ABSTRACT

A three-wavelength photoelasticity method is proposed to simplify the optical setup and speed up data acquisition. By recording six intensity images with circularly polarized illuminations of three close wavelengths, the phase retardation and corresponding inner stress can be computed accurately with a correspondingly developed computational algorithm. Since the mechanical rotations of wave plates and polarizers required by classic photoelasticity techniques are avoided, the data acquisition of this proposed method is very speedy, and measurement of a dynamic sample can be achieved with a very simple and compact optical setup. Besides theoretical analyses, numerical and experimental evidences are also used to confirm the feasibility of this suggested three-wavelength digital photoelasticity method.

ABSTRACT

BACKGROUND: Acute diarrhoea is a common infectious disease among children in many countries and it has different kinds of clinical symptoms including vomiting, abdominal cramps, or fever of 38 °C. Some specific intestinal bacteria and their quantities can result in relevant symptoms. AIM: To analyze the correspondence between enteropathogenic bacteria and acute diarrhoea at family-level using high-throughput sequencing approach. METHODS: Every 30 children of acute diarrhoea with abdominal cramps, vomiting, and fever of 38 °C was regarded as a group, respectively. Stools samples were collected from each group and the DNA of stool was examined by E.Z.N.A.(®) Stool DNA Kit. The 16S rRNA genes sequencing was performed on an Illumina Miseq platform. FINDINGS: The sequencing dataset comprised 65,092 valid reads sequences that affiliated to the 18 phylogenetic families. The four dominant taxonomic groups in all three samples were Streptococcaceae, Veillonellaceae, Enterobacteriaceae and Lactobacillaceae. The stools of children with high fever presented higher pathogenic bacterial diversities and more complex community structures than other two groups. Lactobacillaceae was the enteric predominant microflora that could reduce the severity of acute diarrhoea. CONCLUSION: The reduction of predominant microflora or the aberrant proliferation of sub-dominant microflora can break the intestinal operation mechanism and cause intestinal diseases. What's more, people's living habits are also correlative about acute diarrhoea and parents should prepare light food for their children in order to protect their tender gastrointestinal mucosa.

ABSTRACT

Rapid and effective repair of epithelial tissue is desirable for improving the success rate of operation and reducing postoperative complications. Hydrogel is a widely studied wound repair material, especially as a wound dressing for damaged epithelial tissue. Based on the catalytic effect of thrombin on fibrinogen, in this study, a three-dimensional fibrin gel which of adequate epithelial cell compatibility was constructed by using thrombin and fibrinogen under the cross-linking action of calcium ion. Immunofluorescence staining and hematoxylin-eosin (H&E) staining showed that bone marrow mesenchymal stem cell (BMSC) was embedded in fibrin gel. Furthermore, vascular endothelial growth factor (VEGF) was used to induce BMSC to differentiate into CD31+ and vWF+ endothelial cell (EC) in fibrin gel. The results showed that the fibrin gel surface may effectively promote the adhesion and proliferation of EC and smooth muscle cell (SMC). After 15 days of culture, it was found that the BMSC embedded in the hydrogel had differentiated into EC. The results of in vivo skin wound experiment in rats further proved that the fibrin gel containing BMSC could promote wound healing and repair, and showed the potential to promote neovascularization at the injured site. The construction method of hydrogel materials proposed in this study has potential application value in the field of regenerative medicine.

ABSTRACT

To study the diversity and cultivar-specific of phytochemicals in sweet potato, Liquid Chromatography-Electrospray Ionization-Mass Spectrometry was used to analyze the metabolic profiles of five sweet potato cultivars exhibiting different flesh colors: purple, yellow/orange, and white. A total of 213 metabolites, including 29 flavonoids and 27 phenolic acids, were characterized. The flavonoid profiles of the five different cultivars were distinguished using PCA, the results suggested the flesh color accounted for the observed metabolic differences. In addition to anthocyanins, quinic acids and ferulic acids were the prominent phenolic acids, O-hexoside of quercetin, chrysoeriol were the prominent flavonoids in sweet potato tubers, and they were all higher in the OFSP and PFSP than WFSP. The main differential metabolic pathways between the OFSP, PFSP and the WFSP included those relating to phenylpropanoid and flavonoid biosynthesis. This study provides new insights into the differences in metabolite profiles among sweet potatoes with different flesh colors.

ABSTRACT

According to the logic of the Trivers-Willard hypothesis, in a human population, if socioeconomic status is transmitted across generations to some extent, and if sons of high-status parents tend to have higher reproductive success than daughters, while daughters of low-status parents tend to have higher reproductive success than sons, then we should expect that offspring sex ratio is positively associated with socioeconomic status. This study examines whether the assumptions and prediction of this hypothesis apply to a rural population in northern China. Results show that (1) current family socioeconomic status is positively related to family head's father's socioeconomic status in around 1950, (2) low-status family heads have more grandchildren through their daughters than their sons, whereas high- or middle-status family heads have more grandchildren through sons, and (3) as family heads' status increases, they tend to produce a higher offspring sex ratio. Therefore, the assumptions and prediction of the hypothesis are met in the study population. These results are discussed in reference to past studies on sex ratio manipulation among humans.

ABSTRACT

Significant alteration in the microbial community can occur across reclamation areas suffering subsidence from mining. A reclamation site undergoing fertilization practices and an adjacent coal-excavated subsidence site (sites A and B, respectively) were examined to characterize the bacterial diversity using 454 high-throughput 16S rDNA sequencing. The dominant taxonomic groups in both the sites were Proteobacteria, Acidobacteria, Bacteroidetes, Betaproteobacteria, Actinobacteria, Gammaproteobacteria, Alphaproteobacteria, Deltaproteobacteria, Chloroflexi, and Firmicutes. However, the bacterial communities' abundance, diversity, and composition differed significantly between the sites. Site A presented higher bacterial diversity and more complex community structures than site B. The majority of sequences related to Proteobacteria, Gemmatimonadetes, Chloroflexi, Nitrospirae, Firmicutes, Betaproteobacteria, Deltaproteobacteria, and Anaerolineae were from site A; whereas those related to Actinobacteria, Planctomycetes, Bacteroidetes, Verrucomicrobia, Gammaproteobacteria, Nitriliruptoria, Alphaproteobacteria, and Phycisphaerae originated from site B. The distribution of some bacterial groups and subgroups in the two sites correlated with soil properties and vegetation due to reclamation practice. Site A exhibited enriched bacterial community, soil organic matter (SOM), and total nitrogen (TN), suggesting the presence of relatively diverse microorganisms. SOM and TN were important factors shaping the underlying microbial communities. Furthermore, the specific plant functional group (legumes) was also an important factor influencing soil microbial community composition. Thus, the effectiveness of 454 pyrosequencing in analyzing soil bacterial diversity was validated and an association between land ecological system restoration, mostly mediated by microbial communities, and an improvement in soil properties in coalmining reclamation areas was suggested.
