RESUMEN

Acholeplasma axanthum is one of the few procaryotes, and the only member of the Mollicutes, known to contain phosphosphingolipids. Examination of strain S743 for glycolipids revealed the presence of glucosides of cholesterol and galactosides of glycerol as the predominant glycolipids. The major component is acylated diglucosylcholesterol, followed by monogalactosyldiacylglycerol and monoglucosylcholesterol. The glucose residues of the sterol-based compounds appear to be alpha-linked pyranoses, while the galactose of the glycerol-based lipid is an alpha-linked furanose. The "glycolipid' fraction also contained N-(3-hydroxy)acyl sphinganines with varying degrees of O-acylation. None of these ceramide derivatives was linked to carbohydrate. The major glycolipid, tentatively identified as alpha-D-glucopyranosyl-(1 leads to 3)-(O-acyl)-alpha-D-glucopyranosyl-(1 leads to 3)-cholesterol, along with its deacylated derivative, appears to be the first reported instance of steryl diglycosides among procaryotes, in contrast to the steryl monoglycosides, which are common to other mycoplasmata and some spirochetes.

RESUMEN

A long-chained polyalcohol (polyol) was isolated from the glycolipid fraction of the extreme thermoacidophile Bacillus acidocaldarius. The polyol and its Smith degradation products, as well as the alkanes derived from these compounds were characterized by mobility on thin-layer and gas-liquid chromatography, and by infrared and mass spectrometry. The polyol is proposed to be a fully saturated pentacyclic tetrol (C35H62O4, Mr 546) containing a 1,2,3,4-tetrahydroxy pentane substituted to a hopane-derived pentacyclic triterpene nucleus.

The total lipid content of the extreme thermoacidophile Bacillus acidocaldarius comprises about 8.1% of the cell dry weight. Total lipid had a distribution of 15.7% neutral linique component initially characterized as an N-acylglucosamine beta-linked to the primary hydroxyl of an unusual fully saturated pentacyclic triterpene derived tetrol(C35H62O4, Mr 546), which appears to be a derivative of the pentacylcic triterpene hopane substituted at C-29 with a 1,2,3,4-tetrahydroxy pentane. Other major glycolipids present were partially characterized as O-beta-D-glucopyranosyl-(1 leads to 4)-O-2-acylamido-2-deoxy-beta D-glucopyranosyldiacylglycerol and O-beta-D-glucopyranosyl-(1 leads to 4)-O-2-acylamido-2-deoxy-beta-D-glucopyranosylmonoacylglycerol. Minor components of the glycolipid fraction included O-beta-D-glucopyranosyl-(1 leads to 4)-O-2-acylamido-2-deoxy-beta-D-glucopyranosylglycerol, O-2-amino-2-deoxy-beta-D-glucopyranosyl pentacyclic tetrol and free pentacyclic tetrol. The distributions of esterified and amide-linked fatty acids were similar, being comprised primarily of branched heptadecanoic, 11-cyclohexyundecanoic and 13-cyclohexyltridecanoic acids. The acid lipids were composed of a sulfonoglycosyldiacylglycerol (43.2%), diphosphatidylglycerol (32.3%), lysodiphosphatidylglycerol (5.3%), phosphatidic acid (5.8%) and phosphatidylglycerol (13.4%).

RESUMEN

Three Legionella-like organisms isolated from patients with pneumonia are shown to belong to the species Legionella sainthelensi by DNA hybridization studies and to a new serogroup, serogroup 2, by serological studies (ATCC 49322). L. sainthelensi serogroup 2 and L. santicrucis are indistinguishable by slide agglutination, but are separable on the basis of their cell wall fatty acid profiles.

We compared the cellular fatty acid and protein content of 43 strains of oral fusobacteria isolated from patients with chronic gingivitis, acute necrotizing ulcerative gingivitis, and juvenile and adult periodontitis, as well as from the stump-tailed macaque, to that of eight reference strains of oral and non-oral Fusobacterium species. A gas-liquid chromatographic examination of trimethylsilyl derivatives of the fatty acid methyl esters revealed n14:0, 3-OH-14:0, n16:0, 16:1, 3-OH-16:0, n18:0, and 18:1 in each of the 51 study strains, and a variable occurrence of 14 other fatty acids. The n16:0 to 3-OH-16:0 ratio distinguished between Fusobacterium nucleatum and the non-oral species Fusobacterium varium, Fusobacterium necrophorum, Fusobacterium russii, Fusobacterium necrogenes, Fusobacterium mortiferum, and Fusobacterium naviforme. The soluble protein content, as determined by polyacrylamide gel electrophoresis, varied considerably among the oral fusobacterial strains studied, and underscored the heterogenous nature of these organisms. "Fingerprinting" of oral fusobacteria may be readily accomplished by polypeptide analysis.

How dentists learn about and integrate new procedures and techniques into dental practice are major concerns in innovation research in dentistry. The purposes of this study were to: (1) develop reliable measures of self-reported adherence to different national guidelines; (2) to examine the relationships among the different measures; and (3) to determine if predictors of these adherence behaviours exist. One thousand U.S. general dentists were asked to complete a survey of adherence behaviours to established guidelines in infection control, radiation protection, and antibiotic prophylaxis. Thirty-five percent responded to the mailed survey. Reliable scales were developed in infection control and antibiotic prophylaxis behaviors. Step-wise multiple regression analysis produced several weak predictors of adherence to guidelines in these two areas. Characteristics of early adopters such as office complexity, membership in professional associations, and year of graduation, were consistent with other dental innovation literature. Significant correlations were found between adherence behaviors in infection control and antibiotic prophylaxis. These correlations do support the findings that practitioners who are early adopters of innovation are likely to do so in more than one area.

The purpose of this examiner-blind study was threefold: to compare the microbial counts obtained by two different techniques for assessing bacterial motility, to assess the inter-rater reliability of these two techniques and to evaluate the effect of a water irrigating device (Water Pik) on bacterial motility at 3- and 6-mm probing depths. Subgingival plaque samples were taken from 10 healthy patients having at least two sites that probed greater than 6 mm, (one control, one experimental). Half of the patients were sampled at 3 mm, the other half at 6 mm, both at baseline (Day 0) and at Day 21. Two slides were prepared from each plaque sample, one for dark-field evaluation and one stained with a simplified silver-plating technique for flagella. All slides were read simultaneously by 3 observers, and the per cent motility calculated for spirochetes, motiles and all others. Strong positive inter-rater reliability correlations ranging from r = 0.95 to r = 0.99 were found for both the dark-field and flagella staining techniques. Spirochete counts obtained by both techniques were highly correlated (r = 0.91), whereas counts for motiles resulted in negative correlations between the techniques. Dark-field counts were consistently higher than the flagella stain counts for motile rods. Spirochetes were reduced, but not significantly, after irrigation of both 3-mm and 6-mm sites. Bacterial motility can be evaluated by both dark-field and flagella-staining techniques with a high degree of inter-rater reliability.

L-idaro-1,4-lactone was synthesized by two different, published methods: (1) epimerization of monopotassium D-glucarate by refluxing in aqueous barium hydroxide, and (2) oxidation of L-iditol by heating in dilute nitric acid. The lactone, formed by heat dehydration from aqueous solution at low pH, was purified by paper chromatography, and quantitated by gas-liquid chromatography using inositol as the internal standard. The monolactone inhibited human, seminal-fluid alpha-L-idosiduronase activity, with either phenyl or 4-methylumbelliferyl alpha-L-idosiduronic acid as the substrate, to the same degree as D-glucaro-1,4-lactone inhibits alpha-D-glucosiduronase.

Drawings about the dentist at work were solicited from 1,101 children in grades 2, 4, 6 and 8 in an urban school district. A system to classify the contents of these drawings was developed. The frequency of various items occurring in the children's drawings was determined. The "typical" or most frequent child's drawing of the dentist at work was described. This drawing contained a normal dental chair, dentist, a patient in the chair and dental cabinetry or furniture. The picture was a very matter-of-fact representation. Abnormal or bizarre pictures occurred infrequently. A few children drew pictures that did not relate to dentistry.

701 dental-school outpatients rated the degree of fear occasioned by each of 60 events that occur routinely during dental treatment. The 4,206 ratings then were subjected to principal components factor analysis with oblique rotation. Four meaningful factors were deduced: fear of pain and its antecedents, anticipatory fear, fear of negative evaluation, and fear due to perceived absence of control over the dentist. This outcome is compared and contrasted with results of other attempts to delineate the components of dental fear.

Pediatric dentists are at increasing risk of exposure to the human immunodeficiency virus (HIV). Therefore, a nationwide survey was conducted to describe pediatric dentists' use of barrier techniques and infection-control procedures and to identify factors that might impede their use. Questionnaires were mailed to a random sample of 1581 practicing pediatric dentists. Seven hundred fifty-two completed questionnaires were returned. Results showed that 95.3% of the respondents have changed some aspect of infection control during the past two years; 85.9% routinely wear gloves; 77.9% routinely wear protective glasses; and 45.6% routinely wear facemasks. Results also highlighted several problems created by the use of gloves and facemasks.

A study was done to assess the impact of an inquiry-oriented curriculum in a dental hygiene program. Two instruments, designed to measure student perception of personal and faculty inquiry and disinquiry behavior, were administered to four consecutive classes twice a year between 1978 and 1981. Junior and senior students perceived a significant increase in faculty inquiry behavior and, initially, a decrease in disinquiry behavior. Time in the program affected student disinquiry behavior at a significant level, though no significant change was seen in inquiry behavior. National Board mean scores improved, and several categories of clinical productivity increased significantly during this period. The implications of these findings are discussed.

A differentiated model of self-esteem, based on William James's (1890), C. H. Cooley's (1902/1968), and G. H. Mead's (1925/1968) works and developed by Dr. Susan Harter (1983), is presented as a valuable tool for the occupational therapist to use in evaluation and intervention. Competence and social support are considered important components, and discounting is presented as a mechanism for the maintenance or improvement of self-esteem. Social comparison, social desirability, and defensiveness are discussed as important considerations, particularly regarding children with disabilities.

Negative attitudes exhibited by rehabilitation professionals could have adverse consequences for persons with disabilities in their achievement of desired outcomes. Because occupational therapists are in a position to exert considerable influence in the therapeutic relationship, this study explored entry-level occupational therapy practitioners' equal status contact with and attitudes toward persons with disabilities. METHOD. Questionnaires were sent to 402 entry-level occupational therapists. These questionnaires consisted of: a demographic scale, the Disability Social Distance Scale, the Contact With Disabled Persons Scale, and a Similarity Scale (constructed for this study to evaluate the concept of equal status). Data analysis explored the relationship between 172 respondents' equal status perception and contact with persons with disabilities and their attitudes toward persons with disabilities. RESULTS. Respondents exhibited generally positive attitudes toward persons with disabilities. Perceived equality, personal and professional contact, and the majority of demographic factors were not significantly correlated with their attitudes toward persons with disabilities. Respondents who specified working with persons without disabilities in the wellness capacity exhibited more positive attitudes than those working with persons with biomechanical and neurological disabilities. In addition, respondents who had a greater frequency of professional contact with persons with disabilities also appeared to experience more personal contact with persons with disabilities. CONCLUSION. Entry-level occupational therapists' attitudes toward persons with disabilities may not only be influenced by parameters of the contact variable but also by the profession's holistic philosophy, the occupational therapy educational curricula, and the personal characteristics of those who choose to pursue a career in occupational therapy.

RESUMEN

The purpose of this study was to investigate factors that affected occupational therapy practice in the rural regions of the Rocky Mountains. It compared data from the Rocky Mountain region (Wyoming, Colorado, New Mexico, Utah, and Arizona) to a previous study of the Northwest region of Alaska, Idaho, Montana, and Washington and expanded on the Northwest study. Results indicated that the two areas were similar in their demographics and problems, including need for more occupational therapists. The results also have implications regarding specialized preservice training for rural-based occupational therapists, other professions' availability of continuing education, and other support systems. The authors provide recommendations based on the results, other literature, and conjectures, for recruitment, training, and ongoing support of rural occupational therapists through cooperative consortiums and distance learning.

We tested the following groups for visual pursuit abilities: 54 mentally retarded, 46 cerebral-palsied, and 131 learning-disabled children, 3 to 10 years of age. The resulting five behavioral scores, six directions, and total scores were analyzed for age trends. Comparisons were made between the handicapped groups and between the handicapped and normative samples. The scores were also correlated with other visual and postural abilities. All three handicapped groups scored lower than the normative group and also showed patterns different from the normative group and different from each other. The learning-disabled group most closely approximated the normative group's development patterns. Educational data were not able to be used, and we suggest that future research examine the correlations (both in handicapped and nonhandicapped children) of ocular pursuits and educational achievement, and also obtain more complete data on cerebral-palsied children. Therapists may find the visual pursuits test useful particularly when they want to evaluate a number of aspects of ocular pursuit functioning or when they desire more precision than what is usually available by simple observation.

OBJECTIVES: The purposes of this study were to examine the cross-cultural validity of the Assessment of Motor and Process Skills (AMPS) as well as the reliability of raters from different cultures. METHOD: Six trained raters from diverse cultural backgrounds scored 10 Japanese subjects' performances on familiar and culturally relevant tasks of their own choosing. RESULTS: Results indicated high cross-cultural validity and interrater reliability for the AMPS, as indicated by goodness of fit of subjects and raters via the many-faceted Rasch measurement model. The relative rater severity was also evaluated cross-culturally by the standardized difference (z). The Japanese rater and one of the three American raters varied significantly in severity between their scoring of Japanese subjects versus European subjects. CONCLUSION: The results support the hypothesis that the AMPS can be used as a cross cultural instrumental activities of daily living assessment. Further study is needed to clarify the issue of possible cultural bias in rater severity.

Although play is an important activity of childhood, it is often a neglected modality in occupational therapy. Poverty has been identified as a factor that adversely affects the play of children when a limited number of factors are studied. The purpose of this pilot study was to explore multiple play behaviors that may be related to socioeconomic status. The developmental play ages of 41 middle socioeconomic status preschool children (22 boys, 19 girls) and 43 low socioeconomic status preschool children (24 boys, 19 girls) were assessed with a modified version of the Preschool Play Scale (Bledsoe & Shepherd, 1982; Knox, 1974). Teachers rated the children's play behavior after a minimum of 2 months of interaction in their preschool classrooms. No significant differences between these two groups of children were found for developmental play age, play dimensions, or play categories. It was concluded that the exposure to positive experiences, such as peer interaction and preschool setting, may minimize the effects of poverty conditions on the play behavior of children.

Ocular pursuits have been evaluated by occupational therapists (OTRs) in many settings, but normative information has been lacking. In this study, a standardized method of testing and recording ocular pursuits was developed and pilot tested on both normal and handicapped children. Normative data were then collected by using a cooperative research method in which 28 OTRs in the state of Colorado were trained, in a one-day workshop, to administer and score the test, achieving .98 inter-rater reliability for total test score. Each qualified tester evaluated approximately l5 normal children between 3 and ll years of age. The total sample was 489 subjects. There were some age-related trends, and most behaviors demonstrated a ceiling effect, particularly with older children. The results imply that many normal children have some immaturity in ocular pursuit skills until 6 or 7 years of age.

This study examines the effect of a training program on long-term-care (LTC) surveyors' knowledge of factors affecting oral health and their behavior in citing oral health deficiencies of LTC residents. Twenty surveyors were recruited for the experimental group, along with a non-equivalent comparison group of 31 surveyors. The experimental group participated in a three-hour interactive, case-based training program. The experimental group had a significant increase in total knowledge (p < 0.002) post-test scores with a moderate effect size, eta 2 = 0.115. The training program resulted in a statistically significant increase (p < 0.0001) in oral health citation behavior for the experimental group for six months following training. This study provides evidence that an interactive case-based training program can affect participants' knowledge regarding oral health of LTC residents and participants' oral health citation behavior. As the elderly population experiences an increased life expectancy with increased need for assistance with Activities of Daily Living (ADL), similar training programs will become an important factor in assessing the quality of care provided to LTC residents.
