide, cortisol, glucagon, and catecholamines were not influenced by surgical instrumentation. Mean intraindividual coefficient of variance levels for glucose infusion rates and repeatedly measured insulin, glucose, and tracer enrichment indicated stable clamping conditions. Basal and maximal insulin-stimulated glucose utilization was twice as high as in humans at approximately 5.5 and 21 mg kg(-1) min(-1). Surgical trauma elicited pronounced peripheral and moderate hepatic insulin unresponsiveness (45% lower whole body glucose disposal and 19% less suppressed endogenous glucose release) and apparently diminished metabolic insulin clearance. Step hot-GINF HEC seems suitable for assessment of acute insulin resistance in anesthetized pigs, and combination with invasive investigative methods requiring surgical instrumentation can be accomplished without the premises for utilization of the technique being altered, but attention must be paid to alterations in metabolic insulin clearance.

The abilities of montmorillonite (MMT) and Cu (II)-bearing montmorillonite (Cu-MMT) for adsorption of Cd from aqueous solutions have been studied at different optimized conditions of shaking time, pH value, and initial concentration, respectively. The results showed that the adsorbability of Cu-MMT onto Cd is stronger than the MMT. The amount of Cu-MMT necessary to remove Cd from solution was about 0.5 g for 100-200 mg/L Cd solution at a pH of 4.0. The maximum adsorption (94%) was 20 min of shaking time using Cu-MMT as adsorbent. The maximum adsorption of Cd = 95% at a pH of 4.0. By increasing the initial concentration, the adsorption amount of Cd onto Cu-MMT increased. Animal experiment of this study indicated that the addition of Cu-MMT to the Cd-contaminated diet of pigs reduced significantly the Cd levels in tissues (p < 0.05) and increased contents in feces (p < 0.01), improving the safety of animal products. Thus, the overall results established the use of Cu-MMT as a heavy metal adsorbent in animal's diets, implying a potential protective role against heavy metal toxicities.

Understanding how energy is utilized by the pig, and how the pig responds to changes in dietary energy concentration, is essential information in determining the optimal concentration of dietary energy under farm conditions, which are often highly diverse. The objective of these experiments was to determine how changes in dietary DE concentration, achieved through graded changes in diet composition, would affect the performance and carcass composition of growing pigs. In Exp. 1, which was conducted in a research facility, 300 pigs (31.1 +/- 2.6 kg) were assigned to diets containing 3.09, 3.24, 3.34, 3.42, or 3.57 Mcal of DE/kg. Experiment 2, which was conducted at a commercial swine farm, involved 720 pigs (36.8 +/- 5.9 kg) assigned to diets containing 3.12, 3.30, or 3.43 Mcal of DE/kg. Increased DE concentration was attained by using more wheat, soybean meal, and fat and less barley; true ileal lysine was adjusted as DE increased, and minimal AA:lysine ratios were maintained. In Exp. 1, ADG improved linearly as the energy content of the diet increased (P = 0.03). Feed intake decreased (P < 0.001) and feed efficiency and daily caloric intake improved (P = 0.005) with increased DE content. Variability in growth was not affected by treatment. Carcass index and LM thickness were not affected by increasing dietary DE content; backfat thickness, however, was increased (P < 0.001). In Exp. 2, overall ADG was unaffected by dietary energy content, although an improvement in growth was observed until the pigs reached approximately 80 kg of BW. Overall feed intake decreased with increasing energy content (P = 0.01), although this was not observed during the initial 6 wk of the experiment. Carcass index, lean yield, and backfat were not affected by increasing dietary energy content, whereas LM thickness tended to increase (P = 0.08). The value per pig was unaffected by increasing dietary energy content in both experiments, and returns above feed costs were reduced. Increasing the energy density of the diet for growing pigs through incremental changes in dietary composition had a variable impact on overall growth performance and carcass quality. Increasing the dietary DE had no effect on variations in BW at the time of marketing.
