RESUMEN

DNA from hair that has undergone retort pouch sterilization shows considerably more fragmentation. Assessing the extent of DNA fragmentation using a PCR assay could therefore be applied to infer the sterilization history of a contaminated food sample. Such assessments could make it possible to determine whether a food sample had been contaminated by hair during the production process. To determine the extent of retort pouch sterilization, primers specific for the detection of human DNA were designed to give an amplification product of approx 500 bp. Hair was subjected to retort sterilization as a model of contamination, and PCR was performed using the extracted DNA as a template and the primer set for determining retort pouch sterilization. The results showed that no DNA amplification occurred in retort pouch samples, whereas amplification was observed in samples that were unheated, heated in hot water, or heated in a microwave oven. The present method was thus able to specifically detect thermal decomposition of DNA from hair that had undergone retort sterilization, and will be useful for determining whether hair discovered in a retort pouch was responsible for contamination.

RESUMEN

A method for simultaneous analysis of egg and milk allergens using LC-QTOF-MS was developed. The proteins measured were α-casein, ß-lactoglobulin, and ovalbumin, which are the main protein allergens in milk and eggs. The proteins were digested using trypsin, and the digests were analyzed by LC-QTOF-MS. Sixteen peaks were detected that confirmed the amino acid sequences of the digests, and a MRM method with high resolution (MRM-HR) using product ions of these peaks was applied for quantification. Next, validation studies were performed using beverage products to which milk and egg standard protein solutions had been added. Good linearity was achieved over the concentration range of 1.25 to 20 µg/g of milk and egg protein, and acceptable reproducibility and accuracy were obtained at 10 µg/g. Moreover, good agreement was also observed between LC-QTOF-MS and ELISA. These findings suggest that this LC-QTOF-MS method may be useful for determining the milk and egg protein contents of beverages.

OBJECTIVES: The aim of this study was to determine the incidence rate and clinical features of second primary pancreatic ductal carcinoma (SPPDC) in the remnant pancreas after pancreatectomy for pancreatic ductal carcinoma (PDC). METHODS: Data of patients undergoing R0 resection for PDC at a single high-volume center were reviewed. SPPDC was defined as a tumor in the remnant pancreas after R0 resection for PDC, and SPPDC met at least one of the following conditions: 1) the time interval between initial pancreatectomy and development of a new tumor was 3 years or more; 2) the new tumor was not located in contact with the pancreatic stump. We investigated the clinical features and treatment outcomes of patients with SPPDC. RESULTS: This study included 130 patients who underwent surgical resection for PDC between 2005 and 2014. Six (4.6%) patients developed SPPDC. The cumulative 3- and 5-year incidence rates were 3.1% and 17.7%, respectively. Four patients underwent remnant pancreatectomy for SPPDC. They were diagnosed with the disease in stage IIA or higher and developed recurrence within 6 months after remnant pancreatectomy. One patient received carbon ion radiotherapy and survived 45 months. One patient refused treatment and died 19 months after the diagnosis of SPPDC. CONCLUSIONS: The incidence rate of SPPDC is not negligible, and the cumulative 5-year incidence rate of SPPDC is markedly high. Post-operative surveillance of the remnant pancreas is critical for the early detection of SPPDC, even in long-term survivors after PDC resection.

Food-derived biological signals are transmitted to the brain via peripheral nerves through the paracrine activity of gastrointestinal (GI) hormones. The signal transduction circuit of the brain-gut axis has been analyzed in animals; however, species-related differences and animal welfare concerns necessitate investigation using in vitro human experimental models. Here, we focused on the receptors of five GI hormones (CCK, GLP1, GLP2, PYY, and serotonin (5-HT)), and established human induced pluripotent stem cell (iPSC) lines that functionally expressed each receptor. Compared to the original iPSCs, iPSCs expressing one of the receptors did not show any differences in global mRNA expression, genomic stability, or differentiation capacities of the three germ layers. We induced parasympathetic neurons from these established iPSC lines to assess vagus nerve activity. We generated GI hormone receptor-expressing neurons (CCKAR, GLP1R, and NPY2R-neuron) and tested their responsiveness to each ligand using Ca2+ imaging and microelectrode array recording. GI hormone receptor-expressing neurons (GLP2R and HTR3A) were generated directly by gene induction into iPSC-derived peripheral nerve progenitors. These receptor-expressing neurons promise to contribute to a better understanding of how the body responds to GI hormones via the brain-gut axis, aid in drug development, and offer an alternative to animal studies.

A 66-year-old Japanese woman had been diagnosed with a neuroendocrine tumor of the pancreatic head (G2) 3 years previously and undergone pancreaticoduodenectomy. Nine months postoperatively, recurrence with multiple liver metastases developed and she was referred to our department. A regimen of 10 mg of everolimus for 2 weeks plus 1-week washout was instituted, and no adverse events were observed. Fourteen months after treatment initiation, she developed severe generalized erythema multiforme (EM). Skin biopsy revealed spongiosis in the epidermis and interface change and edema in the superficial dermis. Mast cells were observed from the dermis to the subcutaneous tissue, as well as perivascular eosinophilic infiltration, leading to EM being diagnosed. Oral everolimus was discontinued, and the EM was relieved by treatment including steroid therapy. Everolimus is an inhibitor of the mammalian target of rapamycin, and its indications include neuroendocrine tumors. Skin disorders are commonly seen in the early stages of everolimus treatment, but their severity is almost always mild and never severe. This is the first report on a patient who presented with severe generalized EM more than 1 year after everolimus treatment initiation. Patients on everolimus therapy should be monitored for skin disorders on a long-term basis.

Parturition is an essential process in placental mammals for giving birth to offspring. However, the molecular machineries of parturition are not fully understood. We investigated whether oxytocin plays a crucial role in the progress of parturition in cooperation with the prostaglandin F2α (PGF2α) receptor. We first examined alterations in the expression of uterine contraction-associated genes in uteri of oxytocin receptor-deficient mice (Oxtr-/-) during parturition. We found that induction of cyclooxygenase (COX)-2 and connexin 43 expression was impaired in Oxtr-/-, whereas that of PGF2α receptor expression was not. We next generated mice with double knockout of genes for the oxytocin receptor/oxytocin and PGF2α receptor (Oxtr-/-;Ptgfr-/- and Oxt-/-;Ptgfr-/-) and evaluated their parturition with Oxtr-/-, Oxt-/-, Ptgfr-/-, and wild-type mice. In Oxtr-/-;Ptgfr-/- and Oxt-/-;Ptgfr-/-, pregnancy rates were similar to those of other genotypes. However, normal parturition was not observed in Oxtr-/-;Ptgfr-/- or Oxt-/-;Ptgfr-/- because of persistent progesterone from the corpus luteum, as observed in Ptgfr-/-. We administered RU486, a progesterone antagonist, to Ptgfr-/-, Oxtr-/-;Ptgfr-/-, and Oxt-/-;Ptgfr-/- on gestation day 19. These mice were able to deliver a living first pup and the parturition onset was similar to that in Ptgfr-/-. Meanwhile, unlike Ptgfr-/-, ∼75% of Oxtr-/-;Ptgfr-/- and Oxt-/-;Ptgfr-/- administered RU486 remained in labor at 24 hours after the onset of parturition. All of the pups that experienced prolonged labor died. We thus revealed that the oxytocin receptor is an upstream regulator of COX-2 and connexin 43 in the uterus during parturition and that both oxytocin/oxytocin receptor and PGF2α receptor are major components for successful parturition.

The hexadentate nitrogen ligands 1-isoTQEN ( N,N,N',N'-tetrakis(1-isoquinolylmethyl)ethylenediamine) and 3-isoTQEN ( N,N,N',N'-tetrakis(3-isoquinolylmethyl)ethylenediamine) have been prepared. The structures of these ligands are based on that of TPEN ( N,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine). The introduction of a benzene ring into TPEN affords fluorescence ability upon zinc-ion binding. Compared to the quinoline isomer TQEN, isoquinoline derivatives 1-isoTQEN and 3-isoTQEN exhibit a lower-energy shift in the excitation and emission wavelengths and an enhanced fluorescence intensity, probably because of the energy-transfer mechanism between adjacent isoquinoline rings. Importantly, an increase in the Zn (2+)/Cd (2+) discriminating ability and a reduction in the background fluorescence induced by pH were also achieved for isoquinoline derivatives. The zinc-ion-induced fluorescence of these isoTQENs was not quenched by an addition of TPEN, which demonstrates the significantly high zinc-ion binding ability of these isoTQEN ligands.

RESUMEN

A new method was established for the simultaneous determination of 10 sweeteners and a degradation product in beverages by using LC-MS/MS. An ACQUITY UPLC BEH C18 (2.1 × 100 mm, 1.7 µm) was used as the LC column and 0.1% each of aqueous formic acid and formic acid in acetonitrile were used as the mobile phase. A simple and rapid determination of sweeteners was possible by diluting with a solvent, and in the case of some samples containing a large amount of foreign matter, after pre-treatment by diluting with solvent and clean-up of the sample using an Oasis HLB cartridge. All the validation results were satisfactory. As the regulations and standards for sweeteners vary from country to country, a field survey of 58 beverages marketed in Japan was performed using the present method. No issues concerning the labelling or food sanitation law were found in the tested samples.

Previously, we have reported that 1- and 3-isoTQENs (N,N,N',N'-tetrakis(1- or 3-isoquinolylmethyl)ethylenediamines) exhibit a specific fluorescence enhancement toward zinc ion. In this study, three methoxy-substituted derivatives of 1-isoTQEN were synthesized and their fluorescent response toward zinc ion was studied. The substitution pattern of the methoxy group significantly changes the solubility of compounds in aqueous DMF, λ(max) in the absorption spectra, excitation/emission wavelengths and fluorescence intensity of zinc complexes. In the presence of zinc ion, 7-MeO-1-isoTQEN exhibits higher fluorescence intensity and longer excitation/emission wavelengths (λ(ex) = 342 nm, λ(em) = 526 nm) than 6-MeO-1-isoTQEN (λ(ex) = 303 nm, λ(em) = 469 nm) and 5,6,7-triMeO-1-isoTQEN (λ(ex) = 340 nm, λ(em) = 504 nm). The fluorescence intensity of a zinc complex of 7-MeO-1-isoTQEN (ϕ = 0.122) is four times higher than the parent 1-isoTQEN (ϕ = 0.034) under the same conditions. The crystal structure of 7-MeO-1-isoTQEN-Zn complex reveals that all six nitrogen atoms participate to the metal coordination with ideal octahedral geometry, affording significantly high metal binding affinity comparable with TPEN (N,N,N',N'-tetrakis(2-pyridylmethyl)ethylenediamine). 7-MeO-1-isoTQEN detects zinc ion concentration change in cells by fluorescence microscopic analysis.

A 6-methoxyquinoline conjugated diethylenetriamine derivative, N,N''-bis(6-methoxy-2-quinolylmethyl)diethylenetriamine (6-MeOBQDIEN) has been synthesized and its fluorescent response toward zinc ion was investigated. In the presence of zinc ion, 6-MeOBQDIEN exhibits fluorescence (λ(ex) = 329 nm, λ(em) = 418 nm, φ = 0.039). The fluorescent intensity of the zinc complex of the compound is two times higher than the parent BQDIEN (φ = 0.021) under the same conditions. The crystal structure of 6-MeOBQDIEN-Zn complex shows that all five nitrogen atoms participate to the metal coordination in a distorted square-pyramidal geometry (τ = 0.145) with the aliphatic nitrogen in an apical position. Fluorescent microscopic analysis using 6-MeOBQDIEN reveals the zinc ion concentration change in living cells.

Several bisquinoline derivatives, N,N'-bis(2-quinolylmethyl)-N,N'-dialkylethylnediamines (alkyl=methyl, ethyl, isopropyl and t-butyl), have been synthesized and their fluorescent responses toward zinc ion were investigated. These compounds exhibit zinc ion-induced fluorescence and their intensities decrease as the alkyl groups become larger. The t-butyl derivative (BQDtBEN) exhibited negligible fluorescence even in the presence of zinc ion. The fluorescence intensity of the zinc complex of the bisquinoline derivative (BQDMEN) is higher than that of TQEN (N,N,N',N'-tetrakis(2-quinolylmethyl)ethylenediamine), indicating that the TQEN-Zn complex has an intramolecular quenching mechanism due to the energy transfer among four quinoline rings and the remaining photoinduced electron transfer (PET) mechanism. Introduction of methoxy substituents into the quinoline ring shifted the excitation and emission wavelengths towards a lower-energy direction and increased the fluorescence intensity, which allows N,N'-bis(6-methoxy-2-quinolylmethyl)-N,N'-dimethylethylenediamine (6-MeOBQDMEN) to be used for cellular fluorescent microscopic analysis (lambdaex=331 nm, lambdaem=406 nm and phi=0.28 for 6-MeOBQDMEN-Zn complex).

Quinoline-based, tetradentate nitrogen ligands, N,N'-bis(2-quinolylmethyl)-N,N'-dialkyl-1,2-ethanediamine (alkyl = methyl, bqdmen; ethyl, bqdeen; isopropyl, bqdpen), have been investigated as the supporting ligands for the formation of bis(micro-oxo) dinuclear manganese complexes. Bis(micro-oxo)Mn(2)(iii,iii) complexes and were obtained for bqdmen and bqdeen, respectively, as evidenced by X-ray crystallography, whereas bqdpen did not afford any manganese complexes due to its steric bulk. Complexes and exhibit highly positive Mn(2)(iii,iii)/Mn(2)(iii,iv) and Mn(2)(iii,iv)/Mn(2)(iv,iv) redox couples relative to the corresponding pyridine-ligated (micro-O)(2)Mn(2)(iii,iii) complexes.
