RESUMEN

This study presents the design and implementation of an electronic system aimed at capturing vibrations produced during truck operation. The system employs a graphical interface to display vibration levels, ensuring the necessary comfort and offering indicators as a solution to mitigate the damage caused by these vibrations. Additionally, the system alerts the driver when a mechanical vibration that could potentially impact their health is detected. The field of health is rigorously regulated by various international standards and guidelines. The case of mechanical vibrations, particularly those transmitted to the entire body of a seated individual, is no exception. Internationally, ISO 2631-1:1997/Amd 1:2010 oversees this study. The system was designed and implemented using a blend of hardware and software. The hardware components comprise a vibration sensor, a data acquisition card, and a graphical user interface (GUI). The software components consist of a data acquisition and processing library, along with a GUI development framework. The system underwent testing in a controlled environment and demonstrated stability and robustness. The GUI proved to be intuitive and could be integrated into modern vehicles with built-in displays. The findings of this study suggest that the proposed system is a viable and effective method for capturing vibrations in trucks and informing drivers about vibration levels. This system has the potential to enhance the comfort and safety of truck drivers.

Gold nanostars (GNST), gold nanospheres (GNP) and carbon black (CB) are chosen as alternative nanomaterials to modify carbon screen-printed electrodes (c-SPEs). The resulting three kinds of modified c-SPEs (GNP-SPE, CB-SPE and GNSP-SPE) were electrochemically and microscopically characterized and compared with standardized c-SPEs after pretreatment with phosphate buffer by pre-anodization (pre-SPE). The results show outstanding electrochemical performance of the carbon black-modified SPEs which show low transient current, low capacitance and good porosity. A competitive chronoamperometric immunoassay for the shellfish toxin domoic acid (DA) is described. The performances of the CB-SPE, GNP-SPE and pre-SPE were compared. Hapten-functionalized magnetic beads were used to avoid individual c-SPE functionalization with antibody while enhancing the signal by creating optimum surface proximity for electron transfer reactions. This comparison shows that the CB-SPE biosensor operated best at a potential near - 50 mV (vs. Ag/AgCl) and enables DA to be determined with a detection limit that is tenfold lower compared to pre-SPE (4 vs. 0.4 ng mL-1). These results show very good agreement with HPLC data when analysing contaminated scallops, and the LOD is 0.7 mg DA kg-1 of shellfish. Muscle imbalance and deficit are key parameters for guiding rehabilitation and sports sessions and avoiding injuries. However, the high cost and non-portable nature of most instruments employed for muscle strength assessment frequently hamper an affordable evaluation in field conditions. This study evaluated the validity and reliability of a low-cost digital dynamometer to assess the maximal isometric strength of upper limb muscles. Fourteen physically active volunteers performed the main movements of the upper limb at maximal isometric strength: flexion-extension, internal-external rotation and abduction-adduction of the shoulder; and flexion-extension of both elbow and wrist. Validity was examined by comparing the strength values from the low-cost digital dynamometer and those measured by an isokinetic dynamometer, assumed as the gold standard. The correlation between both devices was perfect (r > 0.913), while Bland-Altman plots showed absolute agreement between both devices, the maximum range of the values of bias was -0.99-1.00 N in wrist extension. Inter-tester and intra-tester reliability were excellent for all movements (ICC ≥ 0.855). The low-cost digital dynamometer showed strong validity and excellent reliability in assessing maximal isometric strength during the main movements of the upper limb. Professionals may use it for an affordable isometric muscle strength assessment in field situations.

Food security (a pressing issue for all nations) faces a threat due to population growth, land availability for growing crops, a changing climate (leading to increases in both abiotic and biotic stresses), heightened consumer awareness of the risks related to the use of agrichemicals, and also the reliance on depleting fossil fuel reserves for their production. Legislative changes in Europe mean that fewer agrichemicals will be available in the future for the control of crop pests and pathogens. The need for the implementation of a more sustainable agricultural system globally, incorporating an integrated approach to disease management, has never been more urgent. To that end, the Valorizing Andean Microbial Diversity (VALORAM) project (http://valoram.ucc.ie), funded under FP7, examined the role of microbial communities in crop production and protection to improve the sustainability, food security, environmental protection, and productivity for rural Andean farmers. During this work, microbial volatile organic compounds (mVOCs) of 27 rhizobacterial isolates were identified using gas chromatography/mass spectrometry (GC/MS), and their antifungal activity against Rhizoctonia solani was determined in vitro and compared to the activity of a selection of pure volatile compounds. Five of these isolates, Pseudomonas palleroniana R43631, Bacillus sp. R47065, R47131, Paenibacillus sp. B3a R49541, and Bacillus simplex M3-4 R49538 trialled in the field in their respective countries of origin, i.e., Bolivia, Peru, and Ecuador, showed significant increase in the yield of potato. The strategy followed in the VALORAM project may offer a template for the future isolation and determination of putative biocontrol and plant growth-promoting agents, useful as part of a low-input integrated pest management system.

This study assessed the ecological health of waters within the Saudi Arabian Exclusive Economic Zone, by utilizing benthic biotic indices with a marine monitoring dataset covering the years 2013 to 2018. This comprehensive evaluation covered a vast expanse, encompassing 67 distinctive sampling locations characterized by a wide range of depth and salinity gradients. The study examined spatial fluctuations in the benthic community and assessed potential correlations with environmental variables, including salinity, depth, sediment texture, total organic carbon, and other relevant factors. The macrobenthic density varied across the study sites, with an average density of 566 ± 120 ind.m-2. The Shannon diversity index ranged from 3.21 and 5.90, with an average of 4.70 ± 0.52. Based on the average AMBI values, all the locations were categorized as either slightly disturbed or undisturbed. Additionally, the M-AMBI analysis indicated that 95.5 % sites were in good or high ecological status.

Phytoplankton monitoring is essential for the global understanding of aquatic ecosystems. The present research studies the phytoplankton community of the Urdaibai estuary, combining microscopy and eDNA metabarcoding for the first time in the area. The main aims were to describe the phytoplankton community composition in relation to the environmental conditions of the estuary, and to compare the two methods used. Diatoms Minutocellus polymorphus and Chaetoceros tenuissimus dominated the outer estuary, being replaced by Teleaulax acuta (cryptophyte), Kryptoperidinium foliaceum (dinoflagellate) and Cyclotella spp. (diatom) towards the inner area. This change was mainly prompted by salinity and nutrients. Metabarcoding revealed the presence of 223 species that were not observed by microscopy in previous studies in the estuary. However, several characteristic species (e.g., K. foliaceum) were only detected with microscopy. Additionally, microscopy covered the limitations of eDNA metabarcoding concerning quantification. Thus, to give a full insight, a combination of techniques is recommended.

Embedding Raman reporters within nanosized gaps of metallic nanoparticles is an attractive route for surface-enhanced Raman spectroscopy (SERS) applications, although often this involves complex synthesis procedures that limit their practical use. Herein, we present the tip-selective direct growth of silver satellites surrounding gold nanostars (AuNSt@AgSAT), mediated by a dithiol Raman reporter 1,4-benzenedithiol (BDT). We propose that BDT is embedded within nanogaps which form between the AuNSt tips and the satellites, and plays a key role in mediating the satellite growth. Not only proposing a rationale for the mechanistic growth of the AuNSt@AgSAT, we also demonstrate an example for its use for the detection of Hg2+ ions in water. The presence of Hg2+ resulted in amalgamation of the AuNSt@AgSAT, which altered both its structural morphology and Raman enhancement properties. This provides a basis for the detection where the Raman intensity of BDT is inversely proportional to the Hg2+ concentrations. As a result, Hg2+ could be detected at concentrations as low as 0.1 ppb. This paper not only provides important mechanistic insight into the tip-selective direct growth of the anisotropic nanostructure but also proposes its excellent Raman enhancement capability for bioimaging as well as biological and chemical sensing applications.

Bacterial pathogens represent a safety concern in the food industry, and this is amplified by the lack of sensing devices that can be applied on-site by non-trained personnel. In this study, peroxidase-mimicking activity of gold nanostars was exploited to develop a user-friendly colourimetric sensor. A smartphone was exploited as an image reader and analyser, empowered with a novel App developed in-house. The mobile App was evaluated and compared with a commercial smartphone App for its capability to quantify generated colourimetric signals. A major obstacle found with sensors relying on gold nanozymes is the fact that modification of the surface of gold nanoparticles with biorecognition elements generally lead to a suppression of their nanozyme activity. This drawback was overcome by introducing an autocatalytic growth step, which successfully restored the peroxidase-mimicking activity through generation of new gold nanoseeds acting as catalytic centres. A proof-of-concept using this sensing mechanism was developed targeting Mycobacterium bovis, a zoonotic pathogen primarily found in cattle but that can be transmitted to humans by consumption of contaminated food and cause tuberculosis disease. The resulting smartphone-based immunological sensor has shown promising results with a linear response between 104 - 106 CFU/mL, enabling detection of M. bovis at concentrations as low as 7.2·103 CFU/mL in buffer conditions. It is anticipated that the concept of the developed approach will have applicability in many fields relying on smartphone-based biosensing.

As the number of introduced species keeps increasing unabatedly, identifying and prioritising current and potential Invasive Alien Species (IAS) has become essential to manage them. Horizon Scanning (HS), defined as an exploration of potential threats, is considered a fundamental component of IAS management. By combining scientific knowledge on taxa with expert opinion, we identified the most relevant aquatic IAS in the Iberian Peninsula, i.e., those with the greatest geographic extent (or probability of introduction), severe ecological, economic and human health impacts, greatest difficulty and acceptability of management. We highlighted the 126 most relevant IAS already present in Iberian inland waters (i.e., Concern list) and 89 with a high probability of being introduced in the near future (i.e., Alert list), of which 24 and 10 IAS, respectively, were considered as a management priority after receiving the highest scores in the expert assessment (i.e., top-ranked IAS). In both lists, aquatic IAS belonging to the four thematic groups (plants, freshwater invertebrates, estuarine invertebrates, and vertebrates) were identified as having been introduced through various pathways from different regions of the world and classified according to their main functional feeding groups. Also, the latest update of the list of IAS of Union concern pursuant to Regulation (EU) No 1143/2014 includes only 12 top-ranked IAS identified for the Iberian Peninsula, while the national lists incorporate the vast majority of them. This fact underlines the great importance of taxa prioritisation exercises at biogeographical scales as a step prior to risk analyses and their inclusion in national lists. This HS provides a robust assessment and a cost-effective strategy for decision-makers and stakeholders to prioritise the use of limited resources for IAS prevention and management. Although applied at a transnational level in a European biodiversity hotspot, this approach is designed for potential application at any geographical or administrative scale, including the continental one.

In the present study, a facile, eco-friendly, and controlled synthesis of gold nanoparticles (Au NPs) using Prunus nepalensis fruit extract is reported. The biogenically synthesized Au NPs possess ultra-active intrinsic peroxidase-like activity for the oxidation of 3,3',5,5'-tetramethylbenzidine (TMB) in the presence of H2O2. Chemical analysis of the fruit extract demonstrated the presence of various bioactive molecules such as amino acids (l-alanine and aspartic acids), organic acids (benzoic acid and citric acid), sugars (arabinose and glucose), phenolic acid, and bioflavonoids (niacin and myo-inositol), which likely attributed to the formation of stable biogenic Au NPs with excellent peroxidase-mimicking activity. In comparison with the natural horseradish peroxidase (HRP) enzyme, the biogenic Au NPs displayed a 9.64 times higher activity with regard to the reaction velocity at 6% (v/v) H2O2, presenting a higher affinity toward the TMB substrate. The Michaelis-Menten constant (KM) values for the biogenic Au NPs and HRP were found to be 6.9 × 10-2 and 7.9 × 10-2 mM, respectively, at the same concentration of 100 pM. To investigate its applicability for biosensing, a monoclonal antibody specific for Mycobacterium bovis (QUBMA-Bov) was directly conjugated to the surface of the biogenic Au NPs. The obtained results indicate that the biogenic Au NPs-QUBMA-Bov conjugates are capable of detecting M. bovis based on a colorimetric immunosensing method within a lower range of 100 to 102 cfu mL-1 with limits of detection of ∼53 and ∼71 cfu mL-1 in an artificial buffer solution and in a soft cheese spiked sample, respectively. This strategy demonstrates decent specificity in comparison with those of other bacterial and mycobacterial species. Considering these findings together, this study indicates the potential for the development of a cost-effective biosensing platform with high sensitivity and specificity for the detection of M. bovis using antibody-conjugated Au nanozymes.

Global ocean warming, wave extreme events, and accelerating sea-level rise are challenges that coastal communities must address to anticipate damages in coming decades. The objective of this study is to undertake a time-series analysis of climate change (CC) indicators within the Bay of Biscay, including the Basque coast. We used an integrated and flexible methodology, based on Generalized Additive Mixed Models, to detect trends on 19 indicators (including marine physics, chemistry, atmosphere, hydrology, geomorphology, biodiversity, and commercial species). The results of 87 long-term time series analysed (~512,000 observations), in the last four decades, indicate four groups of climate regime shifts: 1) A gradual shift associated with CC starting in the 1980s, with a warming of the sea surface down to 100 m depth in the bay (0.10-0.25 °C per decade), increase in air temperature and insolation. This warming may have impacted on benthic community redistribution in the Basque coast, favouring warm-water species relative to cold-water species. Weight at age for anchovy and sardine decreased in the last two decades. 2) Deepening of the winter mixed layer depth in the south-eastern bay that probably led to increases in nutrients, surface oxygen, and chlorophyll concentration. Current increases on chlorophyll and zooplankton (i.e., copepods) biomass are contrary to those expected under CC scenarios in the region. 3) Sea-level rise (1.5-3.5 cm per decade since 1990s), associated with CC. 4) Increase of extreme wave height events of 16.8 cm per decade in the south-eastern bay, probably related to stormy conditions in the last decade, with impacts on beach erosion. Estimating accurate rates of sea warming, sea-level rise, extreme events, and foreseeing the future pathways of marine productivity, are key to define the best adaptation measures to minimize negative CC impacts in the region.

Nanomaterial-based artificial enzymes (or nanozymes) have attracted great attention in the past few years owing to their capability not only to mimic functionality but also to overcome the inherent drawbacks of the natural enzymes. Numerous advantages of nanozymes such as diverse enzyme-mimicking activities, low cost, high stability, robustness, unique surface chemistry, and ease of surface tunability and biocompatibility have allowed their integration in a wide range of biosensing applications. Several metal, metal oxide, metal-organic framework-based nanozymes have been exploited for the development of biosensing systems, which present the potential for point-of-care analysis. To highlight recent progress in the field, in this review, more than 260 research articles are discussed systematically with suitable recent examples, elucidating the role of nanozymes to reinforce, miniaturize, and improve the performance of point-of-care diagnostics addressing the ASSURED (affordable, sensitive, specific, user-friendly, rapid and robust, equipment-free and deliverable to the end user) criteria formulated by World Health Organization. The review reveals that many biosensing strategies such as electrochemical, colorimetric, fluorescent, and immunological sensors required to achieve the ASSURED standards can be implemented by using enzyme-mimicking activities of nanomaterials as signal producing components. However, basic system functionality is still lacking. Since the enzyme-mimicking properties of the nanomaterials are dictated by their size, shape, composition, surface charge, surface chemistry as well as external parameters such as pH or temperature, these factors play a crucial role in the design and function of nanozyme-based point-of-care diagnostics. Therefore, it requires a deliberate exertion to integrate various parameters for truly ASSURED solutions to be realized. This review also discusses possible limitations and research gaps to provide readers a brief scenario of the emerging role of nanozymes in state-of-the-art POC diagnosis system development for futuristic biosensing applications.

OBJECTIVE: Strongyloidiasis is a parasitic infection classified by the World Health Organization as a neglected tropical disease. Although predominantly asymptomatic, it can become a life-threatening disease in an immunocompromised host. Epidemiologic studies in the Western Hemisphere are scarce, but even more scarce are descriptions of the natural course of this disease. Our objectives were to identify the different manifestations and outcomes of Strongyloides stercoralis infection in the Hispanic veteran population in Puerto Rico. We also discuss the importance of pursuing a timely diagnosis in high-risk patients migrating from or traveling to endemic areas, regardless of their symptomatic status. METHODS: This was a single-center, retrospective record review study involving patients diagnosed with S. stercoralis via serology, stool samples, or organ biopsies, from 2008 through 2014. RESULTS: A total of 270 patients tested positive; 210 (77.8%) were asymptomatic. The mean age at diagnosis was 75.4 years old. Symptomatic patients had pulmonary (n = 25), gastrointestinal (n = 21), and dermatologic (n = 5) symptoms; 9 had multiple symptoms. Five had hyperinfection, mostly after treatment with systemic steroids or preexisting immunosuppression. The most common laboratory abnormality was eosinophilia. Reasons for testing were eosinophilia, asthma, diarrhea, screening for parasites, and unexplained skin rash. CONCLUSION: Our study highlights the importance of being aware of this potentially fatal infection, especially when treating patients traveling from endemic countries. It also highlights the importance of timely screening, diagnosis, and treatment of S. stercoralis infection in order to prevent potentially fatal outcomes, especially when considering immunosuppressive drugs.

The Nuffield Council on Bioethics suggests that introgression of genetic material into related species in centres of crop biodiversity is an insufficient justification to bar the use of genetically modified crops in the developing world. They consider that a precautionary approach to forgo the possible benefits invokes the fallacy of thinking that doing nothing is itself without risk to the poor. Here we report findings relevant to this and other aspects of environmental biosafety for genetically modified potato in its main centre of biodiversity, the central Andes. We studied genetically modified potato clones that provide resistance to nematodes, principal pests of Andean potato crops. We show that there is no harm to many non-target organisms, but gene flow occurs to wild relatives growing near potato crops. If stable introgression were to result, the fitness of these wild species could be altered. We therefore transformed the male sterile cultivar Revolucion to provide a genetically modified nematode-resistant potato to evaluate the benefits that this provides until the possibility of stable introgression to wild relatives is determined. Thus, scientific progress is possible without compromise to the precautionary principle.

The feasibility of using lab-cultured amphipods Corophium multisetosum (Stock 1952) to evaluate the toxicity of contaminants present within marine sediments was studied. This was done by comparing the sensitivity of lab-cultured amphipods in a cadmium toxicity test and to toxic sediment samples, during a 10-days bioassay, with field collected individuals. Different responses were observed between field and cultured individuals. Cadmium test indicated high temporal variability in the LC(50) values of field amphipods (2.40-6.55 mg L(-1)). Sensitivity of cultured amphipods was within the seasonal range of the field individuals (5.81 mg L(-1), LC(50)). However, culture amphipods showed much lower sensitivity in toxic sediment samples. Our results indicate that sensitivity should be determined using a sediment matrix, if the assessment of toxicity is based upon bioassays performed with cultured burrower-amphipods.

In recent years, gold nanoparticles have demonstrated excellent enzyme-mimicking activities which resemble those of peroxidase, oxidase, catalase, superoxide dismutase or reductase. This, merged with their ease of synthesis, tunability, biocompatibility and low cost, makes them excellent candidates when compared with biological enzymes for applications in biomedicine or biochemical analyses. Herein, over 200 research papers have been systematically reviewed to present the recent progress on the fundamentals of gold nanozymes and their potential applications. The review reveals that the morphology and surface chemistry of the nanoparticles play an important role in their catalytic properties, as well as external parameters such as pH or temperature. Yet, real applications often require specific biorecognition elements to be immobilized onto the nanozymes, leading to unexpected positive or negative effects on their activity. Thus, rational design of efficient nanozymes remains a challenge of paramount importance. Different implementation paths have already been explored, including the application of peroxidase-like nanozymes for the development of clinical diagnostics or the regulation of oxidative stress within cells via their catalase and superoxide dismutase activities. The review also indicates that it is essential to understand how external parameters may boost or inhibit each of these activities, as more than one of them could coexist. Likewise, further toxicity studies are required to ensure the applicability of gold nanozymes in vivo. Current challenges and future prospects of gold nanozymes are discussed in this review, whose significance can be anticipated in a diverse range of fields beyond biomedicine, such as food safety, environmental analyses or the chemical industry.

The European Water Framework and Marine Strategy Directives relate to the assessment of ecological quality, within estuarine and coastal systems. This legislation requires quality to be defined in an integrative way, using several biological elements (phytoplankton, benthos, algae, phanerogams, and fishes), together with physico-chemical elements (including pollutants). This contribution describes a methodology that integrates all of this information into a unique quality assessment for 51 stations from 18 water bodies, within the Basque Country. These water bodies are distributed into four typologies, including soft-bottom coastal areas and three types of estuaries. For each station, decision trees were used to integrate (i) water, sediment and biomonitor chemical data to achieve an integrated physico-chemical assessment and (ii) multiple biological ecosystem elements into an integrated biological assessment. Depending on the availability of ecological quality ratios or global quality values, different integration schemes were used to combine station assessments into water body assessments on a single scale. Several examples from each element have been selected, to illustrate their responses to different pressures; likewise, to establish how the assessed integrated quality has changed, over time. The results made biological and ecological sense and physico-chemical improvements were often correlated with improvements in the quality of benthos and fishes. These tools permit policy makers and managers to take decisions, based upon scientific knowledge, in water management, regarding the mitigation of human pressures and associated recovery processes.

Naccobus aberrans is a major pest of the potato crop in the Andean regions of Argentina, Bolivia, and Perú. It is endemic in northwest Argentina and is also found in lowlands. The resistance of eleven Andean potato landraces and three accessions of the wild tuber-bearing species Solanum acaule, S. infundibuliforme, and S. megistacrolobum were evaluated against a population of N. aberrans from Coctaca, Jujuy province, while Solanum tuberosum ssp. tuberosum 'Spunta', 'Kennebec', and 'Frital INTA' were evaluated against a population from the southeast of Buenos Aires province. The presence, the number of galls, and the number of individuals were recorded. In addition, a reproduction factor was calculated and races were determined. Results showed that the N. aberrans population from Coctaca corresponded to race 2 and the population from the lowlands belonged to the sugar beet group. Landrace Azul, one genotype of S. megistacrolobum, and two genotypes of S. acaule showed resistance towards the race from Coctaca while no infection was recorded in potato cultivars with the Naccobus race from the lowland area.

Laboratory experiments were carried out over two different periods, with the aim of investigating and utilizing the induction to gonadal maturation of the sea urchin Paracentrotus lividus. The final objective was to obtain viable gametes outside the period of natural spawning in the environment; which would allow the utilization of ecotoxicological bioassays with sea urchin larvae at any time of the year. The experiment consisted of maintaining some sea urchins in tanks and providing them with a natural photoperiod, unlimited food and a constant temperature of 20 degrees C. During days 0, 30, 60 and 90, gonads from 15 of these sea urchins were compared with those collected simultaneously in the natural environment. The gametes obtained were used to carry out fecundations, in order to check their viability. The final results obtained were clearly influenced by the gonadal state of the sea urchins at the initial stage of the experiment. The best results were obtained within a time period of 60 days and when the initial gonad index was low.
