Vitellogenin (Vtg) serves as the precursor of yolk protein and exhibits widespread distribution in tissues, including in the ovary of both vertebrates and invertebrates. Vtg plays a critical role in facilitating oocyte maturation and embryonic development following oviposition. In this study, we have successfully elucidated the complete transcript sequence of TtVtg6-like from an ancient chelicerate Tachypleus tridentatus. The TtVtg6-like transcript encompassed a length of 4887 bp and encoded 1629 amino acids residues. Notably, TtVtg6-like was found to contain 25 exons. Furthermore, the molecular weight and isoelectric point of TtVtg6-like were determined to be 191.6 KDa and 6.73, respectively. Subsequent mRNA expression analysis demonstrated the specific expression of TtVtg6-like in ovary and yellow connective tissue. In addition, TtVtg6-like was located and distributed in both ovary and yellow connective tissue. Intriguingly, employing an siRNA approach to silence TtVtg6-like resulted in a decrease in TtVtg6-like transcription levels. Concomitantly, TtVtg6-like silencing led to increase production of ROS, ultimately resulting in DNA damage and cell apoptosis within the ovarian primary cell. The induction of apoptosis ovarian primary cells due to TtVtg6-like silencing was further corroborated through TUNEL assay and flow cytometry analysis. Overall, our findings underscore the significance of TtVtg6-like in ovarian cell development, revealing its potential association with ovarian cell apoptosis. Consequently, the insights gained from this study contribute to the future exploration of vitellogenesis and ovarian development in T. tridentatus.

CONTEXT: Desloratadine, an H1 receptor antagonist, is suggested as an effective first-line drug for chronic urticarial (CU). However, the efficacy of desloratadine alone is limited, and the recurrence rate of CU is relatively high. OBJECTIVE: We sought to evaluate the efficacy and clinical feasibility of desloratadine in combination with compound glycyrrhizin in the treatment of CU. MATERIALS AND METHODS: A systematic literature search was conducted in the databases of the China National Knowledge Infrastructure Database, VIP, WanFang, PubMed, and Web of Science using subject terms: "Chronic urticaria", "Loratadine", and "Compound glycyrrhizin". Randomised controlled trials (RCTs) that compared the efficiency and safety of the combination treatment with desloratadine alone starting from January 1, 2014 until February 10, 2021 were selected by two co-first authors independently, and the extracted data were analysed using Rev Man 5.3 software. RESULTS: Fourteen RCTs were included in our meta-analysis with a total of 1501 patients. The results showed that the combination treatment yielded a better treatment effect (total response rate: RR = 1.23, 95% CI: 1.17 to 1.29, p < 0.00001; cure rate: RR = 1.50, 95% CI: 1.30 to 1.73, p < 0.00001), lower recurrence rate as well as superior immune improvement than the treatment with desloratadine alone. In addition, there was no significant difference in the safety of the two treatments. DISCUSSION AND CONCLUSION: The combination of desloratadine and compound glycyrrhizin is a promising treatment for CU and is associated with decreased serum IgE level and improved proportions of CD4+ T and CD8+ T cells.

Traditional Chinese medicine (TCM) is a summary of the diagnosis and treatment experience formed by the working people in the long-term struggle against diseases, so it is very important to protect the intangible cultural heritage of TCM. How to extract valuable knowledge accurately and conveniently from the massive medical records of TCM is one of the important issues in the current research on the development of TCM. Due to the large amount of data of TCM medical records, many feature attributes, and diverse patterns, the existing classification technology has high computational complexity, low mining efficiency, and poor universality. Therefore, this paper proposed to quantify the medical records of TCM and obtained the main symptoms according to the improved hierarchical clustering feature selection algorithm. This paper also proposed a support vector machine (SVM) classification method using improved particle swarm algorithm to classify TCM information, which not only improves the efficiency and accuracy of TCM information classification but also discovers the potential dialectical and symptom patterns in diagnosis and treatment, so that the intangible cultural heritage protection of TCM can be developed sustainably. This paper showed that the information acquisition accuracy of the improved algorithm was very high. Before the improved algorithm was used, the accuracy of information mining for TCM was 67.90% at the highest and 65.53% at the lowest, but after using the improved algorithm, the accuracy rate of information mining for TCM was 88.02% at the highest and 82.45% at the lowest. It can be seen that using the improved algorithm to mine TCM information can quickly process effective information.

The potential impact of exposure to toxic elements in rice on human health has become a global public health issue. This study analyzed the pollution characteristics and probabilistic health risks of exposure to iAs, Pb, Cd, Cr, and Hg in rice produced in a typical multi-mining county using Monte Carlo simulation, a geographic information system, and bioavailability analysis. The results showed that the enrichment of As and Cd was prominent in rice, with mean tAs, iAs, and Cd contents of 0.34 ± 0.20, 0.15 ± 0.09, and 0.48 ± 0.50 mg·kg-1, respectively. The probability of non-carcinogenic risk via rice consumption in adults and children exceeding the threshold was 72% and 78%, respectively, and that of carcinogenic risk was as high as 100%. Among toxic elements, Cd and iAs were the main risk factors for health risks. The high-level health-risk areas mainly occurred in the north-eastern and central parts of the study area. Sensitivity analysis highlighted that the top three influential parameters for non-carcinogenic risk in adults were Content(Cd), Content(iAs), and Bioaccessibility(Cd), whereas those in children were ingestion rate of rice, Content(Cd), and Content(iAs). The Content(Cd) was the decisive factor for carcinogenic risk, with a sensitivity coefficient of 78.0% in adults and 64.7% in children. Considering the high risk of ingestion of local rice in this area, it is imperative to place strict supervision and take control measures.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease impacting patients' quality of life and imposing heavy societal and economic burdens. Apoptosis of intestinal epithelial cells (IECs) has been considered an early event during the onset of UC and plays a crucial role in disease development. Thus, effectively inhibiting apoptosis of IECs is of critical significance for the clinical management of UC, presenting a potential direction for the research and development of pharmacotherapeutic agents. In recent years, research on the ameliorative effects of natural products on UC through inhibiting IECs apoptosis has attracted increasing attention and made remarkable achievements in ameliorating UC. In this review, we summarized the currently available research about the anti-apoptotic effects of natural products on UC and its mechanisms involving the death-receptor mediated pathway, mitochondrial-dependent pathway, ERS-mediated pathway, MAPK-mediated pathway, NF-κB mediated pathway, P13k/Akt pathway, JAK/STAT3 pathway, and NLRP3/ASC/Caspase-1 pathway. Hopefully, this review may yield useful information about the anti-apoptotic effects of natural products on UC and their potential molecular mechanisms and provide helpful insights for further investigations.

Background: An enhanced understanding of larval ecology is fundamental to improve the management of locally depleted horseshoe crab populations in Asia. Recent studies in the northern Beibu Gulf, China demonstrated that nesting sites of Asian horseshoe crabs are typically close to their nursery beaches with high-density juveniles distributed around mangrove, seagrass and other structured habitats. Methods: A laboratory Y-maze chamber was used to test whether the dispersal of early-stage juvenile tri-spine horseshoe crab Tachypleus tridentatus is facilitated by chemical cues to approach suitable nursery habitats. The juvenile orientation to either side of the chamber containing controlled seawater or another with various vegetation cues, as well as their movement time, the largest distance and displacement were recorded. Results: The juveniles preferred to orient toward seagrass Halophila beccarii cues when the concentration reached 0.5 g l-1, but ceased at 2 g l-1. The results can be interpreted as a shelter-seeking process to get closer to the preferred settlement habitats. However, the juveniles exhibited avoidance behaviors in the presence of mangrove Avicennia marina and invasive saltmarsh cordgrass Spartina alterniflora at 2 g l-1. The juveniles also spent less time moving in the presence of the A. marina cue, as well as reduced displacement in water containing the S. alterniflora cue at 1 and 2 g l-1. These results may explain the absence of juvenile T. tridentatus within densely vegetated areas, which have generally higher organic matter and hydrogen sulfide. Conclusion: Early-stage juvenile T. tridentatus are capable of detecting and responding to habitat chemical cues, which can help guide them to high-quality settlement habitats. Preserving and restoring seagrass beds in the intertidal areas should be prioritized when formulating habitat conservation and management initiatives for the declining horseshoe crab populations.

INTRODUCTION: Cisplatin, a chemotherapeutic drug, is widely used for the treatment of various malignant tumors with good effects. However, cisplatin-induced nephrotoxicity is a major dose-limiting factor and a significant adverse event. Mannitol is used to reduce cisplatin-induced nephrotoxicity, which is controversial. This study aimed to evaluate the efficacy and safety of a hydration regimen containing mannitol against cisplatin-induced nephrotoxicity through a meta-analysis. METHODS: Potential records from PubMed, EMBASE, Cochrane Library, and ClinicalTrials that met the inclusion criteria were included from inception to May 2021. Cochrane Collaboration tools were used to assess the risk of bias in the included studies. Jadad's and NOS scores were applied to assess the quality of randomized controlled trials (RCTs) and case-control studies. A random-effects model or fixed-effects model was used depending on the heterogeneity. Subgroup analyses were performed to evaluate the potential study characteristics. The pooled odds ratios (ORs) and 95% confidence intervals (CIs) were evaluated. RESULTS: Four RCTs and seven case-control studies involving 4168 patients were included. Pooled results showed that mannitol use could reduce the incidence of cisplatin-induced nephrotoxicity (OR = 0.66, 95% CI [0.45-0.97], p = 0.03), especially reducing grade 3 nephrotoxicity events according to CTCAE 4.0 (OR = 0.37,95% CI [0.16-0.84]). Moreover, mannitol use was not significantly associated with creatinine clearance, serum creatine, and electrolyte disturbance (p > 0.05). Gastrointestinal cancer (OR = 0.36, 95% CI [0.15-0.83], p = 0.02) and urinary tract cancer (OR = 0.32,95% CI [0.14-0.73], p = 0.007) may be more sensitive to mannitol, although the test for overall effect was significantly different (OR = 0.66, 95% CI [0.49-0.89], p = 0.007). For patients with diabetes and hypertension, mannitol may worsen renal function (OR = 1.80, 95% CI [1.18-2.72], p = 0.006; OR = 2.19, 95% CI [1.50, 3.19], p < 0.0001, respectively). Mannitol may have a better protective effect when doses of mannitol were ≥ 25 g (OR = 0.58, 95% CI [0.39-0.88], p = 0.01) and doses of cisplatin < 75 mg/m2 (OR = 0.59, 95% CI [0.36-0.94], p = 0.03). It revealed that mannitol use was likely to cause nausea or vomiting (OR = 1.86, 95% CI [1.20-2.89], p = 0.006). CONCLUSION: Current evidence revealed that mannitol was an effective and safe drug to reduce cisplatin-induced nephrotoxicity events, especially Grade 3 events. However, it may cause more nausea/vomiting events and deteriorate renal function in patients with diabetes or hypertension. We also found that mannitol had the best effect when mannitol was ≥ 25 g in total or cisplatin was < 75 mg/m2. The present review aimed to summarize the effectiveness and features of traditional Chinese medicine (TCM) for the treatment of infectious diseases and to discuss the limitation of the development of TCM. The personalized medicine with TCM exerts a curative effect on viral and bacterial infectious diseases with unique advantages on the improvement of clinical manifestation, pathogen inhibition, and organ recovery during severe and drug-resistant infection. The deficiency of personalized medicine with TCM lies in that the current research design of TCM primarily focuses on the study of the effective components and material basis of Chinese herbs at the cellular, molecular, and genetic level, while ignoring the guidance of the TCM syndrome differentiation theory, which is the core concept of individualized treatment. Personalized medicine with TCM has a broad prospective for infectious diseases due to the specific efficacy and advantages. While the curative effect of individualized treatment with TCM cannot be excluded from the TCM syndrome differentiation theory, the study of personalized medicine with TCM for infectious diseases urgently requires a unified standardization of the clinical syndrome differentiation and the evolution rule of infectious diseases by TCM theory.

Concentrations and distribution characteristics of 2,3,7,8-substituted polychlorinated dibenzo-p-dioxins and dibenzofurans (2,3,7,8-PCDD/Fs) were analyzed in waste water from a paper mill. And concentrations of 2,3,7,8-PCDD/Fs in waste water before and after electron beam irradiation with different doses were compared. The feasibility, mechanism and rates of 2,3,7,8-PCDD/Fs degradation were discussed. The PCDD/Fs concentrations and corresponding I-TEQ (toxic equivalent quantity) values were 239 pg x L(-1) and 41.0 pg x L(-1), respectively, in the waste water. The concentrations of total 2,3,7,8-PCDD/Fs decreased after electron beam radiolysis at a dose of 30 kGy and 60 kGy with degradation rates of 5.27% and 23.6%, respectively.
