The presence of toxic trace elements (TEs) has resulted in a worldwide deterioration in freshwater ecosystem quality. This study aimed to analyze the distribution of TEs, including chromium (Cr), nickel (Ni), arsenic (As), mercury (Hg), cadmium (Cd), and lead (Pb), in water, sediment, and organs of Tilapia (Oreochromis mossambicus) collected from selected inland water bodies in Tamil Nadu, India. The water samples exhibited a range of concentrations for TEs: Cr varied from 0.014 to 5.193 µg/L, Ni ranged from 0.283 to 11.133 µg/L, As ranged from 0.503 to 1.519 µg/L, Cd from 0.001 to 0.616 µg/L, and Pb ranged from non-detectable (ND) to 6.103 µg/L. The concentrations of TEs in sediment were found to vary within the following ranges: 5.259 to 32.621 mg/kg for Cr, 1.932 to 30.487 mg/kg for Ni, 0.129 to 0.563 mg/kg for As, 0.003 to 0.011 mg/kg for Cd, ND to 0.003 mg/kg for Hg, and 0.404 to 1.575 mg/kg for Pb. The study found that the accumulation pattern of TE in fishes across all selected areas was liver > bone > gill > muscle. The organs had TE concentrations of Cr (ND-0.769 mg/kg), Ni (ND-1.053 mg/kg), As (0.002-0.080 mg/kg), Pb (ND-0.411 mg/kg), and Hg (ND-0.067 mg/kg), which was below the maximum residual limit prescribed by EC and FSSAI. The bioconcentration factor (BCF) of TEs exhibited a greater magnitude in comparison with the biota-sediment accumulation factor due to the higher concentration of TEs in fish and lower level in water. The assessment of both carcinogenic and non-carcinogenic risks suggests that the consumption of Tilapia from the study region does not pose any significant risks.

The aim of this study was to assess the levels of heavy metals in both wild and farmed tilapia on Lake Kariba in Zambia and to evaluate the impact of intensive fish farming on wild tilapia. Three sites for wild fish (2 distant and 1 proximal to fish farms) and two fish farms were selected. One hundred fish (52 from distant sites; 20 near fish farms; 28 farmed fish) were sampled and muscle tissues excised for analysis of heavy metals (Mg, Fe, Zn, Al, Cu, Se, Co, Mo, As, Cr, V, Ni, Hg, Pb, Li, Cd, and Ag) by acid (HNO3) digestion and ICP-MS. All metals were found to be below the maximum limits (MLs) set by WHO/EU. Essential metals were higher in farmed tilapia, whereas non-essential metals were higher in wild tilapia. Significantly higher levels of essential metals were found in wild fish near the fish farms than those distant from the farms. Estimated weekly intake (EWI) for all metals were less than the provisional tolerable weekly intakes (PTWI). Target hazard quotients (THQ) and Hazard Indices (HI) were <1, indicating no health risks from a lifetime of fish consumption. Selenium Health Benefit Value (HBVSe) was positive for all locations, indicating protective effects of selenium against mercury in fish. Total cancer risk (CR) due to As, Cr, Cd, Ni and Pb was less than 1 × 10-4, indicating less than 1 in 10,000 carcinogenic risk from a lifetime consumption of tilapia from Lake Kariba. Hg levels (0.021 mg/kg) in wild tilapia at site 1 were higher than the Environmental quality standard (EQS = 0.020 mg/kg) set by EU, indicating possible risk of adverse effects to fish. Except for Hg, levels of metals in fish were safe for human consumption and had no adverse effects on fish.

The antimicrobial residues of aquacultural production is a growing public concern, leading to reexamine the method for establishing robust withdrawal time and ensuring food safety. Our study aims to develop the optimizing population physiologically-based pharmacokinetic (PBPK) model for assessing florfenicol residues in the tilapia tissues, and for evaluating the robustness of the withdrawal time (WT). Fitting with published pharmacokinetic profiles that experimented under temperatures of 22 and 28 °C, a PBPK model was constructed by applying with the Bayesian Markov chain Monte Carol (MCMC) algorithm to estimate WTs under different physiological, environmental and dosing scenarios. Results show that the MCMC algorithm improves the estimates of uncertainty and variability of PBPK-related parameters, and optimizes the simulation of the PBPK model. It is noteworthy that posterior sets generated from temperature-associated datasets to be respectively used for simulating residues under corresponding temperature conditions. Simulating the residues under regulated regimen and overdosing scenarios for Taiwan, the estimated WTs were 12-16 days at 22 °C and 9-12 days at 28 °C, while for the USA, the estimated WTs were 14-18 and 11-14 days, respectively. Comparison with the regulated WT of 15 days, results indicate that the current WT has well robustness and resilience in the environment of higher temperatures. The optimal Bayesian population PBPK model provides effective analysis for determining WTs under scenario-specific conditions. It is a new insight into the increasing body of literature on developing the Bayesian-PBPK model and has practical implications for improving the regulation of food safety.

This study was planned to recycle calcium and the phosphorus-rich Nile tilapia fish scale biowaste into nano-hydroxyapatite (FHAP), using ultrasonic-assisted extraction of calcium and phosphorus from fish scales, which was optimized in term of extraction time, acid concentration, extraction temperature, and ultrasonic power. These two elements were determined simultaneously by inductively coupled plasma atomic emission spectrometry and the FHAP phase was formed upon addition of the extracted element solution in alkaline medium using homogenous precipitation assisted with ultrasound energy. The FHAP adsorbent was characterized by x-ray diffraction, scanning electron microscopy, energy dispersive spectroscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, and Brunauer-Emmett-Teller. A combination of FHAP and the ultrasonic method was then used to remove Hg2+ from aqueous solution. Four significant variables affecting Hg2+ removal, namely, adsorbent dosage, pH, ultrasonic power, and adsorption time, were studied. The results exhibited that the optimal conditions for maximizing the removal of Hg2+ were 0.02 g adsorbent dosage, pH 8, 0.4 kW ultrasonic power, 20 min adsorption time, and 30 °C adsorption temperature. The sorption mechanism of Hg2+ was revealed by isotherm modeling, indicating that FHAP adsorbent has a potential for Hg2+ removal in aqueous media with the maximum adsorption capacity being 227.27 mg g-1. This adsorption behavior is in agreement with the Langmuir model as reflected by a satisfactory R2 value of 0.9967, when the kinetics data were fitted with pseudo-second-order. Therefore, the FHAP could be an alternative adsorbent for the ultrasonic-assisted removal of Hg2+ at very high efficiency and within a very short period of time.

The study objective was to assess the profile of aliphatic and polycyclic aromatic hydrocarbons in the aquatic environment of Lake Manzala and to determine the potential sources and effects of these hydrocarbons. To reach these objectives 24 surface sediment and 24 tilapia fish were collected and analyzed using GC-FID and GC-MS. The highest concentrations for n-alkanes (∑n-C15-35) (19,625 ng g-1) and PAHs (∑45PAHs) (5,153 ng g-1) were found in sediments collected from Bahr Al-Baqar drain while Al-Temsah and Al-Hamra sites contained the lowest concentrations. Different diagnostic molecular ratios for hydrocarbons were used to evaluate sources of the hydrocarbons. The hydrocarbons were found to have differing proportions of petrogenic, pyrogenic, and biogenic sources. Comparison to sediment quality guidelines indicated PAHs were below the effect range low (ERL) with the exception of acenaphthene, fluorine, and ∑45PAHs in Bahr Al-Baqar drain sediments. Tissue samples had moderate PAH concentrations for ∑45PAHs ranging from 302.5 ng g-1 West of Bashteer (S3) to 596 ng g-1 in Legam (S5). Since fish metabolize PAH quickly, their detection suggest continual or very recent exposure. The PAH in sediment and fish from Lake Manzala are at a low or below level of environmental concern.

This study reports the heavy metal (Hg, Cd, Cr, Cu, and Pb) contamination risks to and safety of two species of fresh water fish (tilapia, Oreochromis mossambicus and carp, Cyprinus carpio) that are farmed in the Yonki Reservoir in the Eastern Highlands of Papua New Guinea (PNG). The upper reaches of the reservoir are affected by alluvial and large-scale gold mining activities. We also assessed heavy metal levels in the surface waters and sediments and in selected aquatic plant species from the reservoir and streams that intersect the gold mining areas. The water quality was acceptable, except for the Cr concentration, which exceeded the World Health Organization (WHO) standard for water contamination. The sediments were contaminated with Cd and Cu in most of the sampling stations along the upstream waters and the reservoir. The Cd concentration in the sediments exceeded the US Environmental Protection Agency's Sediment Quality Guideline (SQG) values, and the geoaccumulation index (Igeo) values indicated heavy to extreme pollution. In addition, the Cd, Cu, and Pb concentrations in aquatic plants exceeded the WHO guidelines for these contaminants. Between the fish species, tilapia accumulated significantly higher (P < 0.05) Cu in their organ tissues than carp, confirming the bioaccumulation of some metals in the aquatic fauna. The edible muscles of the fish specimens had metal concentrations below the maximum permissible levels established by statutory guidelines. In addition, a human health risk assessment, performed using the estimated weekly intake (EWI) values, indicated that farmed fish from the Yonki Reservoir are safe for human consumption.

We report results from an evaluation of the levels of heavy metals, i.e., copper (Cu), cadmium (Cd), lead (Pb), nickel (Ni), manganese (Mn), chromium (Cr), and iron (Fe) in sediment and tilapia fish samples from a wide stretch of the Kafue river of Zambia. In sediment samples, the highest Pb and Fe concentrations were recorded at Hippo Dam, i.e., 36.2 ± 0.1 mg/kg dw and 733 ± 37 mg/kg dw at Kafue Town, respectively. Other notably high metal concentrations in sediment were Cr at Kafue Bridge (42.5 ± 0.1 mg/kg dw [dw]), Cu at Mpongwe (233 ± 5 mg/kg dw), and Mn at Kafue Town (133 ± 1 mg/kg dw); it was highest at Ithezi Tezhi Dam at 166 ± 1 mg/kg d. Three fish species, i.e., three-spot bream Tilapia andersonii, red-breasted bream T. rendalli, and nile tilapia Oreochromis niloticus were evaluated for levels of the seven metals. The concentrations of the metals in these fish species afforded estimation of the biota sediment-accumulation factor, which is the ratio of the concentration of the metal in liver to that in the sediment. The coefficients of condition (K) values, which give an indication of the health of the fish, were also estimated. The K values ranged from 2.5 ± 0.5 to 5.1 ± 0.6 in all of the three fish species. Partial least squares analysis showed that heavy metals are generally sequestered evenly in all of the parts of all of the three fish species except for elevated levels of Mn, Cd, and Pb in the liver samples.

The trace metal concentrations in edible muscle of red tilapia (Oreochromis spp.) sampled from a former tin mining pool, concrete tank and earthen pond in Jelebu were analysed with microwave assisted digestion-inductively coupled plasma-mass spectrometry. Results were compared with established legal limits and the daily ingestion exposures simulated using the Monte Carlo algorithm for potential health risks. Among the metals investigated, arsenic was found to be the key contaminant, which may have arisen from the use of formulated feeding pellets. Although the risks of toxicity associated with consumption of red tilapia from the sites investigated were found to be within the tolerable range, the preliminary probabilistic estimation of As cancer risk shows that the 95th percentile risk level surpassed the benchmark level of 10(-5). In general, the probabilistic health risks associated with ingestion of red tilapia can be ranked as follows: former tin mining pool > concrete tank > earthen pond.

OBJECTIVE: to analyse the Redness, Oedema, Ecchymosis, Discharge, Approximation (REEDA) scale reliability when evaluating perineal healing after a normal delivery with a right mediolateral episiotomy. METHOD: observational study based on data from a clinical trial conducted with 54 randomly selected women, who had their perineal healing assessed at four time points, from 6 hours to 10 days after delivery, by nurses trained in the use of this scale. The kappa coefficient was used in the reliability analysis of the REEDA scale. RESULTS: the results indicate good agreement in the evaluation of the discharge item (0.75< Kappa ≥0.88), marginal and good agreement in the first three assessments of oedema (0.16< Kappa ≥0.46), marginal agreement in the evaluation of ecchymosis (0.25< Kappa ≥0.42) and good agreement regarding redness (0.46< Kappa ≥0.66). For the item coaptation, the agreement decreased from excellent in the first assessment to good in the last assessment. In the fourth evaluation, the assessment of all items displayed excellent or good agreement among the evaluators. CONCLUSION: the difference in the scores among the evaluators when applying the scale indicates that this tool must be improved to allow an accurate assessment of the episiotomy healing process. .


OBJETIVO: analisar a confiabilidade da escala REEDA (Redness, Oedema, Ecchymosis, Discharge, Approximation) para avaliar a cicatrização do períneo após parto vaginal com episiotomia médio-lateral direita. MÉTODO: estudo observacional, baseado em dados coletados em ensaio clínico, conduzido com 54 mulheres, selecionadas aleatoriamente. As mesmas tiveram o processo de cicatrização perineal avaliado em quatro momentos (de 6 horas a 10 dias após o parto), por enfermeiras treinadas para o uso da escala. O coeficiente kappa foi usado para análise de confiabilidade da escala REEDA. RESULTADOS: os resultados indicam bom nível de concordância na avaliação do item secreção (0,75< Kappa ≥0,88), concordância boa e marginal em relação ao item equimose (0,25< Kappa ≥0,42), e bom nível de concordância em relação à hiperemia (0,46< Kappa ≥0,66). O nível de concordância referente à avaliação do item coaptação diminuiu de excelente, na primeira avaliação, para bom, na última avaliação. CONCLUSÃO: a diferença entre as pontuações atribuídas pelas avaliadoras na aplicação da escala indica que o instrumento precisa ser melhorado, para permitir avaliações mais precisas do processo de cicatrização da episiotomia. .


OBJETIVO: analizar la confiabilidad de la escala de Enrojecimiento, Edema, Equimosis, Drenaje, Aproximación (REEDA) en la evaluación de la curación perineal tras parto normal con episiotomía mediolateral derecha. MÉTODO: estudio observacional con base en datos de un ensayo clínico conducido con 54 mujeres elegidas de forma aleatoria, con evaluación de su curación perineal en cuatro momentos, entre 6 horas y 10 días después del parto, por enfermeras capacitadas en el uso de esta escala. El coeficiente de kappa fue utilizado en el análisis de confiabilidad de la escala REEDA. RESULTADOS: los resultados indican buena concordancia en la evaluación del ítem drenaje (0,75< Kappa ≥0,88), concordancia marginal y buena en las primeras tres evaluaciones de edema (0,16< Kappa ≥0,46), concordancia marginal en la evaluación de equimosis (0,25< Kappa ≥0,42) y buena concordancia sobre enrojecimiento (0,46< Kappa ≥0,66). Para el ítem coaptación, la concordancia disminuyó de excelente en la primera evaluación hasta buena en la última. En el cuarto momento, la evaluación de todos los ítems mostró concordancia excelente o buena entre los evaluadores. CONCLUSIÓN: la diferencia en las notas entre los evaluadores en la aplicación de la escala indica que esta herramienta debe ser mejorada para permitir una evaluación correcta del proceso de curación de la episiotomía. .

A simple and effective method for the rapid determination of five salicylate and benzophenone-type UV absorbing substances in marketed fish is described. The method involves the use of matrix solid-phase dispersion (MSPD) prior to their determination by on-line silylation gas chromatography tandem mass spectrometry (GC-MS/MS). The parameters that affect the extraction efficiency were optimized using a Box-Behnken design method. The optimal extraction conditions involved dispersing 0.5g of freeze-dried powdered fish with 1.0g of Florisil using a mortar and pestle. This blend was then transferred to a solid-phase extraction (SPE) cartridge containing 1.0g of octadecyl bonded silica (C18), as the clean-up co-sorbent. The target analytes were then eluted with 7mL of acetonitrile. The extract was derivatized on-line in the GC injection-port by reaction with a trimethylsilylating (TMS) reagent. The TMS-derivatives were then identified and quantitated by GC-MS/MS. The limits of quantitation (LOQs) were less than 0.1ng/g.

The purposes of this study were to describe the impact of metal pollution on the main economic fish species Tilapia nilotica and to assess the potential health risk from consuming this contaminated fish in Egypt. Trace metals, including Ag, Al, Cd, Bo, Co, Cr, Cu, Fe, Mn, Mo, Ni, Pb, St, V, Zn, and As, were determined in water, Tilapia nilotica, and sediments from the River Nile, Domiate branch, Egypt. Metal concentrations in fish of Al, Cd, Co, Fe, Mn, Pb, V, and Zn (mg/kg dry weight [dw]) and concentrations in sediment of Cd, Co, Cr, Cu, Mn, Ni, Pb, V, and Zn (mg/kg dw) were above the International Atomic Energy Agency (IAEA-407) levels. However, trace metals in river water were still at permissible levels for Egyptian standards. The hazard index (HI) of estimated metal mixtures for intake of Tilapia nilotica (23.37) demonstrated that intake resulted in higher noncarcinogenic risk. In conclusion, the overall problem of metal contamination in fish collected from the River Nile was more serious than postulated to occur in an industrialized and densely populated area. In the light of known risks to public health, environmental protection laws are needed in Egypt.

Concentrations of the heavy metals copper (Cu), cadmium (Cd), zinc (Zn), lead (Pb) and nickel (Ni) were determined in the liver, gills and muscles of tilapia fish from the Langat River and Engineering Lake, Bangi, Selangor, Malaysia. There were differences in the concentrations of the studied heavy metals between different organs and between sites. In the liver samples, Cu>Zn>Ni>Pb>Cd, and in the gills and muscle, Zn>Ni>Cu>Pb>Cd. Levels of Cu, Cd, Zn and Pb in the liver samples from Engineering Lake were higher than in those from the Langat River, whereas the Ni levels in the liver samples from the Langat River were greater than in those from Engineering Lake. Cd levels in the fish muscle from Engineering Lake were lower than in that from the Langat River. Meanwhile, the Cd, Zn and Pb levels in the fish muscle from the Langat River were lower than in that from Engineering Lake, and the Ni levels were almost the same in the fish muscle samples from the two sites. The health risks associated with Cu, Cd, Zn, Pb and Ni were assessed based on the target hazard quotients. In the Langat River, the risk from Cu is minimal compared to the other studied elements, and the concentrations of Pb and Ni were determined to pose the greatest risk. The maximum allowable fish consumption rates (kg/d) based on Cu in Engineering Lake and the Langat River were 2.27 and 1.51 in December and 2.53 and 1.75 in February, respectively. The Cu concentrations resulted in the highest maximum allowable fish consumption rates compared with the other studied heavy metals, whereas those based on Pb were the lowest. A health risk analysis of the heavy metals measured in the fish muscle samples indicated that the fish can be classified at one of the safest levels for the general population and that there are no possible risks pertaining to tilapia fish consumption.

Unscrupulous inclusion of melamine in fish feeds can be harmful to fish and may be hazardous to human health. An eight-week feeding trial examined the effects of melamine (inclusion levels; 5-30 g kg⁻¹ feed) on the growth performance, feed efficiency, histopathological changes and melamine residues in sex-reversed red tilapia, Oreochromis niloticus (L.) × O.mossambicus (Peters). Fish which received melamine-containing feeds grew less, utilized feeds less efficiently and performed poorly, besides exhibiting defects such as fin erosion, anorexia, sluggish swimming behaviour, paling/darkening of skin and low survival. Melamine concentration in the fish reflected its inclusion level in the feeds, and the content was higher in the viscera than in the fish fillet or whole fish. Histopathological alterations were evident in the kidney, liver and gills of fish subjected to melamine treatment - the severity of lesions corresponded to its dosage. Enlargement of renal tubules was observed in the kidney of fish fed with ≥10 g melamine kg⁻¹ feed, although, crystals were not deposited. Fish subjected to melamine insult had more prominent lesions in liver than in kidney. Toxic effects on the gills manifested as epithelial hyperplasia of the primary and secondary lamellae. The anomalies were severe at higher intake levels of melamine.

Biomarkers measured at the molecular and cellular level in fish have been proposed as sensitive "early warning" tools for biological effect measurements in environmental quality assessments. Lake Beira is a hypertrophic urban water body with a complex mixture of pollutants including polycyclic aromatic hydrocarbons (PAHs) and Microcystins. In this study, a suite of biomarker responses viz. biliary fluorescent aromatic compounds (FACs), hepatic ethoxyresorufin O-deethylase (EROD) and glutathione S-transferase (GST), brain and muscle cholinesterases (ChE), serum sorbitol dehydrogenase (SDH), and liver histology of Oreochromis niloticus, the dominant fish inhabiting this tropical Lake were evaluated to assess the pollution exposure and biological effects. Some fish sampled in the dry periods demonstrated prominent structural abnormalities in the liver and concomitant increase in serum SDH and reduction in hepatic GST activities in comparison to the control fish and the fish sampled in the rainy periods. The resident fish with apparently normal liver demonstrated induction of hepatic EROD and GST activities and increase in biliary FACs irrespective of the sampling period indicating bioavailability of PAHs. Muscle ChE activities of the resident fish were depressed significantly indicating exposure to anticholinesterase substances. The results revealed that fish populations residing in this Lake is under threat due to the pollution stress. Hepatic abnormalities in the fish may be mainly associated with the pollution stress due to recurrent exposure to PAHs and toxigenic Microcystis blooms in the Lake.

The present study was carried out in El-Bahria Oasis desert (Giza-Egypt) for 8 months (March-October) as a new study to assess the culture of tilapia spp. in underground well water. The obtained results showed a significant increase (P<0.01) in the values of plasma total protein, glucose, AST (aspartate aminotransferase), ALT (alanine aminotransferase), creatinine, uric acid, sodium, potassium and magnesium as well as a significant decrease (P<0.01) in plasma total lipids and calcium concentration. Furthermore, iron content in some selected vital organs was increased gradually with time, the studied organs are arranged according to their iron content in the following order: spleen > liver > kidney > gills > muscle. Remarkable changes were observed in the chemical muscle composition where the results showed a significant increase (P<0.01) in muscle water content, total lipids and ash. However, a significant decrease (P<0.01) in muscle total protein at the end of the study was observed. The growth of all male farmed tilapia in well water with a 3.2 mg/l iron concentration was unexpected; despite the presence of this high concentration of iron, the weight gain of cultured fish was 250 +/- 14.5 g. Molecular techniques are used nowadays as a good indicator for assessing the alteration in the genomes. RAPD-PCR technique indicated appearance of some changes in polymorphism band patterns. There also exists a distinct distance between the band patterns of cultured fish (T) and control fish (C). Histopathlogical sections showed pathological alterations in liver, kidney gills and spleen and the obtained results were discussed.

A biologically based risk-assessment model of arsenic (As) exposure evaluated farmed tilapia (Orechromis mossambicus) and human health during tilapia consumption in an area of southwestern Taiwan where blackfoot disease (BFD) occurs. The risk assessment addressed exposures to city residents who lived in Taipei, Taichung, and Kaohsiung, as well as subsistence fishers living in the BFD area who consumed tilapia. The models implemented included a physiologically based toxicokinetic and toxicodynamic (PBTK/TD) model to account for As exposure and dose-response profiles in tilapia and a human health exposure/risk model that accounts for target lifetime risk (TR) and hazard quotient (HQ) for humans consuming farmed tilapia. Results demonstrated that 90th percentiles of TR ranged from 1.53 x 10(-8) to 1.62 x 10(-6) for city residents with farmed tilapia consumption rates of 0.41 to 1.37 g/d. The 90th percentiles ranged from 2.07 x 10(-6) to 7.89 x 10(-5) for subsistence fishers in the BFD area with farmed tilapia consumption rates of 16.80 to 59.15 g/d. All predicted 90th percentiles of HQ were less than 1 for city residents and subsistence fishers in the BFD area, indicating small contributions from farmed tilapia consumption. Critical variables included whole-fish body weight, water As content, and As level in tilapia muscle. Although bioaccumulation of As seems unlikely to result in toxicity to farmed tilapia and humans consuming tilapia, the theoretical human health risks in the BFD area are alarming, using a probabilistic risk-assessment model based on conservative assumptions.

Alcolapia grahami is a unique ureotelic tilapia that lives in the highly alkaline, saline Lake Magadi, Kenya (pH, approximately 10.0; alkalinity, approximately 380 mmol L(-1); Na(+), approximately 350 mmol L(-1); Cl(-), approximately 110 mmol L(-1); osmolality, approximately 580 mosm kg(-1)). The fish survived well upon gradual exposure to dilute lake water (down to 1%, essentially freshwater). Urea excretion continued, and there was no ammonia excretion despite favorable conditions, indicating that ureotelism is obligatory. Levels of most ornithine-urea cycle enzymes in the liver were unchanged relative to controls kept for the same period in 100% lake water. The fish exhibited good abilities for hypo- and hyperregulation, maintaining plasma Na(+), Cl(-), and osmolality at levels typical of marine and freshwater teleosts in 100% and 1% lake water, respectively. Plasma total CO(2) did not change with environmental dilution. Routine oxygen consumption (Mo(2)) was extremely high in 100% lake water but decreased by 40%-68% after acclimation to dilute lake water. At every fixed swimming speed, Mo(2) was significantly reduced (by 50% at high speeds), and critical swimming speed was elevated in fish in 10% lake water relative to 100% lake water. Osmotic and Cl(-) concentration gradients from water to plasma were actually increased, and osmotic and Na(+) gradients were reversed, in 10% and 1% dilutions relative to 100% lake water, whereas acid-base gradients were greatly reduced. We suggest that approximately 50% of the animal's high metabolic demand originates from the cost of acid-base regulation in the highly alkaline Lake Magadi. When this load is reduced by environmental dilution, the energy saved can be diverted to enhanced swimming performance.

The aim of the present project is to determine the feasibility of measuring hepatic cytochrome P4501A1 (CYP1A1) and metallothionein (MT) mRNA in fish as an integrative measurement of persistent organic pollutants (POPs) and heavy metal contamination in sediment arising in Hong Kong. Sediment samples were collected from different sites, including Victoria Harbour (VS6), Yim Tin Tsai (YTT) at Tolo Harbour, Mai Po marshes (MPM) at Deep Bay, and Southern Waters (SS6) of coastal waters. The samples were analyzed for total and extractable concentrations of Cd, Cu, Ni, Zn, and Pb, as well as PCBs and PAHs. In addition, biomarker responses were studied in tilapia exposed experimentally to coastal sediment for 7 days. Using RT-PCR technique, hepatic CYP1A1 and MT mRNA were measured. Three control groups were used, including one negative control group maintained in sea water only; the second and third positive control groups were in sea water but were intraperitoneally injected with either beta-naphthoflavone (40 microg/g body weight) or cadmium chloride (10 microg/g body weight), respectively. The chemical data showed that VS6, YTT, and MPM were classified as Class C sediment according to the sediment quality criteria defined by the Hong Kong Environmental Protection Department, indicating the sites were heavily polluted. The exposure of tilapia to the sediment induced hepatic CYP1A1 (VS6 > YTT > MPM > SS6) and MT (VS6 > MPM > YTT > SS6) levels. The induction patterns were comparable to the levels of POPs and metal contamination in the sediment, indicating that the biomarker responses could be used to differentiate low to high levels of contamination among sediment.
