RESUMEN

Plants are an important source for the discovery of novel natural growth regulators. We used activity screening to demonstrate that treatment of Nipponbare seeds with 25 µg/mL isopimaric acid significantly increased the resulting shoot length, root length, and shoot weight of rice seedlings by 11.37 ± 5.05%, 12.96 ± 7.63%, and 27.98 ± 10.88% and that it has a higher activity than Gibberellin A3 (GA3) at the same concentration. A total of 213 inbred lines of different rice lineages were screened, and we found that isopimaric acid had different growth promotional activities on rice seedlings of different varieties. After induction with 25 µg/mL isopimaric acid, 15.02% of the rice varieties tested showed increased growth, while 15.96% of the varieties showed decreased growth; the growth of the remaining 69.02% did not show any significant change from the control. In the rice varieties showing an increase in growth, the shoot length and shoot weight significantly increased, accounting for 21.88% and 31.25%. The root length and weight significantly increased, accounting for 6.25% and 3.13%. Using genome-wide association studies (GWASs), linkage disequilibrium block, and gene haplotype significance analysis, we identified single nucleotide polymorphism (SNP) signals that were significantly associated with the length and weight of shoots on chromosomes 2 and 8, respectively. After that, we obtained 17 candidate genes related to the length of shoots and 4 candidate genes related to the weight of shoots. Finally, from the gene annotation data and gene tissue-specific expression; two genes related to this isopimaric acid regulation phenotype were identified as OsASC1 (LOC_Os02g37080) on chromosome 2 and OsBUD13 (LOC_Os08g08080) on chromosome 8. Subcellular localization analysis indicated that OsASC1 was expressed in the plasma membrane and the nuclear membrane, while OsBUD13 was expressed in the nucleus. Further RT-qPCR analysis showed that the relative expression levels of the resistance gene OsASC1 and the antibody protein gene OsBUD13 decreased significantly following treatment with 25 µg/mL isopimaric acid. These results suggest that isopimaric acid may inhibit defense pathways in order to promote the growth of rice seedlings.

RESUMEN

A naphthalimide hydroxylamine probe has been designed and synthesized to selectively label the whole natural aldehydes present in DNAs including 5-formylcytosine, 5-formyluracil, and abasic sites. The fluorescence characteristics of the generated nucleosides have been examined in detail, and the reaction activities of hydroxylamine, amine groups toward aldehydes in DNA have been discussed with others, which will be a vital reference for designing chemicals for selective labeling of DNAs.

RESUMEN

The green redevelopment of industrial brownfields (GRIB) is an important means to solve the shortage of urban land resources and realize sustainable urban renewal and green development. The identification and analysis of risk factors in GRIB projects are of immense significance for completing construction tasks and ensuring the planned benefits. In this study, work and risk breakdown structures and expert interviews are used to construct the risk network model of a GRIB project, based on three dimensions: process, subject, and system sources. The software package UCINET is used to conduct social network analysis and determine the key risk factors and relationships. The results of this study suggest that the four risk factors with the most brokerage roles and the highest node betweenness centralities are located at the core of the network; the six risk relationships with the highest line betweenness centralities are those with the strongest transmission capacities; the key risk factors are mostly response and stress risks; the main source is the design unit; and the key risk relationships are the influence of the decision-making stage on the design stage and of the design stage on the construction stage. Surpassing the limitations of traditional linear research, this study explains the internal relationship among the risk factors of GRIB projects and identifies the risk factors that play a brokerage role and the risk relationship that plays a conductive role, providing a theoretical basis for introducing social network analysis tools into the risk assessment of such complex construction projects.

As an innovative technique, laser-GMAW hybrid welding manifests significant superiority in enhancing welding productivity and quality, albeit the optimization of process parameters poses a challenge for practical application. The present manuscript elucidates the influence of process parameters on the dimensional characteristics of the welding seam and the distortion of 8 mm T-joints in the context of laser-GMAW hybrid welding, and channels both simulation and experimentation. The outcomes denote that the dual conical model serves as an efficacious aid for the numerical simulation of T-joint laser-GMAW hybrid welding. Furthermore, the repercussions of process parameters on welding seam dimensional characteristics remain consistently similar in both the simulation and experimental results. From the simulation outcomes, it becomes apparent that the distortion of the base material can be efficiently managed by implementing anti-distortion measures. This inquiry offers both a theoretical and experimental foundation for optimizing process parameters of T-joint laser-GMAW hybrid welding, presenting certain engineering applicability.

BACKGROUND AND AIM: The dual action of microglia in neurodegenerating diseases has been controversial for some time. Recent studies indicate that microglia senescence might be the key determinant. When microglia age, they function abnormally and fail to respond correctly to stimuli, which eventually promotes neurodegeneration. Accumulating evidence has shown a close relationship between inflammation and aging. Since neuroinflammation is characterized by microglia activation, we assessed if the repeated activation of microglia would lead to senescence. METHOD: The microglia cell line BV2 was repeatedly stimulated every 48 h with lipopolysaccharide (LPS; 10 ng/ml) and senescence was evaluated by ß-galactosidase staining and the presence of senescence-associated heterochromatic foci as well as by cell cycle arrest detection by flow cytometry. The senescence-associated protein p53 was also detected by Western blot. RESULTS: ß-galactosidase staining was barely detectable in control cells, while it tended to increase with repeated LPS stimulation and was positive in most cells after stimulation with LPS 6 times. Similarly, senescence-associated heterochromatic foci were most prominent in cells repeatedly stimulated with LPS, while almost undetectable in control cells or cells receiving a single stimulation. p53 expression was highest in the cells that received LPS stimulation 6 times, and the largest number of cells arrested in the G0/G1 phase was observed in this same group. CONCLUSION: Microglial cells tend to undergo senescence after repeated activation, implying that microglia senescence may start after multiple inflammatory challenges.

An azide and hydrazine tethered to a naphthalimide analogue was created to selectively react with 5-formyluracil in one system and fluorogenically label 5-formylcytosine in another system. A biotin group was also introduced by copper-free click chemistry through selective labelling of the aldehyde present in 5-formylpyrimidine.

RESUMEN

5-Formyluracil (5fU) based probes were designed and synthesized to detect Al3+ ions in vitro and in biological systems. These probes were synthesised using furan-2-carbohydrazide (fr) and 5fU derivatives. We also selectively labelled 5-formyluracil in DNA with the help of Al3+.
