RESUMEN

The antiproliferative effect of the fatty acid components of Clerodendrum volubile leaves as well as its antioxidant effect on MCF-7 and MDA-MB-231 human breast cancer cell lines were investigated. Fatty acids extracted from C. volubile leaf oil were subjected to gas chromatography mass spectrometry (GCMS) analysis. The cells were cultured and treated with the fatty acids for 48 h, after which the antiproliferation effect was ascertained via MTT assay and cell viability analysis using BD fluorescence activated cells sorting (FACS) Calibur. Cell cycle was analyzed by flow cytometry on FACS Calibur. Western blotting was used in determining expression of proteins in the cell lines. The treated cell lines were assessed for reduced glutathione level, catalase, superoxide dismutase, and lipid peroxidation. The fatty acids significantly inhibited cell proliferation, arrested G0/G1 phase, downregulated the expression of MMP-9, and attenuated oxidative stress in of MCF-7 cell lines but had little or no effect on MDA-MB-231 cell lines. These results indicate the therapeutic potential of the fatty acids components of the leaves of C. volubile on human breast cancer, which may be explored further in drug development.

Glioblastoma multiforme (GBM) is a malignant primary type of brain cancer with high proliferation and metastasis rates due to involvement of the microglial cell. It is resistant against available chemotherapy. Many strategic protocols have been developed but prognosis and patient life has not improved substantially. In this study, the anti-metastatic and antioxidant effect of fatty acids from Clerodendrum volubile leaves were investigated in U87-MG (Human Glioblastoma Multiforme) cell lines. The extracted fatty acids were incubated with U87-MG cells for 48 h. The anti-proliferative effect was determined by MTT assay, while apoptosis and cell cycle were analyzed with BD FACSCalibur. The transwell assay protocol was utilized in the analysis of cell migration and invasion. The treated cell lines were also assessed for reduced glutathione (GSH) level, catalase, superoxide dismutase (SOD) and lipid peroxidation. The fatty acid extract showed significant inhibitory activity on cell proliferation and cell cycle progression, mitigated oxidative stress, and suppressed migration and invasion in U-87 MG cell lines. These results give credence to the therapeutic potential of this plant against cancer, especially GBM.

The search for novel drugs and alternative medicine has led to increased research in medicinal plants. Among such plants is the orange fruit. Its peels have been utilized for long as an active ingredient in most traditional medicines. This study aims at investigating the chemical properties of the hexane and dichloromethane (DCM) extracts of orange peel as well as their biological potentials. Blended peels were extracted with n-hexane and n-dichloromethane, respectively. The resulting extracts were subjected to gas chromatography mass spectrometry (GCMS) characterization. The extracts were also assayed for free radical scavenging ability against 1,1 -diphenyl -2 picrylhydrazyl (DPPH), antioxidative burst via measuring luminol -amplified chemiluminescence response in human blood, and phytotoxicity against lemna minor. GCMS analysis revealed a predominance of fatty acid methyl esters in the hexane extract, while the DCM extract had more ketone metabolites. The DCM extract had significant (p < .05) higher free radical scavenging and antioxidative burst activities compared to the hexane. Both extracts revealed a significantly (p < .05) high phytotoxicity activity. Results from this study indicated that solvent type played a vital a role in the extraction of secondary metabolites, which are responsible for the observed biological activities. The higher activities by the DCM extract can be attributed to its constituents as revealed by GCMS analysis. There is great need to explore the phytotoxicity potentials of both extracts as natural herbicides.

Africa is rich in a wide range of flora that are exploited as herbal medicines and remedies. Several diseases such as diabetes, diarrhea, dysentery and jaundice have been successfully managed using herbal medicines. Herbal decoctions or concoctions have been used as pain killers, antibiotics, and hematinics. This study evaluated the hematopoietic and biochemical properties of the stem bark of Sterculia setigera Del. in Wistar rats. Results showed that S. setigera decoction has copiously high tannin and cardiac glycoside levels. Ingestion of the decoction by rats over a 16-day period significantly (P < 0.05) increased the body weights of rats by 22.4% in the S. setigera-treated group. Hematological profiles showed raised levels of red blood cells, hemoglobin, packed cell volume, mean corpuscular volume, mean cell hemoglobin, mean cell hemoglobin concentration, and platelets, while biochemical parameters showed lower levels of alanine aminotransferase and aspartate aminotransferase, and slight increase in albumin and TP levels. We posit that the results justify the use of the stem bark of S. setigera as a hematinic by traditional medical practitioners and show its relative safety. Further experiments are needed to evaluate its safety.

The hypoglycemic activities of nine sub-fractions from the methanolic leaf crude extract of Anisopus mannii were investigated in normoglycemic and alloxan-induced diabetic mice. The methanolic sub-fraction M at 400 mg/kg bw showed significantly (p<0.05) high reduction in fasting blood glucose (FBG) at 27.36 and 65.57% in normoglycemic and diabetic mice, respectively. In acute toxicity test, M at 2,000 and 5,000 mg/kg bw showed reduction in blood urea nitrogen and creatinine level, elevations in aspartate transaminase, alanine transaminase and total bilirubin levels, as well as the body weights. The weight-ratios of kidney and liver to the body weight of the mice fed with these doses of M were reduced with no sign of histopathological alteration. The M at 250 mg/kg bw significantly reduced the FBG levels in a postprandial study. The hypoglycemic effect of M was eliminated when co-administered with isosorbide dinitrate or nifedipine indicating the induction of insulin secretion via K+ ATP-dependent channels. The UV/HPLC analysis of M indicated saponin at 7.7 mg/g. This study confirmed the traditional use of A. mannii for diabetes mellitus and the potential for the further development as a novel hypoglycemic drug.

The objective of this paper was to investigate the phytochemistry and hypoglycemic activities of aqueous extracts of Anisopus mannii, Daniella olivieri, Detarium macrocarpum, Leptedenia hastate and Mimosa invisa, traditionally prescribed for diabetes mellitus. The aqueous extracts were tested for phytochemicals and free radical scavenging activity by the DPPH assay. The antidiabetic tests were performed in normoglycemic and alloxan induced diabetic mice. High intensity of saponins, xanthones, tannins and glycosides were detected in A. mannii, D. macrocarpum and M. invisa, respectively. For the free radical scavenging activity, D. macrocarpum showed the highest activity with an IC50 of 0.027 mg/ml which was 2.1 folds of ascorbic acid. All extracts showed potent hypoglycemic effects in alloxan induced diabetic mice with the highest fasting blood glucose reduction of 70.39 percent in A. mannii which was 1.54 and 0.98 fold of glibenclamide and human insulin, respectively. A. mannii showed the potent hypoglycemic activity which was 1.54 and 0.98 fold of glibenclamide and insulin, respectively. This study confirmed the traditional use of these Nigerian medicinal plants in diabetes treatment. These plants showed high potential for further investigation to novel anti-diabetic drugs.
