RESUMEN

Clostridium perfringens is one of the critical causative agents causing diarrhea in piglets, with significant economic losses to the pig industry. Under normal gut microbiota homeostasis and well-managed barns, diarrhea caused by C. perfringens could be controlled. Some reports show that probiotics, such as Bacillus subtilis, are beneficial in preventing necrotic enteritis (NE) in chickens, but few reports on piglets. Clostridium perfringens was found in the piglets' diarrhea with intestinal microbiota dysbiosis in our survey. Bacillus subtilis G2B9-Q, which was isolated from the feces of healthy pigs, was found to have anti-Clostridium activity after screening. Clostridium perfringens was used to challenge mice by intraperitoneal injection for modeling to evaluate the anti-infective activity of cell-free supernatant (CFS) of B. subtilis G2B9-Q and different concentrations of B. subtilis G2B9-Q by oral administration. The results showed that G2B9-Q can mitigate intestinal lesions caused by C. perfringens infection, reduce inflammatory reactions, and modulate intestinal microbiota. The CFS of G2B9-Q can alleviate the pathological damage of intestinal tissues caused by C. perfringens infection, reduce the concentration of TNF-α and IL-10 in the sera of mice, as well as the relative expression levels of alpha toxin (CPA), perfringolysin O (PFO) toxin, IL-10, IL-22, and TNF-α in the jejunum and colon tissues, and alleviate the changes in gut microbiota structure caused by C. perfringens infection, which showed better therapeutic effects and indicated that the metabolites of G2B9-Q are essential mediators for their beneficial effects. Therefore, the CFS of G2B9-Q could potentially replace antibiotics in treating C. perfringens infection.

The blood-brain barrier (BBB) is the main obstacle to hydrophilic and large molecules to enter the brain, maintaining the stability of the central nervous system (CNS). But many environmental factors may affect the permeability and structure of the BBB. Electromagnetic pulses (EMP) irradiation has been proven to enhance the permeability of the BBB, but the specific mechanism is still unclear. To explore the potential mechanism of EMP-induced BBB opening, this study investigated the permeability, fine structure and the proteins expression of the tight junction (TJ) of the BBB in the rats exposed to EMP. Using the leakage of fluorescein isothiocyanate-labeled dextran with different molecular mass under different field intensity of EMP exposure, we found that the tracer passing through the BBB is size-dependent in the rat exposed to EMP as field intensity increased. Transmission electron microscopy showed TJ of the endothelial cells in the EMP-exposed group was open, compared with the sham-irradiated group. But the levels of TJ proteins including ZO-1, claudin-5, or occludin were not changed as indicated by western blot. These data suggest that EMP induce BBB opening in a field intensity-dependent manner and probably through dysfunction of TJ proteins instead of their expression. Our findings increase the understanding of the mechanism for EMP working on the brain and are helpful for CNS protection against EMP.

Active manipulation of terahertz (THz) beam deflection and intensity is highly desired for possible applications in wireless communication, radar, and remote sensing. Here, by integrating the phase-gradient metasurfaces and tunable liquid crystal materials, we demonstrate an active THz beam deflection device based on polarization mode conversion. The resonant modes in the photonic cavity formed by the double-layer metasurface and the tunable anisotropic liquid crystal material in the cavity not only improve the polarization conversion efficiency of the device, but also actively regulate the resonance matching conditions. As a consequence, a beam deflection of 47.5° with 50% diffraction intensity at 0.69 THz is achieved in the x-to-y polarization conversion mode, and this beam can be actively modulated with an ultrahigh modulation depth of 99.6% by rotating the anisotropic optical axis of liquid crystals. Moreover, the proposed device can also work as the deflection of 32.5° in the y-to-x polarization conversion mode at 0.94 THz with a maximum diffraction intensity of 38% and an intensity modulation depth of 97.8%. This work provides a new approach based on liquid crystal photonic devices for wavefront manipulation and active modulation for THz waves.

Recently, research about bound states in the continuum (BICs) has become more and more attractive. Nanostructures with rotational symmetry are usually utilized to realize polarization-independent quasi-BIC resonances. Here, we propose a new, to the best of our knowledge, scheme for a polarization-independent quasi-BIC without the help of rotational symmetry. With the rotation of the polarization direction of the incident light, a quasi-BIC resonance can be consistently observed in a dielectric cubic tetramer metasurface without rotational symmetry. Based on far-field multipolar decomposition and near-field electromagnetic distributions, it is found that different multipoles exhibit different dependences on the polarization direction, and the switch between electric and magnetic quadrupoles results in polarization-independent quasi-BIC resonance. Our findings provide an alternative scheme to design polarization-independent devices and promote wider potential applications.

BACKGROUND: The similarity between Crohn's disease (CD) and non-CD, especially with ulcerative colitis (UC) or intestinal tuberculosis (ITB), makes the diagnostic error rate not low. Therefore, there is an urgent need for an efficient, fast, and simple predictive model that can be applied in clinical practice. The purpose of this study is to establish the risk prediction model for CD based on five routine laboratory tests by logistic-regression algorithm, to construct the early warning model for CD and the corresponding visual nomograph, and to provide an accurate and convenient reference for the risk determination and differential diagnosis of CD, in order to assist clinicians to better manage CD and reduce patient suffering. METHODS: Using a retrospective analysis, a total of 310 cases were collected from 2020 to 2022 at The Sixth Affiliated Hospital, Sun Yat-sen University, who were diagnosed by comprehensive clinical diagnosis, including 100 patients with CD, 50 patients with ulcerative colitis (UC), 110 patients with non-inflammatory bowel disease (non-IBD) diseases (65 cases of intestinal tuberculosis, radioactive enterocolitis 39, and colonic diverticulitis 6), and 50 healthy individuals (NC) in the non-CD group. Risk prediction models were established by measuring ESR, Hb, WBC, ALb, and CH levels in hematology. The models were evaluated and visualized using logistic-regression algorithm. RESULTS: 1) ESR, WBC, and WBC/CH ratios in the CD group were higher than those in the non-CD group, while ALb, Hb, CH, WBC/ESR ratio, and Hb/WBC ratio were lower than those in the non-CD group, and the differences were statistically significant (all p < 0.05). 2) CD occurrence had a strong correlation with the WBC/CH ratio, with the correlation coefficient exceeding 0.4; CD occurrence was correlated with other indicators. 3) A risk prediction model containing age, gender, ESR, ALb, Hb, CH, WBC, WBC/CH, WBC/ESR, and Hb/WBC characteristics was constructed using a logistic-regression algorithm. The sensitivity, specificity, positive predictive value, negative predictive value, and area under the curve of the model were 83.0%, 76.2%, 59.0%, 90.5%, and 0.86, respectively. The model based on the corresponding index also had high diagnostic accuracy (AUC = 0.88) for differentiating CD from ITB. Visual nomograph based on the logistic-regression algorithm was also constructed for clinical application reference. CONCLUSIONS: In this study, a CD risk prediction model was established and visualized by five conventional hema-tological indices: ESR, Hb, WBC, ALb, and CH, in addition to a high diagnostic accuracy for the differential diagnosis of CD and ITB.

The differential microphone array, or differential beamformer, has attracted much attention for its frequency-invariant beampattern, high directivity factor and compact size. In this work, the design of differential beamformers with small inter-element spacing planar microphone arrays is concerned. In order to exactly control the main lobe beamwidth and sidelobe level and obtain minimum main lobe beamwidth with a given sidelobe level, we design the desired beampattern by applying the Chebyshev polynomials at first, via exploiting the structure of the frequency-independent beampattern of a theoretical Nth-order differential beamformer. Next, the so-called null constrained and least square beamformers, which can obtain approximately frequency-invariant beampattern at relatively low frequencies and can be steered to any direction without beampattern distortion, are proposed based on planar microphone arrays to approximate the designed desired beampatterns. Then, for dealing with the white noise amplification at low-frequency bands and beampattern divergence problems at high-frequency bands of the null constrained and least square beamformers, the so-called minimum norm and combined solutions are deduced, which can compromise among the white noise gain, directivity factor and beampattern distortion flexibly. Preliminary simulation results illustrate the properties and advantages of the proposed differential beamformers.

Reperfusion after ischemia would cause massive myocardial injury, which leads to oxidative stress (OS). Calcium homeostasis imbalance plays an essential role in myocardial OS injury. CaV1.2 calcium channel mediates calcium influx into cardiomyocytes, and its activity is modulated by a region of calpastatin (CAST) domain L, CSL54-64. In this study, the effect of Ahf-caltide, derived from CSL54-64, on myocardial OS injury was investigated. Ahf-caltide decreased the levels of LDH, MDA and ROS and increased heart rate, coronary flow, cell survival and SOD activity during OS. In addition, Ahf-caltide permeated into H9c2 cells and increased CaV1.2, CaVß2 and CAST levels by inhibiting protein degradation. At different Ca2+ concentrations (25 nM, 10 µM, 1 mM), the binding of CSL to the IQ motif in the C terminus of the CaV1.2 channel was increased in a H2O2 concentration-dependent manner. CSL54-64 was predicted to be responsible for the binding of CSL to CaV1.2. In conclusion, Ahf-caltide exerted a cardioprotective effect on myocardial OS injury by stabilizing CaV1.2 protein expression. Our study, for the first time, proposed that restoring calcium homeostasis by targeting the CaV1.2 calcium channel and its regulating factor CAST could be a novel treatment for myocardial OS injury.

The development of highly active and low-cost catalysts for use in oxygen reduction reaction (ORR) is crucial to many advanced and eco-friendly energy techniques. N-doped carbons are promising ORR catalysts. However, their performance is still limited. In this work, a zinc-mediated template synthesis strategy for the development of a highly active ORR catalyst with hierarchical porous structures was presented. The optimal catalyst exhibited high ORR performance in a 0.1 M KOH solution, with a half-wave potential of 0.89 V vs. RHE. Additionally, the catalyst exhibited excellent methanol tolerance and stability. After a 20,000 s continuous operation, no obvious performance decay was observed. When used as the air-electrode catalyst in a zinc-air battery (ZAB), it delivered an outstanding discharging performance, with peak power density and specific capacity as high as 196.3 mW cm-2 and 811.5 mAh gZn-1, respectively. Its high performance and stability endow it with potential in practical and commercial applications as a highly active ORR catalyst. Additionally, it is believed that the presented strategy can be applied to the rational design and fabrication of highly active and stable ORR catalysts for use in eco-friendly and future-oriented energy techniques.

Forward prediction of directional scattering from all-dielectric nanostructures by a two-level nested U-shaped convolutional neural network (U2-Net) is investigated. Compared with the traditional U-Net method, the U2-Net model with lower model height outperforms for the case of a smaller image size. For the input image size of 40 × 40, the prediction performance of the U2-Net model with the height of three is enhanced by almost an order of magnitude, which can be attributed to the more excellent capacity in extracting richer multi-scale features. Since it is the common problem in nanophotonics that the model height is limited by the smaller image size, our findings can promote the nested U-shaped network as a powerful tool applied to various tasks concerning nanostructures.

Metasurfaces transform the wavefront by spatially varying the amplitude or phase of the incoming beam. Instead of encoding such variation by subwavelength unit cells, it is achievable over diffraction engineering of supercell structures, which outperforms the unit-cell method when the spatial gradient is large. In addition to tight focusing, here we apply this method to achieve plane wave-to-Bessel beam transformation and point-to-point focusing at terahertz frequencies. The Bessel beam has a small beam waist (0.57λ) and long depth of focus (9.1λ) for subwavelength-resolution imaging over a long distance. The point-to-point focusing changes the divergence angle from 16° to 70°. Both devices are validated by numerical simulations and experimental results with good agreement.

Terahertz (THz) waves have attracted much attention in the field of biosensing due to advantages including non-destructiveness, being label-free, and high-sensitivity detection. Here we have experimentally demonstrated a THz polarization sensing method based on reflective metasurface sensors for detecting concentrations of protein solutions and their crystallization process. The protein with varying concentrations has been detected by five different polarization parameters, which show different spectral responses and sensing sensitivities. The sensing accuracy can reach the order of ng/mm2. Furthermore, the crystallization process of the protein sample from the dissolved state to the crystalline has been dynamically measured by polarization sensing, of which the highest sensitivity can reach 0.67 °/%. Therefore, this new sensing platform can have broad development prospects in the trace matter detection of the biological sample.

To build a portable and sensitive method for monitoring the concentration of the flavonoid rutin, a new electrochemical sensing procedure was established. By using nitrogen-doped carbonized polymer dots (N-CPDs) anchoring few-layer black phosphorene (N-CPDs@FLBP) 0D-2D heterostructure and gold nanoparticles (AuNPs) as the modifiers, a carbon ionic liquid electrode and a screen-printed electrode (SPE) were used as the substrate electrodes to construct a conventional electrochemical sensor and a portable wireless intelligent electrochemical sensor, respectively. The electrochemical behavior of rutin on the fabricated electrochemical sensors was explored in detail, with the analytical performances investigated. Due to the electroactive groups of rutin, and the specific π-π stacking and cation-π interaction between the nanocomposite with rutin, the electrochemical responses of rutin were greatly enhanced on the AuNPs/N-CPDs@FLBP-modified electrodes. Under the optimal conditions, ultra-sensitive detection of rutin could be realized on AuNPs/N-CPDs@FLBP/SPE with the detection range of 1.0 nmol L-1 to 220.0 µmol L-1 and the detection limit of 0.33 nmol L-1 (S/N = 3). Finally, two kinds of sensors were applied to test the real samples with satisfactory results.

Simultaneous control of phase and polarization offers a large degree of freedom to tailor the beam properties, for instance, enabling generation of structured beams such as vector beams and vector vortex beams. Here, we propose an ultrathin freestanding metasurface operating at the terahertz frequency for efficient generation of vector vortex beam with an arbitrarily defined topological charge from linearly polarized excitation. The metasurface is composed of bilayer metallic patterns separated by a thin quartz slab, with one layer determining the transmission polarization and the other controlling the transmission phase. The tightly cascaded two layers form a Fabry-Perot cavity to maximize the efficiency of the polarization and phase control. Two metasurfaces for generation of radially polarized vector beam with uniform phase and vortex phase are fabricated and tested at 0.14 THz. The experimental results successfully demonstrate the generation of high-quality vector beams with the desired phase. In the experiment, the ultrathin and freestanding properties allow the metasurface to be easily combined with other components, which shows great potential for the development of various compact terahertz systems.

Activation and proliferation of cancer stem cells exert an important role in the invasion, metastasis, and recurrence of malignant tumors, including lung cancer. Therefore, exploring molecular targets related to self-renewal and mobility of lung cancer stem cells has important clinical significance. In our present study, we aimed to explore the effects of miR-138-5p on lung cancer stem-like cells and associated regulatory mechanism. In our present study, enhanced self-renewal capacity and elevated expression of cancer stem cells markers CD133, CD44, aldehyde dehydrogenase 1 of lung cancer stem-like cells derived from A549 cells were firstly verified. Then, obviously enhanced autophagy was found in lung cancer stem-like cells compared with parental cells A549. Besides, we found that enhanced autophagy induced by rapamycin promoted self-renewal and cell mobility of lung cancer stem-like cells and suppression of autophagy by 3-methyladenine exerted just opposite effects. In addition, miR-138-5p was found to be downregulated in lung cancer stem-like cells compared with that in parental cell A549. At the same time, overexpression of miR-138-5p by transfected with miR-138-5p mimic was found to effectively suppress self-renewal and invasion of lung cancer stem-like cells. Further study revealed that ATG7 was a target of miR-138-5p and overexpressed miR-138-5p suppressed ATG7-mediated autophagy. In addition, specific small interference RNA-ATG7 strengthened the inhibiting effect of miR-138-5p mimic on self-renewal and invasion of lung cancer stem-like cells. Taken together, we found that autophagy helped to maintain self-renewal and invasion ability of lung cancer stem-like cells and overexpressed miR-138-5p exerted anti-tumor effects by blocking the self-renewal and invasion of lung cancer stem-like cells through suppressing ATG7-mediated autophagy.

A scheme to dynamically control the off-axis directional scattering from a silicon nanodisk is proposed, which is based on focused fields formed by the coherent superposition of radially and linearly polarized beams. When the phase condition of the generalized Kerker conditions is satisfied at a specified wavelength, the amplitude requirement for the off-axis directional scattering along a required direction can be fulfilled by tuning the magnitude ratio of the two focused beams. Therefore, directional control of the off-axis scattering in the meridional plane is achieved without the manipulation of the working wavelength. Our findings provide new possibilities of future potential applications of all-dielectric nanoantennas.

Photonic nanojets (PNJs) generated from a single microsphere illuminated by higher-order radially polarized (RP) beams are investigated. The effects of the size parameters of higher-order RP beams, the refractive index, and radius of the dielectric microsphere on the full width at half-maximum and peak intensity of the PNJ are numerically discussed and qualitatively interpreted. The results show that the minimal width of the PNJ can be obtained by optimally adjusting the size parameter. The PNJ beam waist becomes gradually narrower with increasing the radial mode number. As compared to the case of plane wave illumination, sharper PNJs are more easily generated when irradiated by a higher-order RP beam, even for microspheres with lower refractive indices or larger radii. Our findings can promote potential applications of PNJs in a variety of fields including super-resolution microscopy, nanolithography, and optical data storage.

BACKGROUND: It is proposed a new running suture technique called Needle Adjustment Free (NAF) technique, or PAN suture. The efficiency and the safety were evaluated in laparoscopic partial nephrectomy. METHODS: This new running suture technique avoids the Needle Adjustment method used in traditional techniques. The new continuous suture technique (11 patients) was compared with the traditional continuous suture method (33 patients) used in both transperitoneal and retroperitoneal laparoscopic partial nephrectomy (LPN) in terms of suture time (ST), warm ischemia time (WIT), blood loss (BL), open conversion rate and post-op discharge time, post-op bleeding, post-op DVT, ΔGFR (affected side, 3 months post-op). Differences were considered significant when P < 0.05. RESULTS: ST in the PAN suture group was 30.37 ± 16.39 min, which was significant shorter (P = 0.0011) than in the traditional technique group which was 13.68 ± 3.33 min. WIT in the traditional technique group was 28.73 ± 7.89 min, while in the PAN suture group was 20.64 ± 5.04 min, P = 0.0028. The BL in entirety in the traditional technique group was 141.56 ± 155.23 mL, and in the PAN suture group was 43.18 ± 31.17 mL (P = 0.0017). BL in patients without massive bleeding in the traditional technique group was significantly greater than in the PAN suture group at 101.03 ± 68.73 mL versus 43.18 ± 31.17 mL (P = 0.0008). The open conversion rate was 0 % in both groups. There was no significant difference between the two groups in postoperative discharge time, post-op bleeding, post-op DVT, ΔGFR (affected side, 3 months post-op). CONCLUSIONS: The NAF running suture technique, or PAN suture, leading to less ST, WIT and BL, which was shown to be more effective and safer than the traditional technique used for LPN. A further expanded research with larger sample size is needed.

RESUMEN

With the rapid development of artificial intelligence technology, researchers have applied it to the diagnosis of various tumors in the urinary system in recent years, and have obtained many valuable research results. The article sorted the research status of artificial intelligence technology in the fields of renal tumors, bladder tumors and prostate tumors from three aspects: the number of papers, image data, and clinical tasks. The purpose is to summarize and analyze the research status and find new valuable research ideas in the future. The results show that the artificial intelligence model based on medical data such as digital imaging and pathological images is effective in completing basic diagnosis of urinary system tumors, image segmentation of tumor infiltration areas or specific organs, gene mutation prediction and prognostic effect prediction, but most of the models for the requirement of clinical application still need to be improved. On the one hand, it is necessary to further improve the detection, classification, segmentation and other performance of the core algorithm. On the other hand, it is necessary to integrate more standardized medical databases to effectively improve the diagnostic accuracy of artificial intelligence models and make it play greater clinical value.

The simultaneous determination of monosaccharides present in the activated sludge would be crucial to understand the water treatment mechanism. Herein, an ion chromatography-mass spectrometry (IC-MS) with online pretreatment of column switching technique was proposed to analyze monosaccharides hydrolyzed from extracellular polysaccharides in the activated sludge. When the matrix was eliminated in the first dimension, monosaccharides were immediately identified by IC-MS. The improved ionization efficiency was achieved with the addition of T-joint prior to MS. During the performance test, our established method showed excellent detection limits ranging from 0.34 to 2.15 µg/L for all sugar targets. Great linearity (R ≥ 0.9955) was also achieved using this method in the range from 0.01 to 5 mg/L. Furthermore, the average recoveries were obtained between 84.82 and 113.46%. RSDs for peak areas and retention times were determined as 3.76% and 0.27%, respectively. Finally, this approach provided a rapid, convenient, and practical determination of monosaccharides in the activated sludge, which would be helpful for the analysis of monosaccharides derived from other biological samples.

High-index dielectric metasurfaces are rarely reported around 0.1-0.3 THz, as an extremely large etching depth is needed according to the millimeter-scale wavelength. In this work, we propose an easy solution to sub-THz wideband polarization control by utilizing 3D-printed low-index (n~1.5) metagratings. The metagrating with subwavelength lattice is shown as a very efficient half-wave plate (net polarization conversion of 87%) at 0.14 THz but showing noisy spectrum. The design with superwavelength lattice offers a smooth and wide bandwidth for linear polarization rotation. Study of the mechanism shows that the lattice size slightly above wavelength is a better choice for the low-index metadevice as it maintains high efficiency in the zero diffraction order and wide bandwidth due to the small mode dispersion. Such designs offer a feasible solution especially suitable for sub-THz polarization and phase control, complementary to the existing high-index dielectric and metallic metasurfaces.
