RESUMEN

In this short communication, we describe chewing behaviour in sows with no apparent edible substrate in their mouth as a potential social behaviour. Chewing in sows during interaction with unfamiliar sows was unexpectedly observed in a study assessing social abilities and damaging behaviours in group-housed dry sows. Such chewing was observed frequently and performed by almost all sows. To our knowledge, this behaviour has not been described previously in a social context in pigs, but rather as an oral stereotypic behaviour related to chronic hunger. We describe the behaviour in its social context and speculate on reasons for its occurrence in that context.

The benefits to captive animals of environmental enrichment (EE) are widely recognized. Few studies have, however, studied how to maximise the effect of EE. One issue with EE programs seems to be habituation to the enrichment device. To study the effect of habituation to EE, 14 captive sloth bears (Melursus ursinus) were subjected to two different EE treatments. Treatment one presented EE (logs with honey containing holes) for five consecutive days, whereas treatment two presented EE on intermittent days for five days. Intermittent presentations tended to reduce habituation toward the EE. Both consecutive and intermittent presentations significantly reduced stereotypies; however, the consecutive presentations had a longer-lasting effect. Explorative behaviors increased in both treatments, consistent with earlier findings that EE increase levels of natural behaviors. Other behaviors were unaffected by the EE presentations. The results show that intermittent presentation of EE objects may secure the interest of the animals, but continuous access to enrichment may be more efficient in reducing stereotypies in the long run.

BACKGROUND: Low survival rates in metastatic high-grade osteosarcoma (HGOS) have remained stagnant for the last three decades. This study aims to investigate the role of aminopeptidase N (ANPEP) in HGOS progression and its targeting with a novel lipophilic peptidase-enhanced cytotoxic compound melphalan flufenamide (melflufen) in HGOS. METHODS: Meta-analysis of publicly available gene expression datasets was performed to determine the impact of ANPEP gene expression on metastasis-free survival of HGOS patients. The efficacy of standard-of-care anti-neoplastic drugs and a lipophilic peptidase-enhanced cytotoxic conjugate melflufen was investigated in patient-derived HGOS ex vivo models and cell lines. The kinetics of apoptosis and necrosis induced by melflufen and doxorubicin were compared. Anti-neoplastic effects of melflufen were investigated in vivo. RESULTS: Elevated ANPEP expression in diagnostic biopsies of HGOS patients was found to significantly reduce metastasis-free survival. In drug sensitivity assays, melflufen has shown an anti-proliferative effect in HGOS ex vivo samples and cell lines, including those resistant to methotrexate, etoposide, doxorubicin, and PARP inhibitors. Further, HGOS cells treated with melflufen displayed a rapid induction of apoptosis and this sensitivity correlated with high expression of ANPEP. In combination treatments, melflufen demonstrated synergy with doxorubicin in killing HGOS cells. Finally, Melflufen displayed anti-tumor growth and anti-metastatic effects in vivo. CONCLUSION: This study may pave the way for use of melflufen as an adjuvant to doxorubicin in improving the therapeutic efficacy for the treatment of metastatic HGOS.

Anticipatory behaviours may serve a useful tool in studying positive emotional states in animals. This study aimed to investigate if lambs express anticipatory behaviours for opportunities to play or a food reward and if these behaviours would differ depending on the anticipated event. Forty-two male lambs were allocated into three treatment groups (control, play, food). Play and food lambs were conditioned to anticipate play or food in a holding pen for three minutes prior to accessing a reward arena containing toy objects or concentrate, respectively. Control lambs returned to their home pen following three minutes in the holding pen. Compared to the control lambs, both play and food lambs differed in several behaviours frequencies and durations, e.g. by an increased frequency of behavioural transitions and duration of walking. Following these observations, food lambs received the toy objects when entering the reward arena. The subsequent session in the holding pen resulted in a decrease in number of behavioural transitions, time spent walking and an increase in time standing still. In conclusion, anticipating a positive event resulted in differences in behaviour compared to a control group, and these behaviours are affected when the anticipated event does not fit with the lambs' expectations.
