What we believe is a novel dual-channel whispering gallery mode (WGM) sensor for concurrently measuring bidirectional magnetic field and temperature is proposed and demonstrated. Two sensing microcavities [magnetic fluid (MF)-infiltrated capillary and polydimethylsiloxane (PDMS)-coated microbottle, respectively, referred as Channel 1 (CH1) and Channel 2 (CH2)] are integrated into a silica capillary to facilitate the dual-channel design. Resonant wavelengths corresponding to CH1 and CH2 mainly depend on the change in the magneto-induced refractive index and the change in the thermo-induced parameter (volume and refractive index) of the employed functional materials, respectively. The MF-infiltrated capillary enables bidirectional magnetic field sensing with maximum sensitivities of 46 pm/mT and -3 pm/mT, respectively. The PDMS-coated structure can realize the temperature measurement with a maximum sensitivity of 79.7 pm/°C. The current work possesses the advantage of bidirectionally magnetic tunability besides the temperature response, which is expected to be used in field such as vector magnetic fields and temperature dual-parameter sensing.

A kind of temperature and magnetic field sensor using Fabry-Perot interferometers (FPIs) and Vernier effect to enhance sensitivity is proposed. The sensor structure involves filling the FP air cavities with polydimethylsiloxane (PDMS) and magnetic fluid (MF) to create the PDMS and MF cavities for temperature and magnetic field detection, respectively. The two cavities are reflective structures, which are interconnected in series through a fiber-optic circulator. Experimental data demonstrates that the Vernier effect effectively enhances the sensor sensitivity. The average temperature sensitivity of the sensor is 26765 pm/°C within the range of 35∼39.5°C. The magnetic field intensity sensitivity is obtained to be -2245 pm/mT within the range of 3∼11 mT. The sensitivities of the temperature and magnetic field using the Vernier effect are about five times larger than those of the corresponding single FP cavity counterparts.

This work proposes and investigates a bent multimode-no-core-multimode optical fiber structure for vector magnetic field sensing applications. The bent no-core fiber (NCF) serves as the sensing area, and the gold film is deposited on its surface to excite the surface plasmon resonance effect. Due to the strong evanescent field of the unclad and bent NCF, the as-fabricated sensor exhibits a high sensitivity of 5630 nm/RIU in the refractive index range of 1.36-1.39. Magnetic fluid is employed as the magneto-sensitive material for magnetic field sensing, exhibiting a high magnetic field intensity sensitivity of 5.74 nm/mT and a high magnetic field direction sensitivity of 0.22 nm/°. The proposed sensor features a simple structure, low cost, point sensing, and excellent mechanical performance.

BACKGROUND: As independent and correctable risk factors, disturbances in lipid metabolism are significantly associated with type 2 diabetes mellitus (T2DM). This research investigated the mechanism underlying the lipid-regulating effects of Yam Gruel in diabetic rats. METHODS: First, rats in the control group were given a normal diet, and a diabetic rat model was established via the consumption of a diet that was rich in both fat and sugar for six weeks followed by the intraperitoneal injection of streptozotocin (STZ). After the model was established, the rats were divided into five distinct groups: the control group, model group, Yam Gruel (SYZ) group, metformin (MET) group, and combined group; each treatment was administered for six weeks. The fasting blood glucose (FBG), body and liver weights as well as liver index of the rats were determined. Total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), aspartic acid transaminase (AST), alanine aminotransferase (ALT), and nonesterified fatty acid (NEFA) levels were measured. Oil Red O staining was used to assess hepatic steatosis. In addition, the levels of Phospho-acetyl-CoA carboxylase (p-ACC), acetyl coenzyme A carboxylase (ACC), AMP-activated protein kinase (AMPK), Phospho-AMPK (p-AMPK), carnitine palmitoyl transferase I (CPT-1), and Malonyl-CoA decarboxylase (MLYCD) in liver tissues were measured by real-time PCR (q-PCR) and western blotting. RESULTS: After 6 weeks of treatment, Yam Gruel alone or in combination with metformin significantly reduced FBG level, liver weight and index. The concentrations of lipid indices (TG, TC, NEFA, and LDL-C), the levels of liver function indices (ALT and AST) and the degree of hepatic steatosis was improved in diabetic rats that were treated with Yam Gruel with or without metformin. Furthermore, Yam Gruel increased the protein levels of p-ACC/ACC, p-AMPK/AMPK, MLYCD, and CPT-1, which was consistent with the observed changes in gene expression. Additionally, the combination of these two agents was significantly more effective in upregulating the expression of AMPK pathway-related genes and proteins. CONCLUSIONS: These results demonstrated that Yam Gruel may be a potential diet therapy for improving lipid metabolism in T2DM patients and that it may exert its effects via AMPK/ACC/CPT-1 pathway activation. In some respects, the combination of Yam Gruel and metformin exerted more benefits effects than Yam Gruel alone.

Osteoporosis, in which bones become fragile owing to low bone density and impaired bone mass, is a global public health concern. Bone mineral density (BMD) has been extensively evaluated for the diagnosis of low bone mass and osteoporosis. Circulating monocytes play an indispensable role in bone destruction and remodeling. This work proposed a machine learning-based framework to investigate the impact of circulating monocyte-associated genes on bone loss in osteoporosis patients. Females with discordant BMD levels were included in the GSE56815, GSE7158, GSE7429, and GSE62402 datasets. Circulating monocyte types were quantified via CIBERSORT, with subsequent selection of plasma cell-associated DEGs. Generalized linear models, random forests, extreme gradient boosting (XGB), and support vector machines were adopted for feature selection. Artificial neural networks and nomograms were subsequently constructed for osteoporosis diagnosis, and the molecular machinery underlying the identified genes was explored. SVM outperformed the other tuned models; thus, the expression of several genes (DEFA4, HLA-DPB1, LCN2, HP, and GAS7) associated with osteoporosis were determined. ANNs and nomograms were proposed to robustly distinguish low and high BMDs and estimate the risk of osteoporosis. Clozapine, aspirin, pyridoxine, etc. were identified as possible treatment agents. The expression of these genes is extensively posttranscriptionally regulated by miRNAs and m6A modifications. Additionally, they participate in modulating key signaling pathways, e.g., autophagy. The machine learning framework based on plasma cell-associated feature genes has the potential for estimating personalized risk stratification and treatment vulnerability in osteoporosis patients.

AIMS AND OBJECTIVES: (i) To estimate the national incidence of unplanned removal of peripherally inserted central catheters (PICCs) in China. (ii) To explore the associated risk factors to provide evidence for the prevention. DESIGN: A multi-centre prospective cohort study. METHODS: A representative sample of 3222 Chinese adult patients with successful PICC insertion was recruited for the PICC Safety Management Research (PATH) using a two-stage cluster sampling method from December 2020 to June 2022. Sixty hospitals from seven Chinese provinces representing all geographical regions were selected. Demographic information and PICC characteristics were collected using a standard online case report form. Risk factors for the unplanned removal of PICCs were assessed using a cause-specific hazard model and verified using a sub-distribution hazard model. STROBE guidelines were followed in reporting this study. RESULTS: Three thousand one hundred and sixty-six patients were included in the final analysis with a mean age of 59 years and a total of 344,247 catheter days. The incidence of unplanned removal was 10.04%. Female, with thrombosis history, PICC insertion due to infusion failure, valved catheter and double-lumen catheter were risk factors, whereas longer insertion and exposure length were protective factors in the cause-specific hazard model. Higher BMI became an independent risk factor in the sub-distribution hazard model. CONCLUSIONS: Unplanned removal of PICCs is a serious clinical challenge in China. Our findings call for prevention strategies targeting the identified risk factors. RELEVANCE TO CLINICAL PRACTICE: Our study characterised the epidemiology of unplanned removal of PICCs among Chinese adult inpatients, highlighting the need for prevention among this population and providing a basis for the formulation of relevant prevention strategies. PATIENT OR PUBLIC CONTRIBUTION: Patients contributed through sharing their information required for the case report form. Healthcare professionals who provide direct care to the patient at each medical centre contributed by completing the online case report form.

All-fiber-optic magnetic field sensor integrated with magnetic fluid has been investigated for decades, accompanied by the commitment to vectorization, miniaturization, integration and solving the temperature cross-sensitivity caused by thermo-optic effect of magnetic fluid. A kind of dual-channel-in-one temperature-compensated all-fiber-optic vector magnetic field sensor was proposed and investigated theoretically in this work. Three optical surfaces, including two sensing surfaces (plated with gold film of 40 nm thickness and then coated with magnetic fluid and polydimethylsiloxane, respectively, referred as CH1 and CH2) and one reflective surface, were integrated on a single-mode fiber tip to facilitate the dual-channel-in-one design. The Kretschmann configurations were formed by the waveguide fiber, gold film and functional materials at the sensing surfaces (CH1 and CH2). Surface plasmon resonance was excited in different wavelength bands corresponding to CH1 and CH2. Attenuation wavelengths corresponding to CH1 and CH2 depend on the magneto-induced and temperature-induced refractive index change of functional materials, respectively, which makes the temperature-compensated magnetic field sensing possible. The non-centrosymmetric evanescent field generated by micro-fiber-tip-prism enables the vector magnetic field sensing. Especially, the length of the sensing area is only 115.5 µm, which achieves ultra-integration and miniaturization. The current work provides a novel scheme for designing all-fiber-optic vector magnetic field sensing based on magnetic fluid and demonstrates the realization of lab-on-a-fiber and then promotes the industrial application of all-fiber-optic vector magnetic field sensing devices.

A novel, to the best of our knowledge, vector magnetic field sensor with temperature compensation is proposed and investigated. The proposed sensor is realized by side polishing a multi-mode optical fiber and adopting the surface plasmon resonance (SPR) effect. The side-polished surface is coated with a magnetic fluid (MF) and polydimethylsiloxane (PDMS) successively along the fiber axis. The as-fabricated sensor can be used not only for magnetic field strength and direction sensing, but also for temperature detection. The achieved magnetic field intensity sensitivities are 1720 pm/mT (90° direction) and -710 pm/mT (0° direction), and the temperature sensitivity is -2070 pm/°C. On top of its temperature compensation ability, the easy fabrication and very high sensitivity of the proposed sensor are attractive features for vector magnetic field sensing applications.

BACKGROUND: Using grip strength as a predictor of nutritional risk and early ambulation for gastrointestinal tumor surgery and determining its critical value have not been reported. This study was designed to explore the influencing factors of early postoperative ambulation ability for patients with gastrointestinal tumors who underwent laparoscopic surgery. METHODS: Four-hundred twenty-seven patients with gastrointestinal tumors who underwent laparoscopic surgery at three tertiary A hospitals in Beijing were prospectively enrolled. Subsequently, logistic regression analysis was conducted to determine the independent predictors of early postoperative ambulation. Logistic regression analyses for the different gender were also performed. In addition, the effectiveness of preoperative grip strength measurement in nutritional risk assessment was analyzed by using nutritional risk score 2002 (NRS 2002) as a control. RESULTS: The included cases were comprised of 283 male and 144 female patients, with an age of 59.35 ± 11.70 years. Gender, preoperative grip strength, operative time, and number of indwelling tubes were independent predictors of early postoperative ambulation. In the male group, lower preoperative grip strength and more indwelling tubes were independent risk factors for early postoperative ambulation. In the female group, lower preoperative grip strength and extended operating time were independent risk factors. Moreover, preoperative grip strength (male < 32 kg, female < 21 kg) can be used as a risk predictor for both preoperative nutritional risk and early postoperative ambulation. CONCLUSIONS: As a simple and objective measure of muscle strength, grip strength measurement is expected to be an effective predictor for both early postoperative ambulation ability and nutritional status of patients.

Fiber-optic magnetic field sensors based on magnetic fluid (MF) is encountering with thermal effects and demand for vectorization for several years. A common solution is to use axially processed fiber cascaded with fiber Bragg grating (FBG). However, the length of such sensors is usually in centimeter-level, which restricts the sensing applications in narrow space and gradient field cases. In this work, we present an ultracompact reflection-type dual-channel sensor for vector magnetic field (Channel 1, referred as CH1) and temperature (Channel 2, referred as CH2) monitoring, which is composed of a pair of gold-plated wedge-shaped multimode fiber (MMF) tip and gold-plated multimode-no-core fiber (MNF) tip. The surface plasmon resonance (SPR) effect was adopted. The two sensor probes are coated with magnetic-field-sensitive MF and temperature-sensitive polydimethylsiloxane (PDMS), respectively. The issue of vector magnetic field and temperature cross-sensitivity is tactfully resolved. Importantly, the proposed sensing probes are ultracompact and the spatial resolution is extremely small (615 µm for CH1 based on wedge-shaped fiber tip and 2 mm for CH2 based on MNF), which is very helpful for narrow space and gradient magnetic field detection. The obtained magnetic field intensity sensitivities are 1.10 nm/mT (90° direction) and -0.26 nm/mT (0° direction), and temperature sensitivity is -3.12 nm/°C.

In recent years, the biosensor research community has made rapid progress in the development of nanostructured materials capable of amplifying the interaction between light and biological matter. A common objective is to concentrate the electromagnetic energy associated with light into nanometer-scale volumes that, in many cases, can extend below the conventional Abbé diffraction limit. Dating back to the first application of surface plasmon resonance (SPR) for label-free detection of biomolecular interactions, resonant optical structures, including waveguides, ring resonators, and photonic crystals, have proven to be effective conduits for a wide range of optical enhancement effects that include enhanced excitation of photon emitters (such as quantum dots, organic dyes, and fluorescent proteins), enhanced extraction from photon emitters, enhanced optical absorption, and enhanced optical scattering (such as from Raman-scatterers and nanoparticles). The application of photonic metamaterials as a means for enhancing contrast in microscopy is a recent technological development. Through their ability to generate surface-localized and resonantly enhanced electromagnetic fields, photonic metamaterials are an effective surface for magnifying absorption, photon emission, and scattering associated with biological materials while an imaging system records spatial and temporal patterns. By replacing the conventional glass microscope slide with a photonic metamaterial, new forms of contrast and enhanced signal-to-noise are obtained for applications that include cancer diagnostics, infectious disease diagnostics, cell membrane imaging, biomolecular interaction analysis, and drug discovery. This paper will review the current state of the art in which photonic metamaterial surfaces are utilized in the context of microscopy.

LncRNAs are recently increasingly emerging as molecules that take its part in human carcinogenesis. A large body of literature has identified the functional roles of lncRNAs in the pathophysiology of CRC. The current study was intended to provide new ideas and perspectives for the functional role of lncRNA RAD51-AS1 in regulating CRC progression. Herein, a survey of RAD51-AS1 expression profile in The Cancer Genome Atlas (TCGA)-colon adenocarcinoma (COAD) dataset revealed that RAD51-AS1 was downregulated in COAD specimens. Consistently, RAD51-AS1 expression was observed to be lower in CRC cell lines compared with normal cell line (NCM460). In the meanwhile, both the levels of miR-29b-3p and miR-29c-3p were prominently elevated in CRC cells. Functionally, administration of RAD51-AS1 refrained growth, invasion and migration of CRC cells. Additionally, accumulation of RAD51-AS1 hampered glucose consumption and lactate production, as well as the restraint of hexokinase 2 (HK2) and glucose transporter 1 (GLUT1) levels. More important, RAD51-AS1 functioned as a competing endogenous RNA (ceRNA) for sponging miR-29b-3p and miR-29c-3p, leading to enhancement of their common target N-myc downstream-regulated gene 2 (NDRG2). Mechanistically, the delivery of miR-29b/c-3p mimics or ablation of NDRG2 effectively blunted the salutary effects of RAD51-AS1 on CRC cell behaviors. Moreover, augmentation of RAD51-AS1 inhibited the tumorigenesis of CRC cells in vivo. Collectively, these findings provide comprehensive evidence that RAD51-AS1 repressed cell proliferation, migration, invasion and glycolysis process, ultimately contributing to the progression of CRC by repressing the miR-29b/c-3p/NDRG2 signaling axis, insinuating the putative potential of RAD51-AS1/miR-29b/c-3p/NDRG2 interaction network in unraveling CRC pathology and hopefully contributed to the treatment of CRC patients.

INTRODUCTION: Ginseng is associated to the reduction of the risk of liver-cancer and some time is used as adjuvant therapy to treat liver-cancer, but its outcome remains uncertain. Hence, the present study aimed to determine the association between Ginseng consumption and liver-cancer. METHODS: By a systematic-literature search up to Decamber-2019, 9-studies included 13,766 subjects, 9235 Ginseng consumer. Odds ratio (OR) with 95% confidence intervals (CIs) was determined comparing Ginseng consumption and liver-cancer relationship using the dichotomous method with a fixed-effect or random-effect models. RESULTS: Subjects consuming Ginseng had a significantly lower risk of developing liver-cancer than those not consuming Ginseng (OR, 0.46; 95% CI, 0.40-0.52, p < 0.001). Also, there was a significant relationship between Ginseng consumption as adjuvant-therapy and disease control rate (OR, 4.47; 95% CI, 2.41-8.28, p < 0.001), Karnofsky Performance Scale (OR, 4.31; 95% CI, 1.80-10.36, p = 0.001), response to chemotherapy rate (OR, 1.79; 95% CI, 1.05-3.02, p = 0.03) and decline of leukocyte count (OR, 0.17; 95% CI, 0.07-0.42, p < 0.001). However, there was no significant effect, but relatively favoring Ginseng consumption, between Ginseng consumption as adjuvant-therapy and one year survival rate (OR, 1.48; 95% CI, 0.78-2.81, p = 0.23), two year survival rate (OR, 1.69; 95% CI, 0.87-3.25, p = 0.12) gastrointestinal dysfunction (OR, 0.55; 95% CI, 0.17-1.79, p = 0.32), and the hepatic dysfunction (OR, 1.15; 95% CI, 0.59-2.22, p = 0.68). CONCLUSIONS: Ginseng may have an independent relationship with reducing liver-cancer incidence when administrated to healthy subjects as a supplement and with reducing cancer-chemotherapy related outcomes risk when administrated with chemotherapy as adjuvant therapy.

Effective and rapid assessment of pork freshness is significant for monitoring pork quality. However, a traditional sensory evaluation method is subjective and physicochemical analysis is time-consuming. In this study, the near-infrared spectroscopy (NIRS) technique, a fast and non-destructive analysis method, is employed to determine pork freshness. Considering that commonly used statistical modeling methods require preprocessing data for satisfactory performance, this paper presents a one-dimensional squeeze-and-excitation residual network (1D-SE-ResNet) to construct the complex relationship between pork freshness and NIRS. The developed model enhances the one-dimensional residual network (1D-ResNet) with squeeze-and-excitation (SE) blocks. As a deep learning model, the proposed method is capable of extracting features from the input spectra automatically and can be used as an end-to-end model to simplify the modeling process. A comparison between the proposed method and five popular classification models indicates that the 1D-SE-ResNet achieves the best performance, with a classification accuracy of 93.72%. The research demonstrates that the NIRS analysis technique based on deep learning provides a promising tool for pork freshness detection and therefore is helpful for ensuring food safety.

BACKGROUND: Growing evidence has revealed the involvement of circular RNAs (circRNAs) in numerous carcinogenesis. However, the role of circRNAs in the cancer biology of colorectal cancer (CRC) remains vague. METHODS: Quantitative RT-PCR was used to detect the expression level of circRAE1 in CRC tissues and CRC cell lines. Cell proliferation, migration, and invasion were detected using CCK8 assay, Colony formation assay, wound-healing and Transwell assays. The interaction between circRAE1 and miR-338-3p and TRYO3 was confirmed using dual-luciferase reporter assays. RESULTS: We uncovered a novel circRNA Hsa_circ_0060967 (also known as circRAE1) that was remarkably increased in CRC tissues. The high circRAE1 level was positively associated with advanced tumor stage, lymph node metastasis, and tumor size. The loss-of-function assay showed that circRAE1 accelerated cell proliferation, migration, and invasion. Besides, miR-338-3p was lowly expressed in the CRC tissues and CRC cell lines. The dual-luciferase reporter assays showed that circRAE1 could sponge miR-338-3p, which targeted TRYO3 in CRC cells. Furthermore, the overexpression of circRAE1 could rescue the impaired migration and invasion triggered by miR-338-3p mimics or si-TYRO3 in CRC cells and vice versa. CONCLUSION: We identified the network of circRAE1, miR-338-3p, and TYRO3 in CRC cells and determined that the increase in circRAE1 could serve as an oncogene by sponging miR-338-3p, which resulted in an upregulated TYRO3 expression. The finding suggests that circRAE1 is a potential therapeutic target and diagnostic marker for CRC treatment.

BACKGROUND: A patient with a rare pediatric insulinoma and MEN1 syndrome was treated by robotic enucleation surgery. CASE PRESENTATION: We present a case of a 9-year-old girl presenting with repeated loss of consciousness, concomitant with a pale face, palpitations, and convulsions, which had persisted for 2 years and had been aggravated during the previous 2 months. She was previously misdiagnosed with epilepsy in another hospital. We further examined her while she was hospitalized. By combining her medical history and imaging examination and lab test results, a diagnosis of insulinoma was confirmed. Sanger-directed sequencing on a peripheral blood sample revealed an MEN1 gene mutation, indicating pediatric insulinoma with MEN1 syndrome. The patient underwent minimally invasive insulinoma enucleation surgery under the Da Vinci robot-assisted system with intraoperative ultrasound (IOUS) connected. The surgery was successfully completed within 65 min, and the girl recovered well postoperatively and no longer experienced symptoms of hypoglycemia. CONCLUSION: This is the first report of a case of pediatric insulinoma treated using robotic enucleation. This experience demonstrates the feasibility and safety of combining robotic surgery with the enucleation procedure as an excellent strategy for pediatric insulinoma.

Seizures during sleep increase the probability of complication and sudden death. Effective prediction of seizures in sleep allows doctors and patients to take timely treatments to reduce the aforementioned probability. Most of the existing methods make use of electroencephalogram (EEG) to predict seizures, which are not specific developed for the sleep. However, EEG during sleep has its characteristics compared with EEG during other states. Therefore, in order to improve the sensitivity and reduce the false alarm rate, this paper utilized the characteristics of EEG to predict seizures during sleep. We firstly constructed the feature vector including the absolute power spectrum, the relative power spectrum and the power spectrum ratio in different frequencies. Secondly, the separation criterion and branch-and-bound method were applied to select features. Finally, support vector machine classifier were trained, which is then employed for online prediction. Compared with the existing method that do not consider the characteristics of sleeping EEG (sensitivity 91.67%, false alarm rate 9.19%), the proposed method was superior in terms of sensitivity (100%) and false alarm rate (2.11%). This method can improve the existing epilepsy prediction methods and has important clinical value.

Suaeda salsa(S.salsa) is a typical vegetation of coastal wetland in the north of Liaodong Bay. The S. salsa biomass assessment plays an important role in understanding the ecosystem productivity of coastal wetland and the formation of ecosystem structure and function. Usually the S.salsa coverage is inhomogeneous. The low S.salsa coverage can be found at a natural condition, the soil background has a strong influence on S.salsa spectral data. The Transformed Soil Adjusted Vegetation Index (TSAVI) used as independent variable was derived by the Landsat 8 OLI simulation data. The S.salsa biomass inversion models were built based on the regression analysis of TSAVI and ground measured biomass in this study. The correlation between TSAVI (600~687, 820~880 nm) and biomass was significant, the correlation coefficient was about 0.9, up to 0.92. The results of linear and quadratic models were better than those of logarithmic, exponential and power models, the determination coefficient r2 of linear and quadratic models were 0.83. Combined with F value and operation efficiency, the linear model was the best option for mature S.salsa biomass inversion. The linear model was applied to invert the S.salsa biomass by using the Landsat 8 OLI data in the study area and it was further validated using in-situ data. The correlation coefficient between the in-situ value and retrieved value was 0.962, the relative error was 0.106. For higher S.salsa coverage, the relative error was lower. The relative error of the low-cover S.salsa biomass inversion was around 0.18. The results showed that the established model has good accuracy for different coverage. In addition, with the introduction of ±5% error of soil line parameters a and b, the average relative errors were relatively stable, and the correlation coefficients were reduced, but all the correlative coefficients were above 0.9. The results showed that the established model is stable.

The mTOR pathway is a central control of cell growth, proliferation, metabolism, and survival, and is deregulated in most cancers. Cancer cells are addicted to increased activity of mTOR kinase-mediated signaling pathways, leading to numerous inhibitors of mTOR signaling in preclinic and clinical trials for cancer therapy. Phosphorus-containing sirolimus (FIM-A), which targets mTOR signaling, inhibits cancer cell growth in vitro. Here we report that FIM-A reduces the angiogenesis and proliferation of osteosarcoma both in vitro and in vivo. In cultured osteosarcoma cell lines, FIM-A inhibited cell proliferation and arrested cells in the G1 phase of the cell cycle, accompanied with reduction of VEGF and HIF-1alpha. With in vivo mouse osteosarcoma xenografts, FIM-A treatment resulted in the inhibition of mTORC1 signaling as demonstrated by the decreased phosphorylation of p70S6K1 and 4E-BP1. Consistent with this finding, FIM-A significantly decreased the average tumor volume, nuclei staining of PCNA, and the number of intratumoral microvessels. Our data demonstrated that targeting mTORC1 by FIM-A inhibited the growth of osteosarcoma in vitro and in vivo, providing the basis for further development of FIM-A as a therapy for osteosarcoma patients.

The mechanisms of interaction between pulsed laser and materials are complex and indistinct, severely influencing the stability and quality of laser processing. This paper proposes an intelligent method based on the acoustic emission (AE) technique to monitor laser processing and explore the interaction mechanisms. The validation experiment is designed to perform nanosecond laser dotting on float glass. Processing parameters are set differently to generate various outcomes: ablated pits and irregular-shaped cracks. In the signal processing stage, we divide the AE signals into two bands, main and tail bands, according to the laser processing duration, to study the laser ablation and crack behavior, respectively. Characteristic parameters extracted by a method that combines framework and frame energy calculation of AE signals can effectively reveal the mechanisms of pulsed laser processing. The main band features evaluate the degree of laser ablation from the time and intensity scales, and the tail band characteristics demonstrate that the cracks occur after laser dotting. In addition, from the analysis of the parameters of the tail band very large cracks can be efficiently distinguished. The intelligent AE monitoring method was successfully applied in exploring the interaction mechanism of nanosecond laser dotting float glass and can be used in other pulsed laser processing fields.
