The understanding of the biological mechanisms involved in root resorption in deciduous teeth is important to the future development of preventive measures and treatments of this condition. The aim of the present study was to compare the expression and immunostaining of iNOS, MMP-9, OPG and RANKL in the periodontal ligament (PDL) of deciduous teeth with physiologic root resorption (GI), inflammatory pathological root resorption (GII) and permanent teeth (GIII), the negative control. Teeth in GI (n = 10), GII (n = 10) and (GIII) (n = 10) were submitted to immunohistochemical analysis to determine the expression of iNOS, MMP-9, OPG, and RANKL. The immunostaining was analysed by optical density. Statistical analysis included one-way ANOVA, followed by Student-Newman-Keuls post hoc test (p < 0.05). The results showed that iNOS, MMP-9 and RANKL expression in the PDL was higher in GII compared to GI and GIII (p < 0.05). Moreover, RANKL expression was higher in GI compared to GIII (p < 0.001), while OPG immunolabelling was lower in GII compared to GI and GIII (p < 0.001). The PDL of deciduous teeth bearing inflammatory processed exhibited upregulation of resorption-associated factors as well as enzymes related to tissue degradation which, in turn explains the exacerbation and greater susceptibility of those teeth to root resorption process.

:Machaerium hirtum (Vell.) Stellfeld (Fabaceae) known in Brazil as "jacaranda de espinho" or "espinheira santa nativa" is a medicinal plant commonly used in folk medicine to treat ulcers, cough and diarrhea. This study aimed to investigate the anti-inflammatory and antinociceptive effects of hydroalcoholic extracts from M. hirtum twig (HEMh) using in vivo experimental models of nociception through the involvement of transient receptor potential channels, acid-sensing ion channel (ASIC), nitrergic, opioidergic, glutamatergic, and supraspinal pathways. Our results revealed an antinociceptive effect of HEMh mediated by the opioidergic, L-arginine-nitric oxide and glutamate systems, as well as by interactions with TRPA1/ASIC channels. The anti-inflammatory effect of HEMh evaluated with a xylene-induced ear edema and by the involvement of arachidonic acid and prostaglandin E2 (PGE2) showed involvement of the COX pathway, based on observed decreases in PGE2 levels. A phytochemical investigation of the HEMh led to the isolation of α-amyrin, ß-amyrin, allantoin, apigenin-7-methoxy-6-C-ß-D-glucopyranoside, and apigenin-6-C-ß-D-glucopyranosyl-8-C-ß-D-xylopyranoside. In conclusion, the acute oral administration of HEMh inhibits the nociceptive behavioral response in animals through the nitrergic, opioid, glutamatergic pathways, and by inhibition of the TRPA1 and ASIC channels, without causing locomotor dysfunction. In addition, its anti-inflammatory effect is associated with the COX pathway and decreased PGE2 levels.

BACKGROUND: Orofacial pain of myofascial origin is often associated with temporomandibular joint dysfunction, affects chewing muscles and may lead to functional limitations. Dry needling is an intervention commonly used for inactivating myofascial pain trigger points. OBJECTIVE: To systematically review the effects of dry needling on orofacial pain of myofascial origin in patients with temporomandibular joint dysfunction. METHODS: This systematic review has pain intensity as primary outcome. Searches were conducted on April 13th, 2018 in eight databases, without publication date restrictions. We selected randomized controlled trials published in English, Portuguese, or Spanish, with no restrictions regarding subject ethnicity, age or sex. RESULTS: Seven trials were considered eligible. There was discrepancy among dry needling treatment protocols. Meta-analysis showed that dry needling is better than other interventions for pain intensity as well as than sham therapy on pressure pain threshold, but there is very low-quality evidence and a small effect size. There were no statistically significant differences in other outcomes. CONCLUSION: Clinicians can use dry needling for the treatment of temporomandibular joint dysfunction, nevertheless, due the low quality of evidence and high risk of bias of some included studies, larger and low risk of bias trials are needed to assess the effects of dry needling on orofacial pain associated with temporomandibular joint dysfunction.

(-)-Linalool is a monoterpene alcohol which is present in the essential oils of several aromatic plants. Recent studies suggest that (-)-linalool has anti-inflammatory, antihyperalgesic and antinociceptive properties in different animal models. The present study investigated the contribution of glutamatergic system in the antinociception elicited by (-)-linalool in mice. Nociceptive response was characterized by the time that the animal spent licking the injected hind paw or biting the target organ following glutamate receptor agonist injections. (-)-Linalool administered by intraperitoneal (i.p., 10-200 mg/kg), oral (p.o., 5-100 mg/kg) or intrathecal (i.t., 0.1-3 microg/site) routes dose-dependently inhibited glutamate-induced nociception (20 micromol/paw, pH 7.4) with ID(50) values of 139.1 mg/kg; 34.6 mg/kg; and 0.9 microg/site, with inhibitions of 70+/-4; 72+/-7 and 74+/-8%, respectively. However, the intraplantar injection of (-)-linalool partially (49+/-9%) inhibited glutamate-induced nociception. Furthermore, (-)-linalool (200 mg/kg) given i.p. also reduced significantly the biting response caused by intrathecal injection of glutamate (30 microg/site), AMPA (25 ng/site), SP (135 ng/site), NMDA (25 ng/site) and kainate (23.5 ng/site), with inhibitions of 89+/-6%, 73+/-11%, 85+/-4%, 98+/-2% and 52+/-15%, respectively. However, (-)-linalool did not inhibit nociception induced by intrathecal injection of trans-ACPD (8.6 microg/site). Taken together, these results provide experimental evidences indicating that (-)-linalool produce marked antinociception against glutamate induced pain in mice, possible due mechanisms operated by ionotropic glutamate receptors, namely AMPA, NMDA and kainate.

ABSTRACT BACKGROUND AND OBJECTIVES: Low back pain is considered a public health problem worldwide and has a personal, social, occupational and economic impact. Psychosocial signs such as inappropriate beliefs about pain, fear of movement, anxiety, stress, depression and low job satisfaction are characteristics of individuals with low back pain. These clinical signs are mediators of chronic pain and disability. The present study aimed to assess psychological comorbidities in patients with chronic non-specific low back pain who are undergoing physical therapy and patients awaiting physical therapy; in addition to characterizing the psychosocial profile of these individuals. METHODS: This research was carried out with 31 individuals recruited from physical therapy clinics in the region of greater Florianópolis. They were divided into two groups: Treatment (TG) and non-treatment (CG). The following self-report instruments were applied: Hospital Anxiety and Depression Scale (HADS), Health Status Questionnaire (SF-36V2), Visual Analog Scale (VAS), Oswestry Low Back Pain Disability Index (OLBPDI), Fear-Avoidance Belief Questionnaire (FABQ) and Pain Castatrophizing Scale (PCS). RESULTS: Significant differences (p<0.05) were observed in the scores of the instruments applied between the groups. The CG had higher averages than the GT. CONCLUSION: The results obtained in this study support previous findings about the benefits of physical therapy for individuals with chronic nonspecific low back pain, suggesting that, in addition to reducing pain and disability, there are benefits related to psychosocial factors.


RESUMO JUSTIFICATIVA E OBJETIVOS A dor lombar é considerada um problema de saúde pública em todo o mundo e gera impacto pessoal, social, ocupacional e econômico. Os sinais psicossociais como crenças inapropriadas sobre a dor, medo do movimento, ansiedade, estresse, depressão e baixa satisfação no trabalho são características de indivíduos com lombalgia. Esses sinais clínicos são mediadores da dor crônica e incapacidade. O presente estudo teve como objetivo avaliar as comorbidades psicológicas em pacientes com dor lombar crônica inespecífica que estão em atendimento fisioterapêutico e pacientes que aguardam o atendimento de fisioterapia; além de caracterizar o perfil psicossocial desses indivíduos. MÉTODOS Estudo realizado com 31 indivíduos, recrutados em clínicas de fisioterapia na região da grande Florianópolis. Foram divididos em dois grupos: tratamento (GT/n=16) e não tratamento (GC/n=15). Foram aplicados os seguintes instrumentos de autorrelato: Hospital Anxiety and Depression Scale (HADS), Health Status Questionnaire (SF-36V2), Escala Analógica Visual (EAV), Oswestry Low Back Pain Disability Index (OLBPDI), Fear-Avoidance Belief Questionnaire (FABQ) and Pain Castatrophizing Scale (PCS). RESULTADOS: Foram observadas diferenças significativas (p<0,05) nos escores dos instrumentos aplicados entre os grupos. Sendo que o GC apresentou médias maiores que o GT. CONCLUSÃO Os resultados obtidos neste estudo apoiam descobertas anteriores sobre os benefícios da fisioterapia para indivíduos com dor lombar crônica inespecífica, sugerindo que, além da redução da dor e incapacidade, há benefícios relacionados aos fatores psicossociais.

Traumatic brain injury (TBI) is a devastating disease that commonly causes persistent mental disturbances and cognitive deficits. Although studies have indicated that overproduction of free radicals, especially superoxide (O2(-)) derived from nicotinamide adenine dinucleotide phosphate (NADPH) oxidase is a common underlying mechanism of pathophysiology of TBI, little information is available regarding the role of apocynin, an NADPH oxidase inhibitor, in neurological consequences of TBI. Therefore, the present study evaluated the therapeutic potential of apocynin for treatment of inflammatory and oxidative damage, in addition to determining its action on neuromotor and memory impairments caused by moderate fluid percussion injury in mice (mLFPI). Statistical analysis revealed that apocynin (5mg/kg), when injected subcutaneously (s.c.) 30min and 24h after injury, had no effect on neuromotor deficit and brain edema, however it provided protection against mLFPI-induced object recognition memory impairment 7days after neuronal injury. The same treatment protected against mLFPI-induced IL-1ß, TNF-α, nitric oxide metabolite content (NOx) 3 and 24h after neuronal injury. Moreover, apocynin treatment reduced oxidative damage (protein carbonyl, lipoperoxidation) and was effective against mLFPI-induced Na(+), K(+)-ATPase activity inhibition. The present results were accompanied by effective reduction in lesion volume when analyzed 7days after neuronal injury. These data suggest that superoxide (O2(-)) derived from NADPH oxidase can contribute significantly to cognitive impairment, and that the post injury treatment with specific NADPH oxidase inhibitors, such as apocynin, may provide a new therapeutic approach to the control of neurological disabilities induced by TBI.

Na fisiopatologia da Insuficiência Cardíaca (IC) tem-se dado destaque à hiperatividade simpática e à inflamação sistêmica de baixa intensidade, aspectos também presentes na disfunção erétil (DE). O exercício físico constitui-se estratégia que proporciona melhora desses parâmetros sendo plausível a hipótese de que quando exercido por meio da dança proporcionaria melhores resultados. Ao unir atividade física e música, a dança seria capaz, dentre outros mecanismos favoráveis, aprimorar o funcionamento das redes neurais centrais e periféricas, contribuindo para o restabelecimento da normalidade da função neuro-hormonal e redução da resposta inflamatória, o que aumentaria a eficácia do tratamento tanto da IC quanto da DE.

Although the biological effects of thyroid hormones are mediated by nuclear receptors (genomic mechanisms), interactions with receptors associated with the plasma membrane (non-genomic mechanisms) of target cells are not clear. In this study we investigated the rapid stimulatory effect of thyroxine (T(4)) on (45)Ca(2+) uptake as well as ionic currents and intracellular messengers involved in the stimulatory action of T(4) in amino acid accumulation in immature rat testes. Results indicated that 10(-9)M or 10(-6)M T(4) was able to increase immediately (45)Ca(2+) uptake after 60s of hormone exposure. These results indicate for the first time that voltage-dependent Ca(2+) channels and ATP-dependent K(+) channels can be seen as a set-point in the stimulatory effect of T(4) on amino acid accumulation. Apamin-sensitive small-conductance Ca(2+)-activated K(+) channels (SK(Ca)) and chloride channels were shown to be partially involved in this mechanism. The amino acid accumulation triggered by the PKC pathway suggests a functional link between different ion channel activities and the stimulatory effect of T(4) on amino acid accumulation. In conclusion, we show in this study a rapid and stimulatory effect of T(4) on calcium uptake and on amino acid accumulation, both events initiated at the plasma membrane, which strongly characterizes a non-genomic effect of T(4) in immature rat testes.

An important requirement for endodontic paste with antibiotics placed in direct contact with living tissues is biocompatibility. The aim of this study was to evaluate the paste biocompatibility prepared with zinc oxide (1.25mg), tetracycline (8mg) and thiamphenicol (26.67mg). The paste and its components were implanted separately through polyethylene sterile tubes of 10mm in length and 1.3mm in diameter, in the subcutaneous tissue of rats with the experiment control at intervals of 3, 7, 15 and 30 days. Each day 6 rats were used, being 3 of them with implant of the substances in four sites placed on the back of the animals and 3 sham animals where it was implanted the polyethylene empty tubes. The experimental animals were anesthetized in an intra-peritoneal way with ketamina and xilazina (0.75ml / g body weight). After the experimental periods, the animals were anesthetized with the same anesthetic overdose. It was held an excision biopsy of the implant area with 10 mm to the security limit included in paraffin following a plan of random histological cut and uniformlyisotropic or oriented cuts according to stereological principles, getting a statistical estimative of the relative amount of inflammatory cells or not on the test system, getting as a result the paste biocompatibility, being the zinc oxide the most toxic element for the cell quality found.


Un requisito importante para la pasta endodóntica preparada con antibióticos, que es colocada en contacto directo con los tejidos vivos es la biocompatibilidad. El objetivo de este estudio fue evaluar la biocompatibilidad de la pasta preparada con óxido del zinc (1,25mg), tetraciclina (8mg) y el tiamfenicol (26,67mg). La pasta y sus componentes fueron implantados por separado a través de tubos estériles de polietileno de 10 mm de longitud y de 1,3mm de diámetro en el tejido subcutáneo de ratas en intervalos de 3, 7, 15 y 30 días. Cada día, 6 ratas fueron implantadas en cuatro sitios ubicados en la parte posterior de los animales, 3 de ellas con el implante de las sustancias y 3 fueron implantados con los tubos de polietileno vacíos. Los animales del experimento fueron anestesiados intraperitteonealmen, con ketamina y xilasina (0,75 ml/g peso corporal). Después de los periodos experimentales, los animales fueron anestesiados con la misma sobredosis anestésica. Fue realizada una biopsia exisional del área del implante con 10 mm de límite de seguridad, luego se realizaron cortes histológico al azar uniformemente isotrópicos o orientados según los principios esteriológicos, consiguiendo un estimativo estadístico de la cantidad relativa de células inflamatorias en el sistema de prueba. Se obtuvieron resultados de la biocompatibilidad de la pasta, siendo el óxido del zinc el elemento mas tóxico según la cualidad de las células que fueron encontradas.
