ABSTRACT

The transition to parenthood is a significant period of transformation and adjustment for all members of a new family, reshaping relational dynamics that often endure throughout the initial years of a child's life. This qualitative and longitudinal study aimed to explore coparenting representations held by both partners in a parental couple, along with observations of their family interactions, at three distinct points in time, with the aim to better understand the interaction between these representational and behavioral observations. In-depth interviews were conducted with 17 heterosexual, first-time parents in Santiago de Chile, during pregnancy and at two different times during the first year of their child's life. Family interactions were assessed using the Lausanne Trilogue Play task (LTP) on all occasions. Three main coparenting representation categories emerged: traditional, ambiguous, and co-responsible. On observation, approximately half of the couples showed cooperative coparenting interactions, while the other half had conflictual interactions. Couples generally followed stable trajectories over time. Cooperative couples demonstrated co-responsibility and open dialogue, while conflictual couples tended to follow traditional gender roles with tacit communication styles. This study highlights the importance of promoting co-responsibility and dialogue for fostering cooperative relational dynamics during the critical transition to parenthood.


La transición a la paternidad/maternidad es un período significativo de transformación y ajuste para todos los miembros de una nueva familia, dándole nueva forma a las dinámicas de relación que a menudo se sobrellevan a lo largo de los años iniciales de la vida del niño. Este estudio cualitativo y longitudinal se propuso explorar las representaciones en la crianza compartida que tienen ambos miembros de una pareja de padres, junto con observaciones de sus interacciones familiares, en tres puntos distintivos en el tiempo, con el propósito de comprender mejor la interacción entre estas observaciones representacionales y de comportamiento. Se llevaron a cabo entrevistas profundas con 17 padres primerizos, heterosexuales, en Santiago de Chile, durante el embarazo y en dos momentos diferentes durante el primer año de vida de sus niños. Se evaluaron las interacciones de familia usando la tarea del Juego Tripartito de Lausanne (LTP) en todas las ocasiones. Tres principales categorías representacionales de crianza compartida surgieron: tradicional, ambigua y de responsabilidad compartida. Bajo observación, aproximadamente la mitad de las parejas mostró interacciones cooperadoras de crianza compartida, mientras que la otra mitad tuvo interacciones conflictivas. Las parejas generalmente siguieron trayectorias estables a lo largo del tiempo. Las parejas cooperadoras demostraron responsabilidad compartida y diálogo abierto, mientras que las parejas conflictivas tendieron a seguir papeles tradicionales de género con estilo de comunicación tácitos. Este estudio subraya la importancia de promover la responsabilidad compartida y el diálogo para cultivar relaciones dinámicas de cooperación durante la crucial transición a la paternidad/maternidad.

ABSTRACT

Understanding deficits in recognition of infant emotions in mothers with mental illnesses is limited by the lack of validated instruments. We present the development and content validation of the infant facial emotion recognition tool (InFER) in India to examine the ability of mothers to detect the infants' emotions. A total of 164 images of infant faces in various emotional states were gathered from the parents of four infants (two male and two female: up to 12 months old). Infant emotion in each image was identified by the respective mother. Content validation was carried out by 21 experts. Images with ≥70% concordance among experts were selected. The newly developed tool, InFER, consists of a total 39 infant images representing the six basic emotions. This tool was then administered among mothers during their postpartum period-10 healthy mothers and 10 mothers who had remitted from any schizophrenia spectrum disorder, bipolar affective disorder or major depressive disorder. The mean age and mean years of education for both groups were comparable (age∼25 years, education ∼15 years). A significant difference was found between the two groups in their ability to recognize infant emotions (Mann-Whitney U = 12.5; p = 0.004). InFER is a promising tool in Indian settings for understanding maternal recognition of infant emotions.


La comprensión de los déficits en el reconocimiento de las emociones del infante en madres con enfermedades mentales está limitada por la falta de instrumentos validados. Presentamos el desarrollo y la validación de contenido de una herramienta para reconocer la emoción facial del infante (InFER) en la India. para examinar la habilidad de las madres para detectar las emociones del infante. Se recogió un total de 164 imágenes de caras de infantes en varios estados emocionales, aportadas por los progenitores de 4 infantes (dos varones y dos niñas: de hasta 12 meses de edad). La mamá respectiva identificó la emoción del infante en cada imagen. La validación del contenido la llevaron a cabo 21 expertos. Para la construcción de esta herramienta se seleccionaron las imágenes que presentaban el ≥70% de concordancia entre los expertos. La recientemente desarrollada herramienta, InFER está compuesta de un total de 39 imágenes de infantes las cuales representan las seis emociones básicas. Esta herramienta entonces se puso en práctica entre madres durante su período de postparto ­10 madres saludables y 10 madres que habían sido remitidas en relación con cualquiera de los trastornos de la gama de la esquizofrenia, el trastorno afectivo bipolar o un trastorno depresivo mayor. El promedio de edad y el promedio de años de educación en ambos grupos eran comparables (edad ∼25 años, educación ∼15years). Una diferencia significativa se encontró entre los dos grupos en cuanto a su habilidad de reconocer las emociones del infante (Mann­Whitney U = 12.5; p = 0.004). InFER es una herramienta prometedora en entornos de la India para comprender el reconocimiento materno de las emociones del infante.


La compréhension des déficits dans la reconnaissance des émotions du bébé chez les mères ayant des maladies mentales est limitée par le manque d'instruments validés. Nous présentons le développement et la validation du contenu d'un outil de reconnaissance de l'émotion faciale du nourrisson (InFER) en Inde afin d'examiner la capacité des mères à détecter les émotions du bébé. Un total de 164 images de visages de bébés dans des états émotionnels variés ont été assemblées de parents de 4 bébés (deux garçons et deux filles: jusqu'à l'âge de 12 mois) L'émotion du bébé dans chaque image a été identifiée par la mère respective. La validation du contenu a été faite par 21 experts. Des images avec ≥70% de concordance entre les experts ont été sélectionnées pour la construction de l'outil. Ce nouvel outil, InFER, consiste en un total de 39 images de bébés représentant les six émotions de base. Cet outil a ensuite été utilisé chez des mères durant leur période postpartum ­ 10 mères en bonne santé et 10 mères s'étant remises soit d'un trouble du spectre schizophrénique, soit d'un trouble affectif bipolaire ou d'un trouble dépressif majeur. La moyenne d'âge et la moyenne d'années d'éducation pour les deux groupes étaient comparables (âge∼25ans, éducation ∼15ans). Une différence importante a été trouvée entre les deux groupes dans leur capacité à reconnaître les émotions du bébé (Mann­Whitney U = 12,5; p = 0,004). InFER est un outil prometteur dans les contextes de l'Inde pour comprendre la reconnaissance maternelle des émotions du bébé.

ABSTRACT

Infant and early childhood mental health consultation (IECMHC) in early care and education (ECE) settings is a promising approach to support young children. Although research on the effects of IECMHC is encouraging, it is limited by the complexities of the systems in which IECMHC is implemented and the variability in IECMHC models. The current study aims to clearly articulate a statewide, child-focused, short-term IECMHC model, assess consultee satisfaction, examine the effects of consultation on children's functioning in the school and home settings, and evaluate changes in teacher perceptions associated with expulsion risk following consultation. In total, 268 children ages 1-5 (69% White, 75% male) and their family and school caregivers participated in consultation in a New England state, and 95 children and caregivers were included in an evaluation subsample. Of this subsample, teachers and ECE administrators, but not families, indicated significant improvement in children's functioning from referral to end of consultation. There was also a significant decrease in children's risk of expulsion, as measured by teachers' perceptions associated with expulsion decisions. This study contributes to the IECMHC literature by providing results specific to a child-focused model of consultation and highlighting the possible role of adult attributions for children in ECE.


La consulta de salud mental infantil y la temprana niñez (IECMHC) en los entornos de cuidados y educación tempranos (ECE) es un acercamiento prometedor para apoyar a los niños pequeños. A pesar de que la investigación sobre los efectos de IECMHC es alentadora, está limitada por las complejidades de los sistemas dentro de los cuales se implementa IECMHC y la variabilidad en los modelos de IECMHC. El presente estudio se propone articular claramente un modelo de IECMHC para todo el estado, con enfoque en el niño y a corto plazo, evaluar la satisfacción que quienes participan de la consulta, examinar los efectos de la consulta sobre el funcionamiento de los niños en la escuela y en el entorno del hogar, así como evaluar los cambios en las percepciones de los maestros asociadas con los riesgos de expulsión después de la consulta. En total, 268 niños de edad 1­5 (69% blancos, 75% varones) y sus familias y quienes les cuidaba en la escuela participaron en la consulta en un estado de Nueva Inglaterra, y se incluyó a 95 niños y sus cuidadores en una evaluación de un subgrupo muestra. De este subgrupo muestra, los maestros y los administradores de ECE, pero no las familias, indicaron significativas mejorías en el funcionamiento de los niños desde la referencia hasta el final de la consulta. Se dio también una significativa disminución en el riesgo de expulsión de los niños, tal como se midió por medio de las percepciones de los maestros asociadas con las decisiones de expulsión. Este estudio contribuye a la literatura informativa sobre IECMHC por medio de ofrecer resultados específicos al modelo de consulta enfocado en el niño y subrayar el posible papel de las atribuciones del adulto para los niños en ECE.


La consultation de santé mentale du nourrisson et de la petite enfance (Infant and early childhood mental health consultation abrégé en anglais IECMHC) dans des contextes éducatifs et de crèches (abrégé ici CEC selon le français) est une approche prometteuse pour le soutien aux jeunes enfants. Bien que les recherches sur les effets de l'IECMHC sont encourageantes, elles sont limitées par les complexités des systèmes dans lesquels l'IECMHC est mise en oeuvre et la variabilité des modèles de l'IECMHC. Cette étude s'est donné pour but de clairement articuler un modèle IECMHC au niveau de l'état, focalisé sur l'enfant et à court terme, d'évaluer la satisfaction de la personne consultée, d'examiner les effets de la consultation sur le fonctionnement des enfants à l'école et à la maison, et d'évaluer les changements dans les perceptions de l'enseignant liée au risque d'expulsion après la consultation. En tout 268 enfants âgés de 1­5 ans (69% blancs, 75% garçons) et leur famille et les personnes prenant soin d'eux à l'école ont participé à une consultation dans un état de nous Nouvelle Angleterre (aux Etats­Unis), et 95 enfants et personnes prenant soin d'eux ont été inclus dans un sous­échantillon d'évaluation. Dans ce sous­échantillon les enseignants et administrateurs CEC, mais pas les familles, ont fait état d'une amélioration important dans le fonctionnement des enfants du moment de la référence à la fin de la consultation. Il n'a pas de baisse importante du risque d'expulsion des enfants, mesurée par les perceptions des enseignants liées aux décisions d'expulsion. Cette étude contribue aux recherches sur l'IECMHC en offrant des résultats spécifiques à un modèle de consultation focalisé sur l'enfant et en mettant en lumière le rôle possible des attributions adultes pour les enfants dans les CEC.

ABSTRACT

The zinc-iodine aqueous battery is a promising energy storage device, but the conventional two-electron reaction potential and energy density of the iodine cathode are far from meeting practical application requirements. Given that iodine is rich in redox reactions, activating the high-valence iodine cathode reaction has become a promising research direction for developing high-voltage zinc-iodine batteries. In this work, by designing a multifunctional electrolyte additive trimethylamine hydrochloride (TAH), a stable high-valence iodine cathode in four-electron-transfer I-/I2/I+ reactions with a high theoretical specific capacity is achieved through a unique amine group, Cl bidentate coordination structure of (TA)ICl. Characterization techniques such as synchrotron radiation, in-situ Raman spectra, and DFT calculations are used to verify the mechanism of the stable bidentate structure. This electrolyte additive stabilizes the zinc anode by promoting the desolvation process and shielding mechanism, enabling the zinc anode to cycle steadily at a maximum areal capacity of 57 mAh cm-2 with 97% zinc utilization rate. Finally, the four-electron-transfer aqueous Zn-I2 full cell achieves 5000 stable cycles at an N/P ratio of 2.5. The unique bidentate coordination structure contributes to the further development of high-valence and high capacity aqueous zinc-iodine batteries.

ABSTRACT

The control of cellular zinc (Zn) concentrations by dedicated import and export systems is essential for the survival and virulence of Pseudomonas aeruginosa. The transcription of its many Zn transporters is therefore tightly regulated by a known set of transcription factors involved in either the import or the export of Zn. In this work, we show that the Zur protein, a well-known repressor of Zn import, plays a dual role and functions in both import and export processes. In a situation of Zn excess, Zur represses Zn entry, but also activates the transcription of czcR, a positive regulator of the Zn export system. To achieve this, Zur binds at two sites, located by DNA footprinting in the region downstream the czcR transcription start site. In agreement with this regulation, a delay in induction of the efflux system is observed in the absence of Zur and Zn resistance is reduced. The discovery of this regulation highlights a new role of Zur as global regulator of Zn homeostasis in P. aeruginosa disclosing an important link between Zur and zinc export.

ABSTRACT

The EU In-Vitro Diagnostic Device Regulation (IVDR) aims for transparent risk-and purpose-based validation of diagnostic devices, traceability of results to uniquely identified devices, and post-market surveillance. The IVDR regulates design, manufacture and putting into use of devices, but not medical services using these devices. In the absence of suitable commercial devices, the laboratory can resort to laboratory-developed tests (LDT) for in-house use. Documentary obligations (IVDR Art 5.5), the performance and safety specifications of ANNEX I, and development and manufacture under an ISO 15189-equivalent quality system apply. LDTs serve specific clinical needs, often for low volume niche applications, or correspond to the translational phase of new tests and treatments, often extremely relevant for patient care. As some commercial tests may disappear with the IVDR roll-out, many will require urgent LDT replacement. The workload will also depend on which modifications to commercial tests turns them into an LDT, and on how national legislators and competent authorities (CA) will handle new competences and responsibilities. We discuss appropriate interpretation of ISO 15189 to cover IVDR requirements. Selected cases illustrate LDT implementation covering medical needs with commensurate management of risk emanating from intended use and/or design of devices. Unintended collateral damage of the IVDR comprises loss of non-profitable niche applications, increases of costs and wasted resources, and migration of innovative research to more cost-efficient environments. Taking into account local specifics, the legislative framework should reduce the burden on and associated opportunity costs for the health care system, by making diligent use of existing frameworks.

ABSTRACT

In the genus Pseudomonas, zinc homeostasis is mediated by a complete set of import and export systems, whose expression is precisely controlled by three transcriptional regulators: Zur, CzcR and CadR. In this review, we describe in detail our current knowledge of these systems, their regulation, and the biological significance of zinc homeostasis, taking Pseudomonas aeruginosa as our paradigm. Moreover, significant parts of this overview are dedicated to highlight interactions and cross-regulations between zinc and copper import/export systems, and to shed light, through a review of the literature and comparative genomics, on differences in gene complement and function across the whole Pseudomonas genus. The impact and importance of zinc homeostasis in Pseudomonas and beyond will be discussed throughout this review.

ABSTRACT

Maternal grandmothers play a vital role in the transition to motherhood for their own daughters. The current study adds to this literature by investigating the lived experience of motherhood for women who lacked a meaningful relationship with their mothers. Ten mothers of children under 2 years of age participated in a semi-structured interview to explore their lived experiences of being a mother. Women were recruited from two parent-infant services in Northern Ireland. The interviews were analyzed using Interpretive Phenomenological Analysis (IPA). Three superordinate themes were identified: 'The Birth of a Mother', 'Mourning and Loss' and 'Ghosts in the Nursery'. The first theme captured the significant change of identity women experienced during their transition to motherhood. This identity change shed new light on their own experience of being mothered. The second theme captured the mourning and loss these women felt due to their relationship with their mother. Their lack of meaningful maternal relationships have left a hole impossible to fill. The final theme spoke to the intergenerational element of these mother's experience and their desire to break a cycle of maternal deprivation. The rich content from the interviews highlights the need for services to be aware of this struggle of motherhood.


Las abuelas maternas juegan un papel vital en la transición a la maternidad para sus propias hijas. El presente estudio contribuye a este campo de conocimientos escritos por medio de investigar la experiencia de maternidad vividas por mujeres a quienes les hace falta una significativa relación con sus madres. Diez madres de niños menores de dos años de edad participaron en una entrevista semiestructurada para explorar sus vividas experiencias de ser madre. A las mujeres se les reclutó de dos servicios progenitor-infante en Irlanda del Norte. Se analizaron las entrevistas usando el Análisis Fenomenológico Interpretativo (IPA). Se identificaron tres temas de nivel superior: 'El Nacimiento de una Madre,' 'Lamento y Pérdida' y 'Fantasmas en el Cuarto de la Niña.' El primer tema captó el significativo cambio de identidad que las mujeres experimentan durante su transición a la maternidad. El cambio de identidad arrojó una nueva luz en sus propias experiencias de ser criadas por una madre. El segundo tema captó la lamentación y la pérdida que estas mujeres sentían debido a su relación con sus madres. La falta de significativas relaciones maternas ha dejado un vacío imposible de llenar. El tema final tuvo que ver que el elemento intergeneracional de la experiencia de estas madres y su deseo de romper un ciclo de privación materna. El rico contenido de las entrevistas enfatiza la necesidad de servicios para estar conscientes de esta lucha sobre la maternidad.


Les grands-mères maternelles jouent un rôle vital dans la transition à la maternité de leurs propres filles. Cette étude s'ajoute aux recherches précédentes en enquêtant sur l'expérience vécue de la maternité pour les femmes n'ayant pas eu une relation importante avec leurs mères. Dix mères d'enfant de moins de deux ans ont participé à un entretien semi structuré afin d'explorer leurs expériences vécues du fait d'être maman. Ces femmes ont été recrutées dans deux services parent-bébé en Irlande du Nord. Ces entretiens ont été analysés en utilisant l'Analyse Interprétive Phénoménologique (en anglais Interpretive Phenomenological Analysis, soit IPA). Trois thèmes supérieurs ont été identifiés: 'La Naissance d'une Mère', "Deuil et Perte' et 'Fantômes dans la Chambre d'enfant'. Le premier thème a capture le changement d'identité important que les femmes ont vécu durant leurs transitions à la maternité. Ce changement d'identité a apporté un éclairage nouveau sur leur propre expérience d'avoir été maternées. Le second thème a capturé le deuil et la perte que ces femmes ont ressentis du fait de leur lien à leur mère. Leur manque de relations maternelles importantes a laissé un trou impossible à remplir. Le dernier thème fait référence à l'élément intergénérationnel de l'expérience de ces mères et leur désir de casser un cycle de privation maternelle. Ce contenu riche émanant des entretiens met en lumière la prise de connaissance nécessaire de ces difficultés de la maternité dont doivent faire preuve les services.

For new fathers, parenting stress is a risk factor for impaired early parenting and child maltreatment perpetration. Predictors of parenting stress, including fathers' own experiences of trauma, could be useful intervention targets to support new fathers. We aim to examine associations between new fathers' own histories of child maltreatment, and their perinatal mental health, relationships, and parenting stress. We recruited 298 first-time fathers for a survey that measured child maltreatment history, trauma sequelae including posttraumatic stress disorder (PTSD), major depressive disorder (MDD), interpersonal reactivity, substance use, anger expression, coparenting quality, and parenting stress. On the Parenting Stress Index (PSI) (from 36 to 180), bivariate analysis demonstrated that new fathers who experienced child maltreatment (n = 94) had significantly higher parenting stress (x̅ = 85.3, σ = 18.7) than those who did not (n = 204; x̅ = 76.0, σ = 16.6; P < .000). Hierarchical linear regression modeling indicated that a child maltreatment history, PTSD, and MDD were significantly associated with parenting stress. The strongest predictors of parenting stress were coparenting quality and complex trauma sequelae-interpersonal reactivity and anger expression. Interventions to reduce fathers' parenting stress by targeting known mental health and relationship sequelae of maltreatment are promising avenues to breaking intergenerational transmission of child maltreatment and psychiatric vulnerability.


Para nuevos papás, el estrés de crianza es un factor de riesgo para la deficiente crianza temprana y para cometer maltrato infantil. Los factores de predicción del estrés de crianza, incluyendo las propias experiencias de trauma de los papás, pueden ser útiles metas de intervención para apoyar a los nuevos papás. Nos propusimos examinar las asociaciones entre las propias historias de maltrato de los nuevos papás, y su salud mental perinatal, relaciones y estrés de crianza. Reclutamos 298 papás primerizos para una encuesta que medía la historia de maltrato infantil, la secuela de trauma incluyendo el trastorno de estrés postraumático (PTSD), el trastorno depresivo serio (MDD), la reactividad interpersonal, el uso de sustancias, la expresión de ira, la calidad de la crianza compartida, así como el estrés de crianza. En el Índice de Estrés de Crianza (de 36-180), los análisis bivariantes demostraron que los nuevos papás que habían experimentado maltrato infantil (N = 94) tenían significativamente un mayor estrés de crianza (x̅ = 85.3, σ = 18.7) que aquellos que no habían tenido tal experiencia (N = 204; x̅ = 76.0, σ = 16.6; P<.000). El modelo de regresión lineal jerárquica indicó que una historia de maltrato infantil, PTSD y MDD estaban significativamente asociados con el estrés de crianza. Pour les nouveaux pères le stress de parentage est un facteur de risque pour le parentage précoce compromis et la perpétration de maltraitance de l'enfant. Les prédicteurs de stress de parentage, y compris les propres expériences de trauma des pères, pourraient être des cicles d'intervention utiles afin de soutenir les nouveaux pères. Nous nous sommes donné pour but d'examiner les liens entre le propre passé de maltraitance de l'enfant des nouveaux pères et leur santé mentale périnatale, leurs relations et le stress de parentage. Nous avons recruté 298 nouveaux pères (pères pour la première fois) pour un sondage mesurant l'histoire de la maltraitance de l'enfant, les séquelles de trauma y compris les troubles de stress post-traumatique (TSPT), les troubles dépressifs majeurs (MDD en anglais), la réactivité interpersonnelle, la toxicomanie, l'expression de colère et la qualité du co-parentage ainsi que le stress parental. Pour l'Index de Stress de Parentage (de 36-180), une analyse bivariée a montré que les nouveaux pères qui avaient fait l'expérience de maltraitance de l'enfance (N = 94) avaient un stress de parentage bien plus élevé (x̅ = 85,3, σ = 18,7) que ceux n'en ayant pas fait l'expérience (N = 204; x̅ = 76,0, σ = 16,6; P<,000). Un modèle de régression linéaire hiérarchique a indiqué qu'un passé de maltraitance de l'enfant, le TSPT et le MDD étaient fortement liés au stress de parentage. Les facteurs de prédiction les plus forts de stress de parentage étaient la qualité du co-parentage et les séquelles de trauma complexes - réactivité interpersonnelle et l'expression de la colère. Les interventions pour réduire le stress de parentage des pères en ciblant la santé mentale connue et les séquelles de maltraitance sont un chemin prometteur pour casser la transmission intergénérationnelle de la maltraitance de l'enfant et la vulnérabilité psychiatrique.

ABSTRACT

Previous studies on the context between death anxiety and religion do not provide any clear evidence regarding "anxiety buffer" function. In this explorative study, death anxiety and attitude to death were determined in the context of mood, personality and meaning of life among groups of Muslims (n = 60) and Christian Protestants (n = 60). Death anxiety and attitude to death were assessed using the Bochum questionnaire for recording death anxiety and attitudes to death. Death anxiety was mild to moderate in our healthy Participants of Muslim and Christian faith. Attitude towards death was therefore much more pronounced among Muslim members than Christians. The influence of religious beliefs on the fear of death does not appear to be direct and linear. Sources that provide meaning in life and emotional stability can contribute to a reduction in death anxiety and a less problematic attitude towards death.

ABSTRACT

Zinc is an essential cofactor for many metal enzymes and transcription regulators. Zn2+ availability has long been known to affect antibiotic production and morphological differentiation of Streptomyces species. However, the molecular mechanism whereby zinc regulates these processes remains unclear. We investigated the regulatory roles of the zinc-sensing regulator Zur in Streptomyces avermitilis. Our findings demonstrate that Zur plays an essential role in maintaining zinc homeostasis by repressing the expression of the zinc uptake system ZnuACB and alternative non-zinc-binding ribosomal proteins and promoting the expression of zinc exporter ZitB. Deletion of the zur gene resulted in decreased production of avermectin and oligomycin and delayed morphological differentiation, and these parameters were restored close to wild-type levels in a zur-complemented strain. Zur bound specifically to Zur box in the promoter regions of avermectin pathway-specific activator gene aveR, oligomycin polyketide synthase gene olmA1, and filipin biosynthetic pathway-specific regulatory genes pteR and pteF. Analyses by reverse transcription quantitative PCR and luciferase reporter systems indicated that Zur directly activates the transcription of these genes, i.e., that Zur directly activates biosynthesis of avermectin and oligomycin. Zur positively regulated morphological development by repressing the transcription of differentiation-related genes ssgB and minD2. Our findings, taken together, demonstrate that Zur in S. avermitilis directly controls zinc homeostasis, biosynthesis of avermectin and oligomycin, and morphological differentiation. IMPORTANCE Biosynthesis of secondary metabolites and morphological differentiation in bacteria are affected by environmental signals. The molecular mechanisms whereby zinc availability affects secondary metabolism and morphological differentiation remain poorly understood. We identified several new target genes of the zinc response regulator Zur in Streptomyces avermitilis, the industrial producer of avermectin. Zur was found to directly and positively control avermectin production, oligomycin production, and morphological differentiation in response to extracellular Zn2+ levels. Our findings clarify the regulatory functions of Zur in Streptomyces, which involve linking environmental Zn2+ status with control of antibiotic biosynthetic pathways and morphological differentiation.

ABSTRACT

The present study uses a prospective longitudinal study design to investigate the development of maternal self-efficacy in the transition phase to parenthood, drawing on a large sample of socially and/or culturally disadvantaged families (N = 292). Parity, maternal education, migration, informal and formal social support are considered as potential predictors. Results indicate that previous birth experience, being born abroad, and higher levels of formal and informal social support during pregnancy jointly predict higher levels of maternal self-efficacy three months after birth. First-time mothers and mothers born in Germany (where the study was conducted) benefit more from formal support than mothers with previous experience and mothers born outside of Germany. Overall, maternal self-efficacy increases significantly. Implications for prenatal maternal care are discussed.


El presente estudio usa un diseño de estudio longitudinal potencial para investigar el desarrollo de la auto efectividad materna en la fase de transición a ser madre. Usando una muestra grande de familias desfavorecidas social y/o culturalmente (N = 292). Se consideran como potenciales factores de predicción la paridad, la educación materna, la migración, y el apoyo social informal y formal. Los resultados indican que una previa experiencia de dar a luz, el haber nacido en el extranjero, así como altos niveles de apoyo social formal e informal durante el embarazo predicen en conjunto altos niveles de auto efectividad materna tres meses después del parto. Las madres primerizas y las madres nacidas en Alemania (donde se llevó a cabo el estudio) se beneficiaron más del apoyo formal que las madres con experiencia previa y las madres nacidas fuera de Alemania. En general, la auto efectividad materna aumentó significativamente. Se discuten las implicaciones para el cuidado materno prenatal.


Cette étude utilise un plan d'étude prospectif longitudinal pour se pencher sur le développement de l'auto-efficacité maternelle dans la phase de transition à la parenté, en utilisant un grand échantillon de familles socialement et/ou culturellement défavorisées (n = 292). La parité, l'éducation maternelle, la migration, le soutien social informel et formel sont considérés comme des prédicteurs potentiels. Les résultats indiquent qu'une expérience de la naissance antérieure, le fait d'être née à l'étranger et des hauts niveaux de soutien social formel et informel durant la grossesse prédisent ensemble des niveaux élevés d'auto-efficacité maternelle après la naissance. Les mères étant mères pour la première fois et les mères nées en Allemagne (où l'étude a été faite) ont plus bénéficié d'un soutien formel que les mères ayant déjà donné naissance à un enfant et que les mères nées en dehors de l'Allemagne. Dans l'ensemble l'auto-efficacité maternelle a augmenté de manière importante. Les implications pour le soin maternel prénatal sont discutées.

ABSTRACT

IECMHC can and should be a vehicle that promotes greater equity in access to high quality relationships within an early classroom environment. It is important to consider consultation through a racial equity lens to ensure that it is integrated in all levels of work. The goal of the study was to replicate the results from other IECMHC programs with a diverse, largely Latinx, population. The Jump Start program was given the unique opportunity to adapt the Georgetown Framework of Infant and Early Childhood Mental Health Consultation to a multicultural population in Miami. A total of 88 early learning programs and 244 teachers participated. Services were provided in English, Spanish, and Creole at the program- and classroom-level. Pre- and post-data were collected at both levels. Significant improvements at the program- and classroom-level were found post-consultation. Consultants demonstrated fidelity to core program practices while providing culturally and linguistically competent service. The current study replicated findings regarding the effectiveness of IECMHC while expanding results to a diverse metropolitan community. Key features of program success may be attributed to the use of highly trained consultants, action planning, fidelity monitoring, and enrollment of programs that were ready and have a champion for IECMHC.

ABSTRACT

Infant and Early Childhood Mental Health Consultation (IECMHC) aims to improve early childhood professionals' abilities to promote children's mental health through relationship building and collaboration. Using a longitudinal, matched-comparison group design, a 3-year pilot study of a cross-system, embedded model of IECMHC assessed teachers and home visitors in intervention and comparison programs in reflective capacity, burnout, and perceptions of children's behavior. A sample of 136 staff (n = 72 intervention group; n = 64 comparison group; 21% Black; 51% White; 28% Latina/Hispanic) participated in surveys over a 21-month implementation period. A subsample of staff (n = 26) participated in interviews that included a narrative measure of reflective capacity; and a smaller subsample of teachers only (n = 21) completed assessments of children. Staff in the intervention group significantly increased reflective capacity after 21 months. For the staff interview subsample, receiving the intervention predicted lower levels of burnout at 12-15 months post-baseline. Among teachers completing child assessments, those with higher reflective capacity rated children's behaviors more positively than teachers with lower reflective capacity. We conclude that this IECMHC model successfully improved reflective capacity in staff. Future research should investigate reflective capacity as a potential mechanism of change for IECMHC.


La Consulta de Salud Mental de la Infancia y la Temprana Niñez (IECMHC) se propone mejorar las habilidades de los profesionales de la temprana niñez para promover la salud mental de los niños por medio de establecer relaciones y de la colaboración. Usando un diseño longitudinal, de un grupo de comparación emparejado, un estudio experimental de tres años con un modelo incorporado de IECMHC a lo largo del sistema, evaluó a instructores y visitadores a casa en programas de intervención y comparación en cuanto a la capacidad de reflexión, el agotamiento, así como las percepciones del comportamiento de los niños. Un grupo muestra de 136 miembros del personal (n = 72 en el grupo de intervención; n = 64 en el grupo de comparación; 21% negros; 51% blancos; 28% hispano/latino) participó en entrevistas que incluían una medida narrativa de la capacidad de reflexión; y un más pequeño subgrupo muestra de instructores solamente (n = 21) completaron evaluaciones de los niños. Los miembros del personal en el grupo de intervención significativamente aumentaron la capacidad de reflexión después de 21 meses. Para la entrevista del personal en el subgrupo muestra, recibir la intervención predijo más bajos niveles de agotamiento a los 12 a 15 meses después del punto de partida. De los instructores que completaron las evaluaciones del niño, aquellos con más alta capacidad de reflexión evaluaron el comportamiento de los niños más positivamente que los instructores con una más baja capacidad de reflexión. La consultation de Santé Mentale du Nourrisson et de la Petite Enfance (abrégé IECMHC en anglais) a pour but d'améliorer les compétences des professionnels de la petite enfance à promouvoir la santé mentale des enfants au travers de l'établissement de relations et de la collaboration. En utilisant une conception de groupe témoin apparié, longitudinale, une étude pilote de trois années d'un modèle inter-système et intégré de IECMHC a évalué des enseignants et des visiteurs à domicile dans des programmes d'intervention et de comparaison pour ce qui concerne la capacité de réflexion, le burnout et les perception du comportement des enfants. Un échantillon de 136 employés (n = 72 groupe d'intervention; n = 64 groupe de comparaison; 21% Noirs; 51% Blancs; 28% Hispaniques / Latinas) ont participé à des enquêtes sur une période de mise en place de 21 mois. Un sous-échantillon d'employés (n = 26) a participé à des entretiens qui ont inclus une mesure narrative de capacité de réflexion; et un plus petit sous-échantillon d'enseignants uniquement (n = 21) a rempli des évaluations des enfants. Les employés dans le groupe d'intervention ont fortement augmenté leur capacité de réflexion après 21 mois. Pour le sous-échantillon d'employés, le fait d'avoir reçu l'intervention a prédit des niveaux plus bas de burnout à 12-15 mois après référence. ABSTRACT

When a cell is zinc-deficient, ykgM and ykgO, which encode paralogs of the zinc-binding ribosomal proteins L31 and L36, are expressed from the ykgM operon, which is ordinarily held inactive by the Zur repressor. In ribosomes lacking L31, ribosomal subunit association is weakened, resulting in reduced in vitro translation and the deletion mutants of rpmE, the gene encoding L31, forming small colonies. We isolated four suppressor mutants of ∆rpmE that formed normal colonies. All four mutation sites were located in zur, and ribosomes of zur mutant cells contained one copy of YkgM and had translational activities equivalent to those of ribosomes containing L31. L36 is highly conserved among bacteria, chloroplast and mitochondria. Analysis of a deletion mutant of rpmJ, which encodes L36, suggested that L36 is involved in late assembly of the 50S particle, in vitro translation and cell growth. In zur mutant cells lacking rpmJ, the paralog YkgO was expressed and took over the functions of L36. zur mutant cells contained four types of ribosomes containing combinations of L31 or YkgM, and L36 or YkgO. Copy numbers of L31 and YkgM, and L36 and YkgO, summed to 1, indicating that each paralog pair shares a binding site.

BACKGROUND: Zinc uptake-regulator (Zur)-regulated lipoprotein A (ZrlA) plays a role in bacterial fitness and overcoming antimicrobial exposure in Acinetobacter baumannii. This study further characterized the zrlA gene and its encoded protein and investigated the roles of the zrlA gene in bacterial morphology, antimicrobial susceptibility, and production of outer membrane vesicles (OMVs) in A. baumannii ATCC 17978. RESULTS: In silico and polymerase chain reaction analyses showed that the zrlA gene was conserved among A. baumannii strains with 97-100% sequence homology. Recombinant ZrlA protein exhibited a specific enzymatic activity of D-alanine-D-alanine carboxypeptidase. Wild-type A. baumannii exhibited more morphological heterogeneity than a ΔzrlA mutant strain during stationary phase. The ΔzrlA mutant strain was more susceptible to gentamicin than the wild-type strain. Sizes and protein profiles of OMVs were similar between the wild-type and ΔzrlA mutant strains, but the ΔzrlA mutant strain produced 9.7 times more OMV particles than the wild-type strain. OMVs from the ΔzrlA mutant were more cytotoxic in cultured epithelial cells than OMVs from the wild-type strain. CONCLUSIONS: The present study demonstrated that A. baumannii ZrlA contributes to bacterial morphogenesis and antimicrobial resistance, but its deletion increases OMV production and OMV-mediated host cell cytotoxicity.

This study aimed to explore the factor structure, reliability, and validity of a Korean translation of the Parental Reflective Functioning Questionnaire (PRFQ). The PRFQ consists of three subscales: prementalizing modes, certainty about mental states, and interest and curiosity in mental states. A convenience sample of 163 Korean parents completed the K-PRFQ. Exploratory factor analysis showed three factors mapped on to the original PRFQ factors, but items from the original prementalizing modes subscale clustered into two additional factors. Data from a subsample (n = 67) showed that the certainty about mental states and interest and curiosity in mental states subscales correlated positively with more optimal self-reported parenting. We discuss the validity of using the PRFQ in collectivistic cultures.


Este estudio se propuso explorar la estructura de factores, la confiabilidad y la validez de una traducción al coreano del Cuestionario de Funcionamiento de Reflexión del Progenitor (PRFO; Luyten, Mayes, Nijssens y Fonagy, 2017). El PRFQ consiste en tres escalas: Modos de Pre-mentalización, la Certeza acerca de estados mentales y el Interés y curiosidad por los estados mentales. Un conveniente grupo muestra de 163 progenitores coreanos completó el K-PRFQ. Los exploratorios análisis de factores mostraron tres factores identificados dentro de los factores del PRFQ original, sin embargo, temas de la sub-escala original de Modos de Pre-mentalización se agruparon en dos factores adicionales. Los datos provenientes de un sub-grupo muestra (n = 67) mostraron que la Certeza acerca de estados mentales y el Interés y curiosidad por los estados mentales se correlacionaron positivamente con una auto-reportada más óptima crianza. Discutimos la validez de usar el PRFQ en culturas colectivistas.

Une analyse exploratoire de facteur a montré trois facteurs mappés sur les facteurs d'origine du PRFQl, mais des aspects de la sous-échelle des modes de pré-mentalisation se sont regroupés en deux facteurs de plus. Les données d'un sous-échantillon (n = 67) ont montré que la Certitude sur les états mentaux et les sous-échelles d'Intérêt et la curiosité dans les états mentaux étaient en corrélation positive avec un parentage auto-rapporté optimal. Nous avons discuté la validité de l'utilisation du PRFQ dans les cultures collectivistes.

Young children's social-emotional development is powerfully shaped by their early environments, which for many includes early childhood education (ECE). Infant and Early Childhood Mental Health Consultation (IECMHC) pairs teachers and infant and early childhood mental health (IECMH) consultants to promote teachers' capacity to foster positive social-emotional development in ECE. Although the outcomes of IECMHC have been well studied, little research has investigated how this model leads to changes for teachers and children. According to theory, the quality of the relationship between teachers and IECMH consultants, termed consultative alliance (CA), is a key mechanism of change. This study analyzed the role of CA on 6-month outcomes of IECMHC in a sample of 316 children, 289 teachers, and 62 IECMH consultants. Results from multilevel models suggested that stronger CA predicted greater improvements in teacher-child closeness and teacher-rated child attachment behaviors. In addition, a strong CA was related to greater improvement in classroom climate, teachers' self-efficacy, and teachers' perceptions of their jobs. This study upholds the centrality of relationship-building and parallel process in mental health consultation, and by advancing understanding of the mechanisms of change for IECMHC may provide salient implications for policy and practice.


El desarrollo socio-emocional de los niños pequeños es formado de manera poderosa por sus tempranos ambientes, los cuales, para muchos incluyen la educación en la temprana niñez (ECE). La Asesoría de Salud Mental del Infante y la Temprana Niñez (IECMHC) empareja maestros y Asesores de la Salud Mental del Infante y la Temprana Niñez (IECMH) para promover la capacidad de los educadores de crear un positivo desarrollo socio-emocional en ECE. A pesar de que los resultados de IECMHC han sido bien estudiados, poca investigación se ha enfocado en cómo este modelo conduce a cambios para educadores y niños. De acuerdo con la teoría, la calidad de la relación entre educadores y Asesores de IECMH, llamada Alianza Consultiva (CA), es un mecanismo clave para el cambio. Este estudio analizó el papel de la Alianza Consultiva en resultados de seis meses de IECMHC en un grupo muestra de 316 niños, 289 educadores y 62 Asesores de IECMH. Los resultados de modelos de multiniveles sugieren que una más fuerte CA predijo mayores mejoras en la cercanía entre educador y niño y las conductas de afectividad del niño evaluadas por el educador. Adicionalmente, una más fuerte CA se relacionó con una mayor mejora en el ambiente del aula de clases, la auto-efectividad de los educadores y las percepciones que los educadores tenían de su trabajo. Este estudio apoya la centralidad de establecer una relación y un proceso paralelo en la asesoría de salud mental, y por medio del avance en la comprensión de los mecanismos de cambio para IECMHC, pudiera proveer implicaciones destacadas para la política y la práctica.


Le développement socio-émotionnel des jeunes enfants est fortement formé par leurs premiers environnements, ce qui pour bien d'entre eux inclut l'enseignement préscolaire. L'approche IECMHC (Infant and Early Childhood Mental Health Consultation soit Consultation en Santé Mentale du Nourrisson et de la Petite Enfance) jumelle des enseignants et des consultants IECHM (Santé Mentale du Nourrisson et de la Petite Enfance) afin de promouvoir la capacité des enseignements à cultiver et à favoriser un développement socio-émotionnel positif dans l'enseignement préscolaire. Bien que les résultats de l'approche IECMHC aient été bien étudiés il existe peu de recherches sur la manière dont ce modèle mène à des changements pour les enseignants et les enfants. Selon la théorie, la qualité de la relation entre les enseignants et les Consultants IECMH, appelée Alliance Consultative (AC), est un mécanisme clé de changement. Cette étude a analysé le rôle de l'Alliance Consultative sur les résultats à six mois de la IECMHC chez un échantillon de 316 enfants, 289 enseignants, et 62 Consultants IECMH. Les résultats de modèles multi-niveaux ont suggéré qu'une AC plus forte prédisait des améliorations plus importantes dans le rapprochement enseignant-enfant et dans les comportements d'attachement de l'enfant évalués par l'enseignant. De plus une AC plus forte était liée à une amélioration plus importante du climat de la salle de classe, de l'auto-efficacité des enseignements et des perceptions que les enseignants se faisaient de leur travail. Cette étude maintient la centralité de la construction de la relation et du processus parallèle dans la consultation de santé mentale. Trauma, Psychological Stress and Family Situation of Fathers who Experienced Child and Youth Welfare Services: Implications for Youth Welfare Interventions and Therapy This paper explores the current family situation and psychological stress of fathers who experienced trauma and different kinds of youth welfare services including out-of-home placement during their childhood. The main group consisted of n = 119 fathers and was compared to a group of n = 36 fathers whose families never received services from the child and youth welfare authority. In the main group, a high correlation was found between trauma and current psychological stress, albeit the out-of-home care intervention or intensive home-support by child and youth welfare services during childhood. The higher stress level in the main group of fathers in contrast to the comparison group effects their current family situation, especially concerning the contact between fathers and their children. Fathers with higher stress levels were less likely to be in contact with their children. Acting on the assumption that fathers are important for their children, the results implicate a heightened focus on earlier trauma-informed interventions in families with difficulties and out-of-home care settings, to reduce stress levels and prevent intergenerational transmission of problematic family experiences. Current child and youth welfare services, in particular out-of-home placements, should take these trauma-informed approaches and therapy into account.

ABSTRACT

The Bacillus subtilis MntR and Zur transcriptional regulators control homeostasis of manganese and zinc, two essential elements required in various cellular processes. In this work, we describe the global impact of mntR and zur deletions at the protein level. Using a comprehensive proteomic approach, we showed that 33 and 55 proteins are differentially abundant in ΔmntR and Δzur cells, respectively, including proteins involved in metal acquisition, translation, central metabolism, and cell wall homeostasis. In addition, both mutants showed modifications in intracellular metal ion pools, with significant Mg2+ accumulation in the ΔmntR mutant. Phenotypic and morphological analyses of ΔmntR and Δzur mutants revealed their high sensitivity to lysozyme, beta-lactam antibiotics, and external oxidative stress. Mutant strains had a modified cell wall thickness and accumulated lower levels of intracellular reactive oxygen species (ROS) than the wild-type strain. Remarkably, our results highlight an intimate connection between MntR, Zur, antibiotic sensitivity, and cell wall structure.IMPORTANCE Manganese and zinc are essential transition metals involved in many fundamental cellular processes, including protection against external oxidative stress. In Bacillus subtilis, Zur and MntR are key transcriptional regulators of zinc and manganese homeostasis, respectively. In this work, proteome analysis of B. subtilis wild-type, ΔmntR, and Δzur strains provided new insights into bacterial adaptation to deregulation of essential metal ions. Deletions of mntR and zur genes increased bacterial sensitivity to lysozyme, beta-lactam antibiotics, and external oxidative stress and impacted the cell wall thickness. Overall, these findings highlight that Zur and MntR regulatory networks are connected to antibiotic sensitivity and cell wall plasticity.
