RESUMEN

PURPOSE: Our aim was to design and build a 3T 31P/1H calf coil that is capable of providing both good 31P and 1H transmit and receive performance, as well as being capable of accommodating a near-infrared spectroscopy (NIRS) device for simultaneous NIRS data and MRI/MRS acquisition. METHOD: In this work, we propose a new 3T 31P/1H birdcage combination design consisting of two co-centrically positioned birdcages on the same surface to maximize transmit efficiency and sensitivity for both nuclei. The 31P birdcage is a high-pass birdcage, whereas the 1H birdcage is a low-pass one to minimize coupling. The diameter of the 31P/1H birdcage combination was designed to be large enough to accommodate a NIRS device for simultaneous NIRS data and MRI/MRS acquisition. RESULTS: The one-layer coil structure of the birdcage combination significantly streamlines the mechanical design and coil assembly process. Full-wave simulation results show that the 31P and 1H are very well decoupled with each other, and the 1H and 31P SNR surpasses that of their standalone counterparts in the central area. Experiment results show that the inclusion of a NIRS device does not significantly affect the performance of the coil, thus enabling simultaneous NIRS and MRI readouts during exercise. CONCLUSION: Our findings demonstrate the feasibility and effectiveness of this dual-tuned coil design for combined NIRS and MRS measurements, offering potential benefits for studying metabolic and functional changes in the skeletal muscle in vivo.

RESUMEN

The purpose of this study was to evaluate adipose tissue distributions and hepatic and pancreatic fat contents using a 6-point Dixon MRI technique in type 2 diabetes mellitus (T2DM), and to assess associations between fat distributions and biochemical markers of insulin resistance. Intra-abdominal MRI was investigated in 14 T2DM patients, 13 age- and sex-matched healthy controls (HC) and 11 young HC using a 3 T Prisma MRI scanner. All T2DM subjects completed a fasting comprehensive metabolic panel, and demographic measurements were taken according to standardized methodologies. We observed excellent correlation (R2 = 0.94) between hepatic fat fraction quantified using 6-point Dixon MRI and gold standard MRS, establishing the accuracy and reliability of the Dixon technique. Significantly increased visceral adipose tissue (VAT) volumes were found in T2DM patients compared to age-matched HC (1569.81 ± 670.62 cm3 vs. 1106.60 ± 566.85 cm3, p = .04). We also observed a trend of increasing subcutaneous adipose tissues (SAT), and total abdominal fat (TAT) volumes in T2DM compared to age-matched HC. Hepatic fat fraction percentage (HFF%) was 44.6% higher in T2DM compared to age-matched HC and 64.4% higher compared to young HC. Pancreatic fat fractions in the head and body/tail were higher in T2DM patients compared to both healthy cohorts. We also observed correlations between fat contents of the liver and pancreas in T2DM patients, and association between biochemical markers of T2DM with HFF, indicating a risk for non-alcoholic fatty liver disease among T2DM. In summary, this study provides evidence of T2DM patients having increased liver and pancreatic fat, as well as increased adipose tissues.

Highly sensitive potassium (K)-doped carbon nanotube (CNT) and polypyrrole (PPy) nanocomposite membrane-based enzyme field effect transistor (ENFET) has been fabricated on indium tin oxide (ITO) for detection of cholesterol. P-type graphene has been deposited as substrate on ITO glass electrochemically. N-type graphene has been deposited in source and drain regions. Zirconium dioxide (ZrO2) has been deposited on the channel region as gate insulator. K/PPy/CNT composite has been deposited as sensing membrane on the top of ZrO2 layer; 1 µl of cholesterol oxidase (ChOx) has been immobilized on K/PPy/CNT membrane via physical adsorption technique. The response of K/PPy/CNT/FET has been studied using Agilent 3458A digital multimeter in presence of phosphate buffer saline (PBS) of 50 mM, pH 7.0 and 0.9 % NaCl contained in a glass pot. During measurement, 20 µl cholesterol solutions (0.5 to 25 mM) were poured into the pot through micropipette each time. It has been found that K/PPy/CNT/FET has linearly varied from 0.5 to 20 mM. The sensitivity of this FET has been found to be ~400 µA/mM/mm(2) with regression coefficient (r) ~ 0.998. The proposed ENFET has response time of 1 s and stability up to 6 months. The experiment has been repeated 10 times, and only 2.0 % output variation has been observed. The limit of detection (LoD) and Michaelis-Menten constant (K m) were found to be ~1.4 and 2.5 mM, respectively. The results obtained in this work show negligible interference (3.7 %) with uric acid, glucose and urea.
