ABSTRACT

The aim of this study was to explore the effect of emodin on lipid accumulation and inflammation in hepatocytes. The cell morphology was observed by microscopy. LDH release was detected by the kit. Levels of intracellular lipid droplets were observed by oil red O staining. The contents of TC and TG in cells were detected by the kit. Western blot was used to determine protein expressions of FASN,SREBF2,APOB,IL-6 and p-NF-κB in hepatocytes. The results showed that the levels of L02 cell LDH were significantly increased after being treated with emodin,and the cells showed shrinkage,volume reduction,decrease in quantity with the increase of dose. Red lipid droplets were observed in L02 hepatocytes. Intracellular TC and TG contents of L02 cell increased in a concentrationdependent manner,with significant differences between medium and high-dose groups( P < 0. 05). Protein expressions of FASN,SREBF2,IL-6 and p-NF-κB were significantly higher than those of the control group,and the expression level of APOB was significantly lower than that of the control group( P<0. 05). In conclusion,emodin could induce lipid accumulation and inflammatory damage in hepatocytes in a dose-dependent manner,which in turn could damage liver cells. This process was related to the up-regulation of FASN,SREBF2,IL-6,p-NF-κB,as well as the down-regulation of the protein expression of APOB.

ABSTRACT

OBJECTIVE: Lymphoma is considered to be a kind of malignant tumour. Gene therapy and radiotherapy have been reported as treatment methods for head and neck lymphoma. This study aims to evaluate the efficacy and safety for the treatment of head and neck lymphoma by a combination of recombinant adenovirus p53 (rAd-p53) and radiotherapy. METHODS: A total of 156 patients with head and neck lymphoma were selected. All patients received an intratumor injection of rAd-p53 of four different doses, namely, 0, 1 × 1010 VP, 1 × 1011 VP and 1 × 1012 VP, once a week for 8 weeks, and radiotherapy was administered 3 days after the rAd-p53 injection using the same dosage and method. Four, eight and twelve weeks after treatment, tumor reduction and complete response (CR) rates, special laboratory examination and adverse reaction assessment were detected to evaluate the efficacy and safety of combined treatment with rAd-p53 injection and radiotherapy for head and neck lymphoma. RESULTS: At week 4, 8 and 12 of treatment with rAd-p53 at the 1 × 1010 VP, 1 × 1011 VP and 1 × 1012 VP doses, the average tumour reduction and CR rates were evidently elevated, the anti rAd-p53 antibody in the serum of patients was expressed positively, and the T cell subsets (CD3/CD4/CD8) increased and interleukin 2 receptor (IL-2R) level decreased markedly. Additionally, rAd-p53 was proven to be clinically safe in the treatment. CONCLUSION: Altogether, we conclude that rAd-p53 combined with radiotherapy improves the efficacy and safety in treating head and neck lymphoma, which has a broad scope in future clinical application.

ABSTRACT

Geochemical dynamics of cave water were monitored to unveil its variation and controlling factors from October 2009 to December 2013 in Jiguan Cave,west of Henan province,southeastern coast of the loess plateau. The results showed that: (1) the hydrochemical types of the cave water are HCO(3-)-Ca(2+)-Mg2+ and HCO(3-)-Mg(2+)-Ca2+. HCO(3-) are over 80% of the anions, Ca2+ and Mg2+ are the dominate cations, and ground river keeping in erosion and pool water drips in deposition all the year. (2) Dripping water and pool water in Ji guan cave can respond perfectly to the change of external climate environment, which geochemistry indexes possess the extraordinary seasonal effects. (3) The concentration changes of the Ca2+, Mg2+ , SO4(2-) responded sensitively to annual precipitation change. Ca2+, Mg2+, SO4(2-) rise in waterlogging year and fall in drought year. Because HCO(3-) controlled by CO2 concentration. HCO(3-) concentration showed a unconspicuous response to the change of external climate environment. (4) The concentration changes of Ca2+, Mg2+, SO4(2-) have no obvious seasonal variation and showed a unconspicuous response to the change of external climate environment.
