RESUMEN

Hepatocellular carcinoma (HCC) is highly metastatic and invasive. CircRNA participates in gene regulation of multiple tumor metastases, but little is known whether it is a bystander or an actual player in HCC metastasis. We aim to explore the molecular mechanisms of novel circRNAs in HCC metastasis. RT-qPCR was used to detect the expression of 13 circRNAs derived by the ERBB3 gene. The function of circ_0098823 and DNM1L in HCC cells were estimated by CCK-8, transwell assays, flow cytometry, electron microscope, and in vivo experiments. RNA binding protein of circ_0098823 was confirmed by RNA pull-down, mass spectrometry, and RNA immunoprecipitation. The expression of DNM1L after IGF2BP3 knockdown was detected by RT-qPCR and western blot. Circ_0098823 was significantly up-regulated both in HCC tissues and HGF induced cell lines. Circ_0098823 overexpression significantly enhanced proliferation, migration, and invasion but decreased apoptosis of HCC cells, particularly promoted mitochondrial fission. Compared with the control group, the tumors in the circ_0098823 knockdown mice were significantly smaller and lighter. Circ_0098823 silencing suppressed DNM1L expression, a key molecule for fission, which enhanced proliferation, migration and invasion, and inhibited apoptosis of HCC cell. IGF2BP3 was a binding protein of circ_0098823. The expression and mRNA stability of DNM1L were down-regulated by IGF2BP3 knockdown. IGF2BP3 knockdown significantly alleviated the excessive migration, invasion and apoptosis of HCC cells caused by circ_0098823 overexpression. This study uncovered a novel circ_0098823 with tumor-promoting effect, and the mechanism by which circ_0098823 participates in HCC progression through IGF2BP3-guided DNM1L. Our study broadens molecular understanding of HCC progression.

The reported autofocusing ability of a ring Airyprime beam array reaches up to 8632.40, while the strongest autofocusing ability of a circular Airyprime beam (CAPB) is only 1822.49. How can the autofocusing ability of a single beam reach the autofocusing ability of a beam array? To achieve this goal, a circularly transformed Airyprime beam (CTAPB) is introduced by following two steps. First, a circular equation transformation on the two transverse coordinates in the electric field expression of a propagating Airyprime beam is performed. Then, the electric field expression of a propagating Airyprime beam is integrated over the angle. The intensity profile of a CTAPB on the initial plane changes significantly with varying the primary ring radius r0. With increasing r0, therefore, the autofocusing ability of a CTAPB undergoes a process of first increasing and then decreasing, while the focal length always increases. A CTAPB exhibits more powerful autofocusing ability than a CAPB. The maximum autofocusing ability of a CTAPB can reach up to 8634.76, which is 4.74 times that of a CAPB, while the corresponding focal length is 95.11% of a CAPB. A CTAPB on the initial plane can be approximately characterized by a ring Airyprime beam array with sufficient number of Airyprime beams. Due to the better symmetry, a CTAPB has a slightly stronger autofocusing ability than a ring Airyprime beam array and almost the same focal length as a ring Airyprime beam array. The CTAPB is also experimentally generated, and the experimental results indicate that the CTAPB has powerful autofocusing ability. As a replacement of a CAPB and a ring Airyprime beam array, this introduced CTAPB can be applied to the scenes which involve abruptly autofocusing effect.

C-O bond formation via C-H alkoxylation remains a challenge, especially coupling with a secondary alcohol, due to its low activity and sterically encumbered property. Here, we report a general and effective cobalt-catalyzed oxidative cross-coupling of benzamides with secondary alcohols via C-H alkoxylation reaction under solvothermal conditions, enabled by a salicylaldehyde/cobalt complex. The protocol features easy operation without additives, broad substrate scope, and excellent functional tolerance. The applicability is proven by the gram-scale synthesis and modification of natural products.

BACKGROUND: Several previous studies have examined the association of ambient temperature with drowning. However, no study has investigated the effects of heat-humidity compound events on drowning mortality. METHODS: The drowning mortality data and meteorological data during the five hottest months (May to September) were collected from 46 cities in Southern China (2013-2018 in Guangdong, Hunan and Zhejiang provinces). Distributed lag non-linear model was first conducted to examine the association between heat-humidity compound events and drowning mortality at city level. Then, meta-analysis was employed to pool the city-specific exposure-response associations. Finally, we analysed the additive interaction of heat and humidity on drowning mortality. RESULTS: Compared with wet-non-hot days, dry-hot days had greater effects (excess rate (ER)=32.34%, 95% CI: 24.64 to 40.50) on drowning mortality than wet-hot days (ER=14.38%, 95%CI: 6.80 to 22.50). During dry-hot days, males (ER=42.40%, 95% CI: 31.92 to 53.72), adolescents aged 0-14 years (ER=45.00%, 95% CI: 21.98 to 72.35) and urban city (ER=36.91%, 95% CI: 23.87 to 51.32) showed higher drowning mortality risk than their counterparts. For wet-hot days, males, adolescents and urban city had higher ERs than their counterparts. Attributable fraction (AF) of drowning attributed to dry-hot days was 23.83% (95% CI: 21.67 to 26.99) which was significantly higher than that for wet-hot days (11.32%, 95% CI: 9.64 to 13.48%). We also observed that high temperature and low humidity had an additive interaction on drowning mortality. CONCLUSION: We found that dry-hot days had greater drowning mortality risk and burden than wet-hot days, and high temperature and low humidity might have synergy on drowning mortality.

BACKGROUND: The effects of heavy metal exposure on immunological function have sparked widespread concern, but unequivocal evidence on the association between mixed metal exposure and novel systemic inflammatory indexes remains scarce. OBJECTIVES: This study aimed to analyze the associations of heavy metals with two novel systemic inflammation indexes and the mediated effects of serum albumin. METHODS: Nineteen metals were detected among 4082 U.S. adults based on the NHANES. A linear regression, restricted cubic splines (RCS) regression, weighted quantile sum (WQS), Quantile-based Gcomputation (qgcomp), and Bayesian kernel machine regression (BKMR) were conducted to evaluate the associations of single metal and mixed metals with systemic immune-inflammation index (SII) and systemic inflammation response index (SIRI) levels, respectively. A series of subgroup analyses were used to identify potentially vulnerable populations. Furthermore, we conducted mediation analyses to investigate the mediated effects of serum albumin on the associations of metals with SII and SIRI. RESULTS: In the single-exposure model, exposure to various metals such as urinary Co, As, and serum Zn, Cu was associated with SII and SIRI (PFDR<0.05). Simultaneously, the above metals were linear positively correlated with SII and SIRI. Mixed-exposure analyses consistently showed that overall mixed urinary metal levels were positively pertinent for SII and SIRI levels, and the metal Co played a significant role in the urinary metal mixtures. Subgroup analyses showed that exposure to urinary Cd in men and elderly people increased SII and SIRI levels. The results of mediation analyses suggested the association of urinary metal mixture with SII and SIRI was mediated by albumin, and the proportion of mediation was 14.45% and 9.49%, respectively. CONCLUSIONS: Our findings suggested that metal exposure is strongly associated with the levels of system inflammation indexes and that serum albumin is, in part, a mediator of this association.

Optical event horizons in fibers, driven by coherent pumps, have been a prominent subject of study within the field of nonlinear optics. Previously, optical event horizons involving a potent pump and a linear-wave were interpreted as phase-matching processes wherein new spectral components are derived from the linear-wave due to the influence of the strong pump. This nonlinear interaction, coupled with the wave mixing mechanism, has been elaborated upon in the spectral domain. It's portrayed as a cascaded four-wave mixing process, achieving quasi-phase-matching through intermediate spectral components. Until now, research focused on event horizons or soliton linear-wave interactions has predominantly relied on coherent laser pump sources. However, there has been a recent resurgence in the exploration of incoherently pumped nonlinear optics. While the specific dynamics of incoherent light fields and their subsequent nonlinear processes might be elusive due to their inherent random field fluctuations, their incoherent nature unveils a multitude of statistical dynamics for nonlinear phenomena. In this work, we delve into optical event horizons encountered by linear-waves propelled by an incoherent light field within nonlinear optical fibers. Our numerical analysis scrutinizes the dynamics of linear-waves during optical event horizons under incoherent pumping. We further dissect the temporal statistics of the newly birthed idler-waves emerging from these event horizon processes.

Kerr cavities driven in the normal dispersion regime are known to host switching waves. These consist of a traveling wavefront that connects separate regions associated with high- and low-intensity steady states of the cavity. In this Letter, we drive a 230-m custom built fiber ring cavity with strong normal dispersion using nanosecond pulses, allowing us to directly resolve the fine structure of individual switching waves, including resonant oscillations occurring over periods of the order of ∼10 ps. We demonstrate the intimate connection between the temporal and spectral features of the dispersive waves associated with switching waves, while also investigating how these dispersive waves evolve with cavity parameters, namely the frequency detuning and pump desynchronization. Furthermore, by applying a localized and temporary perturbation to our driving field in the presence of a phase modulation trapping potential, we are able to generate a stable and persistent dark pulse, allowing us to directly observe and model the interlocking of two stationary switching waves under quasi-CW pumping conditions. These results further verify the accuracy of the dispersive wave formalism used, and show that their temporal modulation frequency and decay rate in a pulsed-pumped cavity are accurately captured from theory previously applied to CW-pumped systems.

We demonstrate an all-optical random number generator based on spontaneous symmetry breaking in a coherently driven Kerr resonator. Random bit sequences are generated by repeatedly tuning a control parameter across a symmetry-breaking bifurcation that enacts random selection between two possible steady-states of the system. Experiments are performed in a fiber ring resonator, where the two symmetry-broken steady-states are associated with orthogonal polarization modes. Detrimental biases due to system asymmetries are suppressed by leveraging a recently discovered self-symmetrization phenomenon that ensures the symmetry-breaking dynamics act as an unbiased coin toss, with a genuinely random selection between the two available steady-states. We optically generate bits at a rate of 3 MHz without post-processing and verify their randomness using the National Institute of Standards and Technology and Dieharder statistical test suites.

Although studies have estimated the associations of PM2.5 with total mortality or cardiopulmonary mortality, few have comprehensively examined cause-specific mortality risk and burden caused by ambient PM2.5. Thus, this study investigated the association of short-term exposure to PM2.5 with cause-specific mortality using a death-spectrum wide association study (DWAS). Individual information of 5,450,764 deaths during 2013-2018 were collected from six provinces in China. Daily PM2.5 concentration in the case and control days were estimated by a random forest model. A time-stratified case-crossover study design was applied to estimate the associations (access risk, ER) of PM2.5 with cause-specific mortality, which was then used to calculate the population-attributable fraction (PAF) of mortality and the corresponding mortality burden caused by PM2.5. Each 10 µg/m3 increase in PM2.5 concentration (lag03) was associated with a 0.80 % [95 % confidence interval (CI): 0.73 %, 0.86 %] rise in total mortality. We found greater mortality effect at PM2.5 concentrations < 50 µg/m3. Stratified analyses showed greater ERs in females (1.01 %, 95 %CI: 0.91 %, 1.11 %), children ≤ 5 years (2.17 %, 95 %CI: 0.85 %, 3.51 %), and old people ≥ 70 years. We identified 33 specific causes (level 2) of death which had significant associations with PM2.5, including 16 circulatory diseases, 9 respiratory diseases, and 8 other causes. The PAF estimated based on the overall association between PM2.5 and total mortality was 3.16 % (95 %CI: 2.89 %, 3.40 %). However, the PAF was reduced to 2.88 % (95 %CI: 1.88 %, 3.81 %) using the associations of PM2.5 with 33 level 2 causes of death, based on which 250.15 (95 %CI: 163.29, 330.93) thousand deaths were attributable to short-term PM2.5 exposure across China in 2019. Overall, this study provided a comprehensive picture on the death-spectrum wide association between PM2.5 and morality in China. We observed robust positive cause-specific associations of PM2.5 with mortality risk, which may provide more precise basis in assessing the mortality burden of air pollution.

AIMS AND OBJECTIVES: To establish a set of scientific and practical nursing care quality evaluation indicators for PACU in China. BACKGROUND: No unified nursing care quality evaluation indicators for post-anaesthesia care unit (PACU) were found in China. DESIGN: An international literature review, domestic cross-sectional survey and two-round e-Delphi study. METHODS: The international literature review and domestic cross-sectional survey were conducted to identify the potential nursing care quality evaluation indicators for PACU. A total of 38 and 32 indicators were extracted from 24 articles and the cross-sectional survey respectively. Two rounds of e-Delphi study were conducted to collect opinions from a panel of 20 independent experts and establish the final version of the nursing care quality evaluation indicators for PACU. A GRRAS checklist was used to guide the reporting of this study. RESULTS: Of 20 experts, 19 completed the first round of e-Delphi study and 18 completed the second round, with a response rate of 95% and 90% respectively. The experts' authority coefficient in the two rounds of e-Delphi study was 0.91 and 0.90 respectively. The Kendall W value of the two rounds ranged between 0.108 and 0.385 (p < .01). Four structure indicators, 12 process indicators and seven outcome indicators were included in the nursing care quality evaluation indicators for PACU. CONCLUSIONS: With the methods of literature review, cross-sectional survey and e-Delphi study, we established a set of scientific and practical nursing care quality evaluation indicators for PACU, to improve the nursing care quality, reduce incidence of complications and ensure patient safety. RELEVANT TO CLINICAL PRACTICE: The findings from this study enable nurses and managers in PACU settings to evaluate clinical nursing care quality using a robust framework.

The objective of this study is to observe the effect of high selenium on the antioxidant and immune functions of growing goats based on transcriptome sequencing. Eighteen goats were randomly divided into three groups: (1) the control (CON) group was fed a basal diet, and (2) the treatment 1 group (LS) and treatment 2 group (HS) were fed a basal diet with 2.4 and 4.8 mg/kg selenium-yeast (SY), respectively. The results indicate that HS treatment significantly (p < 0.05) increased the apparent digestibility of either extract and significantly increased (p < 0.05) total antioxidant capacity, whereas it significantly (p < 0.05) decreased plasma aspartate aminotransferase and malondialdehyde relative to the control group. The LS treatment had significantly (p < 0.05) increased glutathione S-transferase and catalase compared to CON. A total of 532 differentially expressed genes (DEGs) between the CON and HS were obtained using transcriptome sequencing. Kyoto Encyclopedia of Genes and Genomes analysis identified upregulated (p < 0.05) DEGs mainly related to vascular smooth muscle contraction, alpha-linolenic acid metabolism, biosynthesis of unsaturated fatty acids, the VEGF signalling pathway, and proteoglycans in cancer; downregulated (p < 0.05) DEGs mainly related to the NOD-like receptor signalling pathway, influenza A, cytokine-cytokine receptor interaction, haematopoietic cell lineage, and African trypanosomiasis. Ontology analyses of the top genes show that the identified DEGs are mainly involved in the regulation of granulocyte macrophage colony-stimulating factor production for biological processes, the external side of the plasma membrane for cellular components, and carbohydrate derivative binding for molecular functions. Seven genes are considered potential candidate genes for regulating antioxidant activity, including selenoprotein W, 1, glutathione peroxidase 1, glutathione S-transferase A1, tumour necrosis factor, tumour necrosis factor superfamily member 10, tumour necrosis factor superfamily member 8, and tumour necrosis factor superfamily member 13b. The experimental observations indicate that dietary supplementation with 4.8 mg/kg SY can enhance antioxidant and immune functions by improving muscle immunity, reducing the concentrations of inflammatory molecules, and modulating antioxidant and inflammatory signalling pathways in growing goats.

In this paper, the lone generation of a new kind of beam named finite energy Airyprime (FEA) beam through the Airy transformation of the coherent superposition of four different elegant Hermite-Gaussian modes is reported for the first time. Analytical expressions of the centroid, the r.m.s beam width, the divergence angle, and the beam propagation factor of the FEA beam are derived in the output plane of Airy transformation, respectively. The effects of the Airy control parameters on the intensity distribution, the centroid, the r.m.s beam width, and the beam propagation factor are examined in detail through numerical examples. Unlike the Airy beam, the FEA beam upon free space propagation will be associated with an additional Airy mode, and the beam pattern of the FEA beams propagating in free space will evolve into a solid beam spot with two tails along two transverse directions, as well as the the intensity of main lobe of the FEA beam decays much slowly during free space propagation. Further, an experiment setup is established to generate the FEA beam via Airy transformation of four mixed elegant Hermite-Gaussian modes. The propagation characteristics such as the intensity distribution, the r.m.s beam width and the beam propagation factor are measured. The experimental results agree well with the theoretical predictions. Our study affords an effective and novel approach to generate the FEA beam, and is beneficial to expand the potential application of the FEA beam.

An analytical expression of a single Airyprime beam propagating in free space is derived. Upon propagation in free space, a single Airyprime beam in arbitrary transverse direction is the coherent superposition of the Airyprime and the Airy-related modes, which results in the interference enhancement effect under the appropriate condition. The Airy-related mode is the conventional propagating Airy mode with an additional π/2 phase shift and a weight coefficient of half the normalized propagation distance. Due to the peak light intensity in the initial plane being set to be 1, the strength of interference enhancement effect is characterized by the maximum light intensity. The maximum light intensity of a single Airyprime beam propagating in free space is independent of the scaling factor and is only decided by the exponential decay factor. When the exponential decay factor is above the saturated value, the interference enhancement effect disappears. When the exponential decay factor decreases from the saturated value, the maximum light intensity of a single propagating Airyprime beam increases, and the position of maximum light intensity is getting farther away. With the increase of the scaling factor, the position of maximum light intensity of a single propagating Airyprime beam is extended. The intensity distribution and the transverse Poynting vector of a single propagating Airyprime beam are demonstrated in different observation planes of free space. The flow direction of transverse energy flux effectively supports the interference enhancement effect of a single propagating Airyprime beam. The Airyprime beam is experimentally generated, and the interference enhancement effect is experimentally confirmed. The interference enhancement effect is conducive to the practical application of a single Airyprime beam.

Soliton microresonator frequency combs (microcombs) have recently emerged as an attractive new type of optical comb source with a wide range applications proposed and demonstrated. To extend the optical bandwidth of these microresonator sources, several previous studies have proposed and studied the injection of an additional optical probe wave into the resonator. In this case, nonlinear scattering between the injected probe and the original soliton enables the formation of new comb frequencies through a phase-matched cascade of four-wave mixing processes. In this work, we expand the relevant analyses to consider soliton-linear wave interactions when the soliton and the probe fields propagate in different mode families. We obtain an expression for the phase-matched idler locations as a function of the dispersion of the resonator and the phase detuning of the injected probe. We confirm our theoretical predictions in experiments performed in a silica waveguide ring microresonator.

Age-specific discrepancy of mortality burden attributed to temperature, measured as years of life lost (YLL), has been rarely investigated. We investigated age-specific temperature-YLL rates (per 100,000) relationships and quantified YLL per death caused by non-optimal temperature in China. We collected daily meteorological data, population data and daily death counts from 364 locations in China during 2006-2017. YLL was divided into three age groups (0-64 years, 65-74 years, and ≥75 years). A distributed lag non-linear model was first employed to estimate the associations of temperature with age-specific YLL rates in each location. Then we pooled the associations using a multivariate meta-analysis. Finally, we calculated age-specific average YLL per death caused by temperature by cause of death and region. We observed greater effects of cold and hot temperature on YLL rates for the elderly compared with the young population by region or cause of death. However, YLL per death due to non-optimal temperature for different regions or causes of death decreased with age, with 2.0 (95 % CI:1.5, 2.5), 1.2 (1.1, 1.4) and 1.0 years (0.9, 1.2) life loss per death for populations aged 0-64 years, 65-74 years and over 75 years, respectively. Most life loss per death results from moderate temperature, especially moderate cold for all age groups. The effect of non-optimal temperature on YLL rates is smaller for younger populations than older ones, while the temperature-related life loss per death was more prominent for younger populations.

Radiation-induced rectal injury is one of the common side effects of pelvic radiation therapy. This study aimed to explore the role of nuclear factor erythroid 2-related factor 2 (Nrf2) in this process. In vivo, knockout (KO) of Nrf2 led to aggravated radiation-induced histological changes in the rectums. In vitro, interference or overexpression of Nrf2 resulted in enhanced or reduced radiosensitivity in human intestinal epithelial crypts (HIEC) cells, respectively. A potential relationship between Nrf2 and necroptosis was identified using RNA sequencing (RNA-seq) and western blotting (WB), which showed that necroptosis-related proteins were negatively correlated with Nrf2. Upon treatment with necrostatin-1 (Nec-1), the increased radiosensitivity, decreased cell viability, increased γH2AX foci formation, and decreased mitochondrial membrane potential (MMP) in Nrf2-interfered HIEC cells were alleviated. A significant recovery in morphological alterations was also observed in Nrf2 KO mice administered with Nec-1. Taken together, our results highlight the important protective effect of Nrf2 in radiation-induced rectal injury through the inhibition of necroptosis, and the physiological significance of necroptosis in radiation-induced rectal injury.

Microcombs-optical frequency combs generated in coherently driven nonlinear microresonators-have attracted significant attention over the last decade. The ability to generate two such combs in a single resonator device has, in particular, enabled a host of applications from spectroscopy to imaging. Concurrently, novel comb generation techniques such as synchronous pulsed driving have been developed to enhance the efficiency and flexibility of microcomb generation. Here, we report on the first, to the best of our knowledge, experimental demonstration of dual-microcomb generation via synchronous pulsed pumping of a single microresonator. Specifically, we use two electro-optically generated pulse trains derived from a common continuous wave laser to simultaneously drive two orthogonal polarization modes of an integrated silica ring resonator, observing the generation of coherent dissipative Kerr cavity soliton combs on both polarization axes. Thanks to the resonator birefringence, the two soliton combs are associated with different repetition rates, thus realizing a dual-microcomb source. To illustrate the source's application potential, we demonstrate proof-of-concept spectroscopic measurements.

Kerr microresonators driven in the normal dispersion regime typically require the presence of localized dispersion perturbations, such as those induced by avoided mode crossings, to initiate the formation of optical frequency combs. In this work, we experimentally demonstrate that this requirement can be lifted by driving the resonator with a pulsed pump source. We also show that controlling the desynchronization between the pump repetition rate and the cavity free-spectral range (FSR) provides a simple mechanism to tune the center frequency of the output comb. Using a fiber mini-resonator with a radius of only 6 cm, we experimentally present spectrally flat combs with a bandwidth of 3 THz whose center frequency can be tuned by more than 2 THz. By driving the cavity at harmonics of its 0.54 GHz FSR, we are able to generate combs with line spacings selectable between 0.54 and 10.8 GHz. The ability to tune both the center frequency and frequency spacing of the output comb highlights the flexibility of this platform. Additionally, we demonstrate that under conditions of large pump-cavity desynchronization, the same cavity also supports a new, to the best of our knowledge, form of Raman-assisted anomalous dispersion cavity soliton.

Modulation instability (MI) in the presence of noise typically leads to an irreversible and complete disintegration of a plane wave background. Here we report on experiments performed in a coherently driven nonlinear optical resonator that demonstrate nonlinear localization of dissipative MI: formation of persisting domains of MI-driven spatiotemporal chaos surrounded by a stable quasi-plane-wave background. The persisting localization ensues from a combination of bistability and complex spatiotemporal nonlinear dynamics that together permit a locally induced domain of MI to be pinned by a shallow modulation on the plane wave background. We further show that the localized domains of spatiotemporal chaos can be individually addressed-turned on and off at will-and we explore their transport behavior as the strength of the pinning is controlled. Our results reveal new fundamental dynamics at the interface of front dynamics and MI, and offer a route for tailored patterns of noiselike bursts of light.

Synteny and collinearity analysis is a standard investigative strategy done in many comparative genomic studies to understand genomic conservation and evolution. Currently, most visualization toolkits of synteny and collinearity do not emphasize the graphical representation of the results, especially the lack of extensible format on vector graphics outputs. This limitation becomes more apparent as 3rd generation sequencing brings high-throughput data, requiring relatively higher resolution for the resulting images. We developed VGSC2, the 2nd version of the web-based vector graph toolkit for genome synteny and collinearity analysis. The updated version enables four types of plots for synteny and collinearity, and three types of plots for gene family evolutionary research. Using web-based technologies, VGSC2 provides an easy-to-use user interface to display the homologous genomic result into vector graphs such as SVG, EPS, and PDF, as well as an online editor. VGSC2 is open source and freely available for use online through the web server available at http://bio.njfu.edu.cn/vgsc2.
