RESUMEN

MOTIVATION: Spatial transcriptomics has greatly contributed to our understanding of spatial and intra-sample heterogeneity, which could be crucial for deciphering the molecular basis of human diseases. Intra-tumor heterogeneity, e.g. may be associated with cancer treatment responses. However, the lack of computational tools for exploiting cross-regional information and the limited spatial resolution of current technologies present major obstacles to elucidating tissue heterogeneity. RESULTS: To address these challenges, we introduce RegionalST, an efficient computational method that enables users to quantify cell type mixture and interactions, identify sub-regions of interest, and perform cross-region cell type-specific differential analysis for the first time. Our simulations and real data applications demonstrate that RegionalST is an efficient tool for visualizing and analyzing diverse spatial transcriptomics data, thereby enabling accurate and flexible exploration of tissue heterogeneity. Overall, RegionalST provides a one-stop destination for researchers seeking to delve deeper into the intricacies of spatial transcriptomics data. AVAILABILITY AND IMPLEMENTATION: The implementation of our method is available as an open-source R/Bioconductor package with a user-friendly manual available at https://bioconductor.org/packages/release/bioc/html/RegionalST.html.

RESUMEN

This paper introduces the concept of electromagnetically induced transparency (EIT) into the permittivity extraction of an anisotropic material-nematic liquid crystal (NLC). A novel two-step strategy is presented to extract the complex permittivity of the NLC at the THz band, which evaluates the relative permittivity tensor from the resonant frequencies and then determines the loss tangent from the quality factor Q of the EIT sensor. The proposed method features high accuracy due to the sharp resonance of the EIT sensor and also high robustness to the thickness of the NLC layer because only amplitude rather than phase information of the transmission coefficients is required. The NLC filled EIT sensor shows a sensitivity of 56.8 µm/RIU (the resonance wavelength shift over the refractive index change unit (RIU)) and Figure of Merit (FoM) of 6.92. The uncertainty of the proposed technique in the relative permittivity and loss tangent is 3% and 8.2%, respectively.

OBJECTIVE: To in vestigate potential gender differences in the odds of overweight/obese, weight-related perceptions, and behaviors among Chinese school children. METHODS: Height, weight, and a survey of weight-related perceptions and behaviors were measured in a nationally representative survey of 12,811 children in primary schools in China. Logistic regression analyses were used to assess gender differences, adjusting for confounders. RESULTS: Boys had higher odds of being overweight/obese compared to girls within both urban [adjusted odds ratio (OR) 2.30, 95% CI 2.00 to 2.65] and rural areas (OR = 1.85, 95% CI 1.55 to 2.20). Girls reported healthier diets (e.g., daily vegetables OR = 0.79, 95% CI 0.73 to 0.85) whereas boys consumed fried food (OR = 1.21, 95% CI 1.06 to 1.38) and sugar-sweetened drinks more often (OR = 1.49, 95% CI 1.34 to 1.65). Gender differences included higher odds of boys perceiving themselves as overweight if they had more highly educated mothers (OR = 1.35, 95% CI 1.09 to 1.68), less educated fathers (OR = 0.79, 95% CI 0.63 to 0.99), and if they frequently consumed carbonated drinks (OR = 1.48, 95% CI 1.07 to 2.05). CONCLUSION: Childhood obesity prevention in China should be gender-focused, particularly for boys who reported an unhealthier diet but were less likely to see they were fat, even though more boys were overweight or obese than girls.

OBJECTIVE: This study aims to assess the status of successful aging (SA) in longevity areas in China and explore multiple factors associated with SA among the young-old and oldest-old. METHODS: A total of 2296 elderly people aged 65 and older were interviewed in the longevity areas sub-sample of the Chinese Longitudinal Healthy Longevity Survey (CLHLS) in 2012. Baseline assessments included a researcher-administered questionnaire, physical examination, and laboratory testing. A logistic regression model was used to identify factors associated with SA. RESULTS: The prevalence of SA was 38.81% in the CLHLS in 2012. There were significant differences between ages groups, with SA compromising 56.85% among ⋝65 years group and 20.31% among ⋝100 years group (χ2trend=126.73, P<0.01). The prevalence of SA among females was 33.59%, which was significantly lower than that among males (45.58%) (χ2gender=33.65, P<0.05). In the regression analysis, having anemia (OR=0.744, 95% CI: 0.609-0.910), poor lifestyle (OR=0.697, 95% CI: 0.568-0.854), poor sleep quality (OR=0.558, 95% CI: 0.456-0.682), and central obesity (OR=0.684, 95% CI: 0.556-0.841) were the main factors associated with SA. The promoting SA rate decreased as age increased, and the group of 65-79 years had higher odds than the other age group. CONCLUSION: Preventing central obesity, improving sleep quality and promoting healthy lifestyle may contribute to achieve SA among the elderly.

Cu-V/Al-Ce with varying ratios of Al2O3/CeO2 were prepared to study the simultaneous catalytic oxidation of toluene and CO. Experimental results show that Cu-V/20Al-80Ce exhibits optimal simultaneous oxidation activity and good durability. This superior performance is related to Cu-Ce, V-Ce, and Al-Ce interactions, which facilitate the exposure of active centers, the creation of oxygen vacanicies, and efficient electron transfer. The mutual influence between toluene and CO during the simultaneous oxidation is then demonstrated. Toluene hinders CO oxidation through the competitive adsorption and the consumption of reactive oxygen species. CO enhances toluene oxidation, which is comprehensively explained by affecting the competition between the desorption and oxidation of benzaldehyde. Despite the mutual influence between toluene and CO, the pathways of CO and toluene oxidation are mutually independent. Toluene oxidation proceeds sequentially from toluene to benzyl alcohol, benzaldehyde, benzoate, and finally to CO2. Before being completely oxidized to CO2, CO is initially converted to carboxylic acid, hydrogen carbonate, free carbonate ion, bidentate formate, and monodentate carbonate.

BACKGROUND: Artificial Light at Night (ALAN) is an emerging health risk factor that has been linked to a wide range of adverse health effects. Recent study suggested that disadvantaged neighborhoods may be exposed to higher levels of ALAN. Understanding how social disadvantage correlates with ALAN levels is essential for identifying the vulnerable populations and for informing lighting policy. METHODS: We used satellite data from the National Aeronautics and Space Administration's (NASA) Black Marble data product to quantify annual ALAN levels (2012-2019), and the Center for Disease Control and Prevention's (CDC) Social Vulnerability Index (SVI) to quantify social disadvantage, both at the US census tract level. We examined the relationship between the ALAN and SVI (overall and domain-specific) in over 70,000 tracts in the Contiguous U.S., and investigated the heterogeneities in this relationship by the rural-urban status and US regions (i.e., Northeast, Midwest, South, West). RESULTS: We found a significant positive relationship between SVI and ALAN levels. On average, the ALAN level in the top 20% most vulnerable communities was 2.46-fold higher than that in the 20% least vulnerable communities (beta coefficient (95% confidence interval) for log-transformed ALAN, 0.90 (0.88, 0.92)). Of the four SVI domains, minority and language status emerged as strong predictors of ALAN levels. Our stratified analysis showed considerable and complex heterogeneities across different rural-urban categories, with the association between greater vulnerability and higher ALAN primarily observed in urban cores and rural areas. We also found regional differences in the association between ALAN and both overall SVI and SVI domains. CONCLUSIONS: Our study suggested ALAN as an environmental justice issue that may carry important public health implications. Funding National Aeronautics and Space Administration.

Artificial light at night (ALAN) is a growing environmental hazard with economic, ecological, and public health implications. Previous studies suggested a higher burden of light pollution and related adverse effects in disadvantaged communities. It is critical to characterize the geographic distribution and temporal trend of ALAN and identify associated demographic and socioeconomic factors at the population level to lay the foundation for environmental and public health monitoring and policy-making. We used satellite data from the Black Marble suite to characterize ALAN in all counties in contiguous US and reported considerable variations in ALAN spatiotemporal patterns between 2012 and 2019. As expected, ALAN levels were generally higher in metropolitan and coastal areas; however, several rural counties in Texas experienced remarkable increase in ALAN since 2012, while population-level ALAN burden also increased substantially in many metropolitan areas. Importantly, we found that during this period, although the overall ALAN levels in the USA declined modestly, the temporal trend of ALAN varied across areas with different racial/ethnic compositions: counties with a higher percentage of racial/ethnic minority groups, particularly Hispanic populations, exhibited significantly less decline. As a result, the differences in ALAN levels, as measured by the Black Marble product, across racial/ethnic groups became larger between 2012 and 2019. In conclusion, our study documented variations in ALAN spatiotemporal patterns across America and identified multiple population correlates of ALAN patterns that warrant further investigations. Future studies should identify underlying factors (e.g., economic development and decline, urban planning, and transition to newer lighting technologies such as light emitting diodes) that may have contributed to ALAN disparities in the USA.

Developing efficient catalysts with good resistance to complex flue gas is essential for VOCs removal in coal-fired flue gas. In this study, by exploring the effect of transition metal oxide additive, metal loading and bimetallic synergy on toluene oxidation in coal-fired flue gas, 10Cu-3V/γ-Al2O3 is identified as the optimal catalyst. It achieves 90% of CO2 generation at 350 â„ƒ, which is decreased by ca. 46 â„ƒ compared with 13Cu/γ-Al2O3. And it also exhibits good resistance to H2O and good stability. ICP-OES, N2 adsorption-desorption isotherms, XRD, TEM, XPS, EPR and H2-TPR analyses were applied to characterize the catalyst composition and physicochemical properties. Doping V into 13Cu/γ-Al2O3 not only leads to better dispersity of CuO and homogeneous elements distribution that benefits to produce more active centers, but also constitutes the redox cycle of V5+ + Cu+ ↔ V4+ + Cu2+ which induces more surface chemical oxygen (Osur). Moreover, since SO2 is the main inhibiting factor in toluene oxidation, the SO2 poisoning mechanism was illustrated by XPS, TG and in situ DRIFT analyses in depth.

Objective: The study aimed to analyze the applicability of the World Health Organization's exclusionary guidelines for Urinary creatinine (Ucr) in the general Chinese population, and to identify Ucr related factors. Methods: We conduct a cross-sectional study using baseline data from 21,167 participants in the China National Human Biomonitoring Program. Mixed linear models and restricted cubic splines (RCS) were used to analyze the associations between explanatory variables and Ucr concentration. Results: The geometric mean and median concentrations of Ucr in the general Chinese population were 0.90 g/L and 1.01 g/L, respectively. And 9.36% samples were outside 0.3-3.0 g/L, including 7.83% below the lower limit and 1.53% above the upper limit. Middle age, male, obesity, smoking, higher frequency of red meat consumption and chronic kidney disease were associated significantly with higher concentrations of Ucr. Results of the RCS showed Ucr was positively and linearly associated with body mass index, inversely and linearly associated with systolic blood pressure, diastolic blood pressure, triglycerides level, and glomerular filtration rate, and were non-linearly associated with triiodothyronine. Conclusion: The age- and gender-specific cut-off values of Ucr that determine the validity of urine samples in the general Chinese population were recommended. To avoid introducing bias into epidemiologic associations, the potential predictors of Ucr observed in the current study should be considered when using Ucr to adjust for variations in urine dilution.

The influences of SO2 on Hg° removal over the 1V-8Ce/AC sorbent were systematically investigated at low temperatures. The experimental results showed that SO2 has a dual effect on Hg° removal, that is, SO2 has both a promoting effect and an inhibiting effect on Hg° removal. The SO2 transient response experiment indicated that SO2 could not only react with Hg° to promote the removal of Hg° but also react with the active components and poison the sorbent. O2 is indispensable for the removal of Hg°, which can offset the adverse effects caused by SO2 and H2O. HCl exhibited an obvious promoting effect on Hg° removal in the presence of SO2. The 1V-8Ce/AC sorbent exhibited good sulfur resistance and excellent stability (EHg = 90.04 %) after a 24 h reaction performed under the 1000 ppm SO2 condition at 150 °C. In addition, the Hg-TPD and XPS methods were used to assist in studying the effect of SO2 on Hg° removal over the 1V-8Ce/AC sorbent. Finally, the mechanism of Hg° removal in an SO2 atmosphere was also explored, which showed that Hg° was removed by two possible pathways over the 1V-8Ce/AC sorbent.

MnxCoy/Zrz-AC prepared by impregnation method was investigated on the simultaneous removal of HCHO and Hg0. The samples were characterized by BET, SEM, XRD, H2 pulse chemisorption, H2-TPR, XPS, Hg-TPD and in-situ DRIFTS. Thereinto, the optimal Mn2/3Co8/Zr10-AC achieved 99.87% HCHO removal efficiency and 82.41% Hg0 removal efficiency at 240 °C, respectively. With increased surface area and pore volume, Zr-AC support facilitated higher dispersion of MnOx-CoOx. Moreover, the co-doping of MnOx-CoOx endowed the sample with more active oxygen species and higher reducibility, which further facilitated the removal of HCHO and Hg0. Chemisorption was proved to predominate in Hg0 removal, and oxidation also worked as Hg2+ was detected in outlet gas. Besides, HCHO predominated in the competition of active oxygen species, especially for lattice oxygen, thus suppressed the Hg0 removal. According to in-situ DRIFTS, HCHO removal proceeded as HCHOads → DOM → formate species → CO2 + H2O, and was boosted by active oxygen species. Furthermore, Mn2/3Co8/Zr10-AC was proved with excellent regeneration performance, indicating its potential in practical application.

BACKGROUND: Lack of biomarkers and in vitro models has contributed to inadequate understanding of the mechanisms underlying the inferior clinical response to immune checkpoint inhibitors (ICIs) in patients with oncogene-driven non-small cell lung cancer (NSCLC). METHODS: The effect of small molecule tyrosine kinase inhibitors (TKIs) on peripheral blood mononuclear cells (PBMCs) in 34 patients with oncogene-driven NSCLC (cohort A) was compared with those from 35 NSCLC patients without oncogene-driven mutations received ICI (cohort B) or from 22 treatment-naïve NSCLC patients (cohort C). Data for each blood biomarker were summarized by mean and standard deviation and compared by Wilcoxon rank sum tests or Kruskal-Wallis tests with significance at 2-sided p value < 0.05. Co-culture of PBMCs and pleural effusion-derived tumor cells from individual patients with oncogene-driven NSCLC was used to determine the in vitro cytotoxicity of TKI and ICI. RESULTS: Except for low CD3% in cohort A, there were no significant differences in other 12 blood biomarkers among the 3 cohorts at baseline. TKI treatment in cohort A was associated with significant increase in CD3% and decrease in total and absolute neutrophils (p < 0.05). In cohort B, patients with good clinical response to ICI treatment (N = 18) had significant increases in absolute lymphocyte counts (ALCs), CD4 and/or CD8 cell counts. Conversely, those patients with poor clinical response to ICI (N = 17) had significant decreases in these cell counts. Of the 27 patients with pre- and post-treatment blood samples in cohort A, 11 had poor clinical response to TKIs and decreased lymphocyte counts. Of the remaining 16 patients who had good clinical response to TKI therapy, 10 (62.5%) patients had decreased, and 6 (37.5%) patients had increased lymphocyte counts. Multicolor immunophenotyping of PBMCs revealed ICI treatment activated additional immune cell types that need further validation. We confirmed that TKI treatment could either antagonize or enhance the effect of ICIs in the co-culture assay using patient's tumor cells and PBMCs. CONCLUSIONS: To the best of our knowledge, this is the first study showing that TKIs can have various effects on blood immune cells, which may affect their response to ICIs. Further validation of the blood biomarker and in vitro assay is warranted.

OBJECTIVE: Evidence regarding alcohol consumption and cognitive impairment is controversial. Whether cessation of drinking alcohol by non-dependent drinkers alters the risk of cognitive impairment remains unknown. This study prospectively evaluated the potential association between the history of lifetime alcohol cessation and risk of cognitive impairment. METHODS: This study included 15,758 participants age 65 years or older, selected from the Chinese Longitudinal Healthy Longevity Survey (CLHLS) that covered 23 provinces in China. Current alcohol use status, duration of alcohol cessation, and alcohol consumption before abstinence were self-reported by participants; cognitive function was evaluated using Mini-mental State Examination (MMSE). Cause-specific hazard models and restricted cubic splines were applied to estimate the effect of alcohol use on cognitive impairment. RESULTS: Among the 15,758 participants, mean (± SD) age was 82.8 years (± 11.9 years), and 7,199 (45.7%) were males. During a mean of 3.9 years of follow-up, 3,404 cases were identified as cognitive impairment. Compared with current drinkers, alcohol cessation of five to nine years [adjusted HR, 0.79 (95% CI: 0.66-0.96)] and more than nine years [adjusted HR, 0.82 (95% CI: 0.69-0.98)] were associated with lower risk of cognitive impairment. CONCLUSION: A longer duration of alcohol cessation was associated with a lower risk of cognitive impairment assessed by MMSE. Alcohol cessation is never late for older adults to prevent cognitive impairment.

PURPOSE: To evaluate the feasibility of Quantitative Ultrashort-Time-to-Echo Contrast-Enhanced (QUTE-CE) MRA using ferumoxytol as a contrast agent for abdominal angiography in the kidney. METHODS: Four subjects underwent ferumoxytol-enhanced MRA with the 3D UTE Spiral VIBE WIP sequence at 3 T. Image quality metrics were quantified, specifically the blood Signal-to-Noise Ratio (SNR), blood-tissue Contrast-to-Noise Ratio (CNR) and Intraluminal Signal Heterogeneity (ISH) from both the aorta and inferior vena cava (IVC). Morphometric analysis of the vessels was performed using manual approach and semi-automatic approach using Vascular Modeling ToolKit (VMTK). Image quality and branching order were compared between QUTE-CE MRA and the Gadolinium (Gd) CEMRA reference image. RESULTS: QUTE-CE MRA provides a bright blood snapshot that delineates arteries and veins equally in the same scan. The maximum SNR and CNR values were 3,282 ± 1,218 and 1,295 ± 580, respectively - significantly higher than available literature values using other CEMRA techniques. QUTE-CE MRA had lower ISH and depicted higher vessel branching order (7th vs 3rd) within the kidney compared to a standard dynamic clinical Gd CEMRA scan. Morphometric analysis yielded quantitative results for the total kidney volume, total cyst volume and for diameters of the branching arterial network down to the 7th branch. Vessel curvature was significantly increased (p < 0.001) in the presence of a renal cyst compared to equivalent vessels in normal kidney regions. CONCLUSION: QUTE-CE MRA is feasible for kidney angiography, providing greater detail of kidney vasculature, enabling quantitative morphometric analysis of the abdominal and intra-renal vessels and yielding metrics relevant to vascular diseases while using a contrast agent ferumoxytol that is safe for CKD patients.

OBJECTIVE: To evaluate the associations of sarcopenia, handgrip strength and calf circumference with cognitive impairment among Chinese older adults. METHODS: Totally 2,525 older adults were recruited from the Healthy Aging and Biomarkers Cohort Study. Cognitive impairment was assessed by the Chinese Mini-Mental State Examination. Handgrip strength was calculated from the means of the right and left hand values. Calf circumference was measured at the site of maximum circumference of the non-dominant leg. The formula developed by Ishii was used to define sarcopenia. Multiple logistic regression was performed to evaluate the associations of sarcopenia, handgrip strength, and calf circumference with cognitive impairment. RESULTS: The prevalence of cognitive impairment was 34.36%. The adjusted odds ratio ( OR) for cognitive impairment in individuals with sarcopenia was 2.55 [95% confidence interval (95% CI): 1.86-3.50]. Compared with individuals in the first quartile (Q 1) of calf circumference, the adjusted ORs in the second, third, and fourth quartiles (Q 2, Q 3, and Q 4) were 0.75 (95% CI: 0.58-0.96), 0.59 (95% CI: 0.44-0.79), and 0.62 (95% CI: 0.45-0.8), respectively. Compared with individuals in Q 1 of handgrip strength, the adjusted ORs for Q 2, Q 3, and Q 4 were 0.49 (95% CI: 0.38-0.62), 0.31 (95% CI: 0.23-0.41), and 0.30 (95% CI: 0.21-0.44), respectively. CONCLUSION: Sarcopenia, identified by low handgrip strength and low calf circumference, was positively associated with cognitive impairment.

RESUMEN

It is necessary to control the emissions of toluene, which is hazardous to both human health and the atmosphere environment and has been classified as a priority pollutant. Manganese oxide-based (Mn-based) catalysts have received increased attention due to their high catalytic performance, good physicochemical characteristic, availability in various crystal structures and morphologies, and being environmentally friendly and low cost. These catalysts can be classified into five categories, namely single manganese oxide, Mn-based composite oxides, Mn-based special oxides, supported Mn-based oxides, and Mn-based monoliths. This review focused on the recent progress on the five types of Mn-based catalysts for catalytic removal of toluene at low temperature and further systematically summarized the strategies improving catalysts, including improving synthetic methods, incorporating MnOx with other metal oxides, depositing Mn-based oxides on proper supports, and tuning the supports. Moreover, the effect of coexisting components, the reaction kinetics, and the oxidation mechanisms toward the removal of toluene were also discussed. Finally, the future research direction of this field was presented.

PURPOSE: Given the unmet need for novel immunotherapy in soft tissue sarcoma (STS), we sought to characterize the phenotype and function of intratumoral natural killer (NK) and T cells to identify novel strategies to augment tumor-infiltrating lymphocyte (TIL) function. EXPERIMENTAL DESIGN: Using prospectively collected specimens from dogs and humans with sarcomas, archived specimens, and The Cancer Genome Atlas (TCGA) data, we evaluated blood and tumor NK and T cell phenotype and function and correlated those with outcome. We then assessed the effects of interleukin 15 (IL-15) stimulation on both NK and T cell activation and TIGIT upregulation. Finally, we evaluated cytotoxic effects of IL-15 combined with TIGIT blockade using a novel anti-TIGIT antibody. RESULTS: TILs were strongly associated with survival outcome in both archived tissue and TCGA, but higher TIL content was also associated with higher TIGIT expression. Compared with blood, intratumoral NK and T cells showed significantly higher expression of both activation and exhaustion markers, in particular TIGIT. Ex vivo stimulation of blood and tumor NK and T cells from patients with STS with IL-15 further increased both activation and exhaustion markers, including TIGIT. Dogs with metastatic osteosarcoma receiving inhaled IL-15 also exhibited upregulation of activation markers and TIGIT. Ex vivo, combined IL-15 and TIGIT blockade using STS blood and tumor specimens significantly increased cytotoxicity against STS targets. CONCLUSION: Intratumoral NK and T cells are prognostic in STS, but their activation is marked by significant upregulation of TIGIT. Our data suggest that combined IL-15 and TIGIT blockade may be a promising clinical strategy in STS.

The citrus leaf beetle, Clitea metallica, is a specialized citrus pest through feeding on fresh leaves by larva and adults, and causes nicks and holes into leaves, leaving only a waxy surface layer. Insect cuticle is a complex exoskeleton that is not only involved in development but also protects the insect from environmental contaminations. Due to these key roles of the cuticle, cuticle-related genes are currently investigated in understanding the insect physiology in adaptation. Therefore, in this study, we built two libraries, transcriptomic (43 million clean reads) and small RNA (17 million clean reads), of C. metallica to identify cuticle-related genes and possibly associated miRNAs, being as an example to explore these data sets. Our results showed that a total of 47 cuticular protein genes were identified and most of these genes harbored a conserved motif (the Rebers and Riddiford motif) and belonged to the CPR family. Unigenes encoding proteins involved in chitin synthesis and degradation were also identified, including chitin synthase (2 unigenes), chitinase (14 unigenes), chitinase-like protein (2 unigenes), and chitin deacetylase (5 unigenes). Based on the small RNA library, we identified 30 miRNAs conserved across insect species. Among these miRNAs, 14 were predicted to be target genes associated with cuticle synthesis and degradation. In summary, 70 cuticle-related genes and 14 cuticle-related miRNAs were identified based on the transcriptome and small RNA library of C. metallica. These data sets will promote the understanding of cuticle molecular regulation in C. metallica as well as provide new potential targets for pest control.
