RESUMEN

ZFP57 is a master regulator of genomic imprinting. It has both maternal and zygotic functions that are partially redundant in maintaining DNA methylation at some imprinting control regions (ICRs). In this study, we found that DNA methylation was lost at most known ICRs in Zfp57 mutant embryos. Furthermore, loss of ZFP57 caused loss of parent-of-origin-dependent monoallelic expression of the target imprinted genes. The allelic expression switch occurred in the ZFP57 target imprinted genes upon loss of differential DNA methylation at the ICRs in Zfp57 mutant embryos. Specifically, upon loss of ZFP57, the alleles of the imprinted genes located on the same chromosome with the originally methylated ICR switched their expression to mimic their counterparts on the other chromosome with unmethylated ICR. Consistent with our previous study, ZFP57 could regulate the NOTCH signaling pathway in mouse embryos by impacting allelic expression of a few regulators in the NOTCH pathway. In addition, the imprinted Dlk1 gene that has been implicated in the NOTCH pathway was significantly down-regulated in Zfp57 mutant embryos. Our allelic expression switch models apply to the examined target imprinted genes controlled by either maternally or paternally methylated ICRs. Our results support the view that ZFP57 controls imprinted expression of its target imprinted genes primarily through maintaining differential DNA methylation at the ICRs.

The mechanical properties of the subgrade have a significant impact on the service life and pavement performance of the superstructure of pavement. By adding admixtures and via other means to strengthen the adhesion between soil particles, the strength and stiffness of the soil can be improved to ensure the long-term stability of pavement structures. In this study, a mixture of polymer particles and nanomaterials was used as a curing agent to examine the curing mechanism and mechanical properties of subgrade soil. Using microscopic experiments, the strengthening mechanism of solidified soil was analyzed with scanning electron microscopy (SEM), energy-dispersive spectroscopy (EDS), Fourier infrared spectroscopy (FTIR), and X-ray diffraction (XDR). The results showed that with the addition of the curing agent, small cementing substances on the surface of soil minerals filled the pores between minerals. At the same time, with an increase in the curing age, the colloidal particles in the soil increased, and some of them formed large aggregate structures that gradually covered the surface of the soil particles and minerals. By enhancing the cohesiveness and integrity between different particles, the overall structure of the soil became denser. Through pH tests, it was found that the age had a certain effect on the pH of solidified soil, but the effect was not obvious. Through the comparative analysis of elements in plain soil and solidified soil, it was found that no new chemical elements were produced in the solidified soil, indicating that the curing agent does not have negative impacts on the environment.

Expansion joint failure is one of the main causes that lead to the damages of concrete pavement. The silicon dioxide/shape memory polyurethane (SiO2/SMPU) is a new kind of sealant which can use its shape memory performance to adapt to the width of the expansion joint with the change of pavement temperature, and it can effectively prolong the service life of the pavement and reduce maintenance costs. In this study, the effects of programming and the addition of SiO2 particles to the thermodynamic properties of the specimens were detected using differential scanning calorimetry (DSC), the optimal shape memory programming temperature of which is 72.9 °C. Combined with scanning electron microscopy (SEM) and shape memory effect test, the particles are evenly distributed between the two phases, and the shape fixation rate (Rf) of 98.15% and the shape recovery rate (Rr) of 97.31% show that the composite has a good shape memory effect. Fourier transform infrared spectroscopy (FTIR) and dynamic infrared dichroism illustrate the change of the hydrogen bond of soft and hard segments with the SiO2 particles in the shape memory cycle, revealing the optimal shape memory programming process. This study provides an insight into the reinforcement mechanism of SiO2 nanoparticles in SMPU matrix and verify whether it can meet the engineering requirements of expansion joints when used as a sealant of concrete pavement.

To explore the effect of different emulsifier contents on the stability performance of biomass-emulsified asphalt, three types of emulsified asphalt with 1%, 3%, and 5% anionic emulsifiers were prepared and analyzed by molecular dynamics simulation and macroscopic experiments. Firstly, we used molecular simulation software (Material Studio, MS) to construct a model of biomass-emulsified asphalt with different emulsifier contents and analyzed the microscopic mechanism of the emulsifier to improve the stability of the emulsified asphalt by the radial distribution function, interaction energy, interfacial layer thickness, and solubility parameters of the emulsified asphalt system with different emulsifier contents. The results were validated by macro and micro tests including storage stability, particle size determination, and infrared spectroscopy. The results show that at low emulsifier contents, the emulsifier can reduce the interfacial tension between the oil-water interface and expand the transition region between the two phases (interfacial layer thickness), which will prevent interparticle agglomeration and reduce the emulsion particle size, thus reducing the settling rate and ensuring the stability of the emulsion. When the emulsifier content is further increased beyond the critical micelle concentration, the emulsifiers will agglomerate with each other and show larger peaks in the radial distribution function, and the phenomenon of emulsifier agglomeration will appear in the five-day storage stability test, resulting in a corresponding decrease in the proximity of the infrared absorption peak area ratio in the same wavelength band of the upper and lower layers of the biomass-emulsified asphalt, and the emulsion stability decreases instead.

Uneven support as result of voids beneath concrete slabs can lead to high tensile stresses at the corner of the slab and eventually cause many forms of damage, such as cracking or faulting. Three-dimensional (3D) finite element models of the concrete pavement with void are presented. Mesh convergence analysis was used to determine the element type and mesh size in the model. The accuracy of the model is verified by comparing with the calculation results of the code design standards in China. The reliability of the model is verified by field measurement. The analysis shows that the stresses are more affected at the corner of the slab than at the edge. Impact of void size and void depth at the slab corner on the slab stress are similar, which result in the change of the position of the maximum tensile stress. The maximum tensile stresses do not increase with the increase in the void size for relatively small void size. The maximum tensile stress increases rapidly with the enlargement in the void size when the size is ≥0.4 m. The increments of maximum tensile stress can reach 183.7% when the void size is 1.0 m. The increase in slab thickness can effectively reduce maximum tensile stress. A function is established to calculate the maximum tensile stress of the concrete slab. The function takes into account the void size, the slab thickness and the vehicle load. The reliability of the function was verified by comparing the error between the calculated and simulated results.

This paper investigated the effects of aggregate mesostructures on permanent deformation behavior of an asphalt mixture using the three-dimensional (3D) discrete element method (DEM). A 3D discrete element (DE) model of an asphalt mixture composed of coarse aggregates, asphalt mastic, and air voids was developed. Mesomechanical models representing the interactions among the components of asphalt mixture were assigned. Based on the mesomechanical modeling, the uniaxial static load creep tests were simulated using the prepared models, and effects of aggregate angularity, orientation, surface texture, and distribution on the permanent deformation behavior of the asphalt mixtures were analyzed. It was proven that good aggregate angularity had a positive effect on the permanent deformation performance of the asphalt mixtures, especially when approximate cubic aggregates were used. Aggregate packing was more stable when the aggregate orientations tended to be horizontal, which improved the permanent deformation performance of the asphalt mixture. The influence of orientations of 4.75 mm size aggregates on the permanent deformation behavior of the asphalt mixture was significant. Use of aggregates with good surface texture benefitted the permanent deformation performance of the asphalt mixture. Additionally, the non-uniform distribution of aggregates had a negative impact on the permanent deformation performance of the asphalt mixtures, especially when aggregates were distributed non-uniformly in the vertical direction.

The main objective of this paper was to develop a mesostructure-based finite element model of rubber modified asphalt mixture to predict both the dynamic modulus master curve and phase angle master curve under a large frequency range. The asphalt mixture is considered as a three-phase material consisting of aggregate, asphalt mortar, and air void. The mesostructure of the asphalt mixture was digitized by a computed tomography (CT) scan and implemented into finite element software. The 2S2P1D model was used to obtain the viscoelastic information of an asphalt mortar under a large range of frequencies and temperatures. The continuous spectrum of the 2S2P1D model was converted to a discrete spectrum and characterized by the generalized Maxwell model for numerical simulation. The Prony series parameters of the generalized Maxwell model and the elastic modulus of the aggregates were inputted into the finite element analysis as material properties. The dynamic modulus tests of a rubber modified asphalt mortar and asphalt mixture were conducted under different temperatures and loading frequencies. The dynamic modulus master curve and phase angle master curve of both asphalt mortar and asphalt mixture were constructed. The frequency of the finite element simulations of the dynamic modulus tests ranged from 10-6 to 104. The dynamic modulus and phase angle of the asphalt mixture was calculated and the master curves were compared with the master curves obtained from the experimental data. Furthermore, the effect of the elastic modulus of aggregates on the master curves was analyzed. Acceptable agreement between dynamic modulus master curves obtained from experimental data and simulation results was achieved. However, large errors between phase angle master curves appeared at low frequencies. A method was proposed to improve the prediction of the phase angle master curve by adjusting the equilibrium modulus of the asphalt mortar.
