RESUMEN

If a congenitally blind person learns to distinguish between a cube and a sphere by touch, would they immediately recognize these objects by sight once their vision is restored? This question, posed by Molyneux in 1688, has puzzled philosophers and scientists since then. To overcome ethical and practical difficulties in the investigation of cross-modal recognition, we studied inexperienced poultry chicks, which can be reared in darkness until the moment of a visual test with no detrimental consequences. After hatching chicks in darkness, we exposed them to either tactile smooth or tactile bumpy stimuli for 24 h. Immediately after the tactile exposure, chicks were tested in a visual recognition task, during their first experience with light. At first sight, chicks that had been exposed in the tactile modality to smooth stimuli approached the visual smooth stimulus significantly more than those exposed to the tactile bumpy stimuli. These results show that visually inexperienced chicks can solve Molyneux's problem, indicating cross-modal recognition does not require previous multimodal experience. At least in this precocial species, supra-modal brain areas appear functional already at birth. This discovery paves the way for the investigation of predisposed cross-modal cognition that does not depend on visual experience.

Young precocial birds benefit from staying close to both their mother and their siblings, while prioritising adults, which provide better care. Which features of the stimuli are used by young birds to prioritise approach and eventually attachment to adults over siblings is unknown. We started to address this question in newly hatched domestic chicks (Gallus gallus), focusing on their spontaneous preferences for visual features that systematically vary between adult and juvenile chickens, and that had previously been identified as attractive: size (larger in adults than in juveniles) and colour (darker and redder in adults than in juveniles). Overall, chicks at their first visual experience, that had never seen a conspecific beforehand, were most attracted to the red and large stimuli (two adult features) and spent more time in close proximity with red stimuli than with yellow stimuli. When tested with red large versus small objects (Exp. 1), chicks preferred the large shape. When tested with yellow large and small objects (Exp. 2), chicks did not show a preference. Chicks had a stronger preference for large red stimuli (vs. small yellow objects) than for small red stimuli (vs. a large yellow object) (Exp. 3). These results suggest that the combination of size and colour form the predisposition that helps chicks to spontaneously discriminate between adult and juvenile features from the first stages of life, in the absence of previous experience, exhibiting a preference to approach stimuli with features associated with the presence of adult conspecifics.

Experiencing inadequate parental care during early-life diminishes adult social competencies. For example, low parental care impairs adult socio-cognitive abilities (e.g., recognizing familiar conspecifics) and affiliation (e.g., close social proximity); outcomes attributed to diminished medial amygdala nonapeptide functioning in rodents. Whether parental care has effects beyond familiarity, and if siblings have similar effects to parents, is unclear. Here, zebra finches were used to explore if parent and/or sibling number shape adult recognition and preference of small versus large flocks and nonapeptide (oxytocin, vasotocin) receptor expression in an avian homologue of the mammalian medial amygdala. Chicks were raised by single mothers or fathers in small broods or paired parents in small or large broods matched to single parents for chicks per nest or per parent, respectively. Pair-raised birds had preferred flock sizes as adults, but birds raised by single parents had equal preference for either size. Oxytocin receptor expression was lower in birds raised by single parents versus paired parents, but vasotocin receptor levels were unaffected. Such results highlight parents as formative antecedents of their offspring's social competencies related to group size preference and their nonapeptide mechanisms, outcomes that influence an animal's ability to live in social groups.

The endocrine system is known to mediate responses to environmental change and transitions between different life stages (e.g., a non-breeding to a breeding life stage). Previous works from the field of environmental endocrinology have primarily focused on changes in circulating hormones, but a comprehensive understanding of endocrine signaling pathways requires studying changes in additional endocrine components (e.g., receptor densities) in a diversity of contexts and life stages. Migratory birds, for instance, can exhibit dramatic changes in their physiology and behavior, and both sex steroids as well as glucocorticoids are proposed mediators of the transition into a migratory state. However, the role of changes in endocrine signaling components within integral target tissues, such as flight muscles, in modulating the transition into a migratory state remains poorly understood. Here, we examined changes in gene expression levels of and correlational patterns (i.e., integration) between 8 endocrine signaling components associated with either glucocorticoids or sex steroid signaling in the pectoralis muscles of a nomadic migratory bird, the pine siskin (Spinus pinus). The pectoralis muscle is essential to migratory flight and undergoes conspicuous changes in preparation for migration, including hypertrophy. We focus on endocrine receptors and enzymes (e.g., 5α-reductase) that modulate the signaling capacity of circulating hormones within target tissues and may influence either catabolic or anabolic functioning within the pectoralis. Endocrine signaling components were compared between captive birds sampled prior to the expression of vernal migratory preparation and during the expression of a vernal migratory state. While birds exhibited differences in the size and color of the flight muscle and behavioral shifts indicative of a migratory state (i.e., zugunruhe), none of the measured endocrine components differed before and after the transition into the migratory state. Patterns of integration amongst all genes did, however, differ between the two life stages, suggesting the contrasting demands of different life stages may shape entire endocrine signaling networks within target tissues rather than individual components. Our work aligns with previous endocrine studies on pine siskins and, viewed together, suggest additional studies are needed to understand the endocrine system's role in mediating the development and progression of the vernal migratory state in this species. Further, the patterns observed in pine siskins, a nomadic migrant, differ from previous studies on obligate migrants and suggest that different mechanisms or interactions between endocrine signaling components may mediate the migratory transition in nomadic migrants.

Urban habitats present animals with persistent disturbances and acute stressors not present in rural habitats or present at significantly lower levels. Differences in the glucocorticoid stress response could underlie colonization of these novel habitats. Despite urban habitats characterization as more stressful, previous comparisons of urban and rural birds have failed to find consistent differences in baseline and stress induced glucocorticoid levels. Another aspect of glucocorticoid regulation that could underlie an animal's ability to inhabit novel habitats, but has yet to be well examined, is more efficient termination of the glucocorticoid stress response which would allow birds in urban habitats to recover more quickly after a disturbance. The glucocorticoid stress response is terminated by negative feedback achieved primarily through their binding of receptors in the hippocampus and hypothalamus and subsequent decreased synthesis and release from the adrenals. We investigated if male song sparrows (Melospiza melodia) in urban habitats show more efficient termination of the glucocorticoid stress response than their rural counterparts using two approaches. First, we measured glucocorticoid receptor, mineralocorticoid receptor and 11ß-HSD2 (an enzyme that inactivates corticosterone) mRNA expression in negative feedback targets of the brain (the hippocampus and hypothalamus) as a proxy measure of sensitivity to negative feedback. Second, we measured plasma corticosterone levels after standardized restraint and again following a challenge with the synthetic glucocorticoid, dexamethasone, as a means of assessing how quickly birds decreased glucocorticoid synthesis and release. Though there were no differences in the hypothalamus of urban and rural song sparrows, urban birds had lower glucocorticoid receptor and 11ß-HSD2 mRNA expression in the hippocampus. Further, urban and rural birds had similar reductions in corticosterone following the dexamethasone challenge, suggesting that they do not differ in how quickly they decrease glucocorticoid synthesis and release. Thus, urban and rural song sparrows display similar termination of the glucocorticoid stress response even though urban birds have decreased hippocampal glucocorticoid receptor and 11ß-HSD2 abundance.

Adolescents are highly motivated to engage in social interactions, and researchers have hypothesized that positive social relationships during adolescence can have long term, beneficial effects on stress reactivity and mental well-being. Studies of laboratory rodents provide the opportunity to investigate the relationship between early social experiences and later behavioral and physiological responses to stressors. In this study, female Lister-hooded rats (N = 12 per group) were either (a) provided with short, daily encounters (10 min/day) with a novel partner during mid-adolescence (postnatal day 34-45; "social experience," SE, subjects) or (b) underwent the same protocol with a familiar cagemate during mid-adolescence ("control experience," CE, subjects), or (c) were left undisturbed in the home cage (non-handled "control," C, subjects). When tested in adulthood, the groups did not differ in behavioral responses to novel environments (elevated plus maze, open field, and light-dark box) or in behavioral and physiological (urinary corticosterone) responses to novel social partners. However, SE females emitted significantly more 50 kHz ultrasonic vocalizations than control subjects both before and after social separation from a familiar social partner, which is consistent with previous findings in male rats. Thus, enhanced adolescent social experience appears to have long-term effects on vocal communication and could potentially modulate adult social relationships.

Adolescent social interactions can have long-term effects on physiological responses to stressors in later-life. A larger adolescent group size can result in higher stressor-induced secretion of glucocorticoids in adulthood. The effect may be due to a socially-mediated modulation of gonadal hormones, e.g. testosterone. However, group size (number of animals) has been conflated with social density (space per animal). Therefore it is hard to determine the mechanisms through which adolescent group size can affect the stress response. The current study aimed to tease apart the effects of group size and social density during adolescence on the physiological stress response and gonadal hormone levels in adulthood. Adolescent zebra finches were housed in groups varying in size (2 vs. 5 birds per cage) and density (0.03m3 vs. 0.06m3 per bird) during early adolescence (day 40-60). Density was only manipulated in birds raised in groups of five. Glucocorticoid concentration secreted in response to a standard capture and restraint stressor was quantified in adolescence (day 55±1) and adulthood (day 100+). Basal gonadal hormone concentrations (male testosterone, female estradiol) were also quantified in adulthood. Female birds housed in larger groups, independent of social density, secreted a higher glucocorticoid concentration 45min into restraint regardless of age, and had higher peak glucocorticoid concentration in adulthood. Adult gonadal hormone concentrations were not affected by group size or density. Our results suggest that group size, not density, is a social condition that influences the development of the endocrine response to stressors in female zebra finches, and that these effects persist into adulthood. The findings have clear relevance to the social housing conditions necessary for optimal welfare in captive animals, but also elucidate the role of social rearing conditions in the emergence of responses to stressors that may persist across the lifespan and affect fitness of animals in wild populations.

Experiencing stress during adolescence can increase neophobic behaviors in adulthood, but most tests have been conducted in the absence of conspecifics. Conspecifics can modulate responses to stressors, for example by acting as 'social buffers' to attenuate the aversive appraisal of stressors. Here, we investigate the long-term effects of adolescent stress on the behavioral responses to novel stimuli (a mild stressor) across social contexts in an affiliative passerine bird, the zebra finch. During early (days 40-60) or late (days 65-85) adolescence the birds (n=66) were dosed with either saline or the hormone corticosterone (CORT). CORT was given in order to mimic a physiological stress response and saline was given as a control. In adulthood, the birds' behavioral responses to a novel environment were recorded in both the presence and absence of conspecifics. An acute CORT response was also quantified in adolescence and adulthood. Our findings show clear evidence of social context mediating any long-term effects of adolescent stress. In the presence of familiar conspecifics no treatment effects were detected. Individually, birds dosed with CORT in early adolescence were slower to enter a novel environment, spent more time perching in the same novel environment, and, if female, engaged in more risk assessment. Birds dosed in late adolescence were unaffected. No treatment effects were detected on CORT, but adolescents had a higher CORT concentration than adults. Our results are the first to suggest that familiar conspecifics in adulthood can buffer the long-term effects of stress that occurred during early adolescence.
